Evidence indicates that cruciferous vegetables are protective against a range of cancers with glucosinolates and their breakdown products considered the biologically active constituents. To date, epidemiological studies have not investigated the intakes of these constituents due to a lack of food composition databases. The aim of the present study was to develop a database for the glucosinolate content of cruciferous vegetables that can be used to quantify dietary exposure for use in epidemiological studies of diet-disease relationships. Published food composition data sources for the glucosinolate content of cruciferous vegetables were identified and assessed for data quality using established criteria. Adequate data for the total glucosinolate content were available from eighteen published studies providing 140 estimates for forty-two items. The highest glucosinolate values were for cress (389 mg/100 g) while the lowest values were for Pe-tsai chinese cabbage (20 mg/100 g). There is considerable variation in the values reported for the same vegetable by different studies, with a median difference between the minimum and maximum values of 5.8-fold. Limited analysis of cooked cruciferous vegetables has been conducted; however, the available data show that average losses during cooking are approximately 36 %. This is the first attempt to collate the available literature on the glucosinolate content of cruciferous vegetables. These data will allow quantification of intakes of the glucosinolates, which can be used in epidemiological studies to investigate the role of cruciferous vegetables in cancer aetiology and prevention.

The effects of ester plasticizers and copolymers on the mechanical properties of the natural biodegradable polymers, poly(3-hydroxybutyrate) [PHB] and poly(lactic acid) [PLA] have been studied after subjecting to melt processing conditions. Ester plasticizers were synthesized from citric, tartaric and maleic acids using various alcohols. A variety of PLA copolymers have also been prepared from poly(ethylene glycol) derivatives using stannous octanoate catalysed ring opening polymerisations of DL-lactide. A novel PLA star copolymer was also prepared from an ethoxylated pentaerythritol. The structures of these copolymers were determined by NMR spectroscopy. The plasticizing effect of the synthesised additives at various concentrations was determined. While certain additives were capable of improving the mechanical properties of PLA, none were effective in PHB. Moreover, it was found that certain combinations of additives exhibited synergistic effects. Possible mechanisms are discussed. Biotic and abiotic degradation studies showed that the plasticizers (esters and copolymers) did not inhibit the biodegradability of PHB or PLA in compost at 60°C. Simple toxicity tests carried out on compost extract and its ability to support the growth of cress seeds was established. PLA was found to be susceptible to limited thermal degradation under melt processing conditions. Conventional phenolic antioxidants showed no significant effect on this process, suggesting that degradation was not predominantly a free radical process. PLA also underwent photo-oxidative degradation with UV light and the process could be accelerated in the presence of a photoactivator such as iron (III) diisononyl dithiocarbamate. The mechanisms for the above processes are discussed. Finally, selected compounds were prepared on a pilot plant scale. Extruded and blown films were prepared containing these additives with conventional polymer processing equipment. The mechanical properties were similar to those obtained with laboratory produced compression moulded films.

This study examined how students leveraged different types of knowledge resources on an outdoor learning trail. We positioned the learning trail as an integral part of the curriculum with a pre- and post-trail phase to scaffold and to support students’ meaning-making process. The study was conducted with two classes of secondary two students. We coded two groups’ discourse to examine the use of knowledge resource types in the meaning-making process in an outdoor learning setting: contextual resource, new conceptual resource, prior knowledge resource, as well as the relationship among these knowledge resource types. Next, we also examined environmental interaction and integration in the students’ use of these knowledge resource types. Analysis showed that contextual resources are chiefly instrumental in fostering students’ capacity to harness new conceptual resource and to activate prior knowledge resource in interacting with and integrating the outdoor learning environment in the meaning-making process.

Unravelling the Musa genome allows genes and alleles linked to desired traits to be identified. Short stature and early flowering are desirable agronomic features of banana, as they are of bread wheat (Triticum aestivum). In wheat they were achieved through knowledge of the physiology and genetics of vernalization and photoperiod during development. Bananas and plantains have a facultative long-day response to photoperiod, as do wheat and wall cress (Arabidopsis thaliana). Using keyword searches of the genome of Musa acuminata 'Pahang' we found homologues of the genes of either T. aestivum or Arabidopsis that govern responses to vernalization and photoperiod. This knowledge needs to be interpreted in the context of plant development. Bananas have juvenile, mid-vegetative and reproductive phases of development. Leaf and bunch 'clocks' operate concurrently throughout the juvenile and mid-vegetative phases. In the mid-vegetative phase the plant becomes sensitive to photoperiod. Increased sensitivity to photoperiod reduces the overall pace of the bunch clock without affecting the leaf clock. Separation of the clocks changes the link between leaf number and time of flowering. The 'critical' quantitative trait for the time of flowering is the pace of the bunch clock up to bunch initiation. For bunch size it is the duration of the subsequent phase of female hand formation. Plants with either a short juvenile phase or a faster bunch clock in the mid-vegetative phase will produce fewer leaves and bunch early. In turn, independent manipulation of hand number per bunch and/or fruit per hand will provide manageable bunches with appropriate fruit size. Using published data we explore relationships between plant height, leaf number, bunch weight and hand number among bananas and plantains. Identifying and then manipulating the appropriate genes in Musa opens opportunities for earlier flowering, leading to plants with desirable agronomic qualities.

The xeroderma pigmentosum complementation group B (XPB) protein is involved in both DNA repair and transcription in human cells. It is a component of the transcription factor IIH (TFIIH) and is responsible for DNA helicase activity during nucleotide (nt) excision repair (NER). Its high evolutionary conservation has allowed identification of homologous proteins in different organisms, including plants. In contrast to other organisms, Arabidopsis thaliana harbors a duplication of the XPB orthologue (AtXPB1 and AtXPB2), and the proteins encoded by the duplicated genes are very similar (95% amino acid identity). Complementation assays in yeast rad25 mutant strains suggest the involvement of AtXPB2 in DNA repair, as already shown for AtXPB1, indicating that these proteins may be functionally redundant in the removal of DNA lesions in A. thaliana. Although both genes are expressed in a constitutive manner during the plant life cycle, Northern blot analyses suggest that light modulates the expression level of both XPB copies, and transcript levels increase during early stages of development. Considering the high similarity between AtXPB1 and AtXPB2 and that both of predicted proteins may act in DNA repair, it is possible that this duplication may confer more flexibility and resistance to DNA damaging agents in thale cress. In this study, four leachates samples from 3 different landfills localized in the north of Portugal were characterized and fractionated, to understand the decomposition degree and to evaluate their potential as an agent for fertilization. Humic substances (HS) were extracted, quantified, chemical characterized and further fractionated in humic acid (HA) and fulvic acid (FA). Keeping in mind the purpose to use these fractions as fertilizers, the phytotoxicity of HS, HA and FA solutions was evaluated on cress seed germination. The HS concentration was similar for all the leachates evaluated and was higher than 780 mg/L of total organic carbon. All the leachates analysed registered higher FA concentration than HA. The chemical characterization indicated that HA had a relatively higher aromatic character than the FA obtained from same sources. These results suggest that the HS from landfill leachates were in an early stage of humification, once the degree of humification increase as the landfilling age increase. Overall, the HS extracts showed absence of phytotoxicity, with germination index greater than 80% for samples treated to achieve low electric conductivity values. This suggests that the HS from the leachate may be used to produce liquid organic fertilizers.
