Spectrophotometric determination of total protein is used in several areas such as clinical analysis, food science and technology, biochemistry, protein chemistry, physiology. Five spectrophotometric methods are mostly used: biuret, Lowry, Bradford, Smith and UV absorption. In this review a general overview of these methods is presented (interferences, applications); other methodologies are also discussed.

Propolis is mostly used as hydroalcoholic extract. Recently there has been a growing number of patents dealing with new solvents for preparing propolis extracts. This study aimed to prepare edible oil propolis extracts and compare their chemical composition and biological activity with ethanolic propolis extracts. ESI-MS and spectrophotometric methods were used for qualitative and quantitative analyses, respectively. Antibacterial activity was evaluated by diffusion in agar. Cytotoxicity was tested by MTT assay using tumor cell lines. The oil is able to extract bioactive compounds from propolis. Further studies are needed to improve extraction efficiency and to characterize the active components.

The main goal of this paper was to study the degradation of synthetic dyes using photoelectrocatalytic properties of particulate films of TiO2 supported on plates of titanium and stimulated by UV-Vis radiation. The dyes decolorizations were measured using spectrophotometric methods to verify which the conditions on Ti/TiO2 electrode was the best for the photoelectrodegradation of them. The results showed that decolorization rates were higher than 90% during a period of 270 min. FT-IR spectroscopy showed that intermediate substances were formed after the decolorization and N=N group/aromatic structures were preserved independently of the specific structure of the dyes.

Spectrophotometric methods of zero order, first and second derived order had been developed for olanzapine determination in tablets using ethanol and isopropanol as solvent. The two solvents revealed to be adequate. For the three methods the calibration curve coefficient of correlation had been greater than 0.9998 with limit of detection varying from 0.068 to 0.190 mg L-1, in ethanol, and 0.026 to 0.205 mg L-1, in isopropanol. The inter-day precision was inferior to 1.1 and 1.9 mg L-1 for ethanol and isopropanol, respectively. The average recoveries varied from 98 to 101%, in ethanol and 99 to 103% in isopropanol.

Two simple and efficient procedures have been developed for the rapid simultaneous determination of compounds with mutual spectral interference (rifampicin (RIF) and isoniazid (INH)). The first method was based on the UV–Vis spectral signal (190–600 nm) of synthetic RIF and INH aqueous solutions, whereas the second method involved the visible spectral signal registered between 350 and 800 nm after the reaction of INH with a Cu2+/neocuproine complex. Both multivariate spectrophotometric methods show excellent prevision capacity, providing results that are statistically equivalent with those provided by the standard chromatographic procedure. The methods were validated according to criteria established by ANVISA, showing precision, accuracy and robustness compatible with the requirements for new analytical methods, additionally allowing the reduction of waste generation.

Two sensitive spectrophotometric methods are described for the determination of simvastatin (SMT) in bulk drug and in tablets. The methods are based on the oxidation of SMT by a measured excess of cerium (IV) in acid medium followed by determination of unreacted oxidant by two different reaction schemes. In one procedure (method A), the residual cerium (IV) is reacted with a fixed concentration of ferroin and the increase in absorbance is measured at 510 nm. The second approach (method B) involves the reduction of the unreacted cerium (IV) with a fixed quantity of iron (II), and the resulting iron (III) is complexed with thiocyanate and the absorbance measured at 470 nm. In both methods, the amount of cerium (IV) reacted corresponds to SMT concentration. The experimental conditions for both methods were optimized. In method A, the absorbance is found to increase linearly with SMT concentration (r = 0.9995) whereas in method B, the same decreased (r = -0.9943). The systems obey Beer's law for 0.6-7.5 and 0.5-5.0 µg mL-1 for method A and method B, respectively. The calculated molar absorptivity values are 2.7 X 10(4) and 1.06 X 10(5) Lmol-1 cm-1, respectively; and the corresponding sandel sensitivity values are 0.0153 and 0.0039µg cm-2, respectively. The limit of detection (LOD) and quantification (LOQ) are reported for both methods. Intra-day and inter-day precision, and accuracy of the methods were established as per the current ICH guidelines. The methods were successfully applied to the determination of SMT in tablets and the results were statistically compared with those of the reference method by applying the Student's t-test and F-test. No interference was observed from the common excipients added to tablets. The accuracy and validity of the methods were further ascertained by performing recovery experiments via standard addition procedure.

A flow injection method for the quantitative analysis of vancomycin hydrochloride, C66H75Cl2N9O24.HCl (HVCM), based on the reaction with copper (II) ions, is presented. HVCM forms a lilac-blue complex with copper ions at pH≅4.5 in aqueous solutions, with maximum absorption at 555 nm. The detection limit was estimated to be about 8.5×10-5 mol L-1; the quantitation limit is about 2.5×10-4 mol L-1 and about 30 determinations can be performed in an hour. The accuracy of the method was tested through recovery procedures in presence of four different excipients, in the proportion 1:1 w/w. The results were compared with those obtained with the batch spectrophotometric and with the HPLC methods. Statistical comparison was done using the Student's procedure. Complete agreement was found at a 0.95 significance level between the proposed flow injection and the batch spectrophotometric methods, which present similar precision (RSD: 2.1 % vs. 1.9%).

The synthesis and structural characterization of a novel oxoperoxovanadium(v) complex [VO(O-2)(PAH)-(phen)] containing the ligands 2-phenylacetohydroxamic acid (PAHH) and 1,10-phenanthroline (phen) has been accomplished. The oxoperoxovanadium(v) complex was found to mimic both vanadate-dependent haloperoxidase (VHPO) activity as well as nuclease activity through effective interaction with DNA. The complex is the first example of a structurally characterized stable oxoperoxovanadium(v) complex with a coordinated bi-dentate hydroximate moiety (-CONHO-) from 2-phenylacetohydroximate (PAH). The oxoperoxovanadium(v) complex has been used as catalyst for the peroxidative bromination reaction of some unsaturated alcohols (e.g. 4-pentene-1-ol, 1-octene-3-ol and 9-decene-1-ol) in the presence of H2O2 and KBr. The catalytic products have been characterized by GC-MS analysis and spectrophotometric methods. The DNA binding of this complex has been established with CT DNA whereas the DNA cleavage was demonstrated with plasmid DNA. The interactions of the complex with DNA have been monitored by electronic absorption and fluorescence emission spectroscopy. Viscometric measurements suggest that the compound is a DNA intercalator. The nuclease activity of this complex was confirmed by gel electrophoresis studies.

The feasibility of the photobleaching of a textile azo dye, reactive orange 16 (C.I. 17757), in aqueous solution using titanium dioxide thin-film electrodes prepared by the sol-gel method was investigated. The best conditions for maximum photoelectrocatalytic degradation were found to be pH > 10 for Na2SO4 medium and pH < 6 for NaCl. In both situations, an applied potential of +1.0 V and low dye concentration are recommended, when 100% of color removal is obtained after 20 min of photoelectrocatalysis. The effects of side reaction pathway on the degradation rate of dye in sulfate and chloride medium were presented and the best performance are optimized to situations closed to that verified in the textile effluent. The influence of variables as applied potential, pH, supporting electrolyte and dye concentration on the kinetics of photoelectrochemical degradation also were investigated. Oxalic acid is identified by HPLC and UV-Vis spectrophotometric methods as the main degradation product generated after 180 min of photoelectrocatalysis of 4 x 10(-5) mol l(-1) dye in sodium sulphate pH 12 and NaCl pH 4.0 and a maximum reduction of 56 and 62% TOC was obtained, respectively. A new method is described for the rapid, sensitive, virtually interference-free, and selective quantitation of cyanogenic glycosides in aqueous extracts using membrane introduction mass spectrometry (MIMS). Selective monitoring, by either conventional MIMS or cryotrap-MIMS, not of HCN but of the co-released ketones (acetone and butan-2-one), when performed for both the crude cassava extracts and the linamarase-NaOH-hydrolyzed extracts, is found to offer an advantageous alternative to classic spectrophotometric methods based on HCN analysis for the selective quantitation of the two cyanogenic glycosides linamarin and lotaustralin expressed as both the free HCN content and the total cyanogenic potential (total HCN).
