LMV is one of the most important pathogens of lettuce worldwide. Based on their ability to overcome the resistance genes mo1¹ and mo1² in lettuce, isolates can be divided in two types: LMV-Most, which can infect and are seed-borne in cultivars containing the mo1 gene and LMV-Common, which do not cause symptoms on these cultivars and are seed transmitted only in susceptible cultivars. To evaluate the occurrence of these two types of LMV isolates, a survey was carried out during 2002-2005 in three lettuce production areas from São Paulo State. Total RNA was used for the diagnosis of LMV isolates by RT-PCR using universal primers for the variable N-terminus of the capsid protein, in the 3' end of the genome. Positives samples were analyzed by a second RT-PCR using specifics primers for LMV-Most isolates designed to amplify a fragment from the central region (CI-VPg) of the genome. A total of 1362 samples showing mosaic symptoms were collected and 504 (37.29 %) were positives for LMV. On susceptible lettuce cultivars, LMV-Common was prevalent (77.3%). LMV-Most was found frequently associated with tolerant (mo1¹) lettuce cultivars. Susceptible cultivars correspond today for most of the area of lettuce production. So, despite the ability of LMV-Most isolates to overcome the resistance provided by the recessive mo1¹ gene, they are not prevalent in the conditions of São Paulo State.

Lettuce big vein associated virus (LBVaV) and Mirafiori lettuce big vein virus (MLBVV) have been found in mixed infection in Brazil causing the lettuce big vein disease. Analysis of part of the coat protein (CP) gene of Brazilian isolates of LBVaV collected from lettuce, showed at least 93% amino acid sequence identity with other LBVaV isolates. Genetic diversity among MLBVV CP sequences was higher when compared to LBVaV CP sequences, with amino acid sequence identity ranging between 91% to 100%. Brazilian isolates of MLBVV belong to subgroup A, with one RsaI restriction site on the coat protein gene. There is no indication for a possible geografical origin for the Brazilian isolates of LBVaV and MLBVV.

Entre os problemas fitossanitários da cultura da alface estão as doenças causadas por vírus. Três vírus causam sintomas de mosaico praticamente indistinguíveis: o Lettuce mosaic virus (LMV, Potyvirus), o Lettuce mottle virus (LeMoV, Sequivirus) e o Bidens mosaic virus (BiMV, Potyvirus). Através de RT-PCR utilizando-se oligonucleotídeos específicos para cada um destes vírus, amostras de alface e plantas invasoras, preferencialmente com sintoma de mosaico, foram coletadas em campos de produção de alface das regiões de Mogi das Cruzes, Campinas e Bauru no Estado de São Paulo e analisadas para a presença dos vírus. Verificou-se que o LeMoV foi o vírus encontrado com maior freqüência, seguido do LMV. A ocorrência de BiMV em alface foi extremamente baixa e restrita às regiões de Campinas e Bauru, onde também foi verificado em plantas invasoras como Bidens pilosa e Galinsoga parviflora. Esta ultima é hospedeira dos três vírus.

Bottom rot, caused by Rhizoctonia solani AG 1-IB, is an important disease affecting lettuce in Brazil, where its biological control with Trichoderma was not developed yet. The present study was carried out with the aim of selecting Trichoderma isolates to be used in the control of lettuce bottom rot. Forty-six Trichoderma isolates, obtained with baits containing mycelia of the pathogen, were evaluated in experiments carried out in vitro and in vivo in a greenhouse in two steps. In the laboratory, the isolates were evaluated for their capabilities of parasitizing and producing toxic metabolic substances that could inhibit the pathogen mycelial growth. In the first step of the in vivo experiments, the number and the dry weight of lettuce seedlings of the cultivar White Boston were evaluated. In the second step, 12 isolates that were efficient in the first step and showed rapid growth and abundant sporulation in the laboratory were tested for their capability of controlling bottom rot in two repeated experiments, and had their species identified. The majority of the isolates of Trichoderma spp. (76%) showed high capacity for parasitism and 50% of them produced toxic metabolites capable of inhibiting 60-100% of R. solani AG1-IB mycelial growth. Twenty-four isolates increased the number and 23 isolates increased the dry weight of lettuce seedlings inoculated with the pathogen in the first step of the in vivo experiments.In both experiments of the second step, two isolates of T. virens, IBLF 04 and IBLF 50, reduced the severity of bottom rot and increased the number and the dry weight of lettuce seedlings inoculated with R. solani AG1-IB. These isolates had shown a high capacity for parasitism and production of toxic metabolic substances, indicating that the in vitro and in vivo steps employed in the present study were efficient in selecting antagonists to be used for the control of lettuce bottom rot.

ABSTRACT An experiment was conducted in a greenhouse at the Federal University of Lavras cultivated with American lettuce, cv. Raider-Plus. The aim of this study was to evaluate the effects of irrigation water depths applied by drip pulses and of soil coverage on crop yields and efficiency of water use. The experimental design used was randomized blocks with eight treatments and three replications, totaling twenty-four plots. The treatments consisted of soil with and without coverage (double-sided white and black plastic) associated with four irrigation management levels. Irrigation management consisted in reposition of irrigation depths based on crop evapotranspiration (ETc) with D1-100% of ETc, applied continuously (control), and D2 - 100% of ETc, D3 - 75% of ETc, and D4-50% of ETc, applied by pulses. Irrigation by pulses consisted in splitting the depths into six irrigation pulses with intervals of fifty minutes of rest. It was observed that pulse irrigation saved 25% of water in treatment without mulching and 50% when plastic mulching was used, contributing substantially to improve irrigation water efficiency.

ABSTRACT Proper irrigation management is important both to enable adequate water supply and to minimize problems with diseases, nutrient leaching and unnecessary water and power wastes. Thus, this study aimed at improving irrigation management in greenhouses. For that reason, we assessed the effect of various water tensions on yield performance of crisphead lettuce, Raider-Plus cv. , grown with mulching. The experiment was conducted in greenhouse at the Federal University of Lavras. Treatments constituted five different soil water tensions, namely 12, 25, 35, 45, and 70 kPa. The results showed that irrigation must be carried out at water soil tension of around 12 kPa to reach satisfying commercial values for both total and commercial yield, 66 and 50 t ha-1, as well as keeping good vegetable quality. At this tension, total water consumption was of 167.25 mm. The water use efficiency demonstrated quadratic response to treatments, with high water consumption efficiency found in intermediate treatments (35 and 45 kPa), achieving values of 579.87 and 471.71 kg ha -1mm-1, respectively.

ABSTRACT The integration of fish farming in intensive system and plant production, called "aquaponics" is practiced successfully in countries like the USA, Australia and Europe. In Brazil, this integration has attracted the attention of researchers and producers. In this context, the aim of this study was to evaluate the effect of two substrates (crushed stone number 3, CS III and flexible polyurethane foam, FPF) on the production of aquaponic lettuce, moreover, to show that the residual water from intensive tilapia production provides sufficient qualitative characteristics for competitive production of lettuce without adding commercial fertilizers. The treatment in which FPF was used provided higher concentrations of macro and micronutrients in the shoots, higher production of fresh matter of shoots (95.48 g plant-1) and a larger number of leaves (14.90) relative to CS III. These results were attributed to the lower post-transplanting stress and the higher water retention time provided by the FPF. The residual water from tilapia intensive farming can provide sufficient nutrients for the production of lettuce, making the supplementary fertilization with commercial products unnecessary. Thus, the FPF presents the most suitable conditions to be used as substrate in aquaponics system with recirculation of the residual water from the intensive tilapia farming.

The purpose of this study is to evaluate the hygienic-sanitary quality of vegetables and irrigation water and assess the effectiveness of lemon juice and vinegar in reducing E. coli strains inoculated on lettuce. One hundred and forty samples of vegetables and 45 samples of irrigation water were investigated for thermotolerant coliforms and Salmonella spp. In order to verify the effectiveness of natural household sanitizers in reducing E. coli in inoculated lettuce, four treatment solutions were tested: fresh lemon juice, alcohol vinegar, lemon juice-vinegar mixture, and lemon juice-vinegar-water mixture. The microbiological analysis revealed high rates of contamination by thermotolerant coliforms and identified the presence of E. coli in 32% of the tested vegetable samples and 56% of the water samples. While no significant statistical difference (p < 0, 05) was identified in the tested solutions, the treatment with a combination of lemon juice and vinegar resulted in the highest Decimal Reductions (DR) of E. coli O157: H7 while the treatment with vinegar alone was the most effective against the indigenous E. coli strain

Butter samples were evaluated for free fatty acids, peroxide value, cholesterol, and fatty acid composition focusing on the trans isomer and conjugated linoleic acid (CLA). Sixty six samples were analyzed. Thirty six were collected in Brazil, eighteen in France, and twelve in Argentina. Samples were evaluated by free fatty acids, peroxide value, total lipid, cholesterol and fatty acid composition. The free fatty acid content varied from 0.16 to 0.46 g.100 g-1 and the peroxides value levels from 0.35 to 1.80 meq.kg-1. The cholesterol content varied from 192.8 to 226.3 g.100 g-1 and the total lipid content varied from 81.8 to 86.8 g.100 g-1. The levels of saturated, monounsaturated, and polyunsaturated fatty acids varied from 43.86 to 52.74, from 21.65 to 23.34, and from 2.11 to 2.89 g.100 g-1, respectively. The conjugated linoleic acid (CLA) content varied from 0.56 to 0.86 g.100 g-1 and the levels of total trans isomer varied from 2.18 to 3.81 g.100 g-1.

The objective of this study was to determine the best lettuce cultivar (American 'Graciosa', 'Vanda', 'Marcela' and 'Lavínia') harvesting method. Therefore, quality and shelf-life were evaluated using sensory analyses. Lettuce heads was harvested with the root on by the producer, so that they were cut in different ways in the laboratory. The samples were stored in a cold chamber at 10 °C and 80% ± 2% of relative humidity for nine days, and the sensorial analyses were performed according to Qualitative Descriptive Analysis method on days 1, 3, 6, and 9 of storage by twelve trained testers. The results were evaluated by variance analysis, principal component analysis, and the Tukey test with a reliability of 95%. The results indicate that there was a reduction in the quality of lettuce between the 1st and the 5th day of storage and that after the sixth day of storage the lettuce samples were considered unfit for consumption, except for the 'Lavínia' lettuce cultivar with root and cut treatment 2. On the ninth day of storage all samples were considered inappropriate for consumption.

Abstract Postharvest lettuce often lose water, thus affecting both its market value and consumer acceptance. However, the mechanism of the water-loss is still waiting well exploration. The aim of the present study was to investigate the effect of a foliar application of ABA on the fresh weight-loss and the chlorophyll content of postharvest lettuce as well as its association with the regulation of stomata. The present data demonstrated that exogenously application of ABA, in a concentration range of 0 to 100 µM, significantly lowered the fresh weight-loss of postharvest lettuce. ABA also delayed chlorophyll reduction during ambient storage, but this protective effect was ABA concentration-dependent. Among the tested ABA concentrations, 50 µM or lower ABA produced an inhibition effect on chlorophyll degradation in postharvest lettuce leaves. The results demonstrated that the exogenous ABA treatment can obviously reduce the transpiration rate of lettuce leaves by promoting the stomatal closure of postharvest lettuce, therefore eventually delay fresh weight-loss. The present study primarily showed that the application of exogenous ABA, which originated from a naturally-produced phytohormone, has a great potential in retaining the freshness of postharvest lettuce that is stored in an ambient condition, although possible practical application still need to be further evaluated.

Abstract Pecan nutshell is a residue from food industry that has potential to be used as biopreservative in foods. The objective of this study was to evaluate the antimicrobial activity of pecan nutshell aqueous extract in vitro and its effectiveness to inhibit spoilage microorganisms on lettuce leaves. The results indicate that the aqueous extract presents inhibitory activity against important foodborne pathogenic bacteria such as Listeria monocytogenes, Salmonella Enteritidis, Staphylococcus aureus, Bacillus cereus, Aeromonas hydrophila and Pseudomonas aeruginosa. Antimicrobial activity was not observed against Corynebacterium fimi, Clostridium perfringens, Escherichia coli, and the phytopathogenic fungi tested. When applied onto lettuce leaves, pecan nutshell extract reduced the counts of mesophilic and psychrotrophic bacteria in 2 and 4 log CFU/g, respectively, during storage of leafy for 5 days at refrigeration temperature (5 °C). The extract was not effective to inhibit yeast on lettuce leaves. Thus, the aqueous extract of pecan shell showed great potential to be used as a natural preservative in foods, acting mainly in the inhibition of spoilage and pathogenic bacteria.

O Lettuce mosaic virus espécie mais importante na cultura de alface (Lactuca sativa L.) no Brasil, causando sintomas de mosaico, clareamento das nervuras, necrose, distorção foliar e redução do crescimento da planta, pode ser transmitido por sementes com uma taxa de 1% a 16%, dependendo da interação dos genótipos de alface com os isolados LMV-Most ou LMV-Common. Neste trabalho, avaliou-se a detecção do LMV por PTA-ELISA, em sementes e plântulas de oito genótipos de alface: 'Vanessa Roxa', 'Baba de Verão', 'Verdinha', 'Maravilha das 4 Estações', 'Evely', 'Marcela', 687 ('Sapore' x 'Vera' ), 784 ('Sapore' x 'Vera'), utilizando anti-soro policlonal específico. O vírus não foi detectado em sementes do genótipo 'Verdinha' e, em plântulas dos genótipos 687, 'Marcela' e 'Evely', após a germinação em papel e 687, 784 e 'Marcela' com gene mo1¹, após a germinação em substrato. A avaliação individual do número de sementes infectadas foi de 100% para 'Vanessa Roxa' e 'Baba de Verão', 87,7% para 'Verdinha', 46,6% para 'Maravilha das 4 Estações' e 16,6% para 'Evely'. Nos genótipos com gene de resistência o percentual foi de 15,6%, 26,6%, 90% em 'Marcela', 687 e 784, respectivamente. A detecção do LMV por PTA-ELISA foi eficiente tanto em sementes quanto em plântulas.
