Background. Melatonin is a free radical scavenger with important actions in the study of renal ischemia and reperfusion (I/R). This study evaluated possible renal protection of high doses of melatonin in an experimental model of I/R in which rats were submitted to acute hyperglycemia under anesthesia with isoflurane.Method. Forty-four male Wistar rats, weighing more than 300 g, were randomly divided into 5 groups: G1, sham (n = 10); G2, melatonin (n = 10; 50 mg.kg(-1)); G3, hyperglycemia (n = 9; glucose 2.5 g.kg(-1)); G4, hyperglycemia/melatonin (n = 10; 2.5 g.kg(-1) glucose + melatonin 50 mg.kg(-1)); and G5, I/R (n = 5). In all groups, anesthesia was induced with 4% isoflurane and maintained with 1.5% to 2.0% isoflurane. Intraperitoneal injection of melatonin (G1, G4), glucose (G3, G4), or saline (G1, G5) was performed 40 minutes before left renal ischemia. Serum plasma values for creatinine and glucose were determined at baseline (M1), immediately following reperfusion (M2), and 24 hours after completion of the experiment (M3). Histological analysis was performed to evaluate tubular necrosis (0-5).Results. Serum glucose was higher at M2 in the groups supplemented with glucose, hyperglycemia (356.00 +/- 107.83), and hyperglycemia/melatonin (445.3 +/- 148.32). Creatinine values were higher at T3 (P = .0001) for I/R (3.6 +/- 0.37), hyperglycemia/melatonin (3.9 +/- 0.46), and hyperglycemia (3.71 +/- 0.69) and lower in the sham (0.79 +/- 0.16) and melatonin (2.01 +/- 1.01) groups, P < .05. Histology showed no necrosis injury in the G1, lesion grade 2 in the G2, and severe acute tubular necrosis in the G3: (grade 4), G4: (grade 5) and G5: (grade 4) groups (P < .0001).Discussion. Melatonin protected the kidneys submitted to I/R in rats without hyperglycemia; however, this did not occur when the I/R lesion was associated with hyperglycemia.Conclusions. Due to its antioxidant and antiapoptotic action, melatonin was able to mitigate, but not prevent acute tubular necrosis in rats with hyperglycemia under anesthesia by isoflurane.

Aims: The study investigated the in vivo antioxidant activity and the in vitro radical scavenging capacity of the Combretum lanceolatum Pohl (Combretaceae) flowers ethanolic extract (ClEtOH) in streptozotocin-diabetic rats. Place and Duration of Study: Department of Chemistry, Federal University of Mato Grosso, Cuiabá, Brazil; between February 2012 and December 2012. Methodology: Male Wistar rats were divided into four groups: Normal rats treated with water/vehicle (N); diabetic rats treated with water (DC); diabetic rats treated with 250 mg/kg (DT250) or with 500mg/kg (DT500) of ClEtOH. After 21 days of treatment, liver samples were used for the analysis of the oxidative stress biomarkers and activity of antioxidant enzymes. In vitro radical scavenger capacity was investigated by the following methods: DPPH radical scavenging, ABTS radical cation decolorization and crocin bleaching assays. Results: Significant oxidative stress was observed in liver of DC, since the malondialdehyde (MDA, biomarker of lipoperoxidation) levels were increased in comparison with N. Increased activities of the antioxidant enzymes superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were also observed in DC, which could represent a compensatory mechanism against oxidative stress. Glutathione (GSH) levels were lower and similar between N and DC. The MDA levels were significantly decreased in liver of rats from DT250 and DT500, reaching levels similar those of N, suggesting that ClEtOH prevented lipoperoxidation. The treatment of diabetic rats with ClEtOH also increased the GSH levels, as well as increased the GSH-Px activity, and did not change the SOD activity. The results of in vitro radical scavenging capacity indicated that ClEtOH is highly active. Conclusion: These findings indicate that ClEtOH has antioxidant properties in liver of diabetic rats, decreasing lipoperoxidation and increasing the endogenous antioxidant responses. Both the antihyperglycemic effect and the capacity to scavenge free radicals may be related to the antioxidant activity of ClEtOH in diabetes.

The protective effect of gallic acid and its esters, methyl, propyl, and lauryl gallate, against 2,2'-azobis(2-amidinopropane)hydrochloride (AAPH)-induced hemolysis and depletion of intracellular glutathione (GSH) in erythrocytes was studied. The inhibition of hemolysis was dose-dependent, and the esters were significantly more effective than gallic acid. Gallic acid and its esters were compared with regard to their reactivity to free radicals, using the DPPH and AAPH/pyranine free-cell assays, and no significant difference was obtained. Gallic acid and its esters not only failed to inhibit the depletion of intracellular GSH in erythrocytes induced by AAPH but exacerbated it. Similarly, the oxidation of GSH by AAPH or horseradish peroxidase/H(2)O(2) in cell-free systems was exacerbated by gallic acid or gallates. This property could be involved in the recent findings on pro-apoptotic and pro-oxidant activities of gallates in tumor cells. We provide evidence that lipophilicity and not only radical scavenger potency is an important factor regarding the efficiency of antihemolytic substances.

Increased reactive oxygen species (ROS) promote matrix metalloproteinase (MMP) activities and may underlie cardiomyocyte injury and the degradation of cardiac troponin I (cTI) during acute pulmonary thromboembolism (APT). We examined whether pretreatment or therapy with tempol (a ROS scavenger) prevents MMP activation and cardiomyocyte injury of APT. Anesthetized sheep received tempol infusion (1.0 mg kg(-1) min(-1), i.v.) or saline starting 30 min before or 30 min after APT (autologous blood clots). Control animals received saline. Hemodynamic measurements were performed. MMPs were studied in the right ventricle (RV) by gelatin zymography, fluorimetric activity assay, and in situ zymography. The ROS levels were determined in the RV and cTI were measured in serum samples. APT increased the pulmonary arterial pressure and pulmonary vascular resistance by 146 and 164 %, respectively. Pretreatment or therapy with tempol attenuated these increases. While APT increased RV + dP/dt (max), tempol infusions had no effects. APT increased RV MMP-9 (but not MMP-2) levels. In line with these findings, APT increased RV MMP activities, and this finding was confirmed by in situ zymography. APT increased the RV ROS levels and tempol infusion, before or after APT, and blunted APT-induced increases in MMP-9 levels, MMP activities, in situ MMP activities, and ROS levels in the RV. cTI concentrations increased after APT, and tempol attenuated these increases. RV oxidative stress after APT increases the RV MMP activities, leading to the degradation of sarcomeric proteins, including cTI. Antioxidant treatment may prevent MMP activation and protect against cardiomyocyte injury after APT.

A highly concentrated aqueous saline-containing solution of phenol, 2,4-dichlorophenoxyacetic acid (2,4-D) and 2,4-dichlorophenol (2.4-DCP) was treated by the photo-Fenton process in a system composed of an annular reactor with a quartz immersion well and a medium-pressure mercury lamp (450 W). The study was conducted under special conditions to minimize the costs of acidification and neutralization, which are usual steps in this type of process. Photochemical reactions were carried out to investigate the influence of some process variables such as the initial concentration of Fe2+ ([Fe2+](0)) from 1.0 up to 2.5 mM, the rate in mmol of H2O2 fed into the system (F-H2O2,F-in) from 3.67 up to 7.33 mmol of H2O2/min during 120 min of reaction time, and the initial pH (pH(0)) from 3.0 up to 9.0 in the presence and absence of NaCl (60.0 g/L). Although the optimum pH for the photo-Fenton process is about 3.0, this particular system performed well in experimental conditions starting at alkaline and neutral pH. The results obtained here are promising for industrial applications, particularly in view of the high concentration of chloride, a known hydroxyl radical scavenger and the main oxidant present in photo-Fenton processes. Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl) and other cyclic nitroxides have been shown to inhibit the chlorinating activity of myeloperoxidase (MPO) in vitro and in cells. To examine whether nitroxides inhibit MPO activity in vivo we selected acute carrageenan-induced inflammation on the rat paw as a model. Tempol and three more hydrophobic 4-substituted derivatives (4-azido, 4-benzene-Sulfonyl, and 4-(4-phenyl-1H-1,2,3-triazol-1-yl)) were synthesized, and their ability to inhibit the in vitro chlorinating activity of MPO and carrageenan-induced inflammation in rat paws was evaluated. All of the tested nitroxides inhibited the chlorinating activity of MPO in vitro with similar IC50 values (between 1.5 and 1.8 mu M). In vivo, the attenuation of carrageenan-induced inflammation showed some correlation with the lipophilicity of the nitroxide at early time points but the differences in the effects were small (< 2-fold) compared with the differences in lipophilicity (> 200-fold). No inhibition of MPO activity in vivo was evident because the levels of MPO activity in rat paws correlated with the levels of MPO protein'. Likewise, paw edema, levels of nitrated and oxidized proteins, and levels of plasma exudation correlated with the levels of MPO protein in the paws of the animals that were untreated or treated with the nitroxides. The effects of the nitroxides in vivo were compared with those of 4-aminobenzoic hydrazide and of colchicine. Taken together, the results indicate that nitroxides attenuate carrageenan-induced inflammation mainly by reducing neutrophil migration and the resulting MPO-mediated damage. Quantum chemical calculations at the B3LYP/6-31G* level of theory were employed for the structure-activity relationship and prediction of the antioxidant activity of edaravone and structurally related derivatives using energy (E), ionization potential (IP), bond dissociation energy (BDE), and stabilization energies(Delta E-iso). Spin density calculations were also performed for the proposed antioxidant activity mechanism. The electron abstraction is related to electron-donating groups (EDG) at position 3, decreasing the IP when compared to substitution at position 4. The hydrogen abstraction is related to electron-withdrawing groups (EDG) at position 4, decreasing the BDECH when compared to other substitutions, resulting in a better antioxidant activity. The unpaired electron formed by the hydrogen abstraction from the C-H group of the pyrazole ring is localized at 2, 4, and 6 positions. The highest scavenging activity prediction is related to the lowest contribution at the carbon atom. The likely mechanism is related to hydrogen transfer. It was found that antioxidant activity depends on the presence of EDG at the C-2 and C-4 positions and there is a correlation between IP and BDE. Our results identified three different classes of new derivatives more potent than edaravone.

Generierung und Prozessierung oxidativer DNA Schäden --- Ziel dieser Arbeit war es, adaptive Antworten der Zellen auf einen DNA Schädigung zu untersuchen. Hierzu wurden Experimente zur Reparatur oxidierter Basen (Substrate der Basen Exzisions Reparatur (BER)) oder von Pyrimidindimeren (Substrate der Nukleotid Exzisions Reparatur (NER)) nach einer Vorbehandlung mit DNA-schädigender Agenzien durchgeführt. Die Ergebnisse zeigten, dass sowohl eine Vorbehandlung mit einer alkylierenden als auch mit einer oxidierenden Substanz zu einer adaptiven Erhöhung des zellulären Glutathionspiegels führte, die 16 h nach der Schädigung ihr Maximum erreichte. Jedoch waren die 8-oxoG Glykosylaseaktivitäten über einen Zeitraum von 18 h konstant. Diese Effekte waren unabhängig davon, ob Maus Embryofibroblasten, primäre oder p53 profiziente menschliche Zellen verwendet wurden. Die BER war ebenfalls in keiner der verschiedenen Zelllinien signifikant verbessert. Die adaptive Antwort bezüglich der Glutathionspiegel war also nicht mit einer entsprechenden Veränderung bei der DNA-Reparatur verbunden. Folglich ist die Reparatur von oxidativen DNA-Schäden durch eine vorausgehende Schädigung nicht induzierbar. Der zweite Teil der Untersuchungen zu der Reparatur beschäftigte sich mit der NER. Hierzu wurde die Reaktivierung eines mit UVB-Strahlung geschädigten Plasmids untersucht. Als Wirtszellen fungierten primäre menschliche Fibroblasten und Keratinozyten, die entweder mit UVB vorbehandelt oder ungeschädigt waren. Auch für die NER konnte keine signifikante Beschleunigung der Reparatur von Pyrimidindimeren durch eine Vorbehandlung festgestellt werden. Die Reaktivierung erfolgte ferner unabhängig vom p53-Status der Zellen, wie Versuche mit p53-siRNA zeigten. Neben der Prozessierung war die Generierung oxidativer DNA Schäden Gegenstand der Arbeit. Die verwendete Substanz Tirapazamin (TPZ) ist ein für hypoxische Zellen selektives, neues Zytostatikum und befindet sich momentan in Phase 2/3 der klinischen Prüfung. Ziel war es die von TPZ verursachten DNA Modifikationen zu charakterisieren, sowie die Toxizität und Genotoxizität zu untersuchen. Da es Hinweise auf eine Aktivierung von TPZ über eine Oxidoreduktase (OR) gab, wurden die Experimente in Wildtyp und hOR überexprimierenden Zellen durchgeführt. Die Quantifizierung der verursachten DNA-Modifikationen zeigte, dass der von TPZ verursachte Schaden in Zellen mit hOR erhöht war. Das erhaltene Schadensprofil der durch TPZ verursachten DNA-Modifikationen war dem Schadensprofil von durch Gamma-Strahlung intrazellulär verursachten Hydroxylradikalen sehr ähnlich. Da es nach der Aktivierung von TPZ durch eine OR zu einer Abspaltung von Hydroxylradikalen kommt, bestätigte dies den vermuteten Mechanismus. Weitere Untersuchungen mit t-Butanol, einem Hydroxylradikal Fänger, ergaben eine verminderte DNA-Schädigung, was ebenfalls für eine DNA-Schädigung durch Hydroxylradikale spricht. Untersuchungen zur Mutagenität zeigten das die Mutationsrate in Zellen mit hOR um das 4 fache erhöht ist. Erstaunlich war jedoch, dass der im gleichen Ausmaß von Gamma-Strahlung verursachte DNA-Schaden für die beobachtete Toxizität dieser verantwortlich war, während bei TPZ unter den gleichen Bedingungen keine Toxizität vorlag. Erklärt werden könnte die erhöhte Toxizität und Mutagenität durch so genannte geclusterte DNA-Schäden, die von Gamma-Strahlen, nicht jedoch von TPZ gebildet werden. Nach einer verlängerten Inkubation wurde sowohl für die Toxizität als auch für die Genotoxizität erneut ein verstärkender Effekt durch die OR bestätigt. Überraschend war weiterhin die von der OR unabhängige Generierung von Doppelstrangbrüchen, für die demnach ein grundsätzlich anderer Mechanismus, wie zum Beispiel eine direkte Interaktion mit der Topoisomerase II, angenommen werden muss.

In der vorliegenden Arbeit werden 52 Verbindungen beschrieben, welche auf COX/LOX-Inhibition mit zusätzlichen Hydroxylradikalfängereigenschaften getestet worden sind. rnEs war möglich eine neue Synthesestrategie für noch nicht beschriebene 4,5-Diarylisoselenazole zu entwickeln und eine vorhandene Synthese für Isothiazoliumchloride von zwei Stufen, mit mäßigen Ausbeuten, auf eine Stufe, mit hoher Ausbeute, zu verkürzen.rnEs wurden mehrere COX-Inhibitoren identifiziert. MSD4a, MSD4h, MSD5a und MSD5h konnten als COX-1-, COX-2- und 5-LOX-Hemmer identifiziert werden. Besonders hervorzuheben ist die Verbindung MSD5h, die zusätzlich zur COX-1-, COX-2- und 5-LOX-Inhibition eine leichte Hemmung im Hydroxylradikalfänger-Assay zeigt, für die ein clog P-Wert von 2,65 berechnet wurde und die im XTT-Zytotoxizitätstestsystem, selbst bei einer Konzentration von 100 µM, kaum toxische Eigenschaften besitzt.rnWeiterhin war es möglich zu zeigen, dass Carbonsäuren gute Hydroxylradikalfängereigenschaften in unserem, auf der Fenton-Reaktion basierenden, Testsystem haben. Die Potenz der Carbonsäuren MSD8b und MSD11j im Vergleich zu den unwirksamen korrespondierenden Ester MSD8a und MSD11i führte zu Untersuchungen mit weiteren Carbonsäuren und deren Ester. Um den Wirkungsmechanismus zu erforschen wurde das Testsystem modifiziert, um eine Komplexierung der Eisenionen durch die Carbonsäuren auszuschließen. An Hand der Substanzen MSD8b und MSD11j wurde nachgewiesen, dass diese mit dem Hydroxylradikal reagieren, ohne zu decarboxylieren oder andere Zerfallsreaktionen einzugehen.rnZusätzlich zu den Untersuchungen der Enzym-Inhibition sowie des Hydroxylradikal-Scavenings wurden Molecular Modelling Studien durchgeführt. Die Ergebnisse der Dockingstudien in COX-1- (1eqg), COX-2- (1cx2) und in COX-1 mutierte COX-2-Kristallstrukturen (1cx2) führen zu einer kritischen Bewertung des folgenden Ansatzes: Es ist nicht unbedingt sinnvoll zuerst Strukturen mit dem Computer zu entwerfen und zu modeln und sie erst dann zu synthetisieren und in Enzym- oder Zellassays zu testen. Die Begründung dafür liegt in der Schwierigkeit einschätzen zu können, wie nah das gewählte Modell der Wirklichkeit ist. In den durchgeführten Dockingstudien konnte der sehr große Einfluss des kokristallisierten Liganden in der als Grundlage dienenden Kristallstruktur auf die Dockingergebnisse gezeigt werden. Durch einen zu kleinen kokristallisierten Liganden in der COX-1-Bindungstasche wurden als Ergebnis der Dockingstudie alle Verbindungen als nicht potent eingestuft, obwohl diese zum Teil im Enzymtestsystem wirksam waren. Dies konnte mit den Mutationsversuchen ausgeglichen werden. rnDeshalb kann man aus diesen Ergebnissen als Fazit ziehen, dass eine Strategie, Strukturen zu synthetisieren, in vitro zu testen und dabei die Strukturentwicklung mit Molecular Modelling Studien zu unterstützen, die Methode der Wahl darstellt.rn

Experimental bacterial meningitis due to Streptococcus pneumoniae in infant rats was associated with a time-dependent increase in CSF and cortical urate that was approximately 30-fold elevated at 22 h after infection compared to baseline. This increase was mirrored by a 20-fold rise in cortical xanthine oxidoreductase activity. The relative proportion of the oxidant-producing xanthine oxidase to total activity did not increase, however. Blood plasma levels of urate also increased during infection, but part of this was as a consequence of dehydration, as reflected by elevated ascorbate concentrations in the plasma. Administration of the radical scavenger alpha-phenyl-tert-butyl nitrone, previously shown to be neuroprotective in the present model, did not significantly affect either xanthine dehydrogenase or xanthine oxidase activity, and increased even further cortical accumulation of urate. Treatment with the xanthine oxidoreductase inhibitor allopurinol inhibited CSF urate levels earlier than those in blood plasma, supporting the notion that urate was produced within the brain. However, this treatment did not prevent the loss of ascorbate and reduced glutathione in the cortex and CSF. Together with data from the literature, the results strongly suggest that xanthine oxidase is not a major cause of oxidative stress in bacterial meningitis and that urate formation due to induction of xanthine oxidoreductase in the brain may in fact represent a protective response.

Since 3-hydroxyanthranilic acid (3HAA), an oxidation product of tryptophan metabolism, is a powerful radical scavenger [Christen, S., Peterhans, E., ; Stocker, R. (1990) Proc. Natl. Acad. Sci. U.S.A. 87, 2506], its reaction with peroxyl radicals was investigated further. Exposure to aqueous peroxyl radicals generated at constant rate under air from the thermolabile radical initiator 2,2'-azobis[2-amid-inopropane] hydrochloride (AAPH) resulted in rapid consumption of 3HAA with initial accumulation of its cyclic dimer, cinnabarinic acid (CA). The initial rate of formation of the phenoxazinone CA accounted for approximately 75% of the initial rate of oxidation of 3HAA, taking into account that 2 mol of 3HAA are required to form 1 mol of CA. Consumption of 3HAA under anaerobic conditions (where alkyl radicals are produced from AAPH) was considerably slower and did not result in detectable formation of CA. Addition of superoxide dismutase enhanced autoxidation of 3HAA as well as the initial rates of peroxyl radical-induced oxidation of 3HAA and formation of CA by approximately 40-50%, whereas inclusion of xanthine/xanthine oxidase decreased the rate of oxidation of 3HAA by approximately 50% and inhibited formation of CA almost completely, suggesting that superoxide anion radical (O2.-) was formed and reacted with reaction intermediate(s) to curtail formation of CA. Formation of CA was also observed when 3HAA was added to performed compound I of horseradish peroxidase (HRPO) or catalytic amounts of either HRPO, myeloperoxidase, or bovine liver catalase together with glucose/glucose oxidase.(ABSTRACT TRUNCATED AT 250 WORDS)

The present study investigated the role of oxygen-derived free radicals as mediators of acute damage to rat gastric mucosae exposed to topically applied absolute ethanol. Although a hydroxyl radical scavenger, Dimethylthiourea, was noted to exhibit profound gastroprotective properties, other pretreatment regimens employing a host of known free radical scavengers, and enzyme inhibitors failed to confirm this hypothesis. Furthermore, no change in mucosal malondialdehyde, an indicator of free radical attack to cell membranes, could be detected in ethanol exposed tissues. Taken together, the present study fails to confirm that oxygen-derived free radicals mediate the gastric damaging effects of topically applied absolute ethanol. ^

Listeria monocytogenes is a Gram-positive facultative intracellular bacteria which infects a wide range of hosts. In ruminants, infection with L. monocytogenes frequently causes encephalitis, which is usually fatal in sheep and goat, while cattle often recover with antibiotic therapy. Since the role of NO in the control of Listeria is controversial, we have studied the expression of iNOS in the brains of cattle, sheep and goats which had succumbed to listeria encephalitis. iNOS was demonstrated in decreasing intensity in the M phi of microabscesses from cattle, sheep and goat. iNOS expression was accompanied by NT in the microabscesses of cattle, but was only present to a low degree in sheep and was absent in goats. This is indirect evidence for differences in the ability to produce NO in the three species. Presence of iNOS and NT were inversely correlated with the numbers of bacteria. While microabscesses of goats contained high amounts of L. monocytogenes they occurred only rarely in cattle. To corroborate our hypothesis that NO is involved in the control of listeria encephalitis a new animal model was developed. Eleven day old infant rats were infected intracisternally with a low dose of L. monocytogenes. This resulted in a transient meningoencephalitis with moderate clinical signs and low mortality. Listeria proliferated strongly in the inflammatory lesions during the first days of infection, reached a peak at day 4 and were eliminated until day 7. The presence of bacteria was closely accompanied by high numbers of iNOS-expressing M phi and the formation of NT. Administration of the iNOS inhibitor L-NIL or the radical scavenger PBN resulted in rapid death of the treated animals. However, the increase in bacterial numbers was one order of magnitude higher for animals treated with PBN compared with L-NIL administration. This shows that NO plays an important role in the control of a brain infection with Listeria, but suggests that reactive oxidants other than NO are also involved. In conclusion, our findings point to a possible involvement of the differences in the ability to express iNOS and subsequent NO production in the different clinical outcome of listeria encephalitis in cattle and small ruminants.
