Este trabalho é uma seleção interessante de imagens dinâmicas do esôfago e de curvas de atividade/tempo, variando da motilidade normal até o extremo oposto, a acalásia em estado avançado. A técnica é a usual: quatro horas de jejum, com restrição de fumo, álcool e cafeína; incidência anterior; imagens de 0,5 segundo durante dois minutos, região da boca ao fundo gástrico, seguida de imagem plana de 20 segundos da mesma região (tempo de trânsito normal: < 10 segundos). A coletânea é baseada em vinte anos de experiência empregando uma sistematização com vários parâmetros de análise que permite discriminar pacientes com tempo total de trânsito normal.

OBJETIVO: avaliar o efeito do uso crônico do nelfinavir sobre o peso de ratas albinas prenhes e seus conceptos, bem como o número de implantações, fetos, placentas, reabsorções e mortalidade materna e fetal. MÉTODOS: 50 ratas albinas EPM-1 Wistar, prenhes, foram aleatoriamente divididas em cinco grupos: 2 controles, Contr1 (controle do estresse) e Contr2 (controle do veículo), e três experimentais, Exp40, Exp120 e Exp360, que receberam, respectivamente, 40, 120 e 360 mg/kg por dia de nelfinavir por via oral. A droga e o veículo (água destilada) foram administrados por gavagem em duas tomadas diárias (12/12 horas), desde o primeiro dia até o dia 20 da prenhez. No último dia do experimento, todos os animais foram anestesiados e sacrificados. Foram avaliados a evolução do peso, número de implantações, reabsorções, fetos, placentas, óbitos intra-uterinos, o peso dos fetos e das placentas e malformações maiores. A análise estatística foi realizada pela análise de variância (ANOVA) completada pelo teste de Kruskal-Wallis. RESULTADOS: em relação ao ganho de peso das ratas, houve ganho normal em todos os grupos, não sendo constatadas diferenças significantes entre eles. ANOVA mostrou ausência de diferenças significativas entre os grupos quanto aos parâmetros estudados. As médias do número de fetos foram: controles = 9,7±0,50; grupos tratados com nelfinavir = 9,7±0,81. Para as médias de números de placentas e implantações, controles = 9,7±0,50; grupos tratados com nelfinavir = 9,7±0,78. Quanto às médias de pesos fetais, controles = 4,04±0,50; grupos tratados com nelfinavir = 3,91±0,33 g. Finalmente, para as médias de pesos de placentas, controles = 0,64±0,02; grupos tratados com nelfinavir = 0,67±0,02 g. Além disto, não foram observadas reabsorções, mortalidade das matrizes, óbitos e malformações fetais. CONCLUSÕES: o nelfinavir, em todas as doses administradas, não influiu no ganho de peso das ratas prenhes e não mostrou efeitos deletérios sobre os conceptos.

A prolactina(PRL) humana circula em múltiplas formas de diferentes tamanhos moleculares, das quais três são detectadas na cromatografia por filtração em gel(CFG): Little ou monomérica(mPRL), Big( bPRL) e Big-Big ou macroprolactina( bbPRL ou macroPRL). Em pessoas normais, a principal forma é a mPRL(85 a 90% do total) com uma pequena proporção de macroPRL. Em algumas pessoas, porém, ocorre uma maior quantidade de macroPRL, um fenômeno denominado Macroprolactinemia, que se sabe estar presente entre 10-25% dos soros hiperprolactinêmicos. É importante a sua identificação para evitar investigação e tratamento desnecessário. O método padrão para sua detecção é a cromatografia por filtração em gel( CFG); porém, a precipitação com polietilenoglicol( PEG) é um método de triagem simples, confiável e de baixo custo. Os testes com PEG originais foram feitos com o ensaio imunofluorimétrico( IFMA) Delfia para PRL. Objetivo: Validar um teste sensível e específico para a triagem de macroPRL baseado no ensaio de PRL quimioluminescente Immulite DPC. Resultados e métodos: Analisamos amostras séricas de 142 pessoas de ambos sexos. Baseado nessas amostras de rotina, dosamos a PRL seguida da precipitação com PEG e cálculo da recuperação de PRL( %R de PRL). Destes soros, 88 foram submetidos a cromatografia com filtração em Gel. Foi definido um ponto-de-corte para a presença de macroPRL, baseado numa curva ROC, ao comparar-se os resultados do teste de precipitação com PEG e os da CFG. O ponto-de-corte foi definido em 60%, com o achado de um teste com sensibilidade de 88,9% e especificidade de 98,6%. Correlacionou-se a dosagem de mPRL com a presença de macroPRL na cromatografia. Conclusão: Validamos um teste de triagem para macroPRL baseado no ensaio quimioluminescente DPC Immulite com sensibilidade de 88,9% e especificidade de 98,6 % para a porcentagem de recuperação PRL de 60%. O achado de uma valor de mPRL menor ou igual a 20 depois da precipitação com PEG vai suportar este diagnóstico. A prevalência( 20,4%) de macroPRL encontrada em nosso estudo, utilizando a metodologia proposta, é semelhante à encontrada na literatura.

Foi testado in vivo a sensibilidade de Giardia muris a quatro drogas comumente usadas no tratamento da giardíase humana. Foram utilizados 7 grupos de animais, com 12 camundongos cada, sendo que o grupo controle recebeu apenas solução salina 0,15M (0,5ml/animal). Os demais grupos receberam em dose única: metronidazole e furazolidone (500mg/kg), tinidazole e secnidazole (200mg/kg). A eficácia das drogas foi avaliada através da contagem de cistos nas fezes e pela ausência de trofozoítos no intestino. O metronidazole foi a droga mais eficaz. Os cortes histológicos mostraram diferenças entre o padrão da mucosa intestinal de animais normais e parasitados. No entanto, não se observou diferença entre o padrão de mucosa de animais infectados tratados e não tratados, o que sugere que estas alterações podem ser causadas pelo parasito e não pelas drogas.

Apoptosis is a form of programmed cell death selectively removes abnormal cells, and thus contributes to maintaining the balance of the dynamics of cell reproduction. Therefore the verification of the occurrence of apoptotic cell death after a pathological stimulus is crucial for the analysis of the maintenance of normal cell cycle of a given tissue or organ. In this experiment were used cells lines human mammary tumor MDAMB231, T47, MCF7, which were irradiated with X-rays at a dose of 5 Gy in a time interval of 15 seconds, and filtration of 1mm aluminum. Samples containing the cells were grown in a specific culture medium, containing fetal bovine serum and growth factor, and two samples were prepared with each of the cell lines, one to be irradiated, and another that has not been irradiated, which denoted by negative control of the irradiation. The primary goal of the experiment was to verify and compare the rates of apoptosis in each cell lines, in which were irradiated and that were not irradiated, using flow cytometry as a method for detecting apoptotic cell death in together with specific markers annexin V and propidium iodide. Data from the readings made by flow cytometry were analyzed and interpreted using the software WinMDI statistical graph. In the last years, the use of industrialized systems of lattice structures of wood as an alternative in the construction processes, is becoming more popular in Brazil, mainly for their commitment to the environment.The industrialized system of wooden lattice structures consists in a production of lattice structures, composed of pieces of wood where their mechanical connections are made with the sheet multi-toothed connectors (CDE).Among the many challenges to make this system competitive, the whole system of the connections between the pieces of wood not only must show functionality, but also speed, strength, versatility and economy. Referenced at Brazilian Standard for Wood Structures (NBR 7190/1997 - Project of timber structures) the sheet multi-toothed connectors, are analyzed using three test methods: tensile strength parallel to grain, tensile strength normal to the fibers and shear strength, all of them in two positions, αCH0=0o e αCH0=90o to four types of wood: Angelim (Vatairea heteroptera Ducke); Red-Angico (Parapiptadenia rigida (Benth) Brenae); Garapa (Apuleia leiocarpa (Vog.) Macbr) and Jatoba (Hymenaea stilbocarpa Hayne), belonging to the Leguminosae family and founded in several regions of Brazil.The purpose of this manuscript consists to analyze the mechanical connections with the sheet multi-toothed connectors through tests from NBR7190/1997

Mining is an activity of great economic and social value, assisting in the development of the country. However, it can be extremely harmful to the environment if no proper waste management study exists as mitigation measure its effects. Sample some of these harmful effects are pollution: water, through the seepage of waste into the groundwater; soil; of fauna and flora; sound (due to the noise of machines); visual from the residue stored in the open, changing the local landscape; and air. One way to mitigate environmental impacts caused by mining is the proper management of their waste through their use on highways. To that end, this paper proposes to give an appropriate destination to grit coming from the beneficiation of scheelite, due to mining activity from mining group Tomaz Salustino in Brejuí mine, located in the city of Currais Novos in the state of Rio Grande do Norte. This work was developed in four stages. The first comprised the chemical and mineralogical tests, DRX and FRX in which they sought to discover the composition of the material studied. The next step involved the physical characteristics of the waste by means of specific tests and grinding the solid mass, LL and LP. The third stage included the specific tests applied to the pavement, with the compaction test and test Index Support California. Finally, the fourth stage was the mechanical characterization, represented by direct shear tests, both in the flooded condition and not in flooded condition. The technical feasibility of using the modified energie compacted in layers of subbase power has been verified. In normal and intermediate energies is feasible to use less noble as layers as the subgrade. The incorporation of the waste in layers of road pavements provide an alternative to conventionally used in paving aggregates, providing a proper disposal of tailings from scheelite, as well as environmental preservation

Cryosurgery is an efficient therapeutic technique used to treat benign and malignant cutaneous diseases. The primary active mechanism of cryosurgery is related to vascular effects on treated tissue. After a cryosurgical procedure, exuberant granulation tissue is formed at the injection site, probably as a result of angiogenic stimulation of the cryogen and inflammatory response, particularly in endothelial cells. To evaluate the angiogenic effects of freezing, as part of the phenomenon of healing rat skin subjected to previous injury. Two incisions were made in each of the twenty rats, which were divided randomly into two groups of ten. After 3 days, cryosurgery with liquid nitrogen was performed in one of incisions. The rats' samples were then collected, cut and stained to conduct histopathological examination, to assess the local angiogenesis in differing moments and situations. It was possible to demonstrate that cryosurgery, in spite of promoting cell death and accentuated local inflammation soon after its application, induces quicker cell proliferation in the affected tissue and maintenance of this rate in a second phase, than in tissue healing without this procedure. These findings, together with the knowledge that there is a direct relationship between mononuclear cells and neovascularization (the development of a rich system of new vessels in injury caused by cold), suggest that cryosurgery possesses angiogenic stimulus, even though complete healing takes longer to occur. The significance level for statistical tests was 5% (p<0,05).

An unfavorable denture-bearing area could compromise denture retention and stability, limit mastication, and possibly alter masticatory motion. The purpose of this study was to evaluate the masticatory movements of denture wearers with normal and resorbed denture-bearing areas. Completely edentulous participants who received new complete dentures were selected and divided into 2 groups (n=15) according to the condition of their denture-bearing areas as classified by the Kapur method: a normal group (control) (mean age, 65.9 ± 7.8 years) and a resorbed group (mean age, 70.2 ± 7.6 years). Masticatory motion was recorded and analyzed with a kinesiographic device. The patients masticated peanuts and Optocal. The masticatory movements evaluated were the durations of opening, closing, and occlusion; duration of the masticatory cycle; maximum velocities and angles of opening and closing; total masticatory area; and amplitudes of the masticatory cycle. The data were analyzed by 2-way ANOVA and the Tukey honestly significant difference post hoc test (α=.05). The group with a resorbed denture-bearing area had a smaller total masticatory area in the frontal plane and shorter horizontal masticatory amplitude than the group with normal denture-bearing area (P<.05). Denture wearers with resorbed denture-bearing areas showed reduced jaw motion during mastication.

The morphological criteria for identification of intercalated duct lesions (IDLs) of salivary glands have been defined recently. It has been hypothesised that IDL could be a precursor of basal cell adenoma (BCA). BCAs show a variety of histological patterns, and the tubular variant is the one that presents the strongest resemblance with IDLs. The aim of this study was to analyse the morphological and immunohistochemical profiles of IDLs and BCAs classified into tubular and non-tubular subtypes, to determine whether or not IDL and tubular BCA represent distinct entities. Eight IDLs, nine tubular BCAs and 19 non-tubular BCAs were studied. All tubular BCAs contained IDL-like areas, which represented 20-70% of the tumour. In non-tubular BCA, IDL-like areas were occasional and small (<5%). One patient presented IDLs, tubular BCAs and IDL/tubular BCA combined lesions. Luminal ductal cells of IDLs and tubular BCAs exhibited positivity for CK7, lysozyme, S100 and DOG1. In the non-tubular BCA group, few luminal cells exhibited such an immunoprofile; they were mainly CK14-positive. Basal/myoepithelial cells of IDLs, tubular BCAs and non-tubular BCAs were positive for CK14, calponin, α-SMA and p63; they were more numerous in BCA lesions. IDL, tubular BCA and non-tubular BCA form a continuum of lesions in which IDLs are related closely to tubular BCA. In both, the immunoprofile of luminal and myoepithelial cells recapitulates the normal intercalated duct. The difference between the adenoma-like subset of IDLs and tubular BCA rests mainly on the larger numbers of myoepithelial cells in the latter. Our findings indicate that at least some BCAs can arise via IDLs.

To assess binocular detection grating acuity using the LEA GRATINGS test to establish age-related norms in healthy infants during their first 3 months of life. In this prospective, longitudinal study of healthy infants with clear red reflex at birth, responses to gratings were measured at 1, 2, and 3 months of age using LEA gratings at a distance of 28 cm. The results were recorded as detection grating acuity values, which were arranged in frequency tables and converted to a one-octave scale for statistical analysis. For the repeated measurements, analysis of variance (ANOVA) was used to compare the detection grating acuity results between ages. A total of 133 infants were included. The binocular responses to gratings showed development toward higher mean values and spatial frequencies, ranging from 0.55 ± 0.70 cycles per degree (cpd), or 1.74 ± 0.21 logMAR, in month 1 to 3.11 ± 0.54 cpd, or 0.98 ± 0.16 logMAR, in month 3. Repeated ANOVA indicated differences among grating acuity values in the three age groups. The LEA GRATINGS test allowed assessment of detection grating acuity and its development in a cohort of healthy infants during their first 3 months of life.

Sugarcane holds an important place in the Brazilian economy. Grate part of the sugarcane harvested still accomplished largely manually. Sugarcane harvesters available in Brazil use the technology to chop the cane into 200 to 300 mm billets to allow on the go cane transferring to transport, contradicting the traditional method of whole stalk sugarcane harvesting system. In order to make whole stalk mechanical harvesting system possible, one of the barriers to be expired is the mechanical removal of the straw. The design of a mechanism that accomplishes this operation depends directly on the knowledge of the mechanical properties of the sugarcane related to its resistance to compression and the forces necessary to remove the leaves from the stalk. Compression tests were conducted using the universal testing machine. For leaves removal test by friction, a special apparatus was designed to allow the registration of the normal and traction force. The sugarcane stalk can resist up to 4.9 MPa. With a normal pressure of 0.8 MPa, which correspond to a friction force of 315 N, it is possible to remove the leaves, independent of its location in the sugarcane stalk.
