Given the exponential growth in the spread of the virus world wide web (Internet) and its increasing complexity, it is necessary to adopt more complex systems for the extraction of malware finger-prints (malware fingerprints - malicious software; is the name given to extracting unique information leading to identification of the virus, equivalent to humans, the fingerprint). The architecture and protocol proposed here aim to achieve more efficient fingerprints, using techniques that make a single fingerprint enough to compromise an entire group of viruses. This efficiency is given by the use of a hybrid approach of extracting fingerprints, taking into account the analysis of the code and the behavior of the sample, so called viruses. The main targets of this proposed system are Polymorphics and Metamorphics Malwares, given the difficulty in creating fingerprints that identify an entire family from these viruses. This difficulty is created by the use of techniques that have as their main objective compromise analysis by experts. The parameters chosen for the behavioral analysis are: File System; Records Windows; RAM Dump and API calls. As for the analysis of the code, the objective is to create, in binary virus, divisions in blocks, where it is possible to extract hashes. This technique considers the instruction there and its neighborhood, characterized as being accurate. In short, with this information is intended to predict and draw a profile of action of the virus and then create a fingerprint based on the degree of kinship between them (threshold), whose goal is to increase the ability to detect viruses that do not make part of the same family

The molecular complex containing the seven transmembrane helix photoreceptor S&barbelow;ensory R&barbelow;hodopsin I&barbelow; (SRI) and transducer protein HtrI (H&barbelow;alobacterial Transducer for SRI&barbelow;) mediates color-sensitive phototaxis responses in the archaeon Halobacterium salinarum. Orange light causes an attractant response by a one-photon reaction and white light (orange + UV light) a repellent response by a two-photon reaction. Three aspects of SRI-HtrI structure/function and the signal transduction pathway were explored. First, the coupling of HtrI to the photoactive site of SRI was analyzed by mutagenesis and kinetic spectroscopy. Second, SRI-HtrI mutations and suppressors were selected and characterized to elucidate the color-sensing mechanism. Third, the signal relay through the transducer-bound histidine kinase was analyzed using an in vitro reconstitution system with known and newly identified taxis components. ^ Twenty-one mutations on HtrI were introduced by site-directed mutagenesis. Several replacements of charged residues perturbed the photochemical kinetics of SRI which led to the finding of a cluster of residues at the membrane/cytoplasm interface in HtrI electrostatically coupled to the photoactive site of SRI. We found by laser-flash kinetic spectroscopy that the transducer and these residues have specific effects on the light-induced proton transfer between the retinal chromophore and the protein. ^ One of the mutations showed an unusual mutant phenotype we called “inverted” signaling, in which the cell produces a repellent response to normally attractant light. Therefore, this mutant (E56Q of HtrI) had lost the color-discrimination by the SRI-HtrI complex. We used suppressor analysis to better understand the phenotype. Certain suppressors resulted in return of attractant responses to orange light but with inversion of the normally repellent response to white light to an attractant response. To explain this and other results, we formulated the Conformational Shuttling model in which the HtrI-SRI complex is poised in a metastable equilibrium of two conformations shifted in opposite directions by orange and white light. We tested this model by behavioral analysis (computerized cell tracking and motion study) of double mutants of inverting and suppressing mutations and the results confirmed the equilibrium-shift explanation. ^ We developed an in vitro system for measuring the effect of purified transducer on the histidine-kinase CheAH that controls the flagellar motor switch. The rate of kinase autophosphorylation was stimulated >2 fold in the reconstitution of the complete signal transduction system from purified components from H. salinarum. The in vitro assay also showed that the kinase activity was reduced in the absence and in the presence of high levels of linker protein CheWH. (Abstract shortened by UMI.) ^

Les dépressions chroniques sont fréquentes et souvent traitées par des approches traditionnelles. Cet article vise à présenter la nature spécifique de la psychopathologie et un traitement spécifiquement adapté à ces patients avec dépression chronique. Nous décrirons d’abord les spécificités psychopathologiques de cette population, en nous référant aux travaux de J. Piaget et de D. Kiesler. À partir de ces théories, nous mettrons en avant le modèle Cognitive Behavioral Analysis System of Psychotherapy (CBASP), selon McCullough. Cet auteur propose deux volets d’interventions spécifiquement adaptées aux patients avec dépression chronique : l’analyse situationnelle et les techniques interpersonnelles basées sur la notion de transfert et de contre-transfert. Nous soulignerons la pertinence de cette approche par le résumé de plusieurs études empiriques ayant établi l’efficacité de ce modèle, sous certaines conditions cliniques. Nous terminerons par une réflexion de l’application de ce modèle au-delà du tableau clinique de la dépression chronique en ajoutant ainsi des arguments supplémentaires en faveur de l’apport du modèle CBASP au champ actuel de la psychothérapie des troubles mentaux.

Self-management is being promoted in cystic fibrosis (CF). However, it has not been well studied. Principal aims of this research were (1) to evaluate psychometric properties of a CF disease status measure, the NIH Clinical Score; (2) to develop and validate a measure of self-management behavior, the SMQ-CF scale, and (3) to examine the relation between self-management and disease status in CF patients over two years.^ In study 1, NIH Clinical Scores for 200 patients were used. The scale was examined for internal consistency, interrater reliability, and content validity using factor analysis. The Cronbach's alpha (.81) and interrater reliability (.90) for the total scale were high. General scale items were less reliable. Factor analysis indicated that most of the variance in disease status is accounted for by Factor 1 which consists of pulmonary disease items.^ The SMQ-CF measures the performance of CF self-management. Pilot testing was done with 98 CF primary caregivers. Internal consistency reliability, social desirability bias, and content validity using factor analysis were examined. Internal consistency was good (alpha =.95). Social desirability correlation was low (r =.095). Twelve factors identified were consistent with conceptual groupings of behaviors. Around two hundred caregivers from two CF centers were surveyed and multivariate analysis of variance was used to assess construct validity. Results confirmed expected relations between self-management, patient age, and disease status. Patient age accounted for 50% and disease status 18% of the variance in the SMQ-CF scale.^ It was hypothesized that self-management would positively affect future disease status. Data from 199 CF patients (control and education intervention groups) were examined. Models of hypothesized relations were tested using LISREL structural equation modeling. Results indicated that the relations between baseline self-management and Time 1 disease status were not significant. Significant relations were observed in self-management behaviors from time 1 to time 2 and patterns of significant relations differed between the two groups.^ This research has contributed to refinements in the ability to measure self-management behavior and disease status outcomes in cystic fibrosis. In addition, it provides the first steps in exploratory behavioral analysis with regard to self-management in this disease. ^

KCNQ1 encodes KCNQ1, which belongs to a family of voltage-dependent K+ ion channel proteins. KCNQ1 associates with a regulatory subunit, KCNE1, to produce the cardiac repolarizing current, IKs. Loss-of-function mutations in the human KCNQ1 gene have been linked to Jervell and Lange–Nielsen Syndrome (JLNS), a disorder characterized by profound bilateral deafness and a cardiac phenotype. To generate a mouse model for JLNS, we created a line of transgenic mice that have a targeted disruption in the Kcnq1 gene. Behavioral analysis revealed that the Kcnq1−/− mice are deaf and exhibit a shaker/waltzer phenotype. Histological analysis of the inner ear structures of Kcnq1−/− mice revealed gross morphological anomalies because of the drastic reduction in the volume of endolymph. ECGs recorded from Kcnq1−/− mice demonstrated abnormal T- and P-wave morphologies and prolongation of the QT and JT intervals when measured in vivo, but not in isolated hearts. These changes are indicative of cardiac repolarization defects that appear to be induced by extracardiac signals. Together, these data suggest that Kcnq1−/− mice are a potentially valuable animal model of JLNS.

While most effects of dopamine in the brain are mediated by the D1 and D2 receptor subtypes, other members of this G protein-coupled receptor family have potentially important functions. D3 receptors belong to the D2-like subclass of dopamine receptors, activation of which inhibits adenylyl cyclase. Using targeted mutagenesis in mouse embryonic stem cells, we have generated mice lacking functional D3 receptors. A premature chain-termination mutation was introduced in the D3 receptor gene after residue Arg-148 in the second intracellular loop of the predicted protein sequence. Binding of the dopamine antagonist [125I]iodosulpride to D3 receptors was absent in mice homozygous for the mutation and greatly reduced in heterozygous mice. Behavioral analysis of mutant mice showed that this mutation is associated with hyperactivity in an exploratory test. Homozygous mice lacking D3 receptors display increased locomotor activity and rearing behavior. Mice heterozygous for the D3 receptor mutation show similar, albeit less pronounced, behavioral alterations. Our findings indicate that D3 receptors play an inhibitory role in the control of certain behaviors.

A natação de águas abertas tem registrado aumento no número de competições e participantes em todo mundo. Acompanhando esta tendência têm sido desenvolvidos estudos para identificar as características físicas e as respostas fisiológicas dos atletas neste tipo de prova. Entretanto, são escassos estudos ao nível de análise comportamental, principalmente, em condições reais de distância e meio ambiente (mar). Foi objetivo deste estudo investigar as características de desempenho e da organização temporal das braçadas de nadadores de águas abertas. Mais especificamente, conhecer quais recursos os atletas de águas abertas lançam mão para atingir sua meta de vencer um percurso no mar no menor tempo possível. A amostra foi constituída por 23 atletas, com média de idade de 26,4(±3,2) anos. A tarefa foi nadar um trajeto de 1500 metros em forma de um circuito em mar aberto. Para a captação das variáveis relacionadas ao desempenho utilizou-se um GPS (Garmin modelo Fênix 3) e um cronômetro (FINIS modelo Accusplit Eagle AX602). O registro das imagens para captação dos dados relacionados à descrição da organização temporal das braçadas ocorreu em três pontos do trajeto: início (I) - 20 a 40 metros, meio (M) - 800 a 820 metros e final (F) - 1450 a 1470 metros. Foi utilizada uma filmadora (Nikon Coolpix S5300) afixada à embarcação. O software Kinovea 8.20 permitiu a análise quadro a quadro das braçadas. Foram consideradas variáveis dependentes relacionadas ao desempenho (tempo, velocidade e distância total percorrida, bem como, a frequência de braçadas em cada um dos três pontos do trajeto); aos aspectos variantes das braçadas (tempo total do ciclo, das braçadas, das fases aérea e aquática) e aos aspectos invariantes das braçadas (timing relativo das fases aérea e aquática e sua variabilidade). A análise de variância de medidas repetidas foi usada para comparar os três momentos da tarefa (I, M e F) para todas as variáveis, e a correlação de Pearson para analisar a magnitude das relações entre as variáveis de desempenho, enquanto o teste t de Student para medidas pareadas foi utilizado para comparar as possíveis diferenças entre os braços direito e esquerdo para cada um dos momentos e determinou-se como significância estatística α≤=0,05. Em relação ao desempenho, os resultados indicaram que os nadadores fizeram uso de frequência de braçada (Fb) diferente para os três momentos, sendo maior no I quando comparada ao M e F, e no M, menor que em F; estas mudanças foram acompanhadas por ajustes nos aspectos variantes como o tempo total do ciclo, das braçadas e das fases aérea e aquática. Ainda, nos três momentos os nadadores apresentaram simetria temporal entre as braçadas dos dois braços, apesar de as diferenças serem evidenciadas entre as fases das braçadas quando comparados os braços. Com relação aos aspectos invariantes detectou-se mudança do padrão de I para M e F da tarefa, sendo que em M e F os atletas utilizaram a mesma estrutura temporal. Quanto à variabilidade dos aspectos variantes e invariantes para as braçadas e as fases das braçadas, observou-se diminuição da magnitude ao longo da tarefa sendo que o braço esquerdo apresentou nos três momentos maior variabilidade que o direito. Assim, diante dos resultados, concluiu-se que os recursos utilizados por nadadores habilidosos para nadar em ambiente pouco estável, em condições reais de distância e meio ambiente (mar) compreendem a alteração do desempenho (Fb) associado a ajustes nos aspectos variantes, concomitantemente à alteração dos aspectos invariantes das braçadas, em função do momento da tarefa
