We have here demonstrated for the first time that nitrate not only accelerates testa rupture of non- AR seeds but also modifies expression pattern of the cell-wall remodeling proteins (mannanases; SoMAN6 and SoMAN7) and key genes belonging to metabolism and signaling of ABA (SoNCED6, SoNCED9, SoCYP707A2 and SoABI5) and GAs (SoGA3ox, SoGA20ox, SoGA2ox and SoRGL2). These results were obtained during Sisymbrium officinale seed imbibition in the absence of endosperm rupture. Exogenous ABA induced a notable inhibition of testa rupture in both absence and presence of nitrate being this effect sharply reversed by GA4+7. However, nitrate was capable to provoke testa rupture in absence of ABA synthesis. The expression of SoMAN6 and SoMAN7 were positively altered by nitrate. Although ABA synthesis seems apparent at the start of non-AR seed imbibition, taken together the results of SoNCED6, SoNCED9 and SoCYP707A2 expression seem to suggest that nitrate leads to a strong net ABA decrease. Likewise, nitrate positively affected the SoABI5 expression when the SoNCED9 expression was also stimulated. By contrast, at the early and final of imbibition, nitrate clearly inhibited the SoABI5 expression. The expression of SoGA2ox6 and SoGA3ox2 are strongly inhibited by nitrate whereas of SoGA20ox6 was stimulated. On the other hand, SoRGL2 transcript level decreased in the presence of nitrate. Taken together, the results presented here suggest that the nitrate signaling is already operative during the non-AR S. officinale seeds imbibition. The nitrate, in cross-talk with the AR network likely increases the favorable molecular conditions that trigger germination.

Ginger (Zingiber officinale, Roscoe), a monocotyledonous, sterile cultigen, is widely used as a spice, flavoring agent, and herbal medicine. The pungency of fresh ginger is due to a series of homologous phenolic ketones of which [6]-gingerol is the major one. The gingerols are thermally unstable and can be converted to their corresponding shogaols, which are present in dried ginger, Fresh rhizomes of 17 clones of Australian ginger, including commercial cultivars and experimental tetraploid clones, were assayed by HPLC for gingerols and shogaols. [6]-Gingerol was identified as the major pungent phenolic compound in all samples, while [8]- and [10]-gingerol occurred in lower concentrations. One cultivar known as Jamaican contained the highest concentrations of all three gingerols and was the most pungent of the clones analyzed. Gingerols were stable in ethanolic solution over a 5-month period when stored at 4 degrees C. Shogaols were not identified in the extracts prepared from fresh rhizomes at ambient temperature, confirming that these compounds are not native constituents of fresh ginger, In contrast to previous findings, this study did not find significant differences in gingerol concentrations between the tetraploid clones and their parent diploid cultivar.

Ginger oil, obtained by steam distillation of the rhizome of Zingiber officinale Roscoe, is used in the beverage and fragrance industries. Ginger oil displays considerable compositional diversity, but is typically characterized by a high content of sesquiterpene hydrocarbons, including zingiberene, arcurcumene, beta-bisabolene, and beta-sesquiphellandrene. Australian ginger oil has a reputation for possessing a particular lemony aroma, due to its high content of the isomers neral and geranial, often collectively referred to as citral. Fresh rhizomes of 17 clones of Australian ginger, including commercial cultivars and experimental tetraploid clones, were steam distilled 7 weeks post-harvest, and the resulting oils were analyzed by GC-MS. The essential oils of 16 of the 17 clones, including the tetraploid clones and their parent cultivar, were found to be of substantially similar composition. These oils were characterized by very high citral levels (51-71%) and relatively low levels of the sesquiterpene hydrocarbons typical of ginger oil. The citral levels of most of these oils exceeded those previously reported for ginger oils. The neral-to-geranial ratio was shown to be remarkably constant (0.61 +/- 0.01) across all 17 clones. One clone, the cultivar Jamaican, yielded oil with a substantially different composition, lower citral content and higher levels of sesquiterpene hydrocarbons. Because this cultivar also contains significantly higher concentrations of pungent gingerols, it possesses unique aroma and flavor characteristics, which should be of commercial interest.

Background Cell-to-cell communication (quorum sensing (QS)) co-ordinates bacterial behaviour at a population level. Consequently the behaviour of a natural multi-species community is likely to depend at least in part on co-existing QS and quorum quenching (QQ) activities. Here we sought to discover novelN-acylhomoserine lactone (AHL)-dependent QS and QQ strains by investigating a bacterial community associated with the rhizosphere of ginger (Zingiber officinale) growing in the Malaysian rainforest. Results By using a basal growth medium containing N-(3-oxohexanoyl)homoserine lactone (3-oxo-C6-HSL) as the sole source of carbon and nitrogen, the ginger rhizosphere associated bacteria were enriched for strains with AHL-degrading capabilities. Three isolates belonging to the generaAcinetobacter (GG2), Burkholderia (GG4) and Klebsiella (Se14) were identified and selected for further study. Strains GG2 and Se14 exhibited the broadest spectrum of AHL-degrading activities via lactonolysis while GG4 reduced 3-oxo-AHLs to the corresponding 3-hydroxy compounds. In GG2 and GG4, QQ was found to co-exist with AHL-dependent QS and GG2 was shown to inactivate both self-generated and exogenously supplied AHLs. GG2, GG4 and Se14 were each able to attenuate virulence factor production in both human and plant pathogens. Conclusions Collectively our data show that ginger rhizosphere bacteria which make and degrade a wide range of AHLs are likely to play a collective role in determining the QS-dependent phenotype of a polymicrobial community.

In Australia, Pythium soft rot (PSR) outbreaks caused by P. myriotylum were reported in 2009 and since then this disease has remained as a major concern for the ginger industry. From 2012 to 2015, a number of Pythium spp. were isolated from ginger rhizomes and soil from farms affected by PSR disease and assessed for their pathogenicity on ginger. In this study, 11 distinct Pythium spp. were recovered from ginger farms in Queensland, Australia and species identification and confirmation were based on morphology, growth rate and ITS sequences. These Pythium spp. when tested showed different levels of aggressiveness on excised ginger rhizome. P. aphanidemartum, P. deliense, P. myriotylum, P. splendens, P. spinosum and P. ultimum were the most pathogenic when assessed in vitro on an array of plant species. However, P. myriotylum was the only pathogen, which was capable of inducing PSR symptoms on ginger at a temperature range from 20 to 35 °C. Whereas, P. aphanidermatum only attacked and induced PSR on ginger at 30 to 35 °C in pot trials. This is the first report of P. aphanidermatum inducing PSR of ginger in Australia at high temperatures. Only P. oligandrum and P. perplexum, which had been recovered only from soils and not plant tissue, appeared non-pathogenic in all assays.

Nos últimos anos, tem-se verificado aumento no uso da adubação verde de inverno na cultura da videira. Essa adubação proporciona ao vinhedo melhor controle da erosão e manutenção da fertilidade, aumento da macro e microporosidade do solo pela penetração e decomposição das raízes e aumento do teor de matéria orgânica, além da possibilidade de diminuição do uso de herbicidas. Algumas espécies vêm sendo utilizadas, mesmo sem se conhecerem os efeitos secundários para a videira, e poucos trabalhos têm sido realizados para avaliação desses efeitos. O objetivo deste trabalho foi verificar o efeito alelopático de diferentes espécies de plantas de cobertura verde de inverno sobre o desenvolvimento do porta-enxerto VR 043-43. O delineamento utilizado foi inteiramente casualizado, com 19 tratamentos e quatro repetições. Foram preparados extratos das partes aérea e radicular de nove espécies, utilizadas como cobertura verde de inverno. As estacas enraizadas foram colocadas em vasos com areia e, durante 45 dias, duas vezes por semana, foram regadas com solução aquosa de cada extrato e fertilizante. Após esse período, foi avaliado o comprimento das raízes, o número de raízes por estaca, a matéria fresca do sistema radicular e a matéria fresca das brotações. Os resultados obtidos permitiram concluir que, dentre os extratos das espécies estudadas, o que apresentou maior efeito alelopático foi o extrato da parte aérea da gorga (Spergula arvensis), seguido pelo extrato da parte aérea do dente-de-leão (Taraxacum officinale L.). O centeio (Secale cereale L.) e o nabo forrageiro (Raphanus sativus L.) não apresentaram efeito alelopático.

Hortaliças in natura, comercializadas na região metropolitana de São Paulo, SP (Brasil), foram analisadas através de metodologia própria, visando à pesquisa e à identificação de formas de transmissão de helmintos intestinais de interesse médico. As hortaliças examinadas, constituídas de 50 amostras de cada variedade, foram de: alface (Lactuca sativa), variedades lisa e crespa, escarola (Chichorium sp) e agrião (Nasturtium officinale). Os resultados evidenciaram elevados percentuais de contaminação em todas as variedades analisadas, porém, as freqüências de helmintos foram maiores no agrião. A escarola apresentou valores médios, geralmente situados entre as alfaces e o agrião. Os números médios de ovos e larvas por 100 gramas de amostra, embora elevados, não apresentaram diferenças estatisticamente significantes entre as quatro variedades de hortaliças estudadas. Uma grande variedade de helmintos, de ocorrência freqüente na população residente na Região Metropolitana de São Paulo, foi observada nas amostras. Os mais freqüentes, no entanto, foram: ancilostomídeos e Ascaris sp. Recuperaram-se também ovos de Toxocara sp, Fasciola sp e de tricostrongilídeos, comprovando a ocorrência de contaminação das hortaliças por fezes de animais domésticos. Considerando-se os resultados obtidos, ressalta-se a importância das hortaliças na transmissão de helmintíases intestinais, bem como a necessidade de medidas que propiciem uma melhoria na qualidade higiênico-sanitária destes alimentos.

Foram analisadas hortaliças "in natura", comercializadas na Região Metropolitana de São Paulo, SP-Brasil, visando à pesquisa e à identificação de cistos de protozoários de interesse médico. As hortaliças, constituídas de 50 amostras de cada variedade, consistiram em: alface (Lactuca sativa), variedades lisa e crespa, escarola (Chichorium sp) e agrião (Nasturtium officinale). Os resultados evidenciaram elevados percentuais de contaminação em todas as variedades de hortaliças analisadas, porém as freqüências de protozoários foram maiores no agrião. As amostras de alfaces apresentaram os menores percentuais de contaminação, enquanto que a escarola apresentou valores geralmente situados entre o agrião e as alfaces. Observou-se uma grande variedade de protozoários, cujas freqüências de ocorrência na população residente na Região Metropolitana de São Paulo são igualmente elevadas. Os mais freqüentes foram Entamoeba sp (com 4 e 8 núcleos) e Giardia sp. Foram também isolados cistos de Iodamoeba sp, Endolimax sp e Chilomastix sp. Os elevados níveis de contaminação fecal encontrados nas amostras analisadas apontam para a importância dos alimentos na transmissão de protozoários intestinais.

O objetivo deste estudo foi avaliar a contaminação por enteroparasitas em hortaliças consumidas cruas comercializadas nas cidades de Niterói e Rio de Janeiro. Foram estudadas 128 amostras de hortaliças3/4alface (Lactuca sativa) e agrião (Nasturtium officinale)3/4provenientes do comércio (supermercados, feiras-livre e quitandas) e de restaurantes tipo self-services. Apenas 6,2% das amostras apresentaram presença de estruturas parasitárias com morfologia semelhante as de espécies parasitas de animais. Foi encontrado presença de contaminantes como ácaros, ovos de ácaros, insetos, larvas de nematóides e protozoários ciliados em quase todas as amostras (96,1%), inclusive nas de restaurantes. Este alto percentual sugere a presença de risco de infecção, pois associado a esses agentes poderiam existir estruturas parasitárias infectantes para o homem.
