The characterization of a coffee gene encoding a protein similar to miraculin-like proteins, which are members of the plant Kunitz serine trypsin inhibitor (STI) family of proteinase inhibitors (PIs), is described. PIs are important proteins in plant defence against insects and in the regulation of proteolysis during plant development. This gene has high identity with the Richadella dulcifica taste-modifying protein miraculin and with the tomato protein LeMir; and was named as CoMir (Coffea miraculin). Structural protein modelling indicated that CoMir had structural similarities with the Kunitz STI proteins, but suggested specific folding structures. CoMir was up-regulated after coffee leaf miner (Leucoptera coffella) oviposition in resistant plants of a progeny derived from crosses between C. racemosa (resistant) and C. arabica (susceptible). Interestingly, this gene was down-regulated during coffee leaf miner herbivory in susceptible plants. CoMir expression was up-regulated after abscisic acid application and wounding stress and was prominent during the early stages of flower and fruit development. In situ hybridization revealed that CoMir transcripts accumulated in the anther tissues that display programmed cell death (tapetum, endothecium and stomium) and in the metaxylem vessels of the petals, stigma and leaves. In addition, the recombinant protein CoMir shows inhibitory activity against trypsin. According to the present results CoMir may act in proteolytic regulation during coffee development and in the defence against L. coffeella. The similarity of CoMir with other Kunitz STI proteins and the role of CoMir in plant development and plant stress are discussed.

This study aimed to examine the sensory characteristics of the grains of 21 cultivars of Coffea arabica L. and Coffea canephora Pierre from the essays of genetic improvement of EPAMIG, located in Patrocinio Municipality, Minas Gerais State, where they were collected through cloths stripping method and washed. Subsequently to dry (11 to 12% moisture b.u.), we obtained the coffee designated as natural. The evaluated varieties were: Acaia Cerrado MG 1474; Bourbon Vermelho DATERRA; Catigua MG 1; Catigua MG 2; Catual Amarelo IAC 62; Catuai Vermelho IAC 15; H 419-3-1-4-2; H 419-6-2 -5-2; H 419-6-2-5-3; H 419-6-2-7-3 Vermelho; H 493-1-2-10; H 514-7-10-1 Vermelho; H 514-7-10-6; H 515-4-2-2; H 518-3-6-1; Icatu Amarelo IAC 3282; Mundo Novo 379-19; Mundo Novo TAO 376-4; Rubi MG 1192; Sacramento MG 1 and Topazio MG 1190, from 2005/2006 and 2006/2007 seasons. The cultivars according to the first principal component with notes above 80 points, regarded as superior drink according to attributes with the highest scores (flavor, sweetness, balance, acidity, clean drink, and aspect) were: Catigua MG2, Rubi MG 1192, 514-7-10-6 H, H 419-3-1-4-2, H 419-6-2-5-2, 493-1-2-10 H, H 514-7-10-1 Vermelho, Catigua MG1, Sacramento MG1, 419-6-2-5-3 H, H 515-9-2-2 and Catuai Amarelo IAC 62.

Fruit anatomy of Neotropical species of Indigofera (Leguminosae, Papilionoideae) with functional and taxonomic implications. J. Torrey Bot. Soc. 136: 203-211. 2009-This work reports on the fruit surface and anatomy of seven Neotropical species of Indigofera (I. campestris Bong. ex Benth., I. hirsuta L., I. lespedeziodes Kunth, I. microcarpa Desv., I. spicata Forssk., I. suffruticosa Mill., and I. truxillensis Kunth) to help species diagnosis and clarify the fruit type classification. Flowers and fruits at several stages of development were removed from living material, fixed, and examined with scanning electron (surface analyses) and light microscopies (histological analyses). Species showed differences in relation to the number of exocarp layers, secretory trichome morphology and distribution, presence of stomata, phenolic idioblast size and distribution in mesocarp, the number and arrangement of endocarp fibers, and the presence of it separation tissue. It is noteworthy that no separation tissue was observed in L microcarpa and I. suffruticosa, although they have dehiscent fruits, which indicates it delayed dehiscence. The present work confirms that fruit anatomical characters can be utilized as it tool for fruit type classification, especially in Indigofera, the third largest genus of Leguminosae.

Rooted in a history that dates back 1500 years, the concentration of economic activity and infrastructure in Colombo represents an extreme case of urban primacy within the national context of Sri Lanka. Located on the West Coast of the country, the Colombo metropolitan area accommodates a quarter of the country's 18.6 million population, and is the economic and political core of the country. However, Colombo is a city of extremes. Its modem and well-serviced core stands in stark contrast to the circumstances of more than half of its population, who live in poorly serviced shack and shanty settlements. The proportion of the population living in these areas continues to expand, notwithstanding a history of innovative and participatory approaches to development planning and management. Complicating these development challenges, the potential of the city is undermined by a civil war that has been under way since the early 1980s, taking an immense toll in loss of life, political polarization, and economic opportunity costs. This paper traces Colombo's historical development, provides a description of contemporary characteristics and challenges faced by the city, and examines the evolution of plans and programmes designed to improve the conditions of low-income settlements in the city. During the second half of 1986 the impact of the improvement of water supply and excreta disposal facilities on diarrheal diseases and intestinal parasitosis was studied in 254 children up to six years of age from two favelas (shanty towns) of Belo Horizonte, Brazil. The estimated incidence of diarrhea was 6.2 episodes/child year and the estimated period prevalence reached 31.0 episode days/ child/ year. The point prevalence of parasitosis was 70.7% (Ascaris lumbricoides: 55.4%, Trichuris trichiura: 19.6%, Giardia lamblia: 17.9%). The estimated prevalence of diarrhea decreased with improvement of water supply and sanitation facilities to 45% and 44% respectively, but no statistically significant impact was observed in the case of parasitosis. School education and weaning practice were found to be other important determinants of diarrhea.

To test the hypothesis that the low socioeconomic population living is shanty towns in Porto Alegre presents different levels of poverty which are reflected on its health status, a cross-sectional study was designed involving 477 families living in Vila Grande Cruzeiro, Porto Alegre, Brazil. The poverty level of the families was measured by using an instrument specifically designed for poor urban populations. Children from families living in extreme poverty (poorest quartile) were found to have higher infant mortality rate, lower birth weights, more hospitalizations, and higher malnutrition rates, in addition to belonging to more numerous families. Thus, the shanty town population of Porto Alegre is not homogeneous, and priority should be given to the more vulnerable subgroups.

Os desafios à engenharia moderna são cada vez maiores, pretendendo-se quase sempre obter estruturas mais leves, com propriedades mecânicas atrativas e muitas vezes com geometrias complexas. Com tais requisitos, um dos materiais que tem vindo a ter uma crescente aplicação é o material compósito. Contudo, no que toca ao cálculo estrutural destes materiais, tudo se torna mais complexo, já que são materiais que geralmente são formados por empilhamento de várias camadas de material heterogéneo, podendo estas encontrarem-se dispostas segundo diferentes orientações. Assim, a utilização de um software que permita a previsão das propriedades mecânicas de uma estrutura em material compósito através da micromecânica, a aplicação da Teoria Clássica dos Laminados e de um critério de rotura, como por exemplo o de Tsai-Hill, é fundamental para agilizar o processo de estudo da estrutura a fabricar. Para dar uma resposta a tal necessidade foi desenvolvida uma aplicação, em MATLAB® GUI, denominada CAFE – Composite Analysis For Engineers, com ambiente gráfico apelativo, que permite determinar todas as variáveis importantes no estudo de estruturas em material compósito. Esta aplicação visa suportar e agilizar a aprendizagem desta área do conhecimento, permitindo também o acesso ao código de cálculo por parte do utilizador, de modo a conhecerem-se as equações utilizadas e, eventualmente, ser alvo de futuros desenvolvimentos. O programa desenvolvido foi alvo de validação, recorrendo-se para tal, a uma comparação dos resultados obtidos entre o respetivo programa e por um outro programa de grande fiabilidade. Assim sendo, concluiu-se que o software CAFE apresenta resultados válidos, encontrando-se apto a ser utilizado.

The occurrence of different viruses in nasopharyngeal secretions from children less than 5 years old with acute respiratory infections (ARI) was investigated over a period of 4 years (1982-1985) in Rio de Janeiro. Of the viruses known to be associated with ARI, all but influenza C and parainfluenza types 1, 2 and 4 were found. Viruses were found more frequently in children attending emergency or pediatric wards than in outpatients. This was clearly related to the high incidence of respiratory syncytial virus (RSV) in the more severe cases of ARI. RSV positive specimens appeared mainly during the fall, over four consecutive years, showing a clear seasonal ocurrence of this virus. Emergency wards provide the best source of data for RSV surveillance, showing sharp increase in the number of positive cases coinciding with increased incidence of ARI cases. Adenovirus were the second most frequent viruses isolated and among these serotypes 1,2 and 7 were predominant. Influenza virus and parainfluenza virus type 3 were next in frequency. Influenza A virus were isolated with equal frequency in outpatient departments, emergency and pediatric wards. Influenza B was more frequent among outpatients. Parainfluenza type 3 caused outbreaks in the shanty town population annually during the late winter or spring and were isolated mainly from outpatients. Herpesvirus, enterovi-rus and rhinovirus were found less frequently. Other viruses than RSV and parainfluenza type 3 did not show a clear seasonal incidence.

Os autores estudam a influência do café Rio em ligas com cafés brasileiros de bebida Mole. Foram ensaiadas porcentagens crescentes de Café Rio: 0,0; 0,5; 1,0; 1,5; 2,0; 2,5; 3,0; 3,5; 4,0; 4,5; 5,0; 7,5; 10,0; 12,5; 15,0; 20,0; 25,0; 30,0; 35,0; 40,0; 50,0. Realizaram-se dois experimentos em blocos incompletos equilibrados, com t = 21 tratamentos (os mencionados acima), k = 3 parcelas por bloco, r = 10 repetições, b = 70 blocos, L = 1. Cada parcela era formada de 3 xícaras, de tipo padrão, sobre as quais cada degustador dava uma só opinião. Cada parcela era provada por 3 degustadores. Os dados coletados são, pois, 630 para cada ensaio (210 parcelas, 3 degustadores). Atribuia-se a cada parcela, para fins de análise estatística, a média das opiniões dos 3 degustadores. Os dois ensaios deram resultados bem concordantes, que levaram às seguintes conclusões: a) Faz-se necessária a transformação dos dados, pois as variâncias relativas aos diversos tratamentos são muito discrepantes. b) A transformação T = √ Y dá resultados satisfatórios. c) O café Rio prejudica sensivelmente a bebida do café Mole, para teores a partir de 2,0%. d) Para teores de 4,5% em diante a liga tem bebida Riada ou Rio. e) A regressão obtida não é estritamente linear, mas a linha reta da uma aproximação razoável. f) Consideradas as porcentagens de 0,0 a 10,0%,a equação de regressão para os dados transformados pela transformação T = √ Y é: T = 1,7045 - 0,127 X, onde X é a porcentagem de café Rio e T dá a bebida, na escala numérica adotada, transformada pela raíz quadrada. g) A equação de regressão para os tratamentos de 0,0 a 10,0% de cafe Rio é Y = 3,0997 - 0,3281 X, isto é, há uma queda de 0,3281 na escala numérica da bebida, para cada unidade de porcentagem de café Rio.

O presente trabalho trata do estudo dos modelos de cristalização do cloreto de cobre (CuCl2.2H(2)0) na presença de extratos de sementes de cafe (Coffea arábica L.), variedade Mundo Novo. Pesquisaram-se 16 concentrações diferentes do sistema água-cloreto de cobre-extrato, com 3 repetições, descrevendo-se individualmente cada urna das series. Os melhores modelos foram obtidos usando-se a técnica de mistura filtrada. Os resultados obtidos permitem recomendar, para cloreto de cobre, concentrações entre 0,5 e 1,0 g/placa e, para extrato, concentrações entre 0,05 e 0,075 g/placa.

A passive sampling device called Monitor of NICotine or "MoNIC", was constructed and evaluated by IST laboratory for determining nicotine in Second Hand Tobacco Smoke (SHTS) or Environmental Tobacco Smoke (ETS). Vapour nicotine was passively collected on a potassium bisulfate treated glass fibre filter as collection medium. Analysis of collected nicotine on the treated filter by gas chromatography equipped with Thermoionic-Specific Detector (GC-TSD) after liquid-liquid extraction of 1mL of 5N NaOH : 1 mL of n-heptane saturated with NH3 using quinoline as internal standard. Based on nicotine amount of 0.2 mg/cigarette as the reference, the inhaled Cigarette Equivalents (CE) by non-smokers can be calculated. Using the detected CE on the badge for non-smokers, and comparing with amount of nicotine and cotinine level in saliva of both smokers and exposed non-smokers, we can confirm the use of the CE concept for estimating exposure to ETS. The regional CIPRET (Center of information and prevention of the addiction to smoking) of different cantons (Valais (VS), Vaud (VD), Neuchâtel (NE) and Fribourg (FR)) are going to organize a big campaign on the subject of the passive addiction to smoking. This campaign took place in 2007-2008 and has for objective to inform clearly the Swiss population of the dangerousness of the passive smoke. More than 3'900 MoNIC badges were gracefully distributed to Swiss population to perform a self-monitoring of population exposure level to ETS, expressed in term of CE. Non-stimulated saliva was also collected to determine ETS biomarkers nicotine/cotinine levels of participating volunteers. Results of different levels of CE in occupational and non-occupational situations in relation with ETS were presented in this study. This study, unique in Switzerland, has established a base map on the population's exposure to SHTS. It underscored the fact that all the Swiss people involved in this campaign (N=1241) is exposed to passive smoke, from <0.2 cig/d (10.8%), 1-2 to more than 10 cig/d (89.2%). In the area of high exposure (15-38 cig/d), are the most workers in public restaurant, cafe, bar, disco. By monitoring ETS tracer nicotine and its biomarkers, salivary nicotine and cotinine, it is demonstrated that the MoNIC badge can serve as indicator of CE passive smoking. The MoNIC badge, accompanied with content of salivary nicotine/cotinine can serve as a tool of evaluation of the ETS passive smoking and contributes to supply useful data for future epidemiological studies. It is also demonstrated that the salivary nicotine (without stimulation) is a better biomarker of ETS exposure than cotinine.

Tämän opinnäytetyön tarkoituksena oli selvittää, kuinka kahvilatyöntekijät suhtautuvat kahviin ja kahvin valmistukseen sekä millaisena työntekijät näkevät kahvin osana kahvilan laatua ja liiketoimintaa. Lisäksi tutkimuksessa haluttiin selvittää, löytyykö suhtautumisessa eroja kahvikoulutuksen saaneiden ja kouluttamattomien työntekijöiden välillä. Tutkimus toteutettiin kyselynä ja kysymyslomakkeet jaettiin Helsingin ydinkeskustan kahvilatyöntekijöille lokakuussa 2007. Lomakkeita jaettiin yhteensä 92 kappaletta, joista 61 saatiin takaisin. Kyselylomake sisälsi 12 väittämää, joihin vastaajan tuli vastata valitsemalla viidestä vastausvaihtoehdosta, kuinka samaa mieltä tai eri mieltä vastaaja on väittämästä. Tutkimukseen vastanneet kahvilatyöntekijät kokivat pystyvänsä omilla toimintatavoillaan vaikuttamaan siihen, miltä kahvi maistuu. He myös kokivat olevansa itse vastuussa laadun toteutumisesta. Työntekijöiden suhtautuminen asiakkaiden pyyntöön saada tuoretta kahvia vanhan kahvin tilalle vaihteli voimakkaasti. Vastaajien käsitys kahvin merkityksestä kahvilan myynnille sekä mielikuva kahvista edullisena ja hyväkatteisena jakoi vastaajien mielipiteitä. Lisäksi suuri osa vastaajista oli vain melko samaa mieltä väittämästä, että kahvi on kahvilan tärkein tuote. Kahvia ei siis koettu ehdottomasti kahvilan tärkeimmäksi tuotteeksi. Kahvikoulutuksen saaneiden ja kouluttamattomien työntekijöiden välillä oli jonkin verran eroavaisuuksia muun muassa käsityksissä kahvilaitteiden puhdistuksesta sekä tuoreen ja vanhan kahvin yhdistämisestä. Suurin ero koulutuksen saaneiden ja kouluttamattomien työntekijöiden välillä oli käsityksessä siitä, pystyykö kuvailemaan tarkasti kahvilassa tarjottavien erikoiskahvien sisältöä ja jopa suosittelemaan asiakkaalle jotain tiettyä kahvijuomaa.

Craving is considered the main variable associated with relapse after smoking cessation. Cue Exposure Therapy (CET) consists of controlled and repeated exposure to drug-related cues with the aim of extinguishing craving responses. Some virtual reality (VR) environments, such as virtual bars or parties, have previously shown their efficacy as tools for eliciting smoking craving. However, in order to adapt this technology to smoking cessation interventions, there is a need for more diverse environments that enhance the probability of generalization of extinction in real life. The main objective of this study was to identify frequent situations that produce smoking craving, as well as detecting specific craving cues in those contexts. Participants were 154 smokers who responded to an ad hoc self-administered inventory for assessing craving level in 12 different situations. Results showed that having a drink in a bar/pub at night, after having lunch/dinner in a restaurant and having a coffee in a cafe or after lunch/dinner at home were reported as the most craving-inducing scenarios. Some differences were found with regard to participants' gender, age, and number of cigarettes smoked per day. Females, younger people, and heavier smokers reported higher levels of craving in most situations. In general, the most widely cited specific cues across the contexts were people smoking, having a coffee, being with friends, and having finished eating. These results are discussed with a view to their consideration in the design of valid and reliable VR environments that could be used in the treatment of nicotine addicts who wish to give up smoking.
