People are alarmingly susceptible to manipulations that change both their expectations and experience of the value of goods. Recent studies in behavioral economics suggest such variability reflects more than mere caprice. People commonly judge options and prices in relative terms, rather than absolutely, and display strong sensitivity to exemplar and price anchors. We propose that these findings elucidate important principles about reward processing in the brain. In particular, relative valuation may be a natural consequence of adaptive coding of neuronal firing to optimise sensitivity across large ranges of value. Furthermore, the initial apparent arbitrariness of value may reflect the brains' attempts to optimally integrate diverse sources of value-relevant information in the face of perceived uncertainty. Recent findings in neuroscience support both accounts, and implicate regions in the orbitofrontal cortex, striatum, and ventromedial prefrontal cortex in the construction of value.

Chinese mitten crab Eriocheir sinensis is one of the most important aquaculture crustacean species in China. A cDNA library was constructed from hemocytes of E. sinensis challenged with the mixture of Listonella anguillarum and Staphylococcus aureus, and randomly sequenced to collect genomic information and identify genes involved in immune defense response. Single-pass 5' sequencing of 10368 clones yielded 7535 high quality ESTs (Expressed Sequence Tags) and these ESTs were assembled into 2943 unigenes. BLAST analysis revealed that 1706 unigenes (58.0% of the total) or 4593 ESTs (61.0% of the total) were novel genes that had no significant matches to any protein sequences in the public databases. The rest 1237 unigenes; (42.0% of the total) were closely matched to the known genes or sequences deposited in public databases, which could be classed into 20 or 23 classifications according to "molecular function" or "biological process" respectively based on the Gene Ontology (GO). And 221 unigenes (7.5% of all 2943 unigenes, 17.9% of matched unigenes) or 969 ESTs (12.9% of all 7535 ESTs, 32.9% of matched ESTs) were identified to be immune genes. The relative higher proportion of immune-related genes in the present cDNA library than that in the normal library of E. sinensis and other crustaceans libraries, and the differences and changes in percentage and quantity of some key immune-related genes especially the immune inducible genes between two E. sinensis cDNA libraries may derive from the bacteria challenge to the Chinese mitten crab. The results provided a well-characterized EST resource for the genomics community, gene discovery especially for the identification of host-defense genes and pathways in crabs as well as other crustaceans. The esrB gene of Edwardsiella tarda, which encodes a regulator protein of the type III secretion system, was mutated by the unmarked deletion method and reintroduced by allelic exchange into the chromosome of E. tarda LSE40 by means of the suicide vector pRE 112. The LSE40 esrB mutant was highly attenuated when inoculated intraperitoneally into turbot Scophthamus maximus L., showing a 50% lethal dose of 10(8.1) cfu/fish. The esrB mutants were not recoverable from the internal organs at 14 days post-inoculation. Vaccination with a single dose of 10(5)-10(7) cfu/fish of the esrB mutant elicited significant protection against the wildtype strain of E. tarda LSE40 (relative percentage survival > 50%). Streptococcus iniae is a severe aquaculture pathogen that can also infect humans and animal. A putative secretory antigen, Slat 0, was identified from a pathogenic S. iniae strain by in vivo-induced antigen technology. Using turbot as an animal model, the immunoprotective effect of Sia10 was examined as a DNA vaccine in the form of plasmid pSia10, which expresses sia10 under the cytomegalovirus immediate-early promoter. In fish vaccinated with pSia10, transcription of sia10 was detected in muscle, liver, spleen, and kidney at 7, 14, 21, 28, 35, 42, and 49 days post-vaccination. In addition, production of Sia10 protein was also detected in the muscle tissues of pSia10-vaccinated fish. Fish vaccinated with pSia10 exhibited a relative percent survival (RPS) of 73.9% and 92.3%, respectively, when challenged with high and low doses (producing a cumulative mortality of 92% and 52%, respectively, in the control groups) of S. iniae. Immunological and transcriptional analyses showed that vaccination with pSia10(i) induced much stronger chemiluminescence response and significantly higher levels of nitric oxide production and acid phosphatase activity in head kidney macrophages; (ii) caused the production of specific serum antibodies, which afforded apparent immunoprotection when transferred passively into naive fish; and (iii) upregulated the expression of the genes encoding proteins that are possibly involved in both innate and adaptive immune responses. Taken together, these results indicated that pSia10 is an effective vaccine candidate and may be used in the control of S. iniae infection in aquaculture. (C) 2010 Elsevier Ltd. All rights reserved.

The embryological features of three species of Swertia (s.l.) - S. erythrosticta, S. franchetiana, and S. tetraptera were characterized, and the observations were used, together with previously gathered data on other species, to evaluate a recently proposed polyphyly, based on molecular data, of Swertia s.l. Comparisons of species within the genus showed that they have diversified embryologically, and there are significant between-species differences. Notable features that vary between species include the number of cell layers that form the anther locule wall, the construction of the wall of the mature anther, tapetum origin, the cell number in mature pollen grains, the structure of the fused margins of the two carpels, the ovule numbers in placental cross-sections, the shape of the mature embryo sac, the degree of ovule curvature, antipodal variation and the presence of a hypostase, and seed appendages. They share characters that are widely distributed in the tribe Gentianeae, such as a dicotyledonous type of anther wall formation, a glandular tapetum with uninucleate cells, simultaneous cytokinesis following the meiosis of the microsporocytes, tetrahedral microspore tetrads, superior, bicarpellary and unilocular ovaries, unitegmic and tenuinucellar ovules, Polygonum-type megagametophytes, progamous fertilization, nuclear endosperm, and Solanad-type embryogeny. The presence of variation in embryological characters amongst the species of Swertia s.l. strongly supports the view that Swertia s.l. is not a monophyletic group. Frasera is better separated from Swertia s.l. as an independent genus, and is only distantly related to Swertia s. s. judging from the numerous differences in embryology. Concrete used for underwater repair is often proportioned to spread readily into place and self-consolidate, and to develop high resistance to segregation and water dilution. An investigation was carried out to determine the effect of the dosage of antiwashout admixture, water-cementitious materials ratio (w/cm), and binder composition on the relative residual strength of highly flowable underwater concrete. Two types of antiwashout admixtures were used: a powdered welan gum at 0.07 and 0.15% by mass of binder, and a liquid-based cellulosic admixture employed at a high dosage of 1 to 1.65 L/100 kg of cementitious materials. The w/cms were set at 0.41 and 0.47 to secure adequate performance of underwater concrete for construction and repair. Four binder compositions were used: a Canadian Type 10 cement; a cement with 10% silica fume replacement; a cement with 50% replacement of granulated blast-furnace slag; and a ternary binder containing 6% silica fume and 20% Class F fly ash. Test results indicate that for a given washout mass loss and slump flow consistency, greater relative residual strength can be secured when the dosage of antiwashout admixture is increased, the w/cm is reduced, and a binary binder with 10% silica fume substitution or the ternary binder are employed. Such mixtures can develop relative residual compressive strengths of 85 and 80%, compared to mixtures cast in air, when the value of washout loss is limited to 4 and 6% for mixtures with slump flow values of 450 and 550 mm, respectively.

This research assesses the effectiveness of current MSc. Construction Project Management programmes within the UK and Ireland. A review of published prospectuses is used to create questionnaires for universities, graduates and employers. Responses provide an insight into programme creation and their relative success in addressing the needs of industry and in achieving other educational objectives. Since the majority of learning institutions have attained professional accreditation, it is useful to review these awards and to assess their potential value to both graduates and industry alike. Interviews are conducted with representatives from the main professional accrediting bodies to understand their procedures and rigour in enforcing standards of education and training. The results show that project management education could be further enhanced by the inclusion of more practical learning and that current programmes place greater emphasis on hard skills at the expense of the softer human skills. There is clearly a need for a closer working relationship between academics and practitioners to tackle the perceived gap between theoretical learning and construction practice. Learning institutions can use the findings to improve their programmes and address the education deficiencies identified by the industry, by the professional institutions and by graduates.

The time period bridging the years 2007 to 2012 will be remembered as one characterised by dramatic changes in the Irish and UK construction industries. Construction companies witnessed unprecedented changes in the environment, namely the coincidence of a sharp economic downturn, the significant decline of public works, a reduction in lending, increased competition, and structural changes in the marketplace. Nevertheless, little has been documented on what response strategies construction companies adopt as a result of an economic recession. Based on four exploratory case studies, a taxonomy framework of the response strategies adopted by Irish and UK construction companies during the 2007 economic recession was developed relative to Porter’s (1980) generic strategies of cost leadership, differentiation, and focus. Porter’s model (1980) is a well known theoretical framework among business strategists and industrial economists worldwide. The analysis provides strong support for the adoption of cost leadership strategies as a means to surviving the 2007 economic recession. The case studies further suggest that cost control initiatives are one of the most important attributes in companies’ responses to the 2007 recession. The findings provide valuable assistance for construction contractors in developing effective strategies and thus reducing business failures during recessionary periods.

A flow injection analysis (FIA) system comprising a cysteine selective electrode as detection system was developed for determination of this amino acid in pharmaceuticals. Several electrodes were constructed for this purpose, having PVC membranes with different ionic exchangers and mediator solvents. Better working characteristics were attained with membranes comprising o-nitrophenyl octyl ether as mediator solvent and a tetraphenylborate based ionic-sensor. Injection of 500 µL standard solutions into an ionic strength adjuster carrier (3x10-3 M) of barium chloride flowing at 2.4mL min-1, showed linearity ranges from 5.0x10-5 to 5.0x10-3 M, with slopes of 76.4±0.6mV decade-1 and R2>0.9935. Slope decreased significantly under the requirement of a pH adjustment, selected at 4.5. Interference of several compounds (sodium, potassium, magnesium, barium, glucose, fructose, and sucrose) was estimated by potentiometric selectivity coefficients and considered negligible. Analysis of real samples were performed and considered accurate, with a relative error to an independent method of +2.7%.

The relative ease to concentrate and purify adenoviruses, their well characterized mid-sized genome, and the ability to delete non-essential regions from their genome to accommodate foreign gene, made adenoviruses a suitable candidate for the construction of vectors. The use of adenoviral vectors in gene therapy, vaccination, and as a general vector system for expressing foreign genes have been documented for some time. In this study, the objective was to rescue a BAV3 E1 or E3 recombinant vector carrying the kanamycin resistant gene, a dominant selectable marker with useful applications in studying vectored gene expression in mammalian cells. To accomplish the objective of this study, more information about BAV3 DNA sequences was required in order to make the manipulation of the virus genome accessible. Therefore, sequencing of the BAV3 genome from 1 1 .7% to 30.8% was carried out. Analysis of the determined sequences revealed the primary structure of important viral gene products coded by E2 including BAV3 DNA pol and precursor to terminal protein. Comparative analysis of these proteins with their counterparts from human and non human adenoviruses revealed important insights as to the evolutionary lineage of BAV3. In order to insert the kanamycin resistance gene in either E1 or E3, it was necessary to delete BAV3 sequences to accommodate the foreign gene so as not to exceed the limit of the packaging capacity of the virus. To construct a recombinant BAV3 in which a foreign gene was inserted in the deleted E1 region, an E1 shuttle vector was constructed. This involved the deletion from the viral sequences a region between 1.3% to 9% and inserting the kanamycin resistance gene to replace the deletion. The E1 shuttle vector contained the left (0%- 53.9%) segment of the genome and was expected to generate BAV3 recombinants that can be grown and propagated in cells that can complement the missing E1 functions. To construct a similar shuttle vector for E3 deletion, DNA sequences extending from 78.9% to 82.5% (1281 bp) were deleted from within the E3 region that had been cloned into a plasmid vector. The deleted region corresponds to those that have been shown to be non-essential for viral replication in cell culture. The resulting plasmid was used to construct another recombinant plasmid with BAV3 DNA sequences extending from 37.1% to 100% and with a deletion of E3 sequences that were replaced by kanamycin resistance gene. This shuttle plasmid was used in cotransfections with digested viral DNA in an attempt to rescue a recombinant BAV3 carrying the kanamycin resistance gene to replace the deleted E3. In spite of repeated attempts of transfection, El or E3 recombinant BAV3 were not isolated. It seems that other approaches should be applied to make a final conclusion on BAV3 infectivity.

Cette étude a pour but spécifique de comprendre le processus de transformation et de construction de l’identité tant personnelle que sociale chez les jeunes parents suite à l’entrée en parentalité, et ce par l’impact qu’a l’événement (devenir parent) sur la perception de soi dans les diverses sphères de l’intime. Cette étude se distingue par son intérêt envers les jeunes couples qui ont grandi avec un modèle parental plus traditionnel, mais difficile à reproduire avec les changements sociaux, dont l'entrée des femmes sur le marché du travail. L'étude a été réalisé à l'aide d'entretiens semi-dirigés auprès de dix participants (cinq couples) répondant aux critères suivants: le couple de parents devait être hétérosexuel et vivre ensemble, les participants devaient être âgés entre 27 et 32 ans, leur premier enfant devait avoir entre 2 et 4 ans, ils devaient résider à l'extérieur de la métropole et au moins un des membres du couple devait avoir un diplôme universitaire. L'étude confirme l'idée que pour les pères, l'environnement familial est un agent motivateur dans l'idée de fonder leur propre famille, mais plusieurs vivent un choc quant au niveau de sacrifices nécessaires à faire en tant que parents. Toutefois, le choc identitaire le plus grand a été vécu par les femmes. Plusieurs d'entre elles ont ressenti une véritable confusion face aux différentes pressions sociales quant au rôle qu'elles ont à jouer en tant que mères ou travailleuses et ont ressenti de la culpabilité lorsqu'elles ont fait leur choix identitaire. C'est également le cercle social des femmes qui a été le plus modifié suite à l'arrivée de leur premier enfant, ces dernières s'entourant de collègues (femmes) qui vivent la même situation, tout en ayant moins de temps à offrir à leurs amis et familles suite à l'arrivée de l'enfant.

Cette étude porte sur le théâtre scolaire dans les établissements éducatifs pour jeunes filles tenus par la Congrégation Notre-Dame entre 1850 et 1920. Nous tentons de comprendre ce phénomène, encore très peu étudié, et son implication dans la construction du genre des couventines. L’étude est divisée en deux parties. La première veut comprendre pourquoi le théâtre a été pratiqué. Nous exposons les objectifs pédagogiques et matériels du théâtre pratiqué à la Congrégation, après avoir tenté de saisir les origines de la pratique en remontant à la fondation de la communauté enseignante par Marguerite Bourgeoys. Malgré les avantages qui lui sont reconnus, le théâtre scolaire féminin aura des opposants. Mgr Édouard-Charles Fabre, en particulier, en conteste la moralité. D’autres évêques se montreront toutefois plus ouverts au développement de l’art dramatique. La deuxième partie de cette étude décrit la manière dont le théâtre a été pratiqué. Nous passons en revue les représentations théâtrales données ainsi que les textes disponibles qui leur sont associés. Nous analysons les thèmes abordés par les pièces et les classons par genres dramatiques, mais aussi leur production en tant que performance à travers une étude des mises en scène, des costumes et des décors. À travers cette lecture, nous dévoilons quels étaient les éléments de genre proposés aux filles et démontrons que le théâtre a contribué à l’intégration du genre dans les couvents.

Toute historiographie se construit sous la forme d’un récit, soutenue par une narration qui, autour d’une intrigue, sélectionne et organise les événements en fonction de leur importance relative à des critères prédéterminés. La période des années 1960, qui est habituellement décrite comme celle de la naissance de la littérature québécoise, est on ne peut plus représentative de cette logique narrative qui sous-tend toute démarche historique. Un événement retenu par cette histoire, la Nuit de la poésie du 27 mars 1970, s’est littéralement élevé au rang de mythe dans l’imaginaire littéraire, mais aussi identitaire des Québécois. Que ce soit à travers les triomphes de Claude Gauvreau et de Michèle Lalonde ou la foule de plusieurs milliers de personnes devant le Gesù qui espérait assister à la soirée, cet événement, mis sur pellicule par Jean-Claude Labrecque et Jean-Pierre Masse, a été conçu par plusieurs comme l’apothéose de « l’âge de la parole ». Une observation minutieuse de ce happening soulève cependant de nombreuses questions concernant la construction de ce rôle dans l’histoire littéraire québécoise. En effet, tant l’étude des conditions d’organisation de la soirée que l’analyse de son mode de transmission révèlent des motivations archivistiques qui laissent transparaître l’objectif de la Nuit à « faire événement », à produire des traces qui participeront à l’inscription historique de la poésie québécoise. Considérant que l’œuvre filmique de Labrecque et de Masse constitue le seul véritable document témoignant de cet événement et que celui-ci a fait l’objet d’un montage ne présentant qu’une facette de la soirée, les enjeux de représentation et de mise en récit inhérents au cinéma documentaire poussent à questionner le portrait désiré de la poésie de l’époque que l’on voulait créer à travers le long-métrage. Véritable plateau de tournage, le spectacle du 27 mars 1970 appelle à être analysé dans les termes d’une conscience historique qui pousse les acteurs du milieu littéraire de l’époque à ériger les monuments qui seront à la base du canon et de l’institution littéraires québécois. L’étude, mobilisant les ressources de l’analyse de texte, de la sociologie de la littérature, de la théorie historiographique et de la théorie du cinéma, portera principalement sur le documentaire La Nuit de la poésie 27 mars 1970, considéré comme véritable recueil de textes, mais aussi de façon tout à fait originale sur les archives inédites rejetées du montage final des réalisateurs de l’ONF. Au terme de ce travail, j’approfondirai la réflexion sur le rôle historique d’un événement emblématique de la littérature québécoise en explicitant la construction historique autour de celui-ci. Il s’agira non seulement de relativiser l’événement en tant que tel, mais aussi de réfléchir sur le grand récit espéré par les artistes de l’époque.
