A series of four calix[5]arenes and three calix[6]arenes (R-calixarene-OCH2COR1) (R = H or Bu-t) with alkyl ketone residues (R-1 = Me or Bu-t) on the lower rim have been synthesized, and their affinity for complexation of alkali cations has been assessed through phase-transfer experiments and stability constant measurements. The conformations of these ketones have been probed by H-1 NMR and X-ray diffraction analysis, and by molecular mechanics calculations. Pentamer 3 (R R-1 = Bu-t) possesses a symmetrical cone conformation in solution and a very distorted cone conformation in the solid state. Pentamer 5 (R = H, R-1 = Bu-t) exists in a distorted 1,2-alternate conformation in the solid state, but in solution two slowly interconverting conformations, one a cone and the other presumed to be 1,2-alternate, can be detected. X-ray structure analysis of the sodium and rubidium perchlorate complexes of 3 reveal the cations deeply encapsulated by the ethereal and carbonyl oxygen atoms in distorted cone conformations which can be accurately reproduced by molecular mechanics calculations. The phase-transfer and stability constant data reveal that the extent of complexation depends on calixarene size and the nature of the alkyl residues adjacent to the ketonic carbonyls with tert-butyl much more efficacious than methyl.

Although the retinoblastoma protein (Rb) functions as a checkpoint in the cell cycle, it also regulates differentiation. It has recently been shown that Rb is acetylated during differentiation; however, the role of this modification has not been identified. Depletion of Rb levels with short hairpin RNA resulted in inhibition of human keratinocyte differentiation, delayed cell cycle exit and allowed cell cycle re-entry. Restoration of Rb levels rescued defects in differentiation and cell cycle exit and re-entry; however, re-expression of Rb with the major acetylation sites mutated did not. During keratinocyte differentiation, acetylation of Rb is mediated by PCAF and it is further shown that PCAF acetyltransferase activity is also required for normal differentiation. The major acetylation sites in Rb are located within the nuclear localization sequence and, although mutation did not alter Rb localization in cycling cells, the mutant is mislocalized to the cytoplasm during differentiation. Studies indicate that acetylation is a mechanism for controlling Rb localization in human keratinocytes, with either reduction of the PCAF or exogenous expression of the deacetylase SIRT1, resulting in mislocalization of Rb. These findings identify PCAF-mediated acetylation of Rb as an event required to retain Rb within the nucleus during keratinocyte differentiation.

Stromal-derived growth factors are required for normal epithelial growth but are also implicated in tumour progression. We have observed inactivation of the retinoblastoma protein (Rb), through phosphorylation, in cancer-associated fibroblasts in oro-pharyngeal cancer specimens. Rb is well known for its cell-autonomous effects on cancer initiation and progression; however, cell non-autonomous functions of Rb are not well described. We have identified a cell non-autonomous role of Rb, using three-dimensional cultures, where depletion of Rb in stromal fibroblasts enhances invasive potential of transformed epithelia. In part, this is mediated by upregulation of keratinocyte growth factor (KGF), which is produced by the depleted fibroblasts. KGF drives invasion of epithelial cells through induction of MMP1 expression in an AKT- and Ets2-dependent manner. Our data identify that stromal fibroblasts can alter the invasive behaviour of the epithelium, and we show that altered expression of KGF can mediate these functions. © European Molecular Biology Organization.

In order to study ultracold charge-transfer processes in hybrid atom-ion traps, we have mapped out the potential-energy curves and molecular parameters for several low-lying states of the Rb, Yb+ system. We employ both a multireference configuration interaction and a full configuration interaction (FCI) approach. Turning points, crossing points, potential minima, and spectroscopic molecular constants are obtained for the lowest five molecular states. Long-range parameters, including the dispersion coefficients, are estimated from our ab initio data. The separated-atom ionization potentials and atomic polarizability of the ytterbium atom (ad=128.4 atomic units) are in good agreement with experiment and previous calculations. We present some dynamical calculations for (adiabatic) scattering lengths for the two lowest (Yb, Rb+) channels that were carried out in our work. However, we find that the pseudopotential approximation is rather limited in validity and only applies to nK temperatures. The adiabatic scattering lengths for both the triplet and singlet channels indicate that both are large and negative in the FCI approximation.

Ultracold hybrid ion–atom traps offer the possibility of microscopic manipulation of quantum coherences in the gas using the ion as a probe. However, inelastic processes, particularly charge transfer can be a significant process of ion loss and has been measured experimentally for the ${\rm Y}{{{\rm b}}^{+}}$ ion immersed in a Rb vapour. We use first-principles quantum chemistry codes to obtain the potential energy curves and dipole moments for the lowest-lying energy states of this complex. Calculations for the radiative decay processes cross sections and rate coefficients are presented for the total decay processes; ${\rm Y}{{{\rm b}}^{+}}(6{\rm s}{{\;}^{2}}{\rm S})+{\rm Rb}(5{\rm s}{{\;}^{2}}{\rm S})\to {\rm Yb}(6{{{\rm s}}^{2}}{{\;}^{1}}{\rm S})+{\rm R}{{{\rm b}}^{+}}(4{{{\rm p}}^{6}}{{\;}^{1}}{\rm S})+h\nu $ and ${\rm Y}{{{\rm b}}^{+}}(6{\rm s}{{\;}^{2}}{\rm S})+{\rm Rb}(5{\rm s}{{\;}^{2}}{\rm S})\to {\rm YbR}{{{\rm b}}^{+}}({{X}^{1}}{{\Sigma }^{+}})+h\nu $. Comparing the semi-classical Langevin approximation with the quantum approach, we find it provides a very good estimate of the background at higher energies. The results demonstrate that radiative decay mechanisms are important over the energy and temperature region considered. In fact, the Langevin process of ion–atom collisions dominates cold ion–atom collisions. For spin-dependent processes [1] the anisotropic magnetic dipole–dipole interaction and the second-order spin–orbit coupling can play important roles, inducing coupling between the spin and the orbital motion. They measured the spin-relaxing collision rate to be approximately five orders of magnitude higher than the charge-exchange collision rate [1]. Regarding the measured radiative charge transfer collision rate, we find that our calculation is in very good agreement with experiment and with previous calculations. Nonetheless, we find no broad resonances features that might underly a strong isotope effect. In conclusion, we find, in agreement with previous theory that the isotope anomaly observed in experiment remains an open question.

White micas in carbonate-rich tectonites and a few other rock types of large thrusts in the Swiss Helvetic fold-and-thrust belt have been analyzed by Ar-40/Ar-39 and Rb/Sr techniques to better constrain the timing of Alpine deformation for this region. Incremental Ar-40/Ar-39 heating experiments of 25 weakly metamorphosed (anchizone to low greenschist) samples yield plateau and staircase spectra. We interpret most of the staircase release spectra result from variable mixtures of syntectonic (neoformed) and detrital micas. The range in dates obtained within individual spectra depends primarily on the duration of mica nucleation and growth, and relative proportions of neoformed and detrital mica. Rb/Sr analyses of 12 samples yield dates of ca. 10-39 Ma (excluding one anomalously young sample). These dates are slightly younger than the Ar-40/Ar-39 total gas dates obtained for the same samples. The Rb/ Sr dates were calculated using initial Sr-87/Sr-86 ratios obtained from the carbonate-dominated host rocks, which are higher than normal Mesozoic carbonate values due to exchange with fluids of higher Sr-87/Sr-86 ratios (and lower O-18/O-16 ratios). Model dates calculated using Sr-87/Sr-86 values typical of Mesozoic marine carbonates more closely approximate the Ar-40/Ar-39 total gas dates for most of the samples. The similarities of Rb/Sr and Ar-40/Ar-39 total gas dates are consistent with limited amounts of detrital mica in the samples. The delta(18)O values range from 24-15%. (VSMOW) for 2-6 mum micas and 27-16parts per thousand for the carbonate host rocks. The carbonate values are significantly lower than their protolith values due to localized fluid-rock interaction and fluid flow along most thrust surfaces. Although most calcite-mica pairs are not in oxygen isotope equilibrium at temperatures of ca. 200-400 degreesC, their isotopic fractionations are indicative of either 1) partial exchange between the minerals and a common external fluid, or 2) growth or isotopic exchange of the mica with the carbonate after the carbonate had isotopically exchanged with an external fluid. The geological significance of these results is not easily or uniquely determined, and exemplifies the difficulties inherent in dating very fine-grained micas of highly deformed tectonites in low-grade metamorphic terranes. Two generalizations can be made regarding the dates obtained from the Helvetic thrusts: 1) samples from the two highest thrusts (Mt. Gond and Sublage) have all of their Ar-40/Ar-39 steps above 20 Ma, and 2) most samples from the deepest Helvetic thrusts have steps (often accounting for more than 80% of Ar-39 release) between 15 and 25 Ma. These dates are consistent with the order of thrusting in the foreland-imbricating system and increase proportions of neoformed to detrital mica in the more metamorphosed hinterland and deeply buried portions of the nappe pile. Individual thrusts accommodated the majority of their displacement during their initial incorporation into the foreland-imbricating system, and some thrusts remained active or were reactivated down to 15 Ma.

In this paper, we report the measurement of Rb(2) molecule formation rate constant due to a two body process in a magneto-optical trap as a function of the sample temperature. The ground state molecules are detected by two-photon ionization, through the intermediate a(3)Sigma(+)(u) -> 2(3)Pi(g) molecular band. Our results show that the Rb(2) molecules formed in the MOT could be due to a wave shape resonance, which enhances the molecule formation rate. This effect may be used to enhance the molecule production; and therefore it maybe important to future experiments involving production and trapping of cold ground state molecules.

Os métodos radiométricos (Rb-Sr e Sm-Nd) têm sido aplicados, com sucesso, em rochas sedimentares visando a obtenção de idades deposicionais e informações sobre proveniência. Em muitos casos, apesar dos resultados geologicamente significativos, ainda persistem dúvidas em relação a extensão e interpretação dos dados obtidos. Isto ocorre porque as rochas sedimentares resultam da mistura de fragmentos detríticos de diversas proveniências com diferentes sistemas isotópicos, conseqüentemente, já contendo um registro isotópico da rocha fonte. No caso do método Rb-Sr, aplicado em rochas sedimentares de granulometria fina, pode-se obter idades absolutas para o evento deposicional. Isto é viável desde que este registro isotópico proveniente das rochas fonte seja apagado no momento da deposição, ou seja, deve ocorrer a homogeneização isotópica do Sr no ambiente sedimentar. Para tanto, devem ser observados alguns pré-requisitos em relação a amostragem, granulometria, ambiente deposicional e composição mineralógica das amostras, entre outros. No entanto, ainda existem questionamentos em relação a esta metodologia especialmente quanto à ocorrência e à extensão do processo de homogeneização isotópica do Sr em ambiente sedimentar.(Continua0 O método Sm-Nd, apesar de ser uma técnica relativamente nova quando aplicada a rochas sedimentares, tem se tornado uma ferramenta fundamental para auxílio na determinação de proveniência de rochas sedimentares. As maiores dificuldades estão na correta interpretação dos valores obtidos e sua associação com um ambiente sedimentar, ou seja, de baixa temperatura. Neste trabalho, foram aplicados os métodos radiométricos Rb-Sr e Sm-Nd em amostras de rochas sedimentares coletadas em diferentes contextos geológicos da Bacia do Paraná. Foram tentativamente testados os diferentes parâmetros que atuam no sentido da homogeneização isotópica do Sr e do comportamento do Nd. Os resultados obtidos permitiram aprimorar a metodologia radiométrica Rb-Sr e Sm-Nd quando aplicada em rochas sedimentares, bem como obter resultados sobre a idade deposicional de diferentes unidades sedimentares e, por vezes, sua proveniência.

Objetivou-se com este trabalho avaliar os efeitos de uma dieta de alto nível de energia e proteína combinada com a aplicação de bST no perfil de expressão dos genes da leptina e de seu receptor Ob-Rb no parênquima mamário de novilhas leiteiras. Foram utilizadas amostras de parênquima mamário de 32 novilhas holandesas distribuídas aleatoriamente em quatro tratamentos (n=8): dieta com alto ou baixo teor de energia e proteína combinada ou não com a aplicação de bST. O delineamento utilizado foi em blocos casualizados com arranjo de tratamentos em esquema fatorial 2 × 2. A extração do RNA total das amostras de tecido foi feita e o nível de expressão gênica foi analisado por qRT-PCR utilizando-se o gene da glicuronidase β como controle, pelo método 2-ΔΔCt. Animais que receberam a dieta com alto conteúdo de energia e proteína apresentaram maior expressão de mRNA de leptina, com aumento de 56%, e menor expressão de mRNA do receptor Ob-Rb, com redução de 18%. Por outro lado, a aplicação de bST resultou em diminuição da expressão do mRNA de leptina e do receptor Ob-Rb em 74% e 23%, respectivamente. Não houve interação entre dieta e aplicação de bST. O aumento na expressão de leptina pode explicar, ao menos em parte, os efeitos negativos da dieta de alta energia e proteína, oferecida no período pré-púbere, sobre a produção de leite de novilhas leiteiras.

Great part of the gold mineralizations are associated with shearing zones through which circulate a great volume of fluids, that interact with the host rocks, originating leaching or precipitation of chemical elements, including gold. The studied mineralizations are inserted in the Seridó Belt. The tungsten mineralization in Brejuí Mine is hosted in calcsilicate rocks from Jucurutu Formation. The São Francisco auriferous mineralization has as host rocks mica-schists from Seridó Formation, while the Ponta da Serra and Fazenda Simpático mineralizations are hosted in orthogneisses of this fold belt basement. The research conducted on these mineralizations had the purpose of integrate the data of chemical elements behavior during the shearing/mineralizing event, and its influence on the isotopic systems Rb-Sr and Sm-Nd. The studies of chemical mobility in the auriferous mineralizations showed that elements that during the shearing displayed in general an immobile behavior were Al, Ti and Zr. Among the elements that were mobilized during the event, K and Rb showed mass gain in ali belts of transformed rocks, while the elements Ca, Na and Sr normally lost mass. Petrographic studies showed that the minerais biotite and plagioclase, in all investigated mineralizations, played an important role in the chemical reactions occurred in the transformed rocks to the generation of muscovite, cordierite and sillimanite, justifying the input of K to the formation of muscovite, and the release of Na and Ca from plagioclase to the fluid phase. In the São Francisco auriferous mineralization, the results of the Rb-Sr isotopic analysis yielded ages of 645 ± 19 Ma and 596 ± 17 Ma, with both samples, from original and transformed rocks. Two ages, 569 ± 20 Ma. and 554 ± 19 Ma., were obtained with samples frem the transformed rocks domain. These ages suggest that there were two metamorphic pulses during the emplacement of the mineralized shearing zone. The Sm-Nd data yielded TDM ages of 1,31 Ga and 1,26 Ga with 3Nd (0,6 Ga) of -0,26 e -0,40 for the original and final transformed rocks, respectively. In case of the orthogneisses of Caicó Complex, e.g. the Ponta da Serra and Fazenda Simpático mineralizations, the Rb-Sr data did not yield ages with geological significance. In the Ponta da Serra mineralization, the Sm-Nd isotopic data yielded T DM ages of 2,56 Ga and 2,63 Ga to the original rocks and of 2,71 Ga to the mineralized sheared rock, and values of 3Nd (2,0 Ga) between -3,70 e -5,42 to the original and sheared rock, respectively. In the Fazenda Simpático, Sm-Nd data yielded TDM between 2,65 and 2,69 Ga with values of 3Nd (2,0 Ga) between -5,25 e -5,52. Considering the Sm-Nd data, the TDM ages may be admitted as the age of the parental magma extraction, producer of the protoliths of the orthogneisses from Ponta da Serra and Fazenda Simpático mineralizations. The chemical mobility studies showed that in the basement hosted mineralizations, Rb achieved mass while Sr lost mass, as Sm as well as Nd were strongly mobilized. The Sm/Nd ratio remained constant, however, confirming the isochemical character of those elements. In the basement mineralizations, Rb-Sr ages are destituted of geological significance, because of the partial opening of the isotopic system during the tectono-metamorphic transformations. In the tungsten mineralization, the diagram Sm-Nd constructed with the whole-rock data of calcsilicatic and the high-temperature paragenesis (garnet, diopside and iron-pargasitic hornblende) indicated an 631 ± 24 Ma age, while with the whole-rock data and low-temperature paragenesis (vesuvianite, epidote and calcite), a 537 ± 107 Ma age was obtained. These ages, associated with the petrographic observations, suggest that there was a time gap among the hydrothernal events responsible by the formation of the high and low temperature paragenesis in the calcsilicatic rocks mineralized in scheelite
