Em duas alíquotas de 208 amostras de fezes foram feitos exames pelo método de Kato-Katz, sendo uma após o emprego de 0,2mg de azida sódica para 200mg e em outra sem o referido conservador. Os resultados percentuais com e sem conservador foram, respectivamente, para os Ancilostomídeos 12,5 e 25,9, para Ascaris lumbricoides 71,6 e 72,5, para Schistosoma mansoni 7,6 e 17,7 e para Trichuris trichiura 86 e 85. A contagem dos ovos com e sem o conservador foi, respectivamente, 264 e 539 para os Ancilostomídeos, 13816 e 33751 para A. lumbricoides, 55,5 e 63,5 para S. mansoni e 1345 e 2068 para T. trichiura. Os autores não confirmaram a vantagem do uso de azida sódica para estudo em áreas endêmicas. Sugerem que o ressecamento das fezes e a baixa intensidade das infecções possam explicar os resultados desfavoráveis do presente ensaio clínico.

O diagnóstico parasitológico deve ser realizado de maneira apropriada, com maior sensibilidade e especificidade para a detecção dos parasitas intestinais, uma vez que dele dependerá o tratamento específico. Foi desenvolvido um estudo comparativo para avaliar a concordância entre os métodos Kato-Katz e coprotest® na detecção de helmintos em 332 indivíduos do município de Pedro de Toledo. Destacou-se uma diferença significativa para Trichuris trichiura, 16,2% no Kato-Katz e 7,5% no coprotest®. Devido a essa diferença compararam-se amostras positivas e negativas do método de coprotest® com número de ovos por grama de fezes (opg) obtido pelo método de Kato-Katz. Quando o método de coprotest® era negativo, contaram-se 65 opg de Trichuris trichiura pelo Kato-Katz e quando o coprotest® era positivo, esse número foi maior, 199 opg. O coprotest® mostrou-se inferior ao Kato-Katz nas infecções de baixa carga parasitária.

This study compared the efficiency of Kato-Katz thick smear and thick smear techniques for the diagnosis of intestinal helminths. The sensitivity of the thick smear technique was higher than that of the Kato-Katz method for the diagnosis of all helminths except Schistosoma mansoni.

INTRODUCTION: The laboratory diagnosis of schistosomiasis is based mainly on the detection of parasite eggs in stool samples through the Kato-Katz (KK) technique, reading one slide by test. However, a widely known limitation of parasitological methods is reduced sensitivity, particularly in low endemic areas. METHODS: To increase sensitivity, we conducted further slide readings from the same stool sample using the parasitological method associated with a serological test. We used the KK method (three slides) and the IgG anti-Schistosoma mansoni-enzyme-linked immunosorbent assay (ELISA) technique to diagnose schistosomiasis in low endemic areas in the Brazilian State of Ceará. Fecal samples and sera from 250 individuals were analyzed. RESULTS: Sixteen percent and 47.2% of samples were positive in parasitological tests and serological tests, respectively. Parasitological methods showed that 32 (80%) individuals tested positive on the first slide, 6 (15%) on the second slide, and 2 (5%) on the third. The performance of the ELISA test in the diagnosis, using the KK method as diagnostic reference, showed a negative predictive value of 100%, with specificity and positive predictive values of 62.8% and 33.9%, respectively. CONCLUSIONS: In this study, the increase from one to three slides analyzed per sample using the KK technique was shown to be a useful procedure for increasing the diagnostic sensitivity of this technique.

We have developed an in vitro model of granuloma formation for the purpose of studying the immunological components of delayed type hypersensitivity granuloma formation in patients infected with Schistosoma mansoni. Our data show that 1) granulomatous hypersensitivity can be studied by examining the cellular reactivity manifested as multiple cell layers surrounding the antigen conjugated beads; 2) this reactivity is a CD4 cell dependent, macrophage dependent, B cell independent response and 3) the in vitro granuloma response is antigenically specific for parasite egg antigens. Studies designed to investigate the immune regulation of granulomatous hypersensitivity using purified populations of either CD4 or CD8 T cells have demonstrated the complexity of cellular interactions in the suppression of granulomatous hypersensitivity. The anti-S. mansoni egg immune responses of individual patients with chronic intestinal schistosomiasis can be classified either as soluble egg antigen (SEA) hypersensitive with maximal granulomatous hypersensitivity or SEA suppressive with activation of the T cell suppressor pathway with effective SEA granuloma modulation. Our data suggest that T cell network interactions are active in the generation of effective granuloma modulation in chronic intestinal schistosomiasis patients.

The association of Lutz/Kato-Katz and Lutz/Bermann-Moraes (adapted techniques was used to improve better results that ranged from 0.4 to 11 times in the search of eggs of Ascaris lumbricoides, Schistosoma mansoni, Trichiuris trichiura, Taenia sp. and larvae of Strongyloides stercoralis.

Existen en el medio teatral varias técnicas de entrenamiento vocal para el actor de teatro. ¿Pueden usarse siempre las mismas técnicas indiferentemente del sujeto? ¿Es posible obtener de un ejercicio vocal siempre los mismos resultados o beneficios? Al analizar las rutinas básicas utilizadas en la preparación vocal para teatro desde las patologías funcionales de la voz, es posible afirmar que no todos los sujetos están en disposición física de cumplir con determinadas exigencias particulares. Este trabajo analiza las diferentes patologías funcionales de la voz partiendo de algunos ejercicios en los entrenamientos de Linklater y Houseman enfocados desde el manejo de patologías.

The high sensitivity and the possibility of automation of the enzyme-linked-immunosorbent-assay (ELISA) has indicated this technique as one of the most useful serological test for epidemiological studies. In the present study, an ELISA for detection of IgG antibodies against adult worm antigens (IgG-ELISA) was investigated for epidemiological purposes, in a rural area of the municipality of Itariri (São Paulo, Brazil). Blood on filter paper (1,180 samples) from about 650 schoolchildren were submitted to ELISA and the data compared to the results of the parasitological method of Kato-Katz and also to the IgM-IFT (immunofluorescence test for IgM antibodies to gut associated antigens). The prevalence rates respectively of 8.5%, 43.0%, and 56.2% by the Kato-Katz, IgG-ELISA, and IgM-IFT methods suggest the poor sensitivity of the parasitological method for detection of Schistosoma mansoni eggs in individuals with low worm burden, situation commonly observed in low endemic areas. These results can partially explain the poor degree of agreement between the IgG-ELISA and the Kato-Katz, as suggested by the Kappa index of 0.170. Otherwise, the Kappa index of 0.675 showed substantial agreement between the two serological tests. Some discrepancy of results between the two serological techniques must be better investigated.

This study compares the diagnostic accuracy of the TF-Test® (TFT) for human parasitosis with results obtained using the traditional Kato-Katz (KK), Hoffman-Pons-Janer (HPJ), Willis and Baermann-Moraes (BM) techniques. Overall, four stool samples were taken from each individual; three alternate-day TFT stool samples and another sample that was collected in a universal container. Stool samples were taken from 331 inhabitants of the community of Quilombola Santa Cruz. The gold standard (GS) for protozoa detection was defined as the combined results for TFT, HPJ and Willis coproscopic techniques; for helminth detection, GS was defined as the combined results for all five coproscopic techniques (TFT, KK, HPJ, Willis and BM). The positivity rate of each method was compared using the McNemar test. While the TFT exhibited similar positivity rates to the GS for Entamoeba histolytica/dispar (82.4%) and Giardia duodenalis (90%), HPJ and Willis techniques exhibited significantly lower positivity rates for these protozoa. All tests exhibited significantly lower positivity rates compared with GS for the diagnosis of helminths. The KK technique had the highest positivity rate for diagnosing Schistosoma mansoni (74.6%), while the TFT had the highest positivity rates for Ascaris lumbricoides (58.1%) and hookworm (75%); HPJ technique had the highest positivity rate for Strongyloides stercoralis (50%). Although a combination of tests is the most accurate method for the diagnosis of enteral parasites, the TFT reliably estimates the prevalence of protozoa and selected helminths, such as A. lumbricoides and hookworm. Further studies are needed to evaluate the detection accuracy of the TFT in samples with varying numbers of parasites.
