Mobile element dynamics in seven alleles of the chalcone synthase D locus (CHS-D) of the common morning glory (Ipomoea purpurea) are analyzed in the context of synonymous nucleotide sequence distances for CHS-D exons. By using a nucleotide sequence of CHS-D from the sister species Ipomoea nil (Japanese morning glory [Johzuka-Hisatomi, Y., Hoshino, A., Mori, T., Habu, Y. & Iida, S. (1999) Genes Genet. Syst. 74, 141–147], it is also possible to determine the relative frequency of insertion and loss of elements within the CHS-D locus between these two species. At least four different types of transposable elements exist upstream of the coding region, or within the single intron of the CHS-D locus in I. purpurea. There are three distinct families of miniature inverted-repeat transposable elements (MITES), and some recent transpositions of Activator/Dissociation (Ac/Ds)-like elements (Tip100), of some short interspersed repetitive elements (SINEs), and of an insertion sequence (InsIpCHSD) found in the neighborhood of this locus. The data provide no compelling evidence of the transposition of the mites since the separation of I. nil and I. purpurea roughly 8 million years ago. Finally, it is shown that the number and frequency of mobile elements are highly heterogeneous among different duplicate CHS loci, suggesting that the dynamics observed at CHS-D are locus-specific.

Small potted trees of Spondias purpurea were monitored to determine the costs and controls of flowering and fruiting. The effect of photoperiod, extremes in moisture and temperature, and defoliation were examined. The carbon exchange rates of the leaves, shoots and fruits were determined. Light response curves and diurnal levels were also investigated. $\sp{13}$Carbon labeling was used to determine which plant parts are carbon sinks. Photoperiod induces dormancy and bud activity. Extremes in soil moisture and temperature induce leaf fall. Flowers, fruits, and roots are carbon sinks. The results were used to develop a phenological model with latitude, soil moisture, and air temperature as variables. ^

Paracoccidioides brasiliensis is characterized by a multiple budding phenotype and a polymorphic cell growth, leading to the formation of cells with extreme variations in shape and size. Since Cdc42 is a pivotal molecule in establishing and maintaining polarized growth for diverse cell types, as well as during pathogenesis of certain fungi, we evaluated its role during cell growth and virulence of the yeast-form of P. brasiliensis. We used antisense technology to knock-down PbCDC42`s expression in P. brasiliensis yeast cells, promoting a decrease in cell size and more homogenous cell growth, altering the typical polymorphism of wild-type cells. Reduced expression levels also lead to increased phagocytosis and decreased virulence in a mouse model of infection. We provide genetic evidences underlying Pbcdc42p as an important protein during host-pathogen interaction and the relevance of the polymorphic nature and cell size in the pathogenesis of P. brasiliensis. Novel macrolides, lobatamides A-F (1-6), have been isolated from shallow water Australian collections of Aplidium lobatum, from a deep water collection of Aplidium sp., and from an unidentified Philippine ascidian. Full details of the isolation and structure elucidation of 1-6 are provided herein, along with results and analyses of the testing of lobatamides A-D (1-4) in the NCI human tumor 60 cell-line screen. The lobatamides share a common core structure with the recently described salicylihalamides, which were isolated from a Haliclona sp. sponge. COMPARE analyses of the mean-graph differential cytotoxicity profiles of the lobatamides and the salicylihalamides showed high correlations with each other but not with members of the NCI's standard agents database. These compounds, therefore, appear to comprise a new mechanistic class, meriting further antitumor investigations.

The sludge produced in sewage treatment plants can contain toxic substances. Among these, the genotoxic substances are of great concern. The present paper aimed at evaluating the genotoxicity of treated sludge samples collected at four different sewage treatment plants (STP) located in the State of Sao Paulo Brazil, using the Trad-MN assay. Another objective of the study was to compare the responses of the Clone #4430 with the Tradescantia pallida. Sludge samples mixed with reference soil in concentrations of 10, 25 and 50% (v/v) were tested in experiments with 3 months exposure in the field. Negative and positive controls (arsenic trioxide) were also tested with both plants. In Clone #4430 two sludge samples induced genotoxicity while in T pallida three were positive, although no clear dose-response were observed for both plants. Results with the negative and positive controls suggest that T pallida presented similar results when compared to the Clone #4430. The protocol using plants chronically exposed to sludge mixed with soil seems to be a promising tool to assess the genotoxicity of sludge although time consuming. (C) 2008 Elsevier B.V. The present study was made to check if the Trad-MCN bioassay, developed with inflorescences of Tradescantia pallida cv. Purpurea, might discriminate genotoxic risks in areas of the city of Santo Andre (SE Brazil) contaminated by different air pollutants, and periods of the year when risks are higher, and to determine if the variations in the frequency of micronuclei (MCN) can be explained by environmental factors that characterize the stressful situation in each site. Potted plants were exposed in sites highly contaminated by ozone (Capuava and School) and in sites reached by high vehicular emissions (downtown and Celso Daniel Park). Pedroso Park, far from the polluted areas, was taken as reference. From September 2003 to September 2004, 20 young inflorescences were collected twice a week from each place and the frequencies of MCN were estimated. The environmental conditions observed in the polluted sites were stressful enough to promote an increase of MCN, mainly in sites reached by high vehicular emissions. But MCN rates in Capuava and at Celso Daniel Park could not be predicted only by pollutants which characterized the air contamination in these sites. More severe weather conditions, mainly low temperature, relative humidity and rainfall, caused an increase of MCN. Improvement of the biomonitoring system is recommended to minimize this negative influence of weather factors. (C) 2008 Elsevier Inc. All rights reserved.

Ergot, caused by Claviceps africana, has emerged as a serious threat to sorghum hybrid seed production worldwide. In the absence of gene-for-gene-based qualitative resistance in commercial cultivars, varieties with high pollen production that can escape ergot infection are preferred. Recent demonstration of differences in ergot susceptibility among male-sterile lines has indicated the presence of partial resistance. Using chitin-specific fluorescin-isothiocyanate-conjugated wheat germ agglutin and callose-specific aniline blue, this study investigated the process of sorghum ovary colonization by C. africana. Conidia germinated within 24 h after inoculation (a.i.); the pathogen was established in the ovary by 79 h a.i., and at least half of the ovary was converted into sphacelial tissue by 120 h a.i. Changes in fungal cell wall chitin content and strategic callose deposition in the host tissue were associated with penetration and invasion of the ovary. The rate of ovary colonization differed in three male-sterile lines that also differed in ergot susceptibility. This work demonstrates a possible histological basis for partial resistance in male-sterile sorghum lines that could lay the foundation for variety improvement through further breeding and selection.

Algumas características de madeiras da região de Manaus, no Amazonas foram determinadas, como identificação anatômica, densidade básica e umidade, visando reconhecer o material para produção de carvão vegetal. Identificou-se 28 famílias botânicas sendo que 50% do material foi representado por Anonaceae, Lecythidaceae e Sapotaceae. Quanto a gênero, encontrou-se 43, destes sobressairam-se Miconia, Guateria, Brasimum Goupiae Eschweilera, representando 38% das madeiras. As espécies Goupia glabra, Aldina heterophylla, Jacaranda copaia, Diplotropis purpurea e Tapirina guianensis foram as de maior ocorrência na madeira estudada. A densidade básica média foi 0,73 g/cm3 e a umidade média, sendo a madeira seca ao ar livre, ficou em 24.16% base úmida. A madeira foi carbonizada em fornos de alvenaria e do carvão obtido determinou-se características físicas e químicas. Os resultados médios foram: tamanho médio 45,30mm; friabilidade 15,46% de finos gerados menor que 13 mm e índice de quebra de 39,13%; tensão máxima de ruptura foi 43,36kgf/cm2; densidade aparente 0,51 e verdadeira 1,48; porosidade 65,28%; umidade 6,80%; matérias voláteis 17,65%; cinzas 2,29%; carbono fixo 80,06% e poder calorífico superior 6.826 Kcal/kg.

This paper deals with anatomical descriptions of some types of nectaries in 27 species of honey plants of Piracicaba, S. P. The material studied was divides in two groups: a) Extra-floral nectaries; b) Floral nectaries. Euphorbia pulcherrima, Willd; showed to belonging to the first group: its nectaries tissue consist of an epidermal layer of cell without stomata and with true gland, with subepidermal cells diferentiated by the thickness of the wall. Among the plants with floral nectaries, the following types has been listed, according the location of the nectary in the flower: 1 - with true glands: a) in sepals, Hibiscus rosa sinensis, L.; Dombeya Wallichii, Bth. e Hk; b) in the stamens tube, Antigonum leptopus, Hook e Arn.; 2 - on the receptacle with nectariferous tissue in the epidermal cell with: a) thickness wall with stomata, Prunus persical, L.; b) thin wall without stomata, Crotalaria paulinia, Shranck; Caesal-pinia sepiaria, Roxb; Aberia caffra; 3 - with a disc located in the receptacle with: epidermal: a) with stomata, Coffea arábica, L. var. semper florens; Citrus aurantifolia, Swing; Cinchona sp.; Pryrostegia ignea, Presl.; b) without stomata and with thin wall, Leojurus sibiricus, L.; Bactocydia unguis, Mart., Ipomoea purpurea, L.; Greviüea Thelemanniana, Hueg.; Dolichos lablab, L.; Vernonia polyanthes, Less., Montanoa bipinatifida, C. Koch., Eruca sativa, L. Brassica Juncea, Co; Eucalyptus tereticomis, Smith.; Eucalyptus rostrata, Schleche; Salvia splendens, Selow.; 4 - in the basal tissues of the ovary, Budleia brasiliensis, Jacq F.; Petrea subserrata, Cham.; 5 - in the base of stamens, Per sea americana, Mill. On the anatomical point of view, most of the types of nectary studied has external nectariferous tissues, located on the epidermal cells with thin periclinal wall and without stomata. The sub-epidermal layer were rich in sugar. Short correlation was found between the structure of the nectary and the amount of nectar secretion. So, in the nectary with true glands, in those with thin wall and without stomata on epidermal cells and in those with stomata, the secretion was higher than in the other types listed.

RESUMEN El aumento del CO2 atmosférico debido al cambio global y/o a las prácticas hortícolas promueve efectos directos sobre crecimiento vegetal y el desarrollo. Estas respuestas pueden ocurrir en ecosistemas naturales, pero también se pueden utilizar para aumentar la producción de algunas plantas y de algunos compuestos secundarios. El actual trabajo intenta estudiar los efectos del enriquecimiento atmosférico del CO2 bajo condiciones de invernadero en el crecimiento y la concentración y la composición de metabolitos secundarios de Taxus bacatta, Hypericum perforatum y Echinacea purpurea en condiciones ambientales mediterráneas. La fertilización del CO2 muestra perspectivas interesantes para la mejorara y aplicabilidad de técnicas hortícolas para aumentar productividad de plantas medicinales, a pesar de diferencias claras entre la especie. En general esta técnica promueve aumentos importantes y significativos en producción primaria y, en algunos casos, también en compuestos secundarios. Esto tiene una gran importancia hortícola porque la productividad a nivel de cosecha total aumenta, directamente porque se aumenta la concentración e indirectamente porque se aumenta la biomasa. SUMMARY The increase of atmospheric CO2 due to global change and/or horticultural practices promotes direct effects on plant growth and development. These responses may occur in natural ecosystems, but also can be used to increase the production of some plants and some secondary compounds. Present work tries to study the effects of atmospheric CO2 enrichment under greenhouse conditions on growth and in the concentration and composition of secondary metabolites of Taxus bacatta, Hypericum perforatum and Echinacea purpurea under Mediterranean environmental conditions. CO2 fertilization shows interesting perspectives to increase and improve horticultural techniques in order to increase plant medicinal productivity, in spite of clear differences among the species. In general this technique promotes important and significant increases in primary productivity and, in some cases, also in secondary compounds. This has a great horticultural relevance because the total productivity of this kind of products increase at crop level, directly because concentration is increased and /or indirectly because biomass is increased. L'actual treball intenta estudiar els efectes de l'enriquiment atmosfèric del CO2 sota condicions d'hivernacle en el creixement i la concentració i la composició de metabòlits secundaris de Taxus bacatta, Hypericum perforatum i Echinacea purpurea en condicions ambientals mediterrànies. La fertilització del CO2 mostra perspectives interessants per a la millora i aplicabilitat de tècniques hortícoles per a augmentar productivitat de plantes medicinals, a pesar de diferències clares entre l'espècie. En general aquesta tècnica promou augments importants i significatius en producció primària i, en alguns casos, també en compostos secundaris. Això té una gran importància hortícola perquè la productivitat a nivell de collita total augmenta, directament perquè s'augmenta la concentració i indirectament perquè s'augmenta la biomassa.

O objetivo deste trabalho foi construir um fenograma de trinta indivíduos de seis espécies de Spondias, amostradas em três diferentes locais, baseado em 120 marcadores AFLP das combinações de primers EcoR1/Mse1 para esclarecer as inter-relações entre as espécies, subsidiar a exploração das espécies como porta-enxerto e fornecer informações genômicas dessas espécies. O fenograma UPGMA do coeficiente de Jaccard apresentou boa definição, com valor co-fenético de 0,925 e localização externa de dois "outgroups" das espécies Mangifera indica e Schinopsis brasiliensis da família Anacardeaceae. Todos os indivíduos de S. cytherea, S. tuberosa e S. purpurea foram agrupados dentro de cada espécie, independentemente do local de amostragem, enquanto quatro dos seis indivíduos de Spondias sp (umbu-cajá) e S. mombin foram agrupadas, e outros dois indivíduos foram posicionados fora dos agrupamentos dessas espécies. Os dois indivíduos de Spondias sp (umbuguela) não se posicionaram próximos no fenograma. A posição do umbu-cajá e da umbuguela (PE) entre umbuzeiro e cajazeira, e similaridades entre 50% e 60% sugerem que as mesmas possam ser híbridas entre as duas espécies. O fenograma sugeriu que S. cytherea foi a espécie mais divergente das estudadas.
