We investigated the relationship between the severity and incidence of resistance among Norway rats (Rattus norvegicus) on a farm in Wales and the subsequent outcome of a practical rodent control operation. Bromadiolone resistance factors were estimated for rats trapped on the farm using the blood clotting response test, and were found to be 2 to 3 for male rats and approximately 6 for females. The incidence of resistance in the rat population was high. Infestation size was estimated by census baiting and tracking, and was found to be substantial, with a maximum of 6.5 kg of bait being eaten on a single night. A proprietary rodenticide (Deadline (TM)), containing 0.005% bromadiolone, was used to control the infestation. The duration of baiting was 35 days and, according to the two methods of assessment used, treatment success was in the region of 87 and 93%. No evidence was observed of a significant impact of resistance on the rat control operation, and the remaining rats of this very heavy infestation would probably have been controlled if baiting had continued for longer.

A new blood clotting response test was used to determine the susceptibility, to coumatetralyl and bromadiolone, of laboratory strains of Norway rat from Germany and the UK (Hampshire), and wild rats trapped on farms in Wales (UK) and Westphalia (Germany). Resistance factors were calculated in relation to the CD strain of Norway rat. An outbred strain of wild rats, raised from rats trapped in Germany, was found to be more susceptible to coumatetralyl by a factor of 0.5-0.6 compared to the CD strain. Homozygous and heterozygous animals of a strain of resistant rats from Westphalia were cross-resistant to coumatetralyl and bromadiolone, with a higher resistance factor for bromadiolone than that found in both UK strains. Our results show that the degree of altered susceptibility and resistance varies between strains of wild rat and between resistance foci. Some wild rat strains may be more susceptible than laboratory rat strains. Even in a well-established resistance area, it may be difficult to find infestations with resistance high enough to suspect control problems with bromadiolone, even after decades of use of this compound.

A sample of 10 Norway rats (Rattus norvegicus) was taken for DNA resistance testing from an agricultural site in Kent where applications of the anticoagulant rodenticide bromadiolone had been unsuccessful. All animals tested were homozygous for the single nucleotide VKORC1 polymorphism tyrosine139phenylalanine, or Y139F. This is a common resistance mutation found extensively in France and Belgium but not previously in the UK. Y139F confers a significant level of resistance to first-generation anticoagulants, such as chlorophacinone, and to the second-generation compound bromadiolone. Another compound widely used in the UK, difenacoum, is also thought to be partially resisted by rats which carry Y139F. A silent VKORC1 mutation was also found in all rats tested. The presence of a third important VKORC1 mutation which confers resistance to anticoagulant rodenticides in widespread use in the UK, the others being Y139C and L120Q, further threatens the ability of pest control practitioners to deliver effective rodent control.

We compared the quantity of wheat bait consumed by Norway rats (Rattus norvegicus) from: (i) wooden bait trays, made as safe as possible from non-target animals using materials available at trial sites, and (ii) three different, proprietary tamper-resistant rat bait boxes. A balanced Latin square experimental design was used to overcome operational biases that occur when baits of different types are applied simultaneously at the same sites. The consumption of bait from the four different types of bait placement differed significantly and accounted for more than 76% of the total variation. The amount of bait eaten by rats from the bait trays was approximately eight times greater than the quantity eaten from the tamper-resistant bait boxes. The three bait box designs appeared to deter bait consumption by rats to a similar extent. Tamper-resistant bait boxes are essential tools in the application of rodenticides in many circumstances but their use should not be mandatory when it is possible to make baits safe from non-target animals by other means.

Resistance baselines were obtained for the first generation anticoagulant rodenticides chlorophacinone and diphacinone using laboratory, caesarian-derived Norway rats (Rattus norvegicus) as the susceptible strain and the blood clotting response test method. The ED99 estimates for a quantal response were: chlorophacinone, males 0.86 mg kg−1, females 1.03 mg kg−1; diphacinone, males 1.26 mg kg−1, females 1.60 mg kg−1. The dose-response data also showed that chlorophacinone was significantly (p<0.0001) more potent than diphacinone for both male and female rats, and that male rats were more susceptible than females to both compounds (p<0.002). The ED99 doses were then given to groups of five male and five female rats of the Welsh and Hampshire warfarin-resistant strains. Twenty-four hours later, prothrombin times were slightly elevated in both strains but all the animals were classified as resistant to the two compounds, indicating cross-resistance from warfarin to diphacinone and chlorophacinone. When rats of the two resistant strains were fed for six consecutive days on baits containing either diphacinone or chlorophacinone, many animals survived, indicating that their resistance might enable them to survive treatments with these compounds in the field.

BACKGROUND: The single nucleotide polymorphism (SNP), and consequent amino acid exchange from tyrosine to cysteine at location 139 of the vkorc1 gene (i.e. tyrosine139cysteine or Y139C), is the most widespread anticoagulant resistance mutation in Norway rats (Rattus norvegicus Berk.) in Europe. Field trials were conducted to determine incidence of the Y139C SNP at two rat infested farms in Westphalia, Germany, and to estimate the practical efficacy against them of applications, using a pulsed baiting treatment regime, of a proprietary bait (KleratTM) containing 50 ppm brodifacoum. RESULTS: DNA analysis for the Y139C mutation showed that resistant rats were prevalent at the two farms, with an incidence of 80.0% and 78.6% respectively. Applications of brodifacoum bait achieved results of 99.2% and 100.0% control at the two farms, when measured by census baiting, although the treatment was somewhat prolonged at one site due to the abundance of attractive alternative food. CONCLUSION: The study showed that 50 ppm brodifacoum bait is fully effective against the Y139C SNP at the Münsterland focus and is likely to be so elsewhere in Europe where this mutation is found. The pulsed baiting regime reduced to relatively low levels the quantity of bait required to control these two substantial resistant Norway rat infestations. Previous studies had shown much larger quantities of bromadiolone and difenacoum baits used in ineffective treatments against Y139C resistant rats in the Münsterland. These results should be considered when making decisions about the use of anticoagulant against resistant Norway rats and their potential environmental impacts.

CONTEXT. Rattus tanezumi is a serious crop pest within the island of Luzon, Philippines. In intensive flood-irrigated rice field ecosystems of Luzon, female R. tanezumi are known to primarily nest within the tillers of ripening rice fields and along the banks of irrigation canals. The nesting habits of R. tanezumi in complex rice–coconut cropping systems are unknown. AIMS. To identify the natal nest locations of R. tanezumi females in rice–coconut systems of the Sierra Madre Biodiversity Corridor (SMBC), Luzon, during the main breeding season to develop a management strategy that specifically targets their nesting habitat. METHODS. When rice was at the booting to ripening stage, cage-traps were placed in rice fields adjacent to coconut habitat. Thirty breeding adult R. tanezumi females were fitted with radio-collars and successfully tracked to their nest sites. KEY RESULTS. Most R. tanezumi nests (66.7%) were located in coconut groves, five nests (16.7%) were located in rice fields and five nests (16.7%) were located on the rice field edge. All nests were located above ground level and seven nests were located in coconut tree crowns. The median distance of nest sites to the nearest rice field was 22.5m. Most nest site locations had good cover of ground vegetation and understorey vegetation, but low canopy cover. Only one nest location had an understorey vegetation height of less than 20 cm. CONCLUSIONS. In the coastal lowland rice–coconut cropping systems of the SMBC, female R. tanezumi showed a preference for nesting in adjacent coconut groves. This is contrary to previous studies in intensive flood-irrigated rice ecosystems of Luzon, where the species nests mainly in the banks of irrigation canals. It is important to understand rodent breeding ecology in a specific ecosystem before implementing appropriate management strategies. IMPLICATIONS. In lowland rice–coconut cropping systems, coconut groves adjacent to rice fields should be targeted for the 20 management of R. tanezumi nest sites during the main breeding season as part of an integrated ecologically based approach to rodent pest management.

Context. Rattus tanezumi (the Asian house rat) is the principal rodent pest of rice and coconut crops in the Philippines. Little is known about the population and breeding ecology of R. tanezumi in complex agroecosystems; thus, current methods of rodent control may be inappropriate or poorly implemented. Aims. To investigate the habitat use, population dynamics and breeding biology of R. tanezumi in complex lowland agroecosystems of the Sierra Madre Biodiversity Corridor, Luzon, and to develop ecologically based rodent management (EBRM) strategies that will target specific habitats at specific times to improve cost-efficiency and minimise non-target risks. Methods. An 18-month trapping study was conducted in rice monoculture, rice adjacent to coconut, coconut groves, coconut-based agroforest and forest habitats. Trapped animals were measured, marked and assessed for breeding condition. Key results. Five species of rodent were captured across all habitats with R. tanezumi the major pest species in both the rice and coconut crops. The stage of the rice crop was a major factor influencing the habitat use and breeding biology of R. tanezumi. In rice fields, R. tanezumi abundance was highest during the tillering to ripening stages of the rice crop and lowest during the seedling stage, whereas in coconut groves abundance was highest from the seedling to tillering stage of nearby rice crops. Peaks in breeding activity occurred from the booting stage of the rice crop until just after harvest, but >10% of females were in breeding condition at each month of the year. Conclusions. In contrast with the practices applied by rice farmers in the study region, the most effective time for lethal management based on the breeding ecology of R. tanezumi is likely to be during the early stages of the rice crop, before the booting stage. Farmers generally apply control actions as individuals. We recommend coordinated community action. Continuous breeding throughout the year may necessitate two community campaigns per rice cropping season. To limit population growth, the most effective time to reduce nesting habitat is from the booting stage until harvest. Implications. By adopting EBRM strategies, we expect a reduction in costs associated with rodent control, as well as improved yield and reduced risk to non-target species.

The ecology of soils associated with dead mammals (i.e. cadavers) is poorly understood. Although temperature and soil type are well known to influence the decomposition of other organic resource patches, the effect of these variables on the degradation of cadavers in soil has received little experimental investigation. To address this, cadavers of juvenile rats (Rattus rattus) were buried in one of three contrasting soils (Sodosol, Rudosol, and Vertosol) from tropical savanna ecosystems in Queensland, Australia and incubated at 29 °C, 22 °C, or 15 °C in a laboratory setting. Cadavers and soils were destructively sampled at intervals of 7 days over an incubation period of 28 days. Measurements of decomposition included cadaver mass loss, carbon dioxide–carbon (CO2–C) evolution, microbial biomass carbon (MBC), protease activity, phosphodiesterase activity, and soil pH, which were all significantly positively affected by cadaver burial. A temperature effect was observed where peaks or differences in decomposition that at occurred at higher temperature would occur at later sample periods at lower temperature. Soil type also had an important effect on some measured parameters. These findings have important implications for a largely unexplored area of soil ecology and nutrient cycling, which are significant for forensic science, cemetery planning and livestock carcass disposal.

Na última década, o uso de sistemas microventilados (sistema VMA) em biotérios possibilitou alojar animais com melhor qualidade sanitária devido ao maior controle ambiental fornecido por esses sistemas. Contudo, ainda é necessário uma melhor avaliação desses sistemas, principalmente no que tange às respostas dos animais. Dessa forma, o objetivo deste trabalho foi avaliar a integridade do epitélio do trato respiratório superior (septo nasal e traquéia) de ratos mantidos em sistema VMA. Cem ratos, da linhagem Wistar-Hannover, heterogênicos, machos, de padrão sanitário convencional, foram mantidos durante 6 meses em dois distintos sistemas de ventilação: ventilação para conforto (grupo VGD, controle) e sistema VMA com intervalos de troca de cama variando de 3, 5, 7 e 9 dias (grupos IT3, IT5, IT7 e IT9, respectivamente). Após a eutanásia dos animais, fragmentos de septo nasal e traquéia foram coletados, processados com rotina de técnica histológica e corados com combinação de ácido periódico de Shiff e azul alciano (PAS-AB) e hematoxilina-eosina (HE). Os parâmetros morfométricos avaliados em septo nasal foram: 1) área de epitélio por unidade de superfície de membrana basal (EP/MB), 2) área de cílios por unidade de superfície de membrana basal (CI/MB), 3) número de núcleos por unidade de superfície de membrana basal (NN/MB), 4) área de muco ácido por unidade de superfície de membrana basal (MAc/MB), 5) área de muco ácido por área de epitélio (MAc/EP) e 6) número de núcleos por área de epitélio (NN/EP). Para NN/MB, o grupo VGD apresentou diferença estatística com os grupos IT3, IT5 e IT9. Para NN/EP, o grupo VGD teve diferença estatística com os todos grupos mantidos em VMA (IT3, IT5, IT7 e IT9). Em AE/MB observou-se diferença significativa do grupo VGD com os grupos IT7 e IT9. Para CI/MB notou-se diferença significativa entre os grupos IT3 e IT7. Embora não foi demonstrada diferença significativa entre os demais grupos, os dados sugerem que o grupo IT3 apresenta proporcionalmente maior área de cílios em relação aos demais grupos. Em MAc/MB e MAc/EP observou-se diferença significativa entre os grupos VGD e IT7. Na avaliação qualitativa da traquéia por escores, predominou a lesão inflamatória crônica, com intenso infiltrado de mononucleares (predominando neutrófilos). A presença e intensidade dessas lesões variaram de acordo com o sistema de ventilação sendo que somente no grupo VGD é que foram encontradas lesões severas (grau III). Com base nos resultados obtidos, os animais mantidos no sistema VMA, independente do intervalo de trocas de cama a que foram submetidos, apresentaram menor intensidade de lesões no septo nasal e na traquéia do que os animais mantidos em VGD. A maior integridade epitelial pode ser relacionado com a qualidade das condições ambientais fornecida aos animais alojados no sistema VMA, confirmando ser este o sistema mais adequado para a manutenção de ratos em experimentação.

A enzima estudada no presente trabalho é delta-aminulevulinato dehidratase (δ-ALA D), uma enzima sufidrílica, cuja atividade pode ser inibida por uma variedade de agentes bloqueadores de grupos tiólicos . A reação catalisada pela δ-ALA D (formação do composto monopirrólico porfobilinogênio) faz parte da rota de síntese de compostos tetrapirrólicos como o grupamento heme, consequentemente a inibição desta enzima implica em alterações patológicas decorrentes da inibição da rota de biossíntese do heme e ainda resultar no acúmulo do substrato ALA, o qual pode ter atividade pró oxidante por estar envolvido na produção de espécies ativas de oxigênio. Avaliou-se a susceptibilidade da δ-ALA D de fígado de peixe e rato frente a inibição por selênio orgânico e inorgânico. Os resultados demostraram claramente que a enzima δ-ALA D de peixes e mamíferos é susceptível a inibição “in vitro” por compostos orgânicos e inorgânicos de selênio. A análise comparativa demostrou que a enzima δ-ALA D de peixes é mais resistente a inibição por selênio em relação a de mamíferos. Todavia não foi possível esclarecer as causas deste diferente comportamento. Os resultados demostraram que o sistema de hidroxilação previamente descrito em ratos que acelera cataliticamente a oxidação de DTT na presença de disselenetos também ocorre em tecidos de peixe. Neste sistema estão envolvidos fatores enzimaticos, uma vez que este efeito foi anulado pela desnaturação térmica do sobrenadante. O presente trabalho mostra que a enzima δ-ALA D de peixes e ratos é sensível a inibição; in vitro, por composto de selênio orgânico e inorgânico. A manutenção dos grupos SH do sítio ativo da enzima no estado reduzido é essencial para a ação catalítica da δ-ALA D. A inibição da δ-ALA D causada por selênio orgânico ( (PhSe2) e (BuSe2)) e selênio inorgânico (selenito de sódio) é prevenida por DTT. Estes resultados indicam que o selenio orgânico e inorgânico inibe a δ-ALA D por oxidação de grupos tióis essenciais da enzima. A inibição desta enzima e conseqüentes alterações na rota de síntese de tetrapirróis podem ser responsáveis pelos efeitos tóxicos de selenetos orgânicos e inorgânicos em peixes e outros animais. Em peixes os efeitos toxicos mais relatados referem-se a deficiências reprodutivas, principalmente na redução da sobrevivência larval. Coincidentemente os ovários são o local de maior acumulação de selênio nos tecido de peixe e o desenvolvimento larval exige intensa atividade da enzima δ-ALA D.

The ultrastructural features of the ganglia of the myenteric plexus exhibit changes according to the animal species. These myenteric ganglia in the duodenum of adult rats of the Wistar strain were characterized ultrastructurally in this work. Those ganglia were depicted as compact structures, composed of neurones and glial cells, forming a dense neuropil surrounded by a continuous basal lamina and collagen fibrils. Glial cell bodies were smaller and apparently more frequent than neuronal cell bodies, being morphologically distinguished by nuclear features. In the neuronal extensions granular and agranular synaptic vesicles of different sizes predominate, in addition to mitochondria and myelinized profiles. Gliofilaments were not observed on the glial extensions of the rats.
