In 221 BC, the battle-hardened warriors of the Qin state, the western frontier state and the most aggressive of the Warring States, subjugated the last of its rival states, thus establishing the Qin dynasty, with itscapital in Xianyang, near the modern Xi’an. The Qin dynasty is customarily regarded by Chinese and Western scholars as the beginning of a new age – the Chinese empire – that lasted until 1911 AD. The dynasty lasted only fifteen years. This study examines the main policies of the Qin dynasty and seeks to address the question what brought the quick downfall of the Qin rule. This paper takes the cultural and political contexts carefully into consideration, and argues that the Qin annexation of its rival states might be better understood as an end of an old era as much as a beginning of a new epoch.

Bei kollinearer Laserspektroskopieverlangt die Bestimmung der Kernladungsradien leichterIsotope aus der Isotopeverschiebung nach einer sehr präzisenKenntnis der Ionenstrahlenergie. Zu diesem Zweck wurde eineneue, auf kollinearer Laserspektroskopie basierende Methodezur Strahlenergiemessung entwickelt und erfolgreich in denExperimenten zu Neon eingesetzt. Die experimentellenErgebnisse werden mit theoretischen Rechnungen im Rahmen desSchalenmodells und von kollektiven Kernmodellen verglichen.

Ein neu entwickeltes globales Atmosphärenchemie- und Zirkulationsmodell (ECHAM5/MESSy1) wurde verwendet um die Chemie und den Transport von Ozonvorläufersubstanzen zu untersuchen, mit dem Schwerpunkt auf Nichtmethankohlenwasserstoffen. Zu diesem Zweck wurde das Modell durch den Vergleich der Ergebnisse mit Messungen verschiedenen Ursprungs umfangreich evaluiert. Die Analyse zeigt, daß das Modell die Verteilung von Ozon realistisch vorhersagt, und zwar sowohl die Menge als auch den Jahresgang. An der Tropopause gibt das Modell den Austausch zwischen Stratosphäre und Troposphäre ohne vorgeschriebene Flüsse oder Konzentrationen richtig wieder. Das Modell simuliert die Ozonvorläufersubstanzen mit verschiedener Qualität im Vergleich zu den Messungen. Obwohl die Alkane vom Modell gut wiedergeben werden, ergibt sich einige Abweichungen für die Alkene. Von den oxidierten Substanzen wird Formaldehyd (HCHO) richtig wiedergegeben, während die Korrelationen zwischen Beobachtungen und Modellergebnissen für Methanol (CH3OH) und Aceton (CH3COCH3) weitaus schlechter ausfallen. Um die Qualität des Modells im Bezug auf oxidierte Substanzen zu verbessern, wurden einige Sensitivitätsstudien durchgeführt. Diese Substanzen werden durch Emissionen/Deposition von/in den Ozean beeinflußt, und die Kenntnis über den Gasaustausch mit dem Ozean ist mit großen Unsicherheiten behaftet. Um die Ergebnisse des Modells ECHAM5/MESSy1 zu verbessern wurde das neue Submodell AIRSEA entwickelt und in die MESSy-Struktur integriert. Dieses Submodell berücksichtigt den Gasaustausch zwischen Ozean und Atmosphäre einschließlich der oxidierten Substanzen. AIRSEA, welches Informationen über die Flüssigphasenkonzentration des Gases im Oberflächenwasser des Ozeans benötigt wurde ausgiebig getestet. Die Anwendung des neuen Submodells verbessert geringfügig die Modellergebnisse für Aceton und Methanol, obwohl die Verwendung einer vorgeschriebenen Flüssigphasenkonzentration stark den Erfolg der Methode einschränkt, da Meßergebnisse nicht in ausreichendem Maße zu Verfügung stehen. Diese Arbeit vermittelt neue Einsichten über organische Substanzen. Sie stellt die Wichtigkeit der Kopplung zwischen Ozean und Atmosphäre für die Budgets vieler Gase heraus.

The OPERA experiment, designed to perform the first observation of νμ→ντ oscillations in appearance mode through the detection of the τ leptons produced in ντ charged current interactions, has collected data from 2008 to 2012. In the present paper, the procedure developed to detect τ particle decays, occurring over distances of the order of 1 mm from the neutrino interaction point, is described in detail. The results of its application to the search for charmed hadrons are then presented as a validation of the methods for ντ appearance detection.

We are studying endoplasmic reticulum–associated degradation (ERAD) with the use of a truncated variant of the type I ER transmembrane glycoprotein ribophorin I (RI). The mutant protein, RI332, containing only the N-terminal 332 amino acids of the luminal domain of RI, has been shown to interact with calnexin and to be a substrate for the ubiquitin-proteasome pathway. When RI332 was expressed in HeLa cells, it was degraded with biphasic kinetics; an initial, slow phase of ∼45 min was followed by a second phase of threefold accelerated degradation. On the other hand, the kinetics of degradation of a form of RI332 in which the single used N-glycosylation consensus site had been removed (RI332-Thr) was monophasic and rapid, implying a role of the N-linked glycan in the first proteolytic phase. RI332 degradation was enhanced when the binding of glycoproteins to calnexin was prevented. Moreover, the truncated glycoprotein interacted with calnexin preferentially during the first proteolytic phase, which strongly suggests that binding of RI332 to the lectin-like protein may result in the slow, initial phase of degradation. Additionally, mannose trimming appears to be required for efficient proteolysis of RI332. After treatment of cells with the inhibitor of N-glycosylation, tunicamycin, destruction of the truncated RI variants was severely inhibited; likewise, in cells preincubated with the calcium ionophore A23187, both RI332 and RI332-Thr were stabilized, despite the presence or absence of the N-linked glycan. On the other hand, both drugs are known to trigger the unfolded protein response (UPR), resulting in the induction of BiP and other ER-resident proteins. Indeed, only in drug-treated cells could an interaction between BiP and RI332 and RI332-Thr be detected. Induction of BiP was also evident after overexpression of murine Ire1, an ER transmembrane kinase known to play a central role in the UPR pathway; at the same time, stabilization of RI332 was observed. Together, these results suggest that binding of the substrate proteins to UPR-induced chaperones affects their half lives.

Memory is a hallmark of immunity. Memory carried by antibodies is largely responsible for protection against reinfection with most known acutely lethal infectious agents and is the basis for most clinically successful vaccines. However, the nature of long-term B cell and antibody memory is still unclear. B cell memory was studied here after infection of mice with the rabies-like cytopathic vesicular stomatitis virus, the noncytopathic lymphocytic choriomeningitis virus (Armstrong and WE), and after immunization with various inert viral antigens inducing naive B cells to differentiate either to plasma cells or memory B cells in germinal centers of secondary lymphoid organs. The results show that in contrast to very low background levels against internal viral antigens, no significant neutralizing antibody memory was observed in the absence of antigen and suggest that memory B cells (i) are long-lived in the absence of antigen, nondividing, and relatively resistant to irradiation, and (ii) must be stimulated by antigen to differentiate to short-lived antibody-secreting plasma cells, a process that is also efficient in the bone marrow and always depends on radiosensitive, specific T help. Therefore, for vaccines to induce long-term protective antibody titers, they need to repeatedly provide, or continuously maintain, antigen in minimal quantities over a prolonged time period in secondary lymphoid organs or the bone marrow for sufficient numbers of long-lived memory B cells to mature to short-lived plasma cells.

Auxin is transported across the plasma membrane of plant cells by diffusion and by two carriers operating in opposite directions, the influx and efflux carriers. Both carriers most likely play an important role in controlling auxin concentration and distribution in plants but little is known regarding their regulation. We describe the influence of modifications of the transmembrane pH gradient and the effect of agents interfering with protein synthesis, protein traffic, and protein phosphorylation on the activity of the auxin carriers in suspension-cultured tobacco (Nicotiana tabacum L.) cells. Carrier-mediated influx and efflux were monitored independently by measuring the accumulation of [14C]2,4-dichlorophenoxyacetic acid and [3H]naphthylacetic acid, respectively. The activity of the influx carrier decreased on increasing external pH and on decreasing internal pH, whereas that of the efflux carrier was only impaired on internal acidification. The efflux carrier activity was inhibited by cycloheximide, brefeldin A, and the protein kinase inhibitors staurosporine and K252a, as shown by the increased capability of treated cells to accumulate [3H]naphthylacetic acid. Kinetics and reversibility of the effect of brefeldin A were consistent with one or several components of the efflux system being turned over at the plasma membrane with a half-time of less than 10 min. Inhibition of efflux by protein kinase inhibitors suggested that protein phosphorylation was essential to sustain the activity of the efflux carrier. On the contrary, the pharmacological agents used in this study failed to inhibit [14C]2,4-dichlorophenoxyacetic acid accumulation, suggesting that rapidly turned-over proteins or proteins activated by phosphorylation are not essential to carrier-mediated auxin influx. Our data support the idea that the efflux carrier in plants constitutes a complex system regulated at multiple levels, in marked contrast with the influx carrier. Physiological implications of the kinetic features of this regulation are discussed.

Defining a consumer interest as a long-term trend or short-lived fad has significant implicatiosn for restauranteurs' management decisions. The terms "trend" and "fad" can be operationally defined for the food service industry. The authors examine today's popular cross-cultural cuisine to determine its trend or fad status and discuss the catalysts that promoted or hindered its trend/fad status, as well as implications for the food service industry.

Mass measurements of exotic nuclei is a fast, developing field which is essential for basic nuclear physics and a wide range of applications. The method of storage ring mass spectrometry has many advantages: (1) a large amount of nuclides can be simultaneously measured; (2) very short-lived (T-1/2 greater than or similar to 50 mu s) and very rare species (yields down to single ions) can be accessed; (3) nuclides in several atomic charge states can be investigated, (4) half-life measurements can be performed with time-resolved mass spectrometry. In this contribution we concentrate on some recent achievements and future perspectives of the storage ring mass spectrometry.

The dissertation presented here deals with high-precision Penning trap mass spectrometry on short-lived radionuclides. Owed to the ability of revealing all nucleonic interactions, mass measurements far off the line of ß-stability are expected to bring new insight to the current knowledge of nuclear properties and serve to test the predictive power of mass models and formulas. In nuclear astrophysics, atomic masses are fundamental parameters for the understanding of the synthesis of nuclei in the stellar environments. This thesis presents ten mass values of radionuclides around A = 90 interspersed in the predicted rp-process pathway. Six of them have been experimentally determined for the first time. The measurements have been carried out at the Penning-trap mass spectrometer SHIPTRAP using the destructive time-of-fligh ion-cyclotron-resonance (TOF-ICR) detection technique. Given the limited performance of the TOF-ICR detection when trying to investigate heavy/superheavy species with small production cross sections (σ< 1 μb), a new detection system is found to be necessary. Thus, the second part of this thesis deals with the commissioning of a cryogenic double-Penning trap system for the application of a highly-sensitive, narrow-band Fourier-transform ion-cyclotron-resonance (FT-ICR) detection technique. With the non-destructive FT-ICR detection method a single singly-charged trapped ion will provide the required information to determine its mass. First off-line tests of a new detector system based on a channeltron with an attached conversion dynode, of a cryogenic pumping barrier, to guarantee ultra-high vacuum conditions during mass determination, and of the detection electronics for the required single-ion sensitivity are reported.
