L’immunité innée est notre premier mécanisme de défense contre l’invasion des pathogènes. Cette défense est basée sur la reconnaissance d’éléments invariables des pathogènes par des récepteurs encodés dans les lignées germinales. Dans la réponse anti-virale, le facteur de transcription Interferon Regulatory Factor 3 (IRF3) joue un rôle clé dans la réponse interféron de type I, combattant ainsi la réplication virale et conférant un état anti-viral aux cellules infectées ainsi qu’aux cellules avoisinantes. IRF3 est une protéine dont l’activation et la phosphorylation sont régulées par les kinases TBK1 et IKKi. Nous proposons ici que l’acétylation est une modification post-traductionnelle importante dans la régulation de l’activité d’IRF3. Nous avons observé par immunobuvardage qu’IRF3 est acétylé de façon basale et que cette acétylation est induite par la présence du co-facteur CBP et est inhibée par la présence de la kinase TBK1. Par spectrométrie de masse, nous avons ensuite identifié huit lysines sujettes à l’acétylation sur IRF3. Aussi, par mutagénèse dirigée, nous avons muté de façon ponctuelle chacun de ces sites et avons déterminé que la mutation de la lysine 87 inhibe la capacité d’IRF3 à s’attacher à l’ADN en EMSA et à transactiver son élément de réponse en essai luciférase. Aussi, nous proposons que l’acétylation masque la charge positive de la lysine 87 et contrôle de façon négative l’activité du facteur de transcription IRF3. Notre groupe démontre ainsi pour la première fois l’acétylation du facteur de transcription dans un modèle cellulaire et propose que ce processus joue un rôle inhibiteur dans la régulation de la protéine.

We previously identified the function of the hepatitis C virus (HCV) p7 protein as an ion channel in artificial lipid bilayers and demonstrated that this in vitro activity is inhibited by amantadine. Here we show that the ion channel activity of HCV p7 expressed in mammalian cells can substitute for that of influenza virus M2 in a cell-based assay. This was also the case for the p7 from the related virus, bovine viral diarrhoea virus (BVDV). Moreover, amantadine was shown to abrogate HCV p7 function in this assay at a concentration that specifically inhibits M2. Mutation of a conserved basic loop located between the two predicted trans-membrane alpha helices rendered HCV p7 non-functional as an ion channel. The intracellular localization of p7 was unaffected by this mutation and was found to overlap significantly with membranes associated with mitochondria. Demonstration of p7 ion channel activity in cellular membranes and its inhibition by amantadine affirm the protein as a target for future anti-viral chemotherapy.

Bunyaviruses are considered to be emerging pathogens facilitated by the segmented nature of their genome that allows reassortment between different species to generate novel viruses with altered pathogenicity. Bunyaviruses are transmitted via a diverse range of arthropod vectors, as well as rodents, and have established a global disease range with massive importance in healthcare, animal welfare and economics. There are no vaccines or anti-viral therapies available to treat human bunyavirus infections and so development of new anti-viral strategies is urgently required. Bunyamwera virus (BUNV; genus Orthobunyavirus) is the model bunyavirus, sharing aspects of its molecular and cellular biology with all Bunyaviridae family members. Here, we show for the first time that BUNV activates and requires cellular potassium (K+) channels to infect cells. Time of addition assays using K+ channel modulating agents demonstrated that K+ channel function is critical to events shortly after virus entry but prior to viral RNA synthesis/replication. A similar K+ channel dependence was identified for other bunyaviruses namely Schmallenberg virus (Orthobunyavirus) as well as the more distantly related Hazara virus (Nairovirus). Using a rational pharmacological screening regimen, twin-pore domain K+ channels (K2P) were identified as the K+ channel family mediating BUNV K+ channel dependence. As several K2P channel modulators are currently in clinical use, our work suggests they may represent a new and safe drug class for the treatment of potentially lethal bunyavirus disease.

Background Ageing increases risk of respiratory infections and impairs the response to influenza vaccination. Pre- and probiotics offer an opportunity to modulate anti-viral defenses and the response to vaccination via alteration of the gut microbiota. This study investigated the effect of a novel probiotic, Bifidobacterium longum bv. infantis CCUG 52486, combined with a prebiotic, gluco-oligosaccharide (B. longum + Gl-OS), on the response to seasonal influenza vaccination in young and older subjects in a double-blind, randomized controlled trial, taking into account the influence of immunosenescence markers at baseline. Results Vaccination resulted in a significant increase in total antibody titres, vaccine-specific IgA, IgM and IgG and seroprotection to all three subunits of the vaccine in both young and older subjects, and in general, the increases in young subjects were greater. There was little effect of the synbiotic, although it tended to reduce seroconversion to the Brisbane subunit of the vaccine and the vaccine-specific IgG response in older subjects. Immunological characterization revealed that older subjects randomized to the synbiotic had a significantly higher number of senescent (CD28-CD57+) helper T cells at baseline compared with those randomized to the placebo, and they also had significantly higher plasma levels of anti-CMV IgG and a greater tendency for CMV seropositivity. Moreover, higher numbers of CD28-CD57+ helper T cells were associated with failure to seroconvert to Brisbane, strongly suggesting that the subjects randomized to the synbiotic were already at a significant disadvantage in terms of likely ability to respond to the vaccine compared with those randomized to the placebo. Conclusions Ageing was associated with marked impairment of the antibody response to influenza vaccination in older subjects and the synbiotic failed to reverse this impairment. However, the older subjects randomized to the synbiotic were at a significant disadvantage due to a greater degree of immunosenscence at baseline compared with those randomized to the placebo. Thus, baseline differences in immunosenescence between the randomized groups are likely to have influenced the outcome of the intervention, highlighting the need for detailed immunological characterization of subjects prior to interventions.

Common cold is one of the most frequent human inflammatory diseases caused by viruses and can facilitate bacterial super-infections resulting in sinusitis or pneumonia. The active ingredient of the drug Soledum, 1,8-cineole, is commonly applied for treating inflammatory diseases of the respiratory tract. However, the potential of 1,8-cineole for treating primary viral infections of the respiratory tract remains unclear. In the present study, we demonstrate for the first time that 1,8-cineole potentiates Poly(I:C)-induced activity of the anti-viral transcription factor Interferon Regulatory Factor 3, while simultaneously reducing pro-inflammatory NF-κB-activity in human cell lines, inferior turbinate stem cells (ITSCs) and ex vivo cultivated human nasal mucosa. Co-treatment of cell lines with Poly(I:C) and 1,8-cineole resulted in significantly increased IRF3 reporter gene activity compared to Poly(I:C) alone, whereas NF-κB-activity was reduced. Accordingly, 1,8-cineole- and Poly(I:C)-treatment led to increased nuclear translocation of IRF3 in ITSCs and a human ex vivo model of rhinosinusitis compared to the Poly(I:C)-treated approach. Nuclear translocation of IRF3 was significantly increased in ITSCs and slice cultures treated with LPS and 1,8-cineole compared to the LPS-treated cells mimicking bacterial infection. Our findings strongly suggest that 1,8-cineole potentiates the antiviral activity of IRF3 in addition to its inhibitory effect on pro-inflammatory NF-κB-signalling and may thus broaden its field of application.

Fucans is a name used for sulfated polysaccharides, which is most characteristic structure of the presence of sulfated L-fucose, are found in brown seaweed (Phaeophyceae) and echinoderms (sea urchins and sea cucumbers). These polysaccharides have been reported to possess anticoagulant, antitumor, anti-viral, anti-proliferative and anti-inflammatory activities. Therefore, in the present study was evaluate the effect of the fucan from the brown seaweed Spatoglossum schroederii in models of peritonitis and non-septic shock induced by zymosan, as well as in a murine model of colitis induces by DSS. So, the mice treatment by intravenous route with the fucan was able to reduce the exudate formation and the cell migration in the model of acute peritonitis induced by zymosan during the kinetic of 6, 24 and 48 hours. Similarly, in the model of non-septic shock induced by zymosan the fucan demonstrated a protector effect to inhibited the cellular migration to the peritoneo, to decrease the levels of IL-6 in the serum and in the peritoneal exudate, to attenuate the lose of weight in the mice; beside to reduce the serum levels of hepatic transaminases and as well as the liver injury. In the model of murine colitis, the treatment with the fucan reduced the lose of weight of the animals, decreased the levels of IL-17 and IFN- produced in the gut and decrease the intestinal lesion induced by DSS. In conclusion, the fucan used in this study presented a significant protector effect in the murine models of inflammation

OBJETIVO: Ação do ácido trissódio-cálcio-dietileno-triaminopentaacético (CaNa3DTPA), quelante de ferro com ação ainda anti-viral, antiinflamatória e imunológica, na atenuação de lesões de reperfusão em músculos esqueléticos de ratos. MÉTODOS: 52 ratos Wistar, pesando 188±22g, foram anestesiados e submetidos a semi-amputação de membro posterior direito (MPD), poupando-se o fêmur, artéria e veia femorais. Foram então randomizados e distribuídos: G1-CTAN - controle anestesia, sem cirurgia e sem isquemia; G2-CTCIR - controle cirurgia, sem isquemia; G3-IRCT e G3-IRDTPA - com isquemia (4 hora) e reperfusão ( 2 horas). O G3-IRCT foi tratado, ao final da isquemia, com cloreto de sódio 0,9% e G3-IRDTPA com (CaNa3DTPA).Parâmetros: Circunferência do pé direito e peso do rato, dosagem sérica de CPK, dosagem de malonaldeído e microscopia óptica de músculos soleus bilateral. RESULTADOS: Aumento da circunferência nos G3-IRCT e G3-IRDTPA (significante no G3-IRCT quando comparado ao G1-CTAN); CPK elevado nos G3-IRCT e G3-IRDTPA comparados aos controles; MDA mais alto no membro contralateral do G3-IRDTPA, comparado ao MPD do G3-IRDTPA e ao G1-CTAN; maior edema intersticial em G3-IRCT, maior infiltrado inflamatório em G3-IRDTPA e recuperação dos níveis de glicogênio semelhantes em G3-IRCT e G3-IRDTPA. CONCLUSÃO: Apesar do menor edema no G3-IRDTPA comparado ao G3-IRCT, o CaNa3DTPA não alterou CPK sérico, MDA muscular e morfologia muscular dos animais.

Guazuma ulmifolia Lam., Sterculiaceae, popularly known as mutamba, has a wide geographical distribution, ranging from Mexico to Southern Brazil. The interest in its chemical properties is due to its popular use for hair loss treatment. This activity can be attributed to tannins that have many pharmacological activities such anti-inflammatory, anti-ulcerogenic, anti-microbial, anti-viral, capillary protective action and radical scavenging properties. In the microscopic analysis of barks and leaves, large secretor ducts could be detected among the parenchyma cells, as well as star shaped trichoms on the epidermis. One of the aims of the anatomical study was to detect tissues with polyphenolic compounds. The results indicated two tissues rich in these compounds: the phloematic region and the periderm. Chemical analyses were carried out on the bark and showed the presence of the following chemical groups: flavonoids, tannins, saponins and mucilages.

Caffeic acid is an ortho-phenol found in vegetable tissues presenting important properties such as carcinogenesis inhibitor, anti-oxidant, anti-viral, anti-inflammatory and anti-rheumatic actions. It was observed that caffeic acid was not degraded in daylight during the adsorption on TiO2 at pH 4.8. The adsorption fit very well to a Brunauer-Emmett-Teller isotherm equation with a monolayer coverage of 68.15 mg(CA) g(TiO2)(-1) and saturation coverage of 195.4 mg(CA) g(TiO2)(-1). A strong adsorption of caffeic acid was verified on TiO2 for the dry solid obtained from the mixture. The Raman and IR spectroscopies revealed that the adsorption should occur through the interaction of the diphenol oxygens with contribution of CC double bond of the acrylic group, however, the carboxylic acid group did not have participation in the adsorption. Friend murine leukemia Virus (FV) infection of immunocompetent mice is a well- established model to acquire further knowledge about viral immune suppression mechanisms, with the aim to develop therapeutics against retrovirus-induced diseases. Interestingly, BALB/c mice are infected by low doses of FV and die from FV-induced erythroleukemia, while C57/BL6 mice are infected by FV only at high viral dose, and remain persistently infected for their whole life. Due to the central role of dendritic cells (DC) in the induction of anti-viral responses, we asked for their functional role in the genotype-dependent sensitivity towards FV infection. In my PhD study I showed that bone marrow (BM)-derived DC differentiated from FV-infected BM cells obtained from FV-inoculated BALB/c (FV susceptible) and C57BL/6 (FV resistant) mice showed an increased endocytotic activity and lowered expression of MHCII and of costimulatory receptors as compared with non-infected control BMDC. FV-infected BMDC from either mouse strain were partially resistant towards stimulation-induced upregulation of MHCII and costimulators, and accordingly were poor T cell stimulators in vitro and in vivo. In addition, FV-infected BMDC displayed an altered expression profile of proinflammator cytokines and favoured Th2 polarization. Ongoing work is focussed on elucidating the functional role of proteins identified as differentially expressed in FV-infected DC in a genotype-dependent manner, which therefore may contribute to the differential course of FV infection in vivo in BALB/c versus C57BL/6 mice. So far, more than 300 proteins have been identified which are differently regulated in FV-infected vs. uninfected DC from both mouse strains. One of these proteins, S100A9, was strongly upregulated specifically in BMDC derived from FV-infected C57BL/6 BM cells. S100A9-/- mice were more sensitive towards inoculation with FV than corresponding wild type (WT) mice (both C57BL/6 background), which suggests a decisive role of this factor for anti-viral defense. In addition, FV-infected S100A9-/- BMDC showed lower motility than WT DC. The future work is aimed to further elucidate the functional importance of S100A9 for DC functions. Weltweit sind über 34 Millionen Menschen mit dem HI-Virus infiziert. Da die Behandlung mit der HAART-Therapie aufgrund der hohen Mutationsrate des Virus oft fehlschlägt, ist die stetige Forschung an neuen, verbesserten Wirkstoffen zwingend nötig. Mit einem neuartigen Therapieansatz, der die Aufrechterhaltung des menschlichen antiretroviralen Schutz-mechanismus durch Hemmung des APOBEC3G-Abbaus zum Ziel hat, existiert eine neue Möglichkeit zur Bekämpfung der Infektion. Im Gegensatz zu den HAART-Virustatika soll hier das menschliche Immunsystem aufrechterhalten werden, sodass es die Abwehr des Virus selbst übernehmen kann. Der durch das Virus induzierte Abbau von APOBEC3G ist gekoppelt an die Bildung eines Komplexes aus mehreren Proteinen, darunter das virale Protein vif (viral infectivity factor) und das humane Protein Elongin–C. Wird eine der Interaktionen dieser Komplexbildung gehemmt, so kann APOBEC3G nicht mehr abgebaut werden und der humane Schutzmechanismus bleibt aufrechterhalten.rnDie vorliegende Arbeit widmete sich in diesem Zusammenhang der Suche nach Inhibitoren der vif-Elongin–C-Interaktion. Nach Dockingstudien wurden potentielle Kandidaten synthetisiert und anschließend zunächst mit Hilfe der Mikrokalorimetrie (ITC) und der Oberflächenplasmonenresonanzspektroskopie (SPR) auf ihre Affinität zu rekombinant exprimiertem Elongin–C getestet. Zusätzlich wurde ein auf der Mikroskalierten Thermo-phorese (MST) basierender Bindungsassay etabliert, und die Substanzen auch mit dieser Methode getestet. Während die Bindung in diesen Assays nicht eindeutig nachgewiesen werden konnte, zeigte sich eine Substanz in In-vitro-Tests auf Hemmung der APOBEC3G- und vif-abhängigen Virusreplikation als sehr vielversprechend. Auch wenn der genaue molekulare Wirkort in weiteren Tests erst noch ermittelt werden muss, stellt diese Molekülstruktur aufgrund der bisherigen Testergebnisse bereits eine vielversprechende Basis für weitere Derivatisierungen dar.rn
