O objetivo deste trabalho foi realizar monitoramento químico de seis substratos utilizados na produção de mudas de cacaueiros. Os substratos receberam 10 volumes (100mL, cada) de água destilada, e no lixiviado coletado foram determinados o pH, a condutividade eltrica (CE) e os teores de sódio (Na) e potássio (K) em solução. Os valores de pH variaram de 5,0 a 8,0 em Plantmax® e composto do tegumento da amêndoa do cacau (CTAC), respectivamente. Verificou-se que, a partir do primeiro volume lixiviado, a CE de todos os substratos foi reduzida. Foram encontradas correlações positivas para a CE em relação ao K e Na da fibra de coco (FC) (r=0,83 e 0,72 P<0,05) e CTAC (r=0,98 e 0,99 P<0,05). Os resultados indicam que a FC e o CTAC deveriam ser usados após pré-lavagens. Verificou-se que, embora a FC apresente na primeira lixiviação valor de CE superior aos demais substratos, o valor ideal da 2,0 dSm-1 é alcançado na segunda lixiviação. Os teores de K e Na são um indicativo do valor da CE em FC e CTAC.

O objetivo do trabalho foi verificar o efeito do ácido indolbutírico (AIB) no enraizamento de estacas do cacaueiro. O delineamento experimental foi o inteiramente casualizado, em esquema fatorial 3 x 5 x 2, envolvendo 3 clones (Cepec 2008, CCN 51 e TSH 1188), 5 concentrações de ácido indolbutírico AIB (0; 1.000; 3.000; 6.000 e 9.000 mg kg-1), duas épocas do ano (verão e inverno),, cinco repetições e 10 estacas por parcela. A avaliação do experimento foi realizada 120 dias após o plantio e analisadas as seguintes variáveis: percentagem de sobrevivência (SOB), número de brotações (NB), matéria seca das brotações (MSB), percentagem de estacas enraizadas (ENR), número de raízes (NR) e matéria seca de raízes (MSR). Os fatores época do ano e concentração de AIB apresentaram efeito significativo para as variáveis estudadas quando isolados e em interação com os clones. Os dados obtidos possibilitaram concluir que o valor médio da concentração ideal de AIB (CI) foi de 4.169 mg kg-1 e 3.985 mg kg-1 no verão e inverno, respectivamente. Verificou-se que os clones apresentam diferentes respostas em relação à CI de AIB e época do ano. de modo geral, as CIs no verão foram maiores que no inverno. Os resultados mostraram que a época de plantio e a concentração de AIB influenciaram de maneira mais expressiva na sobrevivência e enraizamento das estacas dos clones Cepec 2008 e CCN 51 e em menor intensidade no clone TSH 1188. Existe uma concentração ideal de AIB para cada um dos clones estudados.

Resilience research has been applied to socioeconomic as well as for agroecological studies in the last 20 years. It provides a conceptual and methodological approach for a better understanding of interrelations between the performance of ecological and social systems. In the research area Alto Beni, Bolivia, the production of cocoa (Theobroma cacao L.), is one of the main sources of income. Farmers in the region have formed producers’ associations to enhance organic cocoa cultivation and obtain fair prices since the 1980s. In cooperation with the long-term system comparisons by the Research Institute of Organic Agriculture (FiBL) in Alto Beni, aspects of the field trial are applied for the use in on-farm research: a comparison of soil fertility, biomass and crop diversity is combined with qualitative interviews and participatory observation methods. Fieldwork is carried out together with Bolivian students through the Swiss KFPE-programme Echanges Universitaires. For the system comparisons, four different land-use types were classified according to their ecological complexity during a preliminary study in 2009: successional agroforestry systems, simple agroforestry systems (both organically managed and certified), traditional systems and conventional monocultures. The study focuses on interrelations between different ways of cocoa cultivation, livelihoods and the related socio-cultural rationales behind them. In particular this second aspect is innovative as it allows to broaden the biophysical perspective to a more comprehensive evaluation with socio-ecological aspects thereby increasing the relevance of the agronomic field studies for development policy and practice. Moreover, such a socio-ecological baseline allows to assess the potential of organic agriculture regarding resilience-building face to socio-environmental stress factors. Among others, the results of the pre-study illustrate local farmers’ perceptions of climate change and the consequences for the different crop-systems: all interviewees mentioned rising temperatures and/or an extended dry season as negative impacts more with regard to their own working conditions than to their crops. This was the case in particular for conventional monocultures and in plots where slash-and-burn cultivation was practised whereas for organic agroforestry systems the advantage of working in the shade was stressed indicating that their relevance rises in the context of climate change.

The improvement of tropical tree crops using conventional breeding methods faces challenges due to the length of time involved. Thus, like most crops, there is an effort to utilize molecular genetic markers in breeding programs to select for desirable agronomic traits. Known as marker assisted breeding or marker assisted selection, genetic markers associated with a phenotype of interest are used to screen and select material reducing the time necessary to evaluate candidates. As the focus of this research was improving disease resistance in tropical trees, the usefulness of the WRKY gene superfamily was investigated as candidates for generating useful molecular genetic markers. WRKY genes encode plant-specific transcriptional factors associated with regulating plants' responses to both biotic and abiotic stress. ^ One pair of degenerate primers amplified 48 WRKY gene fragments from three taxonomically distinct, economically important, tropical tree crop species: 18 from Theobroma cacao L., 21 from Cocos nucifera L. and 9 from Persea americana Mill. Several loci from each species were polymorphic because of single nucleotide substitutions present within a putative non-coding region of the loci. Capillary array electrophoresis-single strand conformational polymorphism (CAE-SSCP) mapped four WRKY loci onto a genetic linkage map of a T. cacao F2 population segregating for resistance to witches' broom disease. Additionally, PCR primers specific for four T. cacao loci successfully amplified WRKY loci from 15 members of the Byttneriae tribe. A method was devised to allow the reliable discrimination of alleles by CAE-SSCP using only the mobility assigned to the sample peaks. Once this method was validated, the diversity of three WRKY loci was evaluated in a germplasm collection of T. cacao . One locus displayed high diversity in the collection, with at least 18 alleles detected from mobility differences of the product peaks. The number of WRKY loci available within the genome, ease of isolation by degenerate PCR, codominant segregation demonstrated in the F2 population, and usefulness for screening germplasm collections and closely related wild species demonstrates that the WRKY superfamily of genes are excellent candidates for developing a number of genetic molecular markers for breeding purposes in tropical trees. ^

The rehabilitation of the old cocoa ( Theobroma cacao L. ) farms is one of the major challenges for a sustainable cocoa production. A study was carried out to set up a guide which could be used as a decision making tool for a quick and efficient diagnosis of the old cocoa orchards and to choose the appropriate regeneration option (rehabilitation or replanting). A sample of 90 rehabilitated cocoa farms and of 75 replanted cocoa farms was surveyed in 12 regions representing the three main cocoa producing sectors in the country. Data were collected on the key agronomic characteristics of these cacao farms. These were cocoa variety, farm size, age, yield, planting density, number of shade trees and the level of damages caused by insects and diseases. The results showed that age, planting density and yield were the discriminating criteria of these farms. The average values of these criteria were 25 to 30 years for the age, 800 to 1 000 trees ha-1 for the planting density and 250 to 400 kg ha-1 an-1 for the yield. Based on these criteria and their average values, a decision making guide was designed for the diagnosis of cocoa farms and the choice of regeneration option. According to this guide, old cocoa farms (more than 25 years), degraded and unproductive should be replanted. However, younger farms having planting density and yield higher than the average values above should be rehabilitated.

Cocoa ( Theobroma cacao L. ) is an important allogamous tropical tree crop, whose centre of diversity is considered to be in Central America. Dry cocoa beans from five cocoa clones, and their intercrossed hybrids were analysed based on the variation of alkaloids and polyphenolic compounds contents, in order to gain insights on the heterosis and broad-sense heritability. Polyphenols and alkaloids were analysed at 280 nm by HPLC, using a Photodiode Array Detector (PDA); while anthocyanins were separated with the SEP-PAK Vac 6cc 1000 mg (waters) column and measured at 520 nm with a PDA. Dry cocoa beans displayed high content of purine alkaloids (2.1 and 8.8 mg g-1 for caffein and theobromine, respectively), and polyphenols (25 and 2978 µg g-1 for catechin and epicatechin, respectively). Among the five cocoa clones, SNK16 was the highest in purine alkaloid (caffein and theobromin) and flavanol (catechin and epicatechin); while T79/467 possessed the greatest quantity of cyanidin-3-galactoside and cyanidin-3-arabinoside. From all the parameters studied, anthocyanins (Cyanidin-3-galactoside and cyanidin-3-arabinoside) exhibited the highest level of heterosis. Parental genotypes SNK16 and T79/467 showed good aptitudes for the combination of characters because their reciprocal hybrids F5 and F9, distinguished themselves by better levels of mid-parent heterosis values. Besides, the heritability value in strict sense of this Cyanidin-3-galactoside was very high. Absence of significant difference between genotypes, coming from reciprocal crossbreeding for Cyanidin-3-galactoside, suggests that this character in cocoa would be nuclear contrary to purine alkaloids and flavan-3-ols, where their transmission to offsprings can be stated as cytoplasmic.

Vários trabalhos vem sendo desenvolvidos sobre o cultivo in vitro de cacau (T.cacao), mas são raros para a maioria das outras espécies do gênero, como o cupuaçu (T. grandiflorum), cuja a área plantada vem aumentando expressivamente, e outras que poderiam servir de fonte de genes para as espécies economicamente já reconhecidas. Protocolos para obtenção de embriões somáticos in vitro para as espécies T. cacao,T. grandiflorum,T. speciosum e o híbrido T. grandiflorum x T. obovatum foram avaliados a partir de duas fontes de explantes, estaminódios e pétalas (formadas por lgulas e cógulas) cultivados em meio de crescimento primário de calo, consistindo de sais DKW, suplementado com 20 g l<sup>-1 de sacarose, 250 mg l<sup>-1de glutamina, 200 mg l<sup>-1 de mio-inositol, 0,2 mg l<sup>-1 de tiamina-HCl, 0,1 mg l<sup>-1 de ácido nicotínico, 0,2 mg l<sup>-1 de glicina, 2 mg l<sup>-1 de 2,4-D, 2,2 g l<sup>-1 de Gelrite® e pH 5,8. A este meio foram adicionadas diferentes concentrações de tidiazuron (0, 5 e 10 µg l<sup>-1). As culturas foram mantidas no escuro por 14 dias, à temperatura de 25 ± 2 ºC, e então transferidas para meio de crescimento secundário de calo, constituído de sais WPM, vitaminas de Gamborg, 20 g l<sup>-1 de sacarose, 2 mg l<sup>-1 de 2,4 D, 0,3 mg l<sup>-1 de cinetina, 50 ml l<sup>-1 de água de côco, 2,2 g l<sup>-1 de Gelrite® e pH 5,8. A formação de calos ocorreu em todas as espécies. Embriões somáticos foram obtidos somente para T. cacao. A calogênese mostrou-se influenciada pelo genótipo e foi maior nos estaminódios.

Novel species of fungi described in the present study include the following from Australia: Neoseptorioides eucalypti gen. & sp. nov. from Eucalyptus radiata leaves, Phytophthora gondwanensis from soil, Diaporthe tulliensis from rotted stem ends of Theobroma cacao fruit, Diaporthe vawdreyi from fruit rot of Psidium guajava, Magnaporthiopsis agrostidis from rotted roots of Agrostis stolonifera and Semifissispora natalis from Eucalyptus leaf litter. Furthermore, Neopestalotiopsis egyptiaca is described from Mangifera indica leaves (Egypt), Roussoella mexicana from Coffea arabica leaves (Mexico), Calonectria monticola from soil (Thailand), Hygrocybe jackmanii from littoral sand dunes (Canada), Lindgomyces madisonensis from submerged decorticated wood (USA), Neofabraea brasiliensis from Malus domestica (Brazil), Geastrum diosiae from litter (Argentina), Ganoderma wiiroense on angiosperms (Ghana), Arthrinium gutiae from the gut of a grasshopper (India), Pyrenochaeta telephoni from the screen of a mobile phone (India) and Xenoleptographium phialoconidium gen. & sp. nov. on exposed xylem tissues of Gmelina arborea (Indonesia). Several novelties are introduced from Spain, namely Psathyrella complutensis on loamy soil, Chlorophyllum lusitanicum on nitrified grasslands (incl. Chlorophyllum arizonicum comb. nov.), Aspergillus citocrescens from cave sediment and Lotinia verna gen. & sp. nov. from muddy soil. Novel foliicolous taxa from South Africa include Phyllosticta carissicola from Carissa macrocarpa, Pseudopyricularia hagahagae from Cyperaceae and Zeloasperisporium searsiae from Searsia chirindensis. Furthermore, Neophaeococcomyces is introduced as a novel genus, with two new combinations, N. aloes and N. catenatus. Several foliicolous novelties are recorded from La Réunion, France, namely Ochroconis pandanicola from Pandanus utilis, Neosulcatispora agaves gen. & sp. nov. from Agave vera-cruz, Pilidium eucalyptorum from Eucalyptus robusta, Strelitziana syzygii from Syzygium jambos (incl. Strelitzianaceae fam. nov.) and Pseudobeltrania ocoteae from Ocotea obtusata (Beltraniaceae emend.). Morphological and culture characteristics along with ITS DNA barcodes are provided for all taxa.

En los últimos años, el mercado de cacao fino o de aroma ha tenido un crecimiento constante en respuesta a la demanda de chocolates de alto contenido de cacao, origen conocido, sabores distintivos y producidos responsablemente, presionando a las industrias de chocolate premium a proveerse de cacaos diferenciados y de alta calidad. Esto exige a las cadenas agroalimentarias ajustar sus estrategias de producción y mercadeo con el fin de poder aprovechar las oportunidades (demanda creciente del producto, aranceles preferenciales para este país, entre otras) que ofrece el mercado. Sin embargo, Ecuador a pesar de ser el principal productor y exportador de cacao fino o de aroma del mundo (exporta alrededor del 96,3 por ciento de su producción), encuentra dificultades para mejorar la inserción de su producto en el mercado internacional y aprovechar estas oportunidades. Por lo tanto, el siguiente trabajo tiene como objetivo realizar un diagnóstico del Sistema Agroindustrial (SAG) del cacao fino o de aroma del Ecuador, con la finalidad de identificar las oportunidades y las limitaciones en el ambiente institucional, organizacional, tecnolgico y comercial, y de este modo, generar información que contribuya a la mejora de la inserción del producto en el mercado mundial. El trabajo se desarrolló partir de un enfoque fenomenolgico utilizando como herramienta metodolgica el método EPESA. Los resultados del estudio se interpretaron y discutieron a partir de la base conceptual de la Nueva Economía Institucional y los distritos agroindustriales y agrocomerciales. Los resultados obtenidos indicarían que las restricciones institucionales estarían dadas por la ausencia de una ley que proteja los derechos de propiedad a los productores de cacao fino o de aroma. En el ambiente comercial el sistema de formación de precios no está alineado con el tipo de producto que se comercializa (especialidad), generándose alta incertidumbre especialmente para los pequeños y medianos productores que explican el 88 por ciento de los actores de la producción. De este modo aparecen comportamientos oportunistas y se generan cuasi rentas apropiables por el eslabón siguiente de la cadena. Esto desincentiva la inversión en tecnología y la innovación dando por resultado una meseta en la producción y restringiendo la mejora en la inserción del producto en el mercado mundial.

A partir del siglo XIX con el desarrollo de la industria del chocolate en Europa, la producción de cacao se incrementó aceleradamente en Brasil y Ecuador, y más tarde a través de los colonizadores, se promovió el cultivo en África, llegando primero a Ghana y luego a Nigeria, Camerún y Costa de Marfil. El mercado mundial de cacao en grano distingue dos amplias categorías: el cacao fino y de aroma y el cacao común u ordinario. Alrededor del 95 por ciento de la producción mundial es cacao común, el cual procede en su mayoría de África, Asia, América Central y del Sur. El restante de la producción, es decir el 5 por ciento, corresponde a cacao fino y de aroma, el mismo que se produce en Ecuador, Indonesia, Papúa Nueva Guinea, Colombia, Venezuela, Trinidad y Tobago, entre otros (ICCO, 2009), y cuyas características distintivas de aroma y sabor son buscadas principalmente por los fabricantes de chocolates finos, por lo que reciben un plus o premio al precio base en los mercados internacionales. La demanda de cacao fino y de aroma es muy limitada y selectiva, ya que se usa en la elaboración de chocolates de calidad premium o gourmet, los mismos que para ser considerados finos deben elaborarse con más del 75 por ciento de esta variedad. En otros casos, el cacao fino y de aroma se combina con el cacao común para reducir el costo de la materia prima, en la elaboración de chocolates de calidad superior a la estándar como chocolates oscuros, tabletas y coberturas. Sin embargo, en los últimos años, el mercado de estos tipos de chocolate ha crecido en respuesta a los altos estándares de vida en los países consumidores, como son Europa y Estados Unidos. Actualmente, en el ámbito mundial, el grado de exigencia de los consumidores respecto de los alimentos se ha elevado y diversificado, en virtud del aumento de su poder de negociación, de la cantidad de información disponible, y de la oferta de una gran variedad de productos. Los consumidores buscan productos auténticos, genuinos, de identificación cultural, avalados por una tradición del saber hacer, es decir, alimentos que tengan un cuento que contar. De aquí la importancia de la Denominación de Origen como sello de certificación. En el Ecuador existe un tipo de cacao fino y de aroma único en el mundo, conocido en el país con el nombre de "Cacao Arriba" que posee características de sabor y aroma muy distintivas y valoradas a nivel mundial, como notas florales, frutales y a nuez. Este reconocimiento le permite ser competitivo en calidad, mas no en productividad, ya que los países africanos son los mayores productores a nivel mundial, y se han especializado en rendimiento y no en atributos.

DNA- and RNA-based polymerase chain reaction (PCR) systems were used with Cacao swollen shoot virus (CSSV) primers designed from conserved regions of the six published genomic sequences of CSSV to investigate whether the virus is transmissible from infected trees through cross-pollination to seeds and seedlings. Pollen was harvested from CSSV infected cocoa trees and used to cross-pollinate flowers of healthy cocoa trees (recipient parents) to generate enough cocoa seeds for the PCR screening. Adequate precautions were taken to avoid cross-contamination during duplicated DNA extractions and only PCR results accompanied by effective positive and negative controls were scored. Results from the PCR analyses showed that samples of cocoa pod husk, mesocarp and seed tissues (testa, cotyledon and embryo) from the cross-pollinations were PCR negative for CSSV DNA. Sequential DNA samples from new leaves of seedlings resulting from the cross-pollinated trees were consistently PCR negative for presence of portions of CSSV DNA for over 36 months after germination. A reverse transcription-PCR analysis performed on the seedlings showed negative results, indicating absence of functional CSSV RNA transcripts in the seedlings. None of the seedlings exhibited symptoms characteristic of the CSSV disease, and all infectivity tests on the seedlings were also negative. Following these results, the study concluded that although CSSV DNA was detected in pollen from CSSV infected trees, there was no evidence of pollen transmission of the virus through cross-pollination from infected cocoa parents to healthy cocoa trees. We present a palaeoecological investigation of pre-Columbian land use in the savannah “forest island” landscape of north-east Bolivian Amazonia. A 5700 year sediment core from La Luna Lake, located adjacent to the La Luna forest island site, was analysed for fossil pollen and charcoal. We aimed to determine the palaeoenvironmental context of pre-Columbian occupation on the site and assess the environmental impact of land use in the forest island region. Evidence for anthropogenic burning and Zea mays L. cultivation began ~2000 cal a BP, at a time when the island was covered by savannah, under drier-than-present climatic conditions. After ~1240 cal a BP burning declined and afforestation occurred. We show that construction of the ring ditch, which encircles the island, did not involve substantial deforestation. Previous estimates of pre-Columbian population size in this region, based upon labour required for forest clearance, should therefore be reconsidered. Despite the high density of economically useful plants, such as Theobroma cacao, in the modern forest, no direct pollen evidence for agroforestry was found. However, human occupation is shown to pre-date and span forest expansion on this site, suggesting that here, and in the wider forest island region, there is no truly pre-anthropogenic ‘pristine’ forest.
