A fitoterapia constitui uma forma de terapia medicinal que vem crescendo visivelmente ao longo dos anos, no entanto, apesar da extensa utilização dos fitoterápicos, a qualidade destes medicamentos muitas vezes é deficiente e questionável. Dentre as plantas medicinais mais empregadas como fitoterápicos, temos a Calendula officinalis L.(Asteraceae), utilizada pelos seus efeitos antiinflamatórios, antisépticos e cicatrizantes. Sendo assim, o presente estudo objetivou aprimorar e consolidar o emprego de metodologias de tecnologia farmacêutica na área de desenvolvimento de fitoterápicos, por meio da realização de estudos preliminares do planejamento/pré-formulação de uma formulação fitoterápica semi-sólida contendo tintura de C. officinalis L., visando o controle de qualidade das etapas do seu desenvolvimento. Na caracterização física e físico-química do pó e da tintura de calndula foi possível obter especificações farmacognósticas condizentes com as da literatura, alm de constatar a identidade do material vegetal através da detecção do marcador químico rutina por CCD. Por meio da validação do método, que apresentou parâmetros recomendados pela legislação vigente, foram determinados 463 μg/mL de rutina na tintura. Os espectros obtidos na região do infravermelho (IV) mostraram bandas características da rutina no extrato liofilizado, alm de demonstrar a permanência dessas bandas após sua mistura com os excipientes da formulação. As técnicas termoanalticas confirmaram a compatibilidade entre os excipientes e o extrato liofilizado de calndula. Na avaliação da estabilidade preliminar do gel, a formulação permaneceu estável durante a realização dos ciclos em estufa (45 2 0C) e temperatura ambiente (25 2 0C). No estudo preliminar da permeação, o gel apresentou tendência para favorecer a permeação dos flavonóides totais expresso em rutina para a fase receptora. Estes resultados demonstram a importância do emprego de protocolos de controle de qualidade das matérias primas vegetais, alm do estabelecimento de metodologias tecnolgicas para a produção de fitoterápicos.

Polyphenolic compounds present in rosemary were found to have antioxidant properties, anticarcinogenic activity, and to increase the detoxification of pro-carcinogens. The aim of the study was to determine the effect the aqueous extract of rosemary (AER) on mutagenicity induced by methylmethane sulfonate in meristematic cells of Allium cepa, as well as to describe its mode of action. Anti-mutagenicity experiments were carried out with 3 different concentrations of AER, which alone showed no mutagenic effects. In antimutagenicity experiments, AER showed chemopreventive activity in cultured meristematic cells of A. cepa against exposure to methylmethane sulfonate. Additionally, post-treatment and simultaneous treatment using pre-incubation protocols were the most effective. Evaluation of different protocols and the percent reduction in DNA indicated bioantimutagenic as well desmutagenic modes of action for AER. AER may be chemopreventive and antimutagenic.

Abstract Background Phenolic compounds combine antioxidant and hypocholesterolemic activities and, consequently, are expected to prevent or minimize cardiometabolic risk. Methods To evaluate the effect of an aqueous extract (AQ) and non-esterified phenolic fraction (NEPF) from rosemary on oxidative stress in diet-induced hypercholesterolemia, 48 male 4-week old Wistar rats were divided into 6 groups: 1 chow diet group (C) and 5 hypercholesterolemic diet groups, with 1 receiving water (HC), 2 receiving AQ at concentrations of 7 and 140 mg/kg body weight (AQ70 and AQ140, respectively), and 2 receiving NEPF at concentrations of 7 and 14 mg/kg body weight (NEPF7 and NEPF14, respectively) by gavage for 4 weeks. Results In vitro, both AQ and NEPF had remarkable antioxidant activity in the 2,2-diphenyl-1-picrylhydrazyl (DPPH●) assay, which was similar to BHT. In vivo, the group that received AQ at 70 mg/kg body weight had lower serum total cholesterol (−39.8%), non-HDL-c (−44.4%) and thiobarbituric acid reactive substance (TBARS) levels (−37.7%) compared with the HC group. NEPF (7 and 14 mg/kg) reduced the tissue TBARS levels and increased the activity of tissular antioxidant enzymes (superoxide dismutase, catalase and glutathione peroxidase). Neither AQ nor NEPF was able to ameliorate the alterations in the hypercholesterolemic diet-induced fatty acid composition in the liver. Conclusions These data suggest that phenolic compounds from rosemary ameliorate the antioxidant defense in different tissues and attenuate oxidative stress in diet-induced hypercholesterolemic rats, whereas the serum lipid profile was improved only in rats that received the aqueous extract.

Future atmospheric CO2 levels will most likely have complex consequences for marine organisms, particulary photosynthetic calcifying organisms. Corallina officinalis L. is an erect calcifying macroalga found in the inter- and subtidal regions of temperate rocky coastlines and provides important substrate and refugia for marine meiofauna. The main goal of the current study was to determine the physiological responses of C. officinalis to increased CO2 concentrations expected to occur within the next century and beyond. Our results show that growth and production of inorganic material decreased under high CO2 levels, while carbonic anhydrase activity was stimulated and negatively correlated to algal inorganic content. Photosynthetic efficiency based on oxygen evolution was also negatively affected by increased CO2. The results of this study indicate that C. officinalis may become less competitive under future CO2 levels, which could result in structural changes in future temperate intertidal communities.

En el presente estudio se analiza la influencia de la inoculación con Azospirillum brasilense, con Pseudomonas fluorescens y la inoculación conjunta con ambas rizobacterias en las especies de plantas aromáticas Ocimum basilicum var. genovesse, Ocimum basilicum var. minimum, Petroselinum sativum var. lisa y Salvia officinalis. Se evaluará su desarrollo morfolgico, atendiendo a tres parámetros: la longitud del tallo, el peso fresco y la superficie foliar. Así como el posible incremento en el contenido de aceite esencial que pueda tener la planta tratada. El cultivo se llevó a cabo en alveolos de ForestPot® 300, sobre mezcla de turba y vermiculita 3:1, con riego diario y sin adición de fertilizantes. Los resultados indican que en todas las especies y en todos los apartados estudiados, la inoculación de las rizobacterias produjo un incremento del desarrollo y del contenido de aceite esencial en comparación con el tratamiento Control, excepto en el caso de la longitud de las dos variedades de O. basilicum al inocularlas con P. fluorescens, en las que produjo una ligera disminución respecto al Control. En el caso de P. sativum var. lisa, solo las plantas que fueron inoculadas sobrevivieron. A partir de estos resultados, puede decirse que la inoculación con estas rizobacteras promotoras del crecimiento puede tener una gran importancia como sustitución de fertilizantes minerales, obteniéndose de este modo una producción más ecolgica y respetuosa con el medio.

Proliferation of dispersed plant cells in culture is strictly dependent on cell density, and cells in a low-density culture can only grow in the presence of conditioned medium (CM). No known plant hormones have been able to substitute for CM. To quantify the mitogenic activity of CM, we examined conditions for the assay system using mechanically dispersed mesophyll cells of Asparagus officinalis L. and established a highly sensitive bioassay method. By use of this method, the mitogenic activity of CM prepared from asparagus cells was characterized: it was heat-stable, susceptible to pronase digestion, and resistant to glycosidase treatment. On the basis of these results, the mitogenic activity in CM was purified 10(7)-fold by column chromatography, and two factors named phytosulfokine-alpha and -beta (PSK-alpha and PSK-beta) were obtained. By amino acid sequence analysis and mass spectrometry, the structures of these two factors were determined to be sulfated pentapeptide (H-Tyr(SO3H)-Ile-Tyr(SO3H)-Thr-Gln-OH) and sulfated tetrapeptide (H-Tyr(SO3H)-Ile-Tyr(SO3H)-Thr-OH). PSK-alpha and PSK-beta were prepared by chemical synthesis and enzymatic sulfation. The synthetic peptides exhibited the same activity as the natural factors, confirming the structure for PSK-alpha and PSK-beta mentioned above. This is the first elucidation of the structure of a conditioned medium factor required for the growth of low-density plant cell cultures.

The objective of this study was to evaluate different binders when preparing salt cereal bars and to characterize them as physical, physico-chemical and sensory parameters. Four formulations of different cereal bars using binders have been developed. The evaluated binders were collagen, guar gum, xanthan gum and psyllium. The developed cereal bars were evaluated according to their physical characteristics (color and texture), physicochemical (pH, moisture, ash, protein, lipids, Aw, crude fiber) beyond their calorie, fatty acid composition and concentration of the main minerals. Among the four binding agents evaluated, psyllium stood out due to its physicochemical characteristics. A cereal bar high in protein and fiber; low in carbohydrates and water activity. The binding agent guar gum and xanthan showed characteristics similar to psyllium, especially regarding to fiber content. Collagen as binder gave the final product a high level in protein and lipid. The color and texture analyzes showed that the salt cereal bars had the color and crispness characteristics for this type of product. Regarding to the composition in the fatty acid, the developed bars offer a good supply of essential fatty acids to the human body. The same was observed regarding to mineral contents. Sensory, salt cereal bars made with chia showed good acceptability, highlighting the elaborate bar with psyllium binder. Different binders demonstrated technological efficiency in the preparation of salt cereal bars. The binder psyllium agent over others showed better physical-chemical and sensory characteristics. However, in general the product has healthy and nutritional characteristics it may be indicated for a protein diet with high fiber content and free sugars.

Salvia species are used worldwide for medicine purposes. In general, these medicinal plants have high amounts of flavonoids and phenolic acids, that are thought to be closely related to their health properties [1,2]. In this work, the aerial parts of Salvia farinacea, Salvia mexico, Salvia greggii and Salvia officinalis were extracted with hot water [3]. Extracts were evaluated for their total phenolic content by an adaptation of the Folin-Ciocalteu method and further analysed by high performance liquid chromatography associated with electrospray mass spectrometry (HPLC-DAD-ESI-MSn) in the negative ion mode [4], in order to identify their individual phenolic constituents. The aqueous extracts of S. farinacea, S. mexico, S. officinalis and S. greggii contained, respectively, 106±13, 159±38, 175±46 and 136±1 μg GAE/mg of total phenolics. These four species were characterized by a clear prevalence of caffeic acid derivatives, in particular of rosmarinic acid (MW 360), that is generally the most abundant phenolic compound in Salvia species [2,3]. In addition, S. mexico and S. officinalis contained moderate amounts of salvianolic acid B (MW 718). Among these two, S. mexico was richer in O-caffeoylquinic acid (MW 354), while the latter presented high amounts of salvianolic acid K (MW 556) and moderate amounts of its structural isomer. All the extracts were enriched in flavones: S. farinacea and S. officinalis contained high amounts of luteolin-O-glucuronide while S. mexico contained luteolin-C-glucoside with respective characteristic mass spectrometry fragmentation pattern m/z at 461→285 and m/z at 447→357, 327. Similarly, S. greggii extract presented high content of luteolin-7-O-glucoside ([M-H]− at m/z 447→ 285) and luteolin-C-glucoside and moderate quantities of apigenin-C-hexoside ([M-H]− at m/z 431→341, 311). Further studies are being undertaken in order to understand the contribution of these phenolic constituents in the biological activities of Salvia plants.
