In order to examine the effects of alcohol on the hard palatine mucosa of rats, sixty adult female rats (Rattus norvegicus albinus) were divided into two experimental groups. The control group received solid diet (Purina rat chow) and tap water ad libitum. The alcoholic group received the same solid diet and was allowed to drink only sugar cane brandy dissolved in 30% Gay Lussac (v/v). At the end of periods of 90, 180 and 270 days of treatment, the animals at estro were sacrificed and the hard palatine mucosa were prepared for TEM and SEM methods. The basal cells of the alcoholic groups (90, 180 and 270 days of treatment) demonstrated some alterations: the intercellular spaces between these cells were higher, presented cytoplasmatic lipid droplets and autolysis. Also, the connective tissue showed intense lipid droplets accumulation in the alcoholic groups. These modifications suggested that chronic alcohol ingestion was able to modify the integrity of the cells in the rat hard palatine mucosa.

Smoking is considered to be the most albeit preventable cause of diseases and premature deaths in the history of mankind. The local action of tobacco on the oral mucosa can cause precancerous and cancerous lesions. However, there is not enough evidence to establish all the systemic effects caused by nicotine on the organism. Thus, the aim of the present study was to characterize the cellular changes of the cheek mucosa of rats submitted to long-term systemic nicotine treatment. Twenty male rats were divided into two experimental groups: a nicotine group and a control group, each consisting of 10 animals. The nicotine group was injected daily with 0.250 mg of nicotine per 100 g of body weight. All animals received a solid diet and water ad libitum. After 90 days of treatment, all animals were weighed and sacrificed. Samples of cheek mucosa were collected for light and transmission electron microscopy. The results revealed oral epithelium containing atypical cells that were characterized by atrophy, cell membrane disorganization and tissue damage. It was concluded that systemic administration of nicotine damaged the cellular integrity of the oral mucosa, impairing tissue function and predisposing the tissue to the action of different pathogenic agents and also to that of other carcinogenic substances present in tobacco. Submandibular glands of male rats were homogenized with 33 mM sodium potassium phosphate buffer, pH 6.5, containing 1 mM MgCl2 and 0.1 mM DTT and purified with ammonium sulphate, phosphocellulose chromatography, eluted with KC1 0.5 M, followed by Blue Sepharose CL-6B chromatography, eluted with NADH 0.5 mM. The enzyme kepts stable for 60 days when stored at -15-degrees-C in 33 mM phosphate buffer. In other experiment the enzyme was purified by oxamate-agarose chromatography from a crude extract of submandibular gland and the results obtained were better than by phosphocellulose and Sepharose CL-6B chromatography. The Km values for pyruvate. NADH, lactate and NAD+ were established. Sodium oxamate at 0.1 and 0.9 mM concentrations inhibited the LDH activity by 40 and 85%, respectively (competitive); with sodium oxalate the inhibition was of 30% (uncompetitive) and with 3-acetyl pyridine adenine dinucleotide was 80%.

The harmful effects of nicotine on male genital system fertility have been reported in experimental and clinical studies. However, its effects on prostatic cells and glandular pathogenesis remain unclear. The aim of the present study was to analyse the histological, histochemical and ultrastructural alterations, in addition to stereology, of the ventral lobe of the prostate of rats, submitted to chronic nicotine administration, as well as to establish the relationship between these changes and prostate diseases. Twelve male Wistar rats (Rattus norvegicus) were divided into two experimental groups: group I (nicotine) and group II (control). Samples of the ventral prostate were collected, processed and submitted to histological analysis, acid phosphatase histochemistry and ultrastructural analysis by transmission and scanning electron microscopies. The results showed that in the nicotine group, the secretory epithelial cells of the ventral lobe of the prostate were atrophied, and prostatic intraepithelial neoplasia occurred and reduced the expression of acid phosphatase. The disorganisation of organelles involved in the glandular secretory process, accompanied by biomembrane destructuring, was also observed. In conclusion, nicotine causes drastic alterations in the secretory epithelium of the ventral prostate, compromising its function. Furthermore, nicotine also induces premalignant lesions in the prostate gland, thus representing a risk factor in the development of prostate diseases.

Adult male rats (Wistar lineage) were alcoholized with sugar cane liquor diluted at 30° GL during 300 days and sacrificed every 60 days in 5 stages. Samples of choroid plexuses of lateral ventricles were collected and examined at transmission electronic microscope to detect possible ultrastructural alterations and to raise possible pathological correlations. Gradual changes were observed in these animals during all the experiment: dilatation and enlargement of cisternae of Golgi complex, dilatation of RER, presence of digestive vacuoles and a large amount of pinocytic vesicles as well as vesicles with electronlucent content throughout cytoplasm, as well as an enlargement of intercellular space between basolateral interdigitation of the cells and of the connective tissue. The changes observed in the epithelium and connective tissue of choroid plexuses specially in 240 and 300 days of treatment are presumably due to a disturbance in hydroelectrolitic homeostasis, contributing to several morpho-functional disturbs of central nervous system. No changes were observed in the control group animals.

Sixty adult tats (Rattus norvegicus albinus) of the same age (3 months) and with a mean body weight of 228 g were divided into two experimental groups. The control group received solid diet (Purina rat chow) and tap water ad libitum. The other (alcoholic group), received the same solid diet and was allowed to drink only sugar cane brandy dissolved in 30° Gay Lussac (v/v). At the end of periods of 90, 180 and 270 days of treatment, the animals were anaesthetized with ethyl ether during estrus, weighed and sacrificed. The final mean body weights were similar in the control and alcoholic groups. The results showed intense atrophy on the lining epithelium of the endometrium of uterine horns in the alcoholic group. Important ultrastructural epithelial alterations were also observed in the female alcoholic group, such as: intense lipid droplet accumulation, increased rough endoplasmic reticulum cisternae and mitochondrial size and presence of intraepithelial neutrophils. The secretory activity of these rats was reduced. Therefore, we concluded that alcohol acts as a toxin on the epithelial layer of the rat endometrium.

A psicologia da aprendizagem foi marcada pelo debate sobre se a aprendizagem seria um processo gradual ou súbito. Enquanto os associacionistas defendiam a primeira proposta, os gestaltistas afirmavam a existência de situações de aprendizagem abruptas. Dentre os principais autores a defenderem esta possibilidade estava Wolfgang Köhler. Os trabalhos deste autor têm sido apontados como evidência de que a aprendizagem seria um processo súbito. Apesar da relevância destes trabalhos em demonstrar a existência de situações em que uma forma súbita de aprendizagem ocorra, muito se tem questionado sobre as conclusões apresentadas por ele para o porquê da ocorrência deste tipo de fenômeno comportamental. Dentre as muitas críticas feitas, a que tem recebido mais atenção refere-se à ausência de controle da história dos seus sujeitos experimentais, bem como à desconsideração do papel que esta história teria nos resultados encontrados. Estudos que investigaram este papel (Epstein et al., 1984 e Epstein & Medalie 1983, 1985) demonstraram que a resposta típica de insight podia ser o resultado da junção de repertórios aprendidos previamente. Os trabalhos de Epstein foram importantes em demonstrar que o insight seria uma junção de repertórios que se combinam em situações apropriadas pelo possível envolvimento de um processo conhecido como Generalização Funcional. O presente trabalho se propôs a investigar se a Generalização Funcional era realmente responsável pela interconexão dos repertórios que culminariam na resolução da tarefa de um modo considerado como súbito. Para tal foi feita uma replicação dos experimentos de Epstein, utilizando-se ratos como sujeitos. Os resultados mostraram que a Generalização Funcional parece ser um requisito necessário, mas não suficiente para a resolução súbita de problemas, de um modo considerado como insight".

Foi estabelecido um modelo de imunossupressão em roedores por inoculação do agente alquilante Ciclofosfamida (CY). A administração de 50 mg/kg de CY em ratos Wistar provocou uma significante diminuição dos parâmetros de celularidade e peso relativo dos órgãos linfóides. Pela análise da titulação de anticorpos, do ensaio sobre as células formadoras de placa e do teste de hemólise foi comprovada que a imunidade humoral dos roedores sofreu supressão. Foram realizadas quatro inoculações desse imunossupressor e a periodicidade entre as inoculações foi determinada pela recuperação dos níveis de normalidade dos parâmetros supracitados. A alteração na contagem diferencial de células sanguíneas brancas representou o maior efeito adverso da CY, observado nos parâmetros de laboratório analisados nos Cebus apella. Nas duas vezes que foi administrada a droga houve redução no número de linfócitos e posteriormente diminuição de neutrófilos, porém somente na segunda foi observada a imunossupressão. Visto a proximidade filogenética dos primatas não humanos, este desenho experimental será de suma importância para o estudo de tumores em diversas fases do desenvolvimento e principalmente para testes de novos fármacos e esquemas terapêuticos. Com relação às análises de genotoxicidade da CY podemos concluir que em ratos Wistar, as administrações de CY aumentaram significativamente a freqüência de micronúcleos em eritrócitos policromáticos (MN PCEs) e provocaram efeito citotóxico (P<0.05). Em C. apella, os linfócitos do sangue periférico, após o tratamento com CY apresentaram um aumento significativo da media de MN/1000 células em relação aos linfócitos controle (P<0.05). A concentração de CY de 50mg/kg, em C. apella, corresponde à concentração DL50 da droga, visto que 50% desses animais morreram durante o experimento de imunossupressão. Até o desenvolvimento deste trabalho, não se conhecia a concentração correspondente ao DL50 nessa espécie. Ao comparamos as duas espécies de animais utilizadas neste trabalho, os primatas não humanos têm uma recuperação imune mais rápida em relação aos ratos Wistar. Provavelmente a capacidade de metabolização da droga seja mais eficaz em C. apella. Nossos resultados apóiam, portanto, que os primatas não humanos constituem os melhores modelos experimentais devido a sua grande proximidade evolutiva e filogenética com o ser humano.
