The use of physical protectors has been considered as an efficient technique for tillage farming of different species, mainly native ones. Based on the importance of the species, Peltophorum dubium for revegetation of degraded areas, this study evaluated the emergence, survival and initial development of P. dubium seedlings under the influence of physical protectors with different filters. Thus, the following treatments were adopted: absence of physical protector (APP), transparent physical protector (TPP), transparent physical protector + blue cellophane (BPP) and transparent physical protector + red cellophane (RPP). The evaluated characteristics were: emergence velocity index (EVI), seedling survival and emergence percentage, plant height, leaf area and root collar diameter. All of these physical protectors increased the mean values of EVI and survival. In conclusion, the emergence speed and initial development of P. dubium (Spreng.) seedlings grown in the interior of physical protectors, independent on the filters, presented positive results. The reduction on the light intensity interferes positively in the initial growth of these plants.

Two new records of Anopheles homunculus in the eastern part of the Atlantic Forest are reported. This species was found for the first time in Barra do Ouro district, Maquine municipality, Rio Grande do Sul state, located in the southern limit of the Atlantic Forest. The 2nd new record was in the Serra Bonita Reserve, Camacan municipality, southeast Bahia state. These records extend the geographical distribution of An. homunculus, suggesting that the species may be widely distributed in coastal areas of the Atlantic Forest. It is hypothesized that the disjunct distribution of the species may be caused by inadequate sampling, and also difficulties in species identification based only on female external characteristics. Species identification was based on morphological characters of the male, larva, and pupa, and corroborated by DNA sequence analyses, employing data from both 2nd internal transcribed spacer of nuclear ribosomal DNA and of mitochondrial cytochrome c oxidase subunit I.

Mosquitoes are vectors of arboviruses that can cause encephalitis and hemorrhagic fevers in humans. Aedes serratus (Theobald), Aedes scapularis (Rondani) and Psorophora ferox (Von Humboldt) are potential vectors of arboviruses and are abundant in Vale do Ribeira, located in the Atlantic Forest in the southeast of the State of Sao Paulo, Brazil. The objective of this study was to predict the spatial distribution of these mosquitoes and estimate the risk of human exposure to mosquito bites. Results of the analyses show that humans are highly exposed to bites in the municipalities of Cananeia, Iguape and Ilha Comprida. In these localities the incidence of Rocio encephalitis was 2% in the 1970s. Furthermore, Ae. serratus, a recently implicated vector of yellow fever virus in the State of Rio Grande do Sul, should be a target for the entomological surveillance in the southeastern Atlantic Forest. Considering the continental dimensions of Brazil and the inherent difficulties in sampling its vast area, the habitat suitability method used in the study can be an important tool for predicting the distribution of vectors of pathogens.

Background: Anopheles (Kerteszia) cruzii is a primary vector of Plasmodium parasites in Brazil's Atlantic Forest. Adult females of An. cruzii and An. homunculus, which is a secondary malaria vector, are morphologically similar and difficult to distinguish when using external morphological characteristics only. These two species may occur syntopically with An. bellator, which is also a potential vector of Plasmodium species and is morphologically similar to An. cruzii and An. homunculus. Identification of these species based on female specimens is often jeopardised by polymorphisms, overlapping morphological characteristics and damage caused to specimens during collection. Wing geometric morphometrics has been used to distinguish several insect species; however, this economical and powerful tool has not been applied to Kerteszia species. Our objective was to assess wing geometry to distinguish An. cruzii, An. homunculus and An. bellator. Methods: Specimens were collected in an area in the Serra do Mar hotspot biodiversity corridor of the Atlantic Forest biome (Cananeia municipality, State of Sao Paulo, Brazil). The right wings of females of An. cruzii (n= 40), An. homunculus (n= 50) and An. bellator (n= 27) were photographed. For each individual, 18 wing landmarks were subjected to standard geometric morphometrics. Discriminant analysis of Procrustean coordinates was performed to quantify wing shape variation. Results: Individuals clustered into three distinct groups according to species with a slight overlap between representatives of An. cruzii and An. homunculus. The Mahalanobis distance between An. cruzii and An. homunculus was consistently lower (3.50) than that between An. cruzii and An. bellator (4.58) or An. homunculus and An. bellator (4.32). Pairwise cross-validated reclassification showed that geometric morphometrics is an effective analytical method to distinguish between An. bellator, An. cruzii and An. homunculus with a reliability rate varying between 78-88%. Shape analysis revealed that the wings of An. homunculus are narrower than those of An. cruzii and that An. bellator is different from both of the congeneric species. Conclusion: It is possible to distinguish among the vectors An. cruzii, An. homunculus and An. bellator based on female wing characteristics.

Background: The most substantial and best preserved area of Atlantic Forest is within the biogeographical sub-region of Serra do Mar. The topographic complexity of the region creates a diverse array of microclimates, which can affect species distribution and diversity inside the forest. Given that Atlantic Forest includes highly heterogeneous environments, a diverse and medically important Culicidae assemblage, and possible species co-occurrence, we evaluated mosquito assemblages from bromeliad phytotelmata in Serra do Mar (southeastern Brazil). Methods: Larvae and pupae were collected monthly from Nidularium and Vriesea bromeliads between July 2008 and June 2009. Collection sites were divided into landscape categories (lowland, hillslope and hilltop) based on elevation and slope. Correlations between bromeliad mosquito assemblage and environmental variables were assessed using multivariate redundancy analysis. Differences in species diversity between bromeliads within each category of elevation were explored using the Renyi diversity index. Univariate binary logistic regression analyses were used to assess species co-occurrence. Results: A total of 2,024 mosquitoes belonging to 22 species were collected. Landscape categories (pseudo-F value = 1.89, p = 0.04), bromeliad water volume (pseudo-F = 2.99, p = 0.03) and bromeliad fullness (Pseudo-F = 4.47, p < 0.01) influenced mosquito assemblage structure. Renyi diversity index show that lowland possesses the highest diversity indices. The presence of An. homunculus was associated with Cx. ocellatus and the presence of An. cruzii was associated with Cx. neglectus, Cx. inimitabilis fuscatus and Cx. worontzowi. Anopheles cruzii and An. homunculus were taken from the same bromeliad, however, the co-occurrence between those two species was not statistically significant. Conclusions: One of the main findings of our study was that differences in species among mosquito assemblages were influenced by landscape characteristics. The bromeliad factor that influenced mosquito abundance and assemblage structure was fullness. The findings of the current study raise important questions about the role of An. homunculus in the transmission of Plasmodium in Serra do Mar, southeastern Atlantic Forest.

Abstract Background The molecular phylogenetic relationships and population structure of the species of the Anopheles triannulatus complex: Anopheles triannulatus s.s., Anopheles halophylus and the putative species Anopheles triannulatus C were investigated. Methods The mitochondrial COI gene, the nuclear white gene and rDNA ITS2 of samples that include the known geographic distribution of these taxa were analyzed. Phylogenetic analyses were performed using Bayesian inference, Maximum parsimony and Maximum likelihood approaches. Results Each data set analyzed septely yielded a different topology but none provided evidence for the seption of An. halophylus and An. triannulatus C, consistent with the hypothesis that the two are undergoing incipient speciation. The phylogenetic analyses of the white gene found three main clades, whereas the statistical parsimony network detected only a single metapopulation of Anopheles triannulatus s.l. Seven COI lineages were detected by phylogenetic and network analysis. In contrast, the network, but not the phylogenetic analyses, strongly supported three ITS2 groups. Combined data analyses provided the best resolution of the trees, with two major clades, Amazonian (clade I) and trans-Andean + Amazon Delta (clade II). Clade I consists of multiple subclades: An. halophylus + An. triannulatus C; trans-Andean Venezuela; central Amazonia + central Bolivia; Atlantic coastal lowland; and Amazon delta. Clade II includes three subclades: Panama; cis-Andean Colombia; and cis-Venezuela. The Amazon delta specimens are in both clades, likely indicating local sympatry. Spatial and molecular variance analyses detected nine groups, corroborating some of subclades obtained in the combined data analysis. Conclusion Combination of the three molecular markers provided the best resolution for differentiation within An. triannulatus s.s. and An. halophylus and C. The latest two species seem to be very closely related and the analyses performed were not conclusive regarding species differentiation. Further studies including new molecular markers would be desirable to solve this species status question. Besides, results of the study indicate a trans-Andean origin for An. triannulatus s.l. The potential implications for malaria epidemiology remain to be investigated.

The scope of my research project is to produce and characterize new crystalline forms of organic compounds, focusing the attention on co-crystals and then transferring these notions on APIs to produce co-crystals of potential interest in the pharmaceutical field. In the first part of this work co-crystallization experiments were performed using as building blocks the family of aliphatic dicarboxylic acids HOOC-(CH2)n-COOH, with n= 2-8. This class of compounds has always been an object of study because it is characterized by an interesting phenomenon of alternation of melting points: the acids with an even number of carbon atoms show a melting point higher than those with an odd one. The acids were co-crystallized with four dipyridyl molecules (formed by two pyridine rings with a different number of bridging carbon atoms) through the formation of intermolecular interactions N•••(H)O. The bases used were: 4,4’-bipyridine (BPY), 1,2-bis(4-pyridyl)ethane (BPA), 1,2-(di-4-pyridyl)ethylene (BPE) and 1,2-bis(4-pyridyl)propane (BPP). The co-crystals obtained by solution synthesis were characterized by different solid-state techniques to determine the structure and to see how the melting points in co-crystals change. In the second part of this study we tried to obtain new crystal forms of compounds of pharmaceutical interest. The APIs studied are: O-desmethylvenlafaxine, Lidocaine, Nalidixic Acid and Sulfadiazine. Each API was subjected to Polymorph Screening and Salt/Co-crystal Screening experiments to identify new crystal forms characterized by different properties. In a typical Salt/Co-crystal Screening the sample was made to react with a co-former (solid or liquid) through different methods: crystallization by solution, grinding, kneading and solid-gas reactions. The new crystal forms obtained were characterized by different solid state techniques (X-ray single crystal diffraction, X-ray powder diffraction, Differential Scanning Calorimetry, Thermogravimetric Analysis, Evolved gas analysis, FT-IR – ATR, Solid State N.M.R).

Bacteriophage BPP-1, which infects Bordetella species, can switch its specificity by mutations to the ligand-binding surface of its major tropism-determinant protein, Mtd. This targeted mutagenesis results from the activity of a phage-encoded diversity-generating retroelement. Purified Mtd binds its receptor with low affinity, yet BPP-1 binding and infection of Bordettella cells are efficient because of high-avidity binding between phage-associated Mtd and its receptor. Here, using an integrative approach of three-dimensional (3D) structural analyses of the entire phage by cryo-electron tomography and single-prticle cryo-electron microscopy, we provide direct localization of Mtd in the phage and the structural basis of the high-avidity binding of the BPP-1 phage. Our structure shows that each BPP-1 particle has a T = 7 icosahedral head and an unusual tail apparatus consisting of a short central tail "hub," six short tail spikes, and six extended tail fibers. Subtomographic averaging of the tail fiber maps revealed a two-lobed globular structure at the distal end of each long tail fiber. Tomographic reconstructions of immuno-gold-labeled BPP-1 directly localized Mtd to these globular structures. Finally, our icosahedral reconstruction of the BPP-1 head at 7A resolution reveals an HK97-like major capsid protein stabilized by a smaller cementing protein. Our structure represents a unique bacteriophage reconstruction with its tail fibers and ligand-binding domains shown in relation to its tail apparatus. The localization of Mtd at the distal ends of the six tail fibers explains the high avidity binding of Mtd molecules to cell surfaces for initiation of infection.

En este artículo se presentan datos experimentales de resistencia a flexión y a compresión de morteros de cemento Portland con adición y sustitución de breas de petróleo y de alquitrán de carbón, que son subproductos de la industria del carbón o del petróleo. Los materiales estudiados son breas de alquitrán de carbón A (BACA) y B (BACB), y dos breas de petróleo (BPP) y (BPT). Los datos demuestran la viabilidad del uso de estas breas en la fabricación de morteros con menores contenidos de cemento, permitiendo diseñar un nuevo material sostenible con el medio ambiente y que contribuya a reducir el impacto ambiental de los materiales de construcción, hecho que permite abrir una nueva vía de valorización de estos subproductos.

This thesis is concerned with investigations of the effects of molecular encounters on nuclear magnetic resonance spin-lattice relaxation times, with particular reference to mesitylene in mixtures with cyclohexane and TMS. The purpose of the work was to establish the best theoretical description of T1 and assess whether a recently identified mechanism (buffeting), that influences n.m.r. chemical shifts, governs Tl also. A set of experimental conditions are presented that allow reliable measurements of Tl and the N. O. E. for 1H and 13C using both C. W. and F.T. n.m.r. spectroscopy. Literature data for benzene, cyclohexane and chlorobenzene diluted by CC14 and CS2 are used to show that the Hill theory affords the best estimation of their correlation times but appears to be mass dependent. Evaluation of the T1 of the mesitylene protons indicates that a combined Hill-Bloembergen-Purcell-Pound model gives an accurate estimation of T1; subsequently this was shown to be due to cancellation of errors in the calculated intra and intemolecular components. Three experimental methods for the separation of the intra and intermolecular relaxation times are described. The relaxation times of the 13C proton satellite of neat bezene, 1,4 dioxane and mesitylene were measured. Theoretical analyses of the data allow the calculation of Tl intra. Studies of intermolecular NOE's were found to afford a general method of separating observed T1's into their intra and intermolecular components. The aryl 1H and corresponding 13C T1 values and the NOE for the ring carbon of mesitylene in CC14 and C6H12-TMS have been used in combination to determine T1intra and T1inter. The Hill and B.P.P. models are shown to predict similarly inaccurate values for T1linter. A buffeting contribution to T1inter is proposed which when applied to the BPP model and to the Gutowsky-Woessner expression for T1inter gives an inaccuracy of 12% and 6% respectively with respect to theexperimentally based T1inter.

A model of an overall telecommunication network with virtual circuits switching, in stationary state, with Bernoulli-Poisson-Pascal (BPP) input flow, repeated calls, limited number of homogeneous terminals and 8 types of losses is considered. One of the main problems of network redimensioning is estimation of the traffic offered in the network because it reflects on finding of necessary number of equivalent switching lines on the basis of the consideration of detailed users behavior and target Quality of Service (QoS). The aim of this paper is to find a new solution of Network Redimensioning Task (NRDT) [4], taking into account the inconvenience of necessary measurements, not considered in the previous research [5]. The results are applicable for redimensioning of every (virtual) circuit switching telecommunication system, both for wireline and wireless systems (GSM, PSTN, ISDN and BISDN). For packet - switching networks proposed approach may be used as a comparison basis and when they work in circuit switching mode (e.g. VoIP).

Une taxonomie révisée et une connaissance des limites d’espèces demeurent toujours importantes dans les points chauds en biodiversité comme les Antilles où de nombreuses espèces endémiques sont retrouvées. Des limites d’espèces divergentes impliquent un différent nombre d’espèces retrouvées dans un écosystème, ce qui peut exercer une influence sur les décisions prises face aux enjeux de conservation. Les genres Gesneria et Rhytidophyllum qui forment les principaux représentants de la famille des Gesneriaceae dans les Antilles comprennent plusieurs taxons aux limites d’espèces ambigües et quelques espèces qui ont des sous-espèces reconnues. C’est le cas de Gesneria viridiflora (Decne.) Kuntze qui comprend quatre sous-espèces géographiquement isolées et qui présentent des caractères végétatifs et reproducteurs similaires et variables. Une délimitation d’espèces approfondie de ce complexe d’espèce est effectuée ici à partir d’une approche de taxonomie intégrative considérant des données morphologiques, génétiques et bioclimatiques. Les données morphologiques quantitatives et qualitatives obtenues à partir de spécimens d’herbier sont utilisées pour délimiter des groupes morphologiques à l’aide d’une analyse en coordonnées principales. Ces groupes sont ensuite testés à l’aide de séquences d’ADN de quatre régions nucléaires en utilisant une méthode bayesienne basée sur la théorie de la coalescence. Finalement, les occurrences et les valeurs de variables de température et de précipitation qui y prévalent sont utilisées dans une analyse en composantes principales bioclimatique pour comparer les groupes délimités morphologiquement et génétiquement. Les résultats de l’analyse morphologique multivariée supportent la distinction entre les groupes formés par les sous-espèces actuellement reconnues de G. viridiflora. Les résultats, incluant des données génétiques, suggèrent une distinction jusqu’ici insoupçonnée des populations du Massif de la Hotte au sud-ouest d’Haïti qui sont génétiquement plus rapprochées des populations de Cuba que de celles d’Hispaniola. Bioclimatiquement, les groupes délimités par les analyses morphologiques et génétiques sont distincts. L’approche de taxonomie intégrative a permis de distinguer cinq espèces distinctes plutôt que les quatre sous-espèces acceptées jusqu’à aujourd’hui. Ces espèces sont : G. acrochordonanthe, G. quisqueyana, G. sintenisii, G. sylvicola et G. viridiflora. Une carte de distribution géographique, un tableau de la nouvelle taxonomie applicable et une clé d’identification des espèces sont présentés. La nouvelle taxonomie déterminée dans cette étude démontre un endémisme insoupçonné dans plusieurs régions du point chaud en biodiversité des Antilles et souligne l’importance d’investiguer les limites d’espèces dans les groupes diversifiés comprenant des taxons aux limites d’espèces incomprises.

O trabalho teve por objetivo caracterizar o estoque de anchoita (Engraulis anchoita) capturado na região sul do Brasil, visando à utilização deste recurso de alto valor biológico no desenvolvimento de produtos semi-prontos e de fácil preparo, tipo empanado. Os experimentos foram conduzidos com anchoita resultante de cruzeiros realizados pelo Navio Oceanográfico Atlântico Sul da Universidade Federal do Rio Grande (FURG), RS, Brasil. Os exemplares foram capturados entre a cidade de Rio Grande (32ºS, RS-Brasil) e 51ºW. Após captura, o pescado foi armazenado a bordo em mistura de gelo e água do mar, na razão 1:1. As amostras foram transportadas para o laboratório de Biotecnologia da FURG e mantidas sob congelamento a -18°C, até a realização das análises. O trabalho está constituído por uma revisão bibliográfica, que enfatiza a importância do recurso pesqueiro em estudo como potencial a ser explorado, discorre sobre ácidos graxos e perfil de voláteis, bem como, o desenvolvimento de produtos à base de pescado. O desenvolvimento do trabalho é expresso por quatro artigos. O primeiro teve como objetivo caracterizar o estoque de anchoita segundo a biometria, rendimento, composição proximal, compostos nitrogenados e ácidos graxos. O rendimento, a composição proximal e o perfil de ácidos graxos foram realizados nas três frações que compõe o peixe: músculo claro, escuro e vísceras. A análise dos resultados demonstrou a variabilidade dos componentes em função das frações avaliadas e da época de captura, o que pode contribuir para a escolha do processo tecnológico a ser aplicado no desenvolvimento de produtos de alto valor agregado a partir dessa matéria-prima. No segundo artigo foi determinado o perfil de ácidos graxos da anchoita e avaliado o comportamento destes compostos durante o armazenamento congelado, bem como, dos voláteis gerados. Os resultados demonstraram a influência do armazenamento na modificação dos ácidos graxos, em especial, EPA e DHA, e que os voláteis gerados podem ser um índice em potencial para avaliar a qualidade da anchoita congelada. No terceiro artigo objetivou-se selecionar e treinar julgadores para avaliação do odor a pescado utilizando os padrões referência obtidos a partir do perfil de voláteis. Neste sentido, foi levantada a terminologia que descreve o odor da anchoita, definido padrões referência, bem como, selecionado e treinado uma equipe de julgadores. Foram utilizados 20 candidatos, deste total, 9 foram selecionados pelo método das amplitudes. Os julgadores selecionados foram submetidos ao treinamento no uso de escala não estruturada e na avaliação da intensidade do odor a pescado. O desempenho dos julgadores foi definido utilizando como amostra solução de lavagem resultante do processo de obtenção de base protéica de anchoita. Os resultados foram avaliados com base no poder de discriminação, repetibilidade das respostas e concordância entre julgadores, segundo análise de variância, com duas fontes de variação (amostra e repetições). Foram obtidos os valores de Famostra e Frepetição, para cada julgador. Os julgadores com o valor de Famostra significativo (p≤0,30) e Frepetição não significativo (p>0,05), bem como, concordância de médias com os demais julgadores foram considerados treinados. Segundo esse processo a equipe foi constituída por 8 julgadores selecionados e treinados na avaliação do odor a pescado. Finalmente, no quarto artigo foi avaliada a possibilidade de uso de base protéica (BPP) de anchoita na elaboração de massa base de empanados, bem como, em substituição a farinha de cobertura. Para obtenção das BPPs, foram testadas duas soluções extratoras (3 ciclos de extração com ácido fosfórico 0,05% e 1 ciclo de ácido fosfórico seguido de 2 ciclos com água). A BPP obtida na melhor condição utilizada foi seca a 70°C e submetida ao processo de moagem em moinho de facas para ser utilizada como farinha de cobertura. Formulações de empanado utilizando diferentes concentrações (25, 50, 75 e 100%) de anchoita desidratada na cobertura foram testadas no produto frito e forneado. Um teste de preferência com consumidores em potencial foi aplicado às diferentes formulações. Os resultados indicaram que a melhor condição de lavagem para obtenção das BPPs testadas foi quando são utilizados 3 ciclos de extração com ácido fosfórico. A avaliação da preferência junto ao consumidor em potencial demonstrou que a anchoita desidratada pode ser utilizada como farinha de cobertura em empanados na concentração de até 75%.

Une taxonomie révisée et une connaissance des limites d’espèces demeurent toujours importantes dans les points chauds en biodiversité comme les Antilles où de nombreuses espèces endémiques sont retrouvées. Des limites d’espèces divergentes impliquent un différent nombre d’espèces retrouvées dans un écosystème, ce qui peut exercer une influence sur les décisions prises face aux enjeux de conservation. Ces espèces sont : G. acrochordonanthe, G. quisqueyana, G. sintenisii, G. sylvicola et G. viridiflora. Une carte de distribution géographique, un tableau de la nouvelle taxonomie applicable et une clé d’identification des espèces sont présentés. La nouvelle taxonomie déterminée dans cette étude démontre un endémisme insoupçonné dans plusieurs régions du point chaud en biodiversité des Antilles et souligne l’importance d’investiguer les limites d’espèces dans les groupes diversifiés comprenant des taxons aux limites d’espèces incomprises.

The human activities responsible for the ambient degradation in the modern world are diverse. The industrial activities are preponderant in the question of the impact consequences for brazilian ecosystems. Amongst the human activities, the petroliferous industry in operation in Potiguar Petroliferous Basin (PPB) displays the constant risk of ambient impacts in the integrant cities, not only for the human populations and the environment, but also it reaches the native microorganisms of Caatinga ground and in the mangrove sediment. Not hindering, the elaboration of strategies of bioremediation for impacted areas pass through the knowledge of microbiota and its relations with the environment. Moreover, in the microorganism groups associated to oil, are emphasized the sulfate-reducing prokaryotes (SRP) that, in its anaerobic metabolism, these organisms participate of the sulfate reduction, discharging H2S, causing ambient risks and causing the corrosion of surfaces, as pipelines and tanks, resulting in damages for the industry. Some ancestries of PRS integrate the Archaea domain, group of microorganisms whose sequenced genomes present predominance of extremophilic adaptations, including surrounding with oil presence. This work has two correlated objectives: i) the detection and monitoring of the gene dsrB, gift in sulfate-reducing prokaryotes, through DGGE analysis in samples of mDNA of a mangrove sediment and semiarid soil, both in the BPP; ii) to relate genomic characteristics to the ecological aspects of Archaea through in silico studies, standing out the importance to the oil and gas industry. The results of the first work suggest that the petrodegraders communities of SRP persist after the contamination with oil in mangrove sediment and in semiarid soil. Comparing the populations of both sites, it reveals that there are variations in the size and composition during one year of experiments. In the second work, functional and structural factors are the probable cause to the pressure in maintenance of the conservation of the sequences in the multiple copies of the 16S rDNA gene. Is verified also the discrepancy established between total content GC and content GC of the same gene. Such results relating ribosomal genes and the ambient factors are important for metagenomic evaluations using PCR-DGGE. The knowledge of microbiota associated to the oil can contribute for a better destination of resources by the petroliferous industry and the development of bioremediation strategies. Likewise, search to lead to the best agreement of the performance of native microbiota in biogeochemical cycles in Potiguar Petroliferous Basin ecosystem
