Avaliou-se o efeito da centrifugação sobre a viabilidade do sêmen canino e compararam-se três meios de diluição pré-centrifugação. Utilizaram-se 10 ejaculados completos de 10 cães que, após a avaliação inicial, foram divididos em quatro porções (grupos). Uma das amostras, não centrifugada, formou o grupo-controle; as outras foram diluídas em três diferentes meios e centrifugadas a 800 x g por 15 minutos, formando os grupos: CPSA - constituído por sêmen centrifugado em plasma seminal autólogo; CLG - sêmen centrifugado em meio à base de leite desnatado e glicose (LG); e CPer- sêmen centrifugado em gradientes de Percoll (45% e 90%). Após a centrifugação e a eliminação do sobrenadante, procedeu-se à ressuspensão de todas as porções do ejaculado em LG e à imediata avaliação quanto à motilidade, vigor, aglutinação espermática e integridade das membranas espermáticas. Todas as suspensões foram, então, incubadas a 37ºC por 30 minutos e reavaliadas. O processo de centrifugação não causou danos aos espermatozoides e a centrifugação em meio LG melhorou a viabilidade espermática.

Para estudar o efeito da idade sobre características ovulatórias e de fertilidade, 147 ciclos estrais de 99 éguas foram agrupados em classes por idade (1 - 3-6 anos; 2 - 7-10 anos; 3 - 11-14 anos; e 4 - 15-19 anos), de acordo com o ano do nascimento. As éguas foram inseminadas com sêmen diluído, resfriado e transportado de apenas um garanhão, três vezes por semana (segundas, quartas e sextas-feiras).O controle folicular, por meio da palpação retal, e a rufiação foram realizados diariamente, durante todo o período experimental. Utilizou-se, para o transporte, o sêmen diluído no diluidor de leite desnatado-glicose no conteiner Celle modificado, sendo a dose inseminante de 400 x 10(6) espermatozóides móveis, no momento da diluição final, pré-resfriamento. O tempo médio da coleta do sêmen à inseminação artificial foi de 3,5 horas e a temperatura final do sêmen, no momento da inseminação, de 14ºC. Não houve influência da idade sobre a velocidade de crescimento folicular e o tamanho do folículo ovulatório. A fertilidade decaiu após os 15 anos de idade, traduzida pela diminuição da taxa de concepção/ciclo e eficiência de prenhez, entretanto, não foram observadas características indicativas de senescência até os 19 anos de idade.

Estudou-se o efeito do número de inseminações sobre a fertilidade de éguas inseminadas, três vezes/semana (segundas, quartas e sextas-feiras), com sêmen diluído, resfriado e transportado, de apenas um garanhão. As éguas foram inseminadas a partir da detecção, pela palpação retal, de um folículo de 3,0 a 3,5 cm de diâmetro, em um dos ovários, até a ovulação. Utilizaram-se para o transporte o sêmen diluído no diluidor leite desnatado-glicose e o conteiner Celle modificado, sendo a dose inseminante de 400 x 10(6) espermatozóides móveis, no momento da diluição final, pré-resfriamento. de acordo com o número de inseminações artificiais (IA) utilizadas/ciclo, os resultados de 148 ciclos, de 100 éguas, foram agrupados em: 1 IA, 2IA, 3 IA e 4 ou mais IA. A eficiência de prenhez foi de 4,29; 5,04; 5,67; e 3,43, para 1, 2, 3 e 4 ou mais IA, respectivamente. As características foliculares diferiram em relação à freqüência de inseminações com os maiores valores observados no grupo de éguas inseminadas quatro ou mais vezes. Concluiu-se que menor velocidade de crescimento folicular e maior diâmetro do folículo ovulatório estiveram associados a maior número de inseminações artificiais/ciclo. Na presença de uma concentração espermática adequada, o número de inseminações/ciclo não exerceu influência sobre a fertilidade.

Curtobacterium wilt has become an important disease of beans in several localities in the country. Its causal agent, Curtobacterium flaccumfacciens pv. flaccumfaciens (Cff), survives and is disseminated through seeds. To date, few studies have been conducted with the objective of developing an effective and low-cost culture medium to isolate this bacterium from bean seeds, for health analysis purposes. Usually, the culture media employed for coryneform bacteria isolation contain specific carbon sources and antimicrobial products not available in the Brazilian market. A culture medium known as MSCFF was developed (peptone - 5 g, meat extract - 3 g, sucrose - 5 g, agar 15 g, skim milk powder* - 5 g. Congo red* - 0.05 g-, chlorothalonil* - 0.01 g, thiophanate methyl* - 0.01 g, nalidixic acid* - 0.01 g, nitrofurantoin* - 0.01 g. oxacillin* 0.001 g, sodium azide* - 0.001 g and distilled water q.s. 1L; *added after autoclaving the basal medium), which has the ability to inhibit growth of a large amount of saprophytic bacteria, but with low supressivity to Cff isolates. The MSCFF medium was highly effective for Cff isolation from naturally infected bean seeds and could be used for routine detection of this bacterium in bean seeds.

One hundred forty seven cycles of mares were allocated in a completely randomized experiment, with different number of replications and divided in four treatments (T1 = 24 hours preovulation; T2 = 48 hours preovulation; T3 = 48 hours preovulation and in tbe same day of ovulation; T4 = 72 hours preovulation and in the same day of ovulation), in order to study the effect of AI/ovulation interval on mare fertility. The mares were inseminated three times for wek (monday, wednesday and friday), with semen of only one stallion diluted in extender skim milk-glucose, using a volume of 15ml, with 400 x 10(6) sptz viable, cooled at 14 degrees C/3.6 hours, and transported in modified container Celle. Conception rates were not different according to the treatments. So, observed spermatic survival for 60 hours showed the practicability of the inseminations on monday, wednesday and friday.

The effect of milk processing on rheological and textural properties of probiotic low-fat yogurt (fermented by two different starter cultures) was studied. Skim milk fortified with skim milk powder was subjected to three treatments: (1) thermal treatment at 85C for 30 min; (2) high hydrostatic pressure (HHP) at 676 MPa for 5 min; and (3) combined treatments of HHP (676 MPa for 5 min) and heat (85C for 30 min). The processed milk was fermented using two different starter cultures containing Streptococcus thermophilus, Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus acidophilus and Bifidobacterium longum at inoculation rates of 0.1 and 0.2%. Rheological parameters were determined and a texture profile analysis was carried out. Yogurts presented different rheological behaviors according to the treatment used, which could be attributed to structural phenomena. The combined HHP and heat treatment of milks resulted in yogurt gels with higher consistency index values than gels obtained from thermally treated milk. The type of starter culture and inoculation rate, providing different fermentation pathways, also affected the consistency index and textural properties significantly. The combined HHP and heat treatment of milks before fermentation, and an inoculation rate of 0.1% (for both cultures), led to desirable rheological and textural properties in yogurt, which presented a creamy and thick consistency that does not require the addition of stabilizers.

The effect of milk processing on the microstructure of probiotic low-fat yogurt was studied. Skim milk fortified with skim milk powder was subjected to three treatments prior to innoculation: thermal treatment at 85 degrees C for 30 min, high hydrostatic pressure at 676 MPa for 5 min, and combined treatments of high hydrostatic pressure (HHP) and heat. The processed milk was then fermented by using two different starter cultures containing Streptococcus thermophilus, Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus acidophilus, and Bifidobacterium longum. The microstructure of heat-treated milk yogurt had fewer interconnected chains of irregularly shaped casein micelles, forming a network that enclosed the void spaces. on the other hand, microstructure of HHP yogurt had more interconnected clusters of densely aggregated protein of reduced particle size, with an appearance more spherical in shape, exhibiting a smoother more regular surface and presenting more uniform size distribution. The combined HHP and heat milk treatments led to compact yogurt gels with increasingly larger casein micelle clusters interspaced by void spaces, and exhibited a high degree of cross-linking. The rounded micelles tended to fuse and form small irregular aggregates in association with clumps of dense amorphous material, which resulted in improved gel texture and viscosity. Diferentes fontes proteicas em rações de leitões sobre atividade da tripsina e parâmetros sangüíneos

Were used 64 piglets submitted eight treatments: ration with skim milk (SM), three rations with crescent levels of swine plasma (SP), three rations with whole egg (WE) and a ration with high inclusion of soybean meal (SB). Were monitored the blood parameters (BP) in pigs at 27 and at 34 days of age. The piglets were slaugther at 28 and at 35 days of age, for collections pancreas and posterior mensurements of absolut (AW) and relative weigth (RW) of pancreas and trypsin activity (TA). Treatments not influencied AW and TA. Significant effect of the crescent levels was verified of SP, with lineal reduction of the leukocytes and increase of the globular volume, to the 27 days; while to the 34 days, lineal increase of the hematias was observed. At 27 days, animals feds rations with crescent levels of SP have inferior percentage of eosinophils than others that consumed crescent levels of WE. The utilization of SP promoted smaller stimulus to the immune reply, while the use of WE promoted larger humoral reply of the piglets.

Were used 64 weaned pigs, distributed in eight treatments: ration with skim milk (SM), three rations with crescent levels of swine plasma (SP), three rations with whole egg (WE) and a ration with high inclusion of soybean meal (SB). Daily weight gain (DWG), daily feed intake (DFI) and feed conversion (FC) were evaluated. The animals that consumed ration SB have the mean of DWG higher that other treatments, and inferior FC, from 35 to 48 days of age. The crescent levels of SP promoted linear reduction in DWG, from 21 to 35, and from 35 to 48 days of age, in DFI from 48 to 138 days of age and linear increased in FC from 21 to 35 of age. Considering the weaned phase until finishing, initial rations can be formulated with SP and WE substituted partially the CP of skim milk in 25 and 45%, respectively.

This study evaluated whether pentoxifylline (PTX) present in the flushing extender influenced the function of equine epididymal spermatozoa after recovery and after thawing. For this experiment, 58 testicles from 29 Brazilian Jumping Horses were used. Cauda epididymides of each stallion were separated and flushed with a skim milk extender, with or without 7.18 mM PTX and then subjected to the freezing process. Samples flushed with the extender containing PTX showed a significant increase in total motility, progressive motility, straight line velocity, curvilinear velocity, and percentage of rapid sperm immediately after the recovery of epididymal sperm and after 15 minutes of incubation at 37°C (P < .05). However, the presence of PTX in the flushing extender did not affect the post-thaw motility parameters or plasma membrane integrity (P > .05). The results of this study showed that the PTX present in the flushing extender improved motility parameters of recently recovered epididymal sperm and had no deleterious effects on plasma membrane integrity and freezability of equine epididymal sperm. © 2013 Elsevier Inc.
