Genetic markers that distinguish fungal genotypes are important tools for genetic analysis of heterokaryosis and parasexual recombination in fungi. Random amplified polymorphic DNA (RAPD) markers that distinguish two races of biotype B of Colletotrichum gloeosporioides infecting the legume Stylosanthes guianensis were sought. Eighty-five arbitrary oligonucleotide primers were used to generate 895 RAPD bands but only two bands were found to be specifically amplified from DNA of the race 3 isolate. These two RAPD bands were used as DNA probes and hybridised only to DNA of the race 3 isolate. Both RAPD bands hybridised to a dispensable 1.2 Mb chromosome of the race 3 isolate. No other genotype-specific chromosomes or DNA sequences were identified in either the race 2 or race 3 isolates. The RAPD markers hybridised to a 2 Mb chromosome in all races of the genetically distinct biotype A pathogen which infects other species of Stylosanthes as well as S. guianensis. The experiments indicate that RAPD analysis is a potentially useful tool for obtaining genotype-and chromosome-specific DNA probes in closely related isolates of one biotype of this fungal pathogen.

Two biotypes (A and B) of Colletotrichum gloeosporioides infect the tropical legumes Stylosanthes spp. in Australia. These biotypes are asexual and vegetatively incompatible. However, field isolates of biotype B carrying a supernumerary 2-Mb chromosome, thought to originate from biotype A, have been reported previously. We tested the hypothesis that the 2-Mb chromosome could be transferred from biotype A to biotype B under laboratory conditions. Selectable marker genes conferring resistance to hygromycin and phleomycin were introduced into isolates of biotypes A and B, respectively. A transformant of biotype A, with the hygromycin resistance gene integrated on the 2-Mb chromosome, was cocultivated with phleomycin-resistant transformants of biotype B. Double antibiotic-resistant colonies were obtained from conidia of these mixed cultures at a frequency of approximately 10(-7). Molecular analysis using RFLPs, RAPDs, and electrophoretic karyotypes showed that these colonies contained the 2-Mb chromosome in a biotype B genetic background. In contrast, no double antibiotic colonies developed from conidia obtained from mixed cultures of phleomycin-resistant transformants of biotype B with biotype A transformants carrying the hygromycin resistance gene integrated in chromosomes >2 Mb in size. The results demonstrated that the 2-Mb chromosome was selectively transferred from biotype A to biotype B. The horizontal transfer of specific chromosomes across vegetative incompatibility barriers may explain the origin of supernumerary chromosomes in fungi.

Large numbers of adults of certain species of butterfly flying in an apparently 'purposeful' manner are often noted by entomologists and the general public. Occasionally, these are recorded in the literature. Using these records we summarise information regarding the direction of movement in Australian butterflies and test whether there are consistent patterns that could account for known seasonal shifts in geographical range. The data were analysed using contingency tables and directionality statistics. Vanessa itea, Vanessa kershawi, Danaus plexippus, Danaus chrysippus and Badamia exclamationis flew predominately south in the spring-summer and north in the autumn-winter. Tirumala hamata has a strong southern component to its flight in spring but, as in Euploea core, appears non-directional in the autumn. For many supposedly known migratory species, the number of literature records are few, particularly in one season (mainly autumn). Thus, for Appias paulina, four of seven records were south in the spring-summer, as were six of nine records for Catopsilia pomona, and three of five for Zizina labradus. For Belenois java, flight records were only available for the spring and these showed geographical differences; predominantly north-west in northern Australia (Queensland) and south-west in southern Australia (Victoria, New South Wales). There were too few records for Papilio demoleus in the literature (four only) to draw any conclusions. Major exceptions to the seasonal trend of south in the spring and north in the autumn were Junonia villida, which showed a predominant north-westward direction in both seasons, and Eurema smilax, with a predominant southern or western flight in both seasons. We discuss these species specific trends in migration direction in relation to seasonal shifts in suitable habitat conditions, possible cues used in orientation and in timing changes in direction.

Previous research has indicated that biotypes A and B of Colletotrichum gloeosporioides that infect Stylosanthes spp. in Australia are asexual and vegetatively incompatible. Selectable marker genes conferring resistance either to hygromycin or phleomycin were introduced into isolates of these biotypes. Vectors conferring resistance to hygromycin and carrying telomeric sequences from Fusarium oxysporum replicated autonomously in C. gloeosporioides and gave frequencies of transformation 100-times higher than vectors that integrated into the genome. Monoconidial colonies resistant to both antibiotics were recovered when hygromycin-resistant biotype-A transformants carrying an autonomously replicating vector were paired in culture with a phleomycin-resistant biotype-B transformant carrying integrative vector sequences. Molecular analysis of double antibiotic-resistant progeny indicated that they contained the autonomous vector in a biotype-B genetic background, Results indicate that transfer of the autonomous vector had occurred from biotype A to biotype B, demonstrating the potential for transfer of genetic information between these biotypes.

23 isolates from root nodules of 13 leguminous species were tested in laboratory for cultural and physiological characteristics. The following conclusions can be drawn: 1 - The cultural and physiological characters were not enough to identify the different isolates. 2 - The data from cultural and physiological tests help the identification of the bacteria, when checked with data from cross-inoculation tests reported elsewhere (GALLI, 1958). The bacteria studied herein may be grouped as the following: a) nodule bacteria from Vigna sinensis, Calopogonium muconoides, Centrosema pubescens, Desmodium adscendens, D. discolor, Indigofera mucronata, I. subulata, I. sumatrana, Stylosanthes guyanensisA Tephrosia cândida and Teramnus uncinatus: bacteria with scanty growth; alcaline reaction in milk-brom cresol purple; no acid production in medium with any of the carbohydrates used. b) nodule bacteria from pea and from Vicia obscura: bacteria with moderate growth; alcaline reaction, with serum zone, in milk-brom cresol purple; acid production in medium with all the carbohydrates used. c) nodule bacteria from Leucaena glauca: bacteria with moderate growth; alcaline reaction, with serum zone, in milk-brom cresol purple; acid production from rafinose and manitol. d) nodule bacteria from Cratylia floribunda: bacteria with scanty growth; alcaline reaction in milk-brom cresol purple; acid production from mannose and ramnose.

Quatro leguminosas forrageiras tropicais (Galactia striata (Jacq) Urb. , Glycine wightii Verde. cv. Tinaroo, Macvoptilium atropurpureus cv. Siratro e Stylosanthes guianensis (Avbl) Swartz cv. IRI 1022) foram cultivadas em solução nutritiva recebendo doses de alumínio de 0,5 , 10 e 20 ppm, com o objetivo de se verificarem os efeitos das doses crescentes do elemento sobre as díferentes espécies. Submetendo-se os dados de produção de matéria seca e de concentrações de macro e micronutrientes à análise estatística, observou-se o seguinte: a) O estilosantes e a galactia mostraram-se mais tolerantes. A galactia apresentou a máxima produção em presença de 5 ppm de Alumínio, não sendo afetada pelas concentrações mais elevadas. O estilosantes apresentou a máxima produção em presença de 20 ppm de Alumínio. b) A soja e o siratro mostram-se sensíveis, notando-se uma queda na produção de matéria seca em presença de 20 ppm de Alumínio na solução. A soja mostrou sintomas mais graves de toxidez, apresentando seu sistema radicular totalmente danificado na concentração mais alta de Alumínio na solução. c) A adição de Alumínio provocou uma diminuição nas concentrações de fósforo, cálcio e magnésio nas raízes ou partes aéreas das plantas e de manganês nas raízes e uma elevação na concentração de Alumínio nas raízes.

IRI 1022) foram cultivadas em solução nutritiva recebendo doses de manganês de 0, 5, 10 e 20 ppm, com o objetivo de se verificarem os efeitos das doses crescentes do elemento sobre as diferentes espécies. Submetendo-se os dados de produção de matéria seca e de concentrações de macro e micronutrientes à análise estatística, observou-se o seguinte: a) O estilosantes foi o que se mostrou mais tolerante, não apresentando variação significativa na produção de matéria seca com o aumento dos teores de manganês. Não se observaram sintomas severos nas folhas ou nas raízes. A galactia também mostrou-se tolerante, apresentando queda significativa de produção em presença de 100 ppm de manganês e sentomas mais severos de toxidez que os apresentados pelo estilosantes. b) O Siratro e a soja mostraram-se sensíveis ao excesso de manganês. A soja mostrou a mínima produção em presença de 25 ppm do elemento não havendo modificação significativa nas concentrações mais elevadas. O siratro apresentou a mínima produção de matéria seca total em presença de 50 ppm de manganês. Os sintomas de toxidez apresentados pela soja foram mais severos que os apresentados pelas plantas de siratro. c) A adição de manganês provocou uma diminuição nas concentrações de cálcio, potássio e magnésio nas partes aéreas ou raízes das plantas estudadas; elevação dos teores de zinco e cobre nas aéreas ou raízes e de manganês na planta toda.
