Any image processing object detection algorithm somehow tries to integrate the object light (Recognition Step) and applies statistical criteria to distinguish objects of interest from other objects or from pure background (Decision Step). There are various possibilities how these two basic steps can be realized, as can be seen in the different proposed detection methods in the literature. An ideal detection algorithm should provide high recognition sensitiv ity with high decision accuracy and require a reasonable computation effort . In reality, a gain in sensitivity is usually only possible with a loss in decision accuracy and with a higher computational effort. So, automatic detection of faint streaks is still a challenge. This paper presents a detection algorithm using spatial filters simulating the geometrical form of possible streaks on a CCD image. This is realized by image convolution. The goal of this method is to generate a more or less perfect match between a streak and a filter by varying the length and orientation of the filters. The convolution answers are accepted or rejected according to an overall threshold given by the ackground statistics. This approach yields as a first result a huge amount of accepted answers due to filters partially covering streaks or remaining stars. To avoid this, a set of additional acceptance criteria has been included in the detection method. All criteria parameters are justified by background and streak statistics and they affect the detection sensitivity only marginally. Tests on images containing simulated streaks and on real images containing satellite streaks show a very promising sensitivity, reliability and running speed for this detection method. Since all method parameters are based on statistics, the true alarm, as well as the false alarm probability, are well controllable. Moreover, the proposed method does not pose any extraordinary demands on the computer hardware and on the image acquisition process.

The orphan nuclear receptor steroidogenic factor 1 (SF-1) is expressed in the adrenal cortex and gonads and regulates the expression of several P450 steroid hydroxylases in vitro. We examined the role of SF-1 in the adrenal glands and gonads in vivo by a targeted disruption of the mouse SF-1 gene. All SF-1-deficient mice died shortly after delivery. Their adrenal glands and gonads were absent, and persistent Mullerian structures were found in all genotypic males. While serum levels of corticosterone in SF-1-deficient mice were diminished, levels of adrenocorticotropic hormone (ACTH) were elevated, consistent with intact pituitary corticotrophs. Intrauterine survival of SF-1-deficient mice appeared normal, and they had normal serum level of corticosterone and ACTH, probably reflecting transplacental passage of maternal steroids. We tested whether SF-1 is required for P450 side-chain-cleavage enzyme (P450scc) expression in the placenta, which expresses both SF-1 and P450scc, and found that in contrast to its strong activation of the P450scc gene promoter in vitro, the absence of SF-1 had no effect on P450scc mRNA levels in vivo. Although the region targeted by our disruption is shared by SF-1 and by embryonal long terminal repeat-binding protein (ELP), a hypothesized alternatively spliced product, we believe that the observed phenotype reflects absent SF-1 alone, as PCR analysis failed to detect ELP transcripts in any mouse tissue, and sequences corresponding to ELP are not conserved across species. These results confirm that SF-1 is an important regulator of adrenal and gonadal development, but its regulation of steroid hydroxylase expression in vivo remains to be established.

Since the conservative Justice and Development Party (AKP) took power in Turkey in 2002, it has enjoyed a constant winning streak: it won each election (with a support level of 49.83% in 2011), subordinated the army (which had de facto stood above the civilian government) and was reforming the country. The situation in the country was stable (especially when compared to the crises and restlessness in the 1990s), the economy was booming, Turkey’s position in regional politics was strengthening, and Ankara’s significance on the international arena was growing. This encouraged the ruling class to make long-term plans, leading up to the hundredth anniversary of the republic in 2023. In the coming decade, Turkey governed by the AKP was to become one of the global economic and political centres, a full member of the EU and at the same time a political and economic leader in the Middle East. However, the negative trends in the situation both domestically (mass public protests, the deadlocked Kurdish issue and the unsuccessful attempt to amend the constitution) and abroad (the war in Syria and the coup in Egypt) seen over the past few months have laid bare the limitations of the AKP’s rule and have affected the government’s democratic mandate, prestige and credibility on the international arena, as well as peace and order and domestic security. When compared to the beginning of 2013, the way the situation will develop in Turkey is at this moment definitely less predictable; and the possible scenarios include both relative peace (however, with socio-political tension present in the background) and the threat of destabilisation. Therefore, although the AKP will still remain the sole major political force, this party will have to face challenges which will decide not only its political future but also the directions the country will be developing in. However, a comprehensive solution of the accumulated problems and a simple return to the status quo ante, convenient to the government, seem unlikely in the foreseeable future.

A critical gene involved in mammalian sex determination and differentiation is the Sty-related gene Sox9. In reptiles, Sox9 resembles that of mammals in both structure and expression pattern in the developing gonad, but a causal role in male sex determination has not been established. A closely related gene, Sox8, is conserved in human, mouse, and trout and is expressed in developing testes and not developing ovaries in mouse. In this study, we tested the possibility of Sox8 being important for sex determination or sex differentiation in the red-eared slider turtle Trachemys scripta, in which sex is determined by egg incubation temperature between stages 15 and 20. We cloned partial turtle Sox8 and anti-Mullerian hormone (Amh) cDNAs, and analyzed the expression patterns of these genes in developing gonads by reverse transcriptase-polymerase chain reaction and whole-mount in situ hybridization. While Amh is expressed more strongly in males than in females at stage 17, Sox8 is expressed at similar levels in males and females throughout the sex-determining period. These observations suggest that differential transcription of Sill is not responsible for regulation of Amh, nor responsible for sex determination in turtle. (C) 2004 Wiley-Liss, Inc.

The promoter regions of plant pararetroviruses direct transcription of the full-length viral genome into a pregenomic RNA that is an intermediate in the replication of the virus. It serves as template for reverse transcription and as polycistronic mRNA for translation to viral proteins. We have identified functional promoter elements in the intergenic region of the Cavendish isolate of Banana streak virus (BSV-Cav), a member of the genus Badnavirus. Potential binding sites for plant transcription factors were found both upstream and downstream of the transcription start site by homology search in the PLACE database of plant cis-acting elements. The functionality of these putative cis-acting elements was tested by constructing loss-of-function and regain-of-function mutant promoters whose activity was quantified in embryogenic sugarcane suspension cells. Four regions that are important for activity of the BSV-Cav promoter were identified: the region containing an as-l-like element, the region around-141 and down to -77, containing several putative transcription factor binding sites, the region including the CAAT-box, and the leader region. The results could help explain the high BSV-Cav promoter activity that was observed previously in transgenic sugarcane plants and give more insight into the plant cell-mediated replication of the viral genome in banana streak disease. The phenomenon of B6-Y-DOM sex reversal arises when certain variants of the Mus domesticus Y chromosome are crossed onto the genetic background of the C57BL/6J (136) inbred mouse strain, which normally carries a Mus musculus-derived Y chromosome. While the sex reversal has been assumed to involve strain-specific variations in structure or expression of Sry, the actual cause has not been identified. Here we used in situ hybridization to study expression of Sry, and the critical downstream gene Sox9, in strains containing different chromosome combinations to investigate the cause of B6-Y-DOM sex reversal. Our findings establish that a delay of expression of Sry(DOM) relative to Sry(B6) underlies B6-Y-DOM sex reversal and provide the first molecular confirmation that Sry must act during a critical time window to appropriately activate Sox9 and effect male testis determination before the onset of the ovarian-determining pathway. During mammalian sexual development, the SOX9 transcription factor up-regulates expression of the gene encoding anti-Mullerian hormone (AMH), but in chickens, Sox9 gene expression reportedly occurs after the onset of Amh expression. Here, we examined expression of the related gene Sox8 in chicken embryonic gonads during the sex-determining period. We found that cSox8 is expressed at similar levels in both sexes at embryonic day 6 and 7, and only at the anterior tip of the gonad, suggesting that SOX8 is not responsible for the sex-specific increase in cAmh gene expression at these stages. We also found that several other chicken Sox genes (cSox3, cSox4 and cSox11) are expressed in embryonic gonads, but at similar levels in both sexes. Our data suggest that the molecular mechanisms involved in the regulation of Amh genes of mouse and chicken are not conserved, despite similar patterns of Amh expression in both species.

The Blue Streak, named for the Sandusky, Ohio athletic teams, opened in 1964. It was the first new wooden roller coaster at the park since the Cyclone in 1929. A classic out-and-back coaster with a fast and hilly ride, the 78-foot-high Blue Streak is a long time guest favorite. It celebrated its 50th anniversary in 2014.

A cultura da banana tem baixa diversidade genética, tornando a espécie susceptível a doenças dizimadoras como a Sigatoka negra. No entanto, a adoção de novas variedades necessita de avaliações agronômicas e físico-químicas. Neste estudo, as variedades de banana, resistentes à Sigatoka negra, foram caracterizadas e comparadas com a variedade tradicional (Grand Naine). Cada variedade foi avaliada considerando-se critérios relevantes para a agroindústria, como pH, sólidos solúveis totais, acidez total titulável, relação SST/ATT, açúcares totais, açúcares redutores e não redutores, umidade, sólidos totais e rendimento no processamento. A variedade Thap Maeo apresentou-se como a variedade mais potencial para substituição da Gran Naine na indústria, com altos teores de sólidos solúveis totais, açúcares redutores, açúcares totais e umidade. As variedades Caipira e FHIA 2 também podem substituir a Grand Naine. Na análise de agrupamentos, verificou-se que a variedade Grand Naine esteve muito próxima das variedades do subgrupo Gros Michel (Bucaneiro, Ambroisa e Calipso) e também da variedade Caipira, apresentando no seu genoma o grupo AAA. Conclui-se que há opções de variedades resistentes para substituição da variedade tradicional, nas regiões afetadas pela Sigatoka-negra.

In 2004, Costa-Santos and cols. reported 24 patients from 19 Brazilian families with 17α-hydroxylase deficiency and showed that p.W406R and p.R362C corresponded to 50% and 32% of CYP17A1 mutant alleles, respectively. The present report describes clinical and molecular data of six patients from three inbred Brazilian families with 17α-hydroxlyse deficiency. All patients had hypogonadism, amenorrhea and hypertension at diagnosis. Two sisters were found to be 46,XY with both gonads palpable in the inguinal region. All patients presented hypergonadotrophic hypogonadism, with high levels of ACTH (> 104 ng/mL), suppressed plasmatic renin activity, low levels of potassium (< 2.8 mEq/L) and elevated progesterone levels (> 4.4 ng/mL). Three of them, including two sisters, were homozygous for p.W406R mutation and the other three (two sisters and one cousin) were homozygous for p.R362C. The finding of p.W406R and p.R362C in the CYP17A1 gene here reported in additional families, confirms them as the most frequent mutations causing complete combined 17α-hydroxylase/17,20-lyase deficiency in Brazilian patients.

FISH has been used as a complement to classical cytogenetics in the detection of mosaicism in sex chromosome anomalies. The aim of this study is to describe three cases in which the final diagnosis could only be achieved by FISH. Case 1 was an 8-year-old 46,XY girl with normal female genitalia referred to our service because of short stature. FISH analysis of lymphocytes with probes for the X and Y centromeres identified a 45,X/46,X,idic(Y) constitution, and established the diagnosis of Turner syndrome. Case 2 was a 21-month-old 46,XY boy with genital ambiguity (penile hypospadias, right testis, and left streak gonad). FISH analysis of lymphocytes and buccal smear identified a 45,X/46,XY karyotype, leading to diagnosis of mixed gonadal dysgenesis. Case 3 was a 47,XYY 19-year-old boy with delayed neuromotor development, learning disabilities, psychological problems, tall stature, small testes, elevated gonadotropins, and azoospermia. FISH analysis of lymphocytes and buccal smear identified a 47,XYY/48,XXYY constitution. Cases 1 and 2 illustrate the phenotypic variability of the 45,X/46,XY mosaicism, and the importance of detection of the 45,X cell line for proper management and follow-up. In case 3, abnormal gonadal function could be explained by the 48,XXYY cell line. The use of FISH in clinical practice is particularly relevant when classical cytogenetic analysis yields normal or uncertain results in patients with features of sex chromosome aneuploidy. Deficiency of the enzyme P450 oxidoreductase is a rare form of congenital adrenal hyperplasia with characteristics of combined and partial impairments in steroidogenic enzyme activities, as P450 oxidoreductase transfers electrons to CYP21A2, CYP17A1, and CYP19A1. It results in disorders of sex development and skeletal malformations similar to Antley-Bixley syndrome. We report the case of a 9-year-old girl who was born with virilized genitalia (Prader stage V), absence of palpable gonads, 46,XX karyotype, and hypergonadotropic hypogonadism. During the first year of life, ovarian cyst, partial adrenal insufficiency, and osteoarticular changes, such as mild craniosynostosis, carpal and tarsal synostosis, and limited forearm pronosupination were observed. Her mother presented severe virilization during pregnancy. The molecular analysis of P450 oxidoreductase gene revealed compound heterozygosis for the nonsense p.Arg223*, and the novel missense p.Met408Lys, inherited from the father and the mother, respectively. O objetivo deste trabalho foi caracterizar histologicamente o desenvolvimento ovariano pré-natal em fetos eqüinos sem raça definida (SRD) nos trópicos, ressaltando surgimento e quantificação de ovogônias e folículos. Foram obtidos 131 fetos em frigorífico, os quais foram mensurados quanto ao comprimento (cm) equivalente à distância cefalococcígea (CR), seguido de corte das gônadas e fixação das amostras em Bouin por 24 horas. As idades foram estimadas por regressão. Foram preparados cortes histológicos ovarianos de 7µm de espessura, os quais foram corados com hematoxilina-eosina. O surgimento de ovogônias, folículos primordiais, em crescimento e antrais foi verificado, respectivamente, em fetos com CR entre 4,1 e 6,5cm (40 a 49 dias de gestação (DG)); 9,1 e 12,0cm (60 a 69 DG); 23,1 e 29,0cm (116 a 135 DG) e 50,1 e 65,0cm (201 a 245 DG). A quantidade máxima de ovogônias, folículos primordiais, em crescimento e antrais foi observada em fetos com CR entre, 4,1 e 6,0cm (40 a 49 DG); 16,1 e 23,0cm (90 a 115 DG); 29,1 e 36,0cm (136 a 155 DG) e 50,1 e 97,0cm (201 a 320 DG), respectivamente. Os resultados sugerem que o desenvolvimento morfológico dos ovários fetais de eqüinos SRD nos trópicos é mais precoce que o observado nos relatos de literatura referentes a estudos de eqüinos de diversas raças em regiões temperadas.
