Considerando a crescente importância que o iogurte vem assumindo no mercado nacional, inúmeras pesquisas têm sido executadas para melhoria da sua qualidade e outros produtos fermentados. A presença de contaminantes constitui, hoje, um dos grandes problemas para a indústria, causando a perda do produto em função das alterações de sabor, cor e também estufamento de embalagens nas prateleiras refrigeradas de comercialização. O principal objetivo deste trabalho foi realizar levantamento das características microbiológicas dos iogurtes, por meio da enumeração de fungos e leveduras encontrados, relacionando-os com a vida-de-prateleira do iogurte de diferentes marcas. Setenta e duas amostras de iogurtes comercializados foram submetidos à análise microbiológica da população de Lactobacillus/Streptococcus, contagem de leveduras e fungos e análises químicas (pH, % ácido láctico). Os iogurtes foram produzidos por 4 produtores diferentes e comercializados de Lavras - MG, Brasil. Lactobacillus delbrueckii spp. bulgaricus e Streptococcus thermophilus foram encontrados na proporção de 1:2, respectivamente, em 80% das amostras analisadas. Não foi encontrada diferença significativa no valor de pH e na concentração de ácido láctico nas amostras. Em 28% das amostras foi encontrada população de leveduras acima de 100 UFC/g, enquanto em 7% foram obtidas contagens de leveduras maiores que 1000 UFC/g. Os resultados indicam que, apesar de todas as inovações técnicas e cuidados durante e depois da fabricação do iogurte, o produto ainda pode estar sujeito à contaminação microbiana, quando não atendidas às condições de higiene e sanidade.

Os objetivos deste trabalho foram a elaboração de um processo de fermentação a partir do mosto de polpa de cajá, Spondias mombin, para a obtenção de uma bebida alcoólica, bem como a avaliação da aceitação da mesma. As polpas das frutas utilizadas foram quimicamente analisadas (açúcares, acidez, pectina, vitamina C, pectinases, amido e fenólicos). A polpa de cajá foi chaptalizada a 24°Brix, constituindo 20L de mosto. O mosto foi desacidificado, com CaCO3, a pH 3,8, para ser submetido ao tratamento enzimático com UltrazymR AFP-L (Novo DK). Foi utilizado SO2 como agente inibidor do crescimento bacteriano e como antioxidante. O mosto foi clarificado com bentonite. Posteriormente, o mosto foi inoculado com Saccharomyces cerevisiae tipo selvagem na concentração de 10(7) células/mL. A fermentação foi conduzida a 22°C durante 10 dias, com acompanhamento diário do grau Brix e da atividade fermentativa pela liberação de CO2. Ao final da fermentação, o mosto foi armazenado a 10°C por 10 dias e foi feita a primeira trasfega. A segunda trasfega ocorreu 30 dias após a primeira, antes da filtração. Na bebida elaborada foram feitas análises de etanol, glicerol, ácidos orgânicos, álcoois superiores, metanol, ésteres e acetaldeído. Observou-se alta concentração de álcoois superiores, os quais são normalmente responsáveis pela formação do sabor e aroma em bebidas alcóolicas. A aceitação da bebida foi avaliada por 45 provadores não treinados, utilizando-se escala hedônica de 9 pontos. Os dados mostraram que o fermentado de cajá foi bem aceito, podendo ser uma nova fonte de investimento para indústrias ou pequenos produtores.

Os cogumelos do gênero Pleurotus normalmente crescem bem em substratos mais pobres em nitrogênio, ao contrário dos cogumelos Agaricus que requerem substratos com relação C/N mais estreita. Por outro lado, os valores nutricionais do cogumelo dependem da composição química do substrato utilizado e das condições de cultivo. Este trabalho teve como objetivo avaliar o teor de proteína dos corpos de frutificação do cogumelo Pleurotus sajor-caju cultivado em capim coast-cross, bagaço de cana-de-açúcar, farelo de trigo e diferentes teores de nitrogênio. Apenas os substratos com teores de nitrogênio de 0,65 a 1,30% foram colonizados, enquanto que nos substratos com 1,75 e 2,20% de nitrogênio não houve colonização. Não houve diferença significativa na produção de cogumelos, porém o teor de proteína dos cogumelos produzidos no substrato com 1,30% de N foi significativamente superior em relação aos substratos com menor teor de N.

The common bean (Phaseolus vulgaris L.) is a staple food in the Brazilian diet and represents the major source of dietary protein and other micronutrients and minerals. Despite the considerable protein concentration in beans, the food is considered of low biological value when compared to animal proteins and other plant protein sources. To improve the availability of protein in beans, enzymatic treatments were performed in four cultivars (ON, OPNS, TAL and VC3). The approach was a completely randomized design with four replicates. We used a 4 × 3 factorial arrangement (four cultivars and three treatments: treatment 1-addition of commercial protease (Trypsin 250, Difco), treatment 2-addition of protease from Bacillus sp., and treatment 3:-control without enzyme addition). The enzyme: substrate ratio was 5% w/w (amount of enzyme per total protein in bean flour). The approach was a completely randomized design with four replicates. A 4 × 3 factorial arrangement (four cultivars and three treatments, the same as those mentioned above) was used. The concentration of total protein (g.100 g-1 of dry matter) in the samples ranged from 16.94 to 18.06%, while the concentration of total phenolics was between 0.78 and 1.12% (g Eq. tannic acid.100 g-1 dry matter). The in vitro protein digestibility of enzymatically untreated bean flour (control) ranged from 47.30 to 56.17% based on the digestibility of casein. Concentrations of P, K, Ca, Mg, and Zn observed in the four cultivars tested were within the average values available in the literature. Treatment 2 with protease from Bacillus sp. induced decreases in the levels of Cu and Mn. The average Fe content increased in all bean flour samples when treated with proteases, reaching a maximum increase of 102% in the TAL flour treated with protease from Bacillus sp. The digestibility of all beans tested was significantly increased (p < 0.05) after the enzyme treatment. The greatest change was observed in the OPNS cultivar treated with protease from Bacillus sp., which increased its digestibility from 54.4% (control treatment) to 81.6%.

Com a crescente exigência do mercado consumidor por produtos de melhor qualidade, busca-se o constante aprimoramento da produção de cachaça, uma vez que todas as etapas da cadeia produtiva de bebidas fermento-destiladas são importantes. O objetivo deste trabalho foi acompanhar o processo de fermentação para produção de cachaça, utilizando diferentes isolados de Saccharomyces cerevisiae a partir da quantificação de metabólitos secundários por Cromatografia Gasosa. O acompanhamento do processo deu-se desde o preparo do inóculo até o final do processo fermentativo. O estudo foi conduzido na Universidade Federal de Lavras (UFLA). Foram utilizados 8 isolados de Saccharomyces cerevisiae inoculados em caldo de cana, dos quais foram retiradas amostras durante a fase de crescimento em sistema de batelada alimentada e fermentação. As amostras foram analisadas quanto à taxa de floculação, ºBrix e álcoois superiores. Os parâmetros avaliados apresentaram diferenças para cada isolado. O melhor isolado para a produção de cachaça foi o isolado UFLA CA116 por apresentar alto número de células viáveis, maior taxa de floculação, ausência 1-propanol, presença de 1,3 butanediol.

To investigate microbial diversity and identify spoilage bacteria in fresh pork sausages during storage, twelve industrial pork sausages of different trademarks were stored at 4 ºC for 0, 14, 28 and 42 days, 80% relative humidity and packaged in sterile plastic bags. Microbiological analysis was performed. The pH and water activity (a w) were measured. The culture-independent method performed was the Polymerase Chain Reaction - Denaturing Gradient Gel Electrophoresis (PCR-DGGE). The culture-dependent method showed that the populations of mesophilic bacteria and Lactic Acid Bacteria (LAB) increased linearly over storage time. At the end of the storage time, the average population of microorganisms was detected, in general, at the level of 5 log cfu g-1. A significant (P < 0.005) increase was observed in pH and a w values at the end of the storage time. The PCR-DGGE allowed a rapid identification of dominant communities present in sausages. PCR-DGGE discriminated 15 species and seven genera of bacteria that frequently constitute the microbiota in sausage products. The most frequent spoilage bacteria identified in the sausages were Lactobacillus sakei and Brochothrix thermosphacta. The identification of dominant communities present in fresh pork sausages can help in the choice of the most effective preservation method for extending the product shelf-life.

This paper reviews the current knowledge of climatic risks and impacts in South Asia associated with anthropogenic warming levels of 1.5°C to 4°C above pre-industrial values in the 21st century. It is based on the World Bank Report “Turn Down the Heat, Climate Extremes, Regional Impacts and the Case for Resilience” (2013). Many of the climate change impacts in the region, which appear quite severe even with relatively modest warming of 1.5–2°C, pose significant hazards to development. For example, increased monsoon variability and loss or glacial meltwater will likely confront populations with ongoing and multiple challenges. The result is a significant risk to stable and reliable water resources for the region, with increases in peak flows potentially causing floods and dry season flow reductions threatening agriculture. Irrespective of the anticipated economic development and growth, climate projections indicate that large parts of South Asia’s growing population and especially the poor are likely to remain highly vulnerable to climate change.

Tick-borne relapsing fever in western North America is a zoonosis caused by spirochetes in the genus Borrelia that are transmitted by argasid ticks of the genus Ornithodoros (1). Human disease occurs in many focal areas and is associated with infections of Borrelia hermsii, B. turicatae, and possibly B. parkeri (2,3). Although the ecologic parameters that maintain B. hermsii and B. turicatae differ, human infections usually occur in rustic cabins (B. hermsii) and caves (B. turicatae) inhabited by ticks and their terrestrial vertebrate hosts (1). Recently, Gill et al. (4) provided evidence that the argasid bat tick, Carios kelleyi, feeds upon humans. Subsequently, Loftis et al. (5) used PCR analysis and DNA sequencing to detect in C. kelleyi an unidentifi ed Borrelia species that was closely related to B. turicatae and B. parkeri.

Spin coherence generation in an ensemble of negatively charged (In,Ga)As/GaAs quantum dots was investigated by picosecond time-resolved pump-probe spectroscopy measuring ellipticity. Robust coherence of the ground-state electron spins is generated by pumping excited charged exciton (trion) states. The phase of the coherent state, as evidenced by the spin ensemble precession about an external magnetic field, varies relative to spin coherence generation resonant with the ground state. The phase variation depends on the pump photon energy. It is determined by (a) pumping dominantly either singlet or triplet excited states, leading to a phase inversion, and (b) the subsequent carrier relaxation into the ground states. From the dependence of the precession phase and the measured g factors, information about the quantum dot shell splitting and the exchange energy splitting between triplet and singlet states can be extracted in the ensemble.

In this thesis we present techniques that can be used to speed up the calculation of perturbative matrix elements for observables with many legs ($n = 3, 4, 5, 6, 7, ldots$). We investigate several ways to achieve this, including the use of Monte Carlo methods, the leading-color approximation, numerically less precise but faster operations, and SSE-vectorization. An important idea is the use of enquote{random polarizations} for which we derive subtraction terms for the real corrections in next-to-leading order calculations. We present the effectiveness of all these methods in the context of electron-positron scattering to $n$ jets, $n$ ranging from two to seven.

Background Sedation prior to performance of diagnostic esophagogastroduodenoscopy (EGDE) is widespread and increases patient comfort. But 98% of all serious adverse events during EGDEs are ascribed to sedation. The S3 guideline for sedation procedures in gastrointestinal endoscopy published in 2008 in Germany increases patient safety by standardization. These new regulations increase costs because of the need for more personnel and a prolonged discharge procedure after examinations with sedation. Many patients have difficulties to meet the discharge criteria regulated by the S3 guideline, e.g. the call for a second person to escort them home, to resign from driving and working for the rest of the day, resulting in a refusal of sedation. Therefore, we would like to examine if an acupuncture during elective, diagnostic EGDEs could increase the comfort of patients refusing systemic sedation. Methods/Design A single-center, double blinded, placebo controlled superiority trial to compare the success rates of elective, diagnostic EGDEs with real and placebo acupuncture. All patients aged 18 years or older scheduled for elective, diagnostic EGDE who refuse a systemic sedation are eligible. 354 patients will be randomized. The primary endpoint is the rate of successful EGDEs with the randomized technique. Intervention: Real or placebo acupuncture before and during EGDE. Duration of study: Approximately 24 months. Discussion Organisation/Responsibility The ACUPEND - Trial will be conducted in accordance with the protocol and in compliance with the moral, ethical, and scientific principles governing clinical research as set out in the Declaration of Helsinki (1989) and Good Clinical Practice (GCP). The Interdisciplinary Endoscopy Center (IEZ) of the University Hospital Heidelberg is responsible for design and conduct of the trial, including randomization and documentation of patients' data. Data management and statistical analysis will be performed by the independent Institute for Medical Biometry and Informatics (IMBI) and the Center of Clinical Trials (KSC) at the Department of General, Visceral and Transplantation Surgery, University of Heidelberg.

Buttressing is a trait special to tropical trees but explanations for its occurrence remain inconclusive. The two main hypotheses are that they provide structural support and/or promote nutrient acquisition. Studies of the first are common but the second has received much less attention. Architectural measurements were made on adult and juvenile trees of the ectomycorrhizal species Microberlinia bisulcata, in Korup (Cameroon). Buttressing on this species is highly distinctive with strong lateral extension of surface roots of the juveniles leading to a mature buttress system of a shallow spreading form on adults. This contrasts with more vertical buttresses, closer to the stem, found on many other tropical tree species. No clear relationship between main buttress and large branch distribution was found. Whilst this does not argue against the essential structural role of buttresses for these very large tropical trees, the form on M. bisulcata does suggest a likely second role, that of aiding nutrient acquisition. At the Korup site, with its deep sandy soils of very low phosphorus status, and where most nutrient cycling takes place in a thin surface layer of fine roots and mycorrhizas, it appears that buttress form could develop from soil-surface root exploration for nutrients by juvenile trees. It may accordingly allow M. bisulcata to attain the higher greater competitive ability, faster growth rate, and maximum tree size that it does compared with other co-occurring tree species. For sites across the tropics in general, the degree of shallowness and spatial extension of buttresses of the dominant species is hypothesized to increase with decreasing nutrient availability.
