Espécies do gênero Psidium, como goiabeira e araçazeiros, são economicamente importantes e têm como área de diversidade genética primária o Brasil. Foram determinadas as relações genéticas, com base no marcador AFLP, para acessos do Banco Ativo de Germoplasma (BAG) de Psidium da Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA) Semiárido, para orientar trabalhos de melhoramento e de manejo de recursos genéticos do gênero. Foram analisados 88 acessos, sendo 64 de goiabeira e 24 de araçazeiros, coletados em dez Estados brasileiros, adotando-se para agrupamento o dendrograma UPGMA, considerando a matriz de similaridade do coeficiente de Jaccard de 149 bandas polimórficas de AFLP de 16 combinações dos iniciadores EcoRI e MseI. A análise da variância de dados moleculares foi realizada considerando a variação entre e dentro das populações de goiabeira dos dez Estados. O dendrograma apresentou boa definição, com coeficiente de correlação cofenética de 0,94. Foram observados dois grandes grupos: um formado por acessos de goiabeira e outro com acessos de araçazeiros, com inclusão de alguns acessos de goiabeira. Foram observados agrupamentos específicos no dendograma apenas para os indivíduos de goiabeira coletados em Goiás e Roraima. Os acessos estudados apresentaram similaridade variando de 28 a 98%, evidenciando a alta variabilidade genética dos mesmos. A variação entre acessos foi estimada em 0,16 (ΦST), indicando diferenciação genética moderada entre as populações de goiabeira dos dez Estados. Para aumentar a variabilidade genética do BAG estudado, sugere-se a coleta de um número maior de acessos nos Estados de Goiás e Roraima, dado o alto índice de similaridade entre os acessos provenientes destes Estados, bem como coletas amplas em outros Estados brasileiros. Sugerem-se ainda cruzamentos entre os poucos acessos de goiabeira posicionados no grupo dos araçazeiros para o desenvolvimento de híbridos interespecificos no gênero Psidium.

Eucalyptus breeding is typically conducted by selection in open-pollinated progenies. As mating is controlled only on the female side of the cross, knowledge of outcrossing versus selling rates is essential for maintaining adequate levels of genetic variability for continuous gains. Outcrossing rate in an open-pollinated breeding population of Eucalyptus urophylla was estimated by two PCR-based dominant marker technologies, RAPD and AFLP, using 11 open-pollinated progeny arrays of 24 individuals. Estimated outcrossing rates indicate predominant outcrossing and suggest maintenance of adequate genetic variability within families. The multilcous outcrossing rate (t(m)) estimated from RAPD markers (0.93 +/- 0.027), although in the same range, was higher (alpha > 0.01) than the estimate based on AFLP (0.89 +/- 0.033). Both estimates were of similar magnitude to those estimated for natural populations using isozymes. The estimated Wright's fixation index was lower than expected based on t, possibly resulting from selection against selfed seedlings when sampling plants for the study. An empirical analysis suggests that 18 is the minimum number of dominant marker loci necessary to achieve robust estimates of t,. This study demonstrates the usefulness of dominant markers, both RAPD and AFLP, for estimating the outcrossing rate in breeding and natural populations of forest trees. We anticipate an increasing use of such PCR-based technologies in mating-system studies, in view of their high throughput and universality of the reagents, particularly for species where isozyme systems have not yet been optimized.

Bark extracts of Stryphnodendron adstringens (Mart) Coville a Leguminosae species, well known in Brazil as barbatimao, are popularly used as healing agent. The objective of this work was to determine the genetic diversity of S. adstringens populations and to correlate genetic distances to the production of tannins. S. adstringens accessions from populations found in Cerrado regions in the states of Goias, Minas Gerais and São Paulo were analyzed using the AFLP (Amplified Fragment Length Polymorphism) technique. A total of 236 polymorphic bands were scored and higher proportion of genetic diversity was found inter populations (70.9%), rather than intra populations (29.1%). F-ST value was found to be significantly greater than zero (0.2906), demonstrating the complex genetic structure of S. adstringens populations. Accessions collected in Cristalina, GO, showed higher percentage of polymorphic loci (87.3%) and the highest genetic diversity. The lowest genetic variability was detected among accessions from the population growing in Caldas Novas, GO. The genetic distance among populations was estimated using the Unweighted Pair Group Method with Arithmetic Mean (UPGMA), which grouped populations into 3 clusters. Moreover, chemotypes with tannin concentration above 40% showed higher genetic similarity. AFLP analysis proved to be an efficient gene mapping technique to determine the genetic diversity among remaining populations of S. adstringens. Obtained results may be employed to implement further strategies for the conservation of this medicinal plant. (C) 2011 Elsevier Ltd. All rights reserved.

Levels of genetic variability for in situ and ex situ genetic conservation were estimated in a population of Myracrodruon urundeuva using the PCR (polymerase chain reaction) technique with the AFLP (Amplified fragment-length polymorphism) genetic marker. Seeds for progeny tests were collected from 30 open-pollination trees (matrices) at Paulo de Faria Ecological Station - SP. From this genetic material, three progeny tests were installed on the Teaching and Research Farm of Ilha Solteira Faculty of Engineering - University of São Paulo State (UNESP), which is located in Selvlria - MS, Brazil. The analysis by genetic marker was conducted with three combinations of different starters EcoRl-Msel, resulting in a total number of 137 polymorphic bands, thus forming a table of binary data. These data were used for the analysis of genetic divergence and distance between progenies. High levels of genetic divergence were observed among families. Based on the Analysis of Molecular Variance (AMOVA), it was shown that 16.2% of genetic diversity is found among progenies and 83.8% within progenies, which suggests deviances of random matings. The grouping of progenies, based on genetic distances, suggests that progenies deriving from trees which are close to each other tend to be more similar. This, in turn, indicates that the population originating the seeds may be genetically structured.

Haemophilus parasuis infection in pigs is characterized by fibrinous polyserositis, arthritis and meningitis. Despite the fact that traditional diagnosis is based on herd history, clinical signs, bacterial isolation and serotyping, molecular-based methods are alternatives for species-specific tests and epidemiological studies. The aim of this study was to characterize H. parasuis field strains from different states of Brazil, employing serotyping and genotyping methods. Serotyping revealed that serovar 4 was the most prevalent (26.1%), followed by serovars 5(17.4%), 14(8.7%), 13 (4.4%) and 2 (4.4%), whereas 39% of the strains were considered as untypeable. AFLP with a single enzyme and PFGE were able to type all isolates tested, generating 34 and 20 different profiles, respectively, including untypeable strains. Besides the slightly higher discrimination index presented by AFLP, PFGE with Not I restriction enzyme showed a better correlation with epidemiological data, grouping strains of the same serovar, animal or farm origin. The results indicated AFLP and PFGE as valuable tools for typing H. parasuis isolates collected in Brazil. (C) 2011 Elsevier Ltd. All rights reserved.

Studies involving amplified fragment length polymorphism (cDNA-AFLP) have often used polyacrylamide gels with radiolabeled primers in order to establish best primer combinations, to analyze, and to recover transcript-derived fragments. Use of automatic sequencer to establish best primer combinations is convenient, because it saves time, reduces costs and risks of contamination with radioactive material and acrylamide, and allows objective band-matching and more precise evaluation of transcript-derived fragments intensities. This study aimed at examining the gene expression of commercial cultivars of P. guajava subjected to water and mechanical injury stresses, combining analyses by automatic sequencer and fluorescent kits for polyacrylamide gel electrophoresis. Firstly, 64 combinations of EcoRI and MseI primers were tested. Ten combinations with higher number of polymorphic fragments were then selected for transcript-derived fragments recovering and cluster analysis, involving 45 saplings of P. guajava. Two groups were obtained, one composed by the control samplings, and another formed by samplings undergoing stress, with no clear distinction between stress treatments. The results revealed the convenience of using a combination of automatic sequencer and fluorescent kits for polyacrylamide gel electrophoreses to examine gene expression profiles. The Unweighted Pair Group Method with Arithmetic Mean analysis using Euclidean distances points out a similar induced response mechanism of P. guajava undergoing water stress and mechanical injury.

Im Rahmen dieser Arbeit wurde eine Routinemethode zur Differenzierung und Identifizierung von Unterlagssorten in jedem Verarbeitungsstadium, wie Holz, Pfropfrebe, bereits im Weinberg gepflanzte Rebe, entwickelt. Hierfür wurde eine Methode erarbeitet, die es ermöglicht, DNA aus Blättern, Holz und Wurzeln gleichermaßen zu extrahieren. Vermischungen von Unterlagssorten in einem Unterlagenholzbündel konnten bis zu 10% Fremd-Unterlagenholz durch eine RAPD-PCR nachgewiesen werden. Mit den 12mer Primer #722b und #722c wurden sortenspezifische Banden für die Unterlagssorten Börner, 8B, 3309C und 5BB festgestellt. Der Primers # 751 war in der Lage von 151 Unterlagssorten und Wildarten 144 Genotypen zu unterschieden. Mit Hilfe der Optimierung von RAMP-Zeiten konnten die Bandenmuster der sieben in Deutschland am häufigsten verwendeten Unterlagssorten auf zwei unterschiedlichen Thermocyclern reproduziert werden. Aufgrund der Optimierung der RAPD-PCR war es möglich, die zur Unterscheidung notwendigen Banden durch eine lineare Transformation anhand einer ermittelten Referenzbande mathematisch und graphisch darzustellen. Klone der Unterlagssorten SO4, 125AA und 5C, sowie die Unterlagssorte Binova, wurden auf die Unterscheidungsmöglichkeit hin mit RAPD, AFLP und SAMPL untersucht. Innerhalb der AFLP-/SAMPL-Methode bildeten die zu einer Sorte gehörenden Unterlagenklone ein Cluster, wobei Binova innerhalb der SO4 Klone zu finden war. Es wurden ‚unterlagssortenspezifische Banden’, ‚wiederholende Banden’ und ‚Einzelbanden’ gefunden.
