本论文以沈阳张士污灌区土壤为例,首次采用传统微生物生态学与现代微生物分子生态学相结合的研究方法系统地研究了污灌区长期重金属污染胁迫下原位农田土壤微生物特征。结果表明,虽然已经停止污灌十多年,张士灌区土壤耕作层(0~30 cm)仍然存在普遍的Cd污染,灌区土壤Cd含量高达1.75~3.89 mg kg-1。部分区域土壤Cd呈现向下迁移的趋势,且同时伴随有Cu、Zn复合污染。灌区土壤Cd含量较高时清水灌溉能降低土壤表层Cd含量,灌区土壤Cd含量下降到一定程度(约2 mg kg-1)后,清水灌溉对消除土壤表层Cd污染的作用消失。重金属元素中Cd对土壤微生物的影响最突出,在三个不同季节中土壤Cd与土壤微生物生物量(MBC)和微生物商(qM)呈显著负相关,与土壤微生物代谢商(qCO2)呈显著正相关。所检测的微生物指标中qM和qCO2与多种重金属元素呈显著相关性,可作为评价一定程度重金属污染的微生物指标。土壤营养元素(除P外)与微生物特征呈显著正相关性,土壤营养元素对微生物的刺激作用有可能在某种程度上掩盖了重金属对土壤微生物的负面影响。 用16S rDNA-PCR-DGGE方法,研究了不同浓度Cd胁迫下土壤Cd抗性细菌群落结构的动态变化,结果表明在Cd的胁迫下Cd抗性细菌多样性显著增加,不同土壤样品中Cd抗性细菌群落结构向相似的方向偏移,群落结构最终将可能趋向一致。Cd胁迫使敏感菌Pontibacter消失,而伯克氏菌(Burkholderia)、罗尔斯通氏菌(Ralstonia)、芽孢杆菌(Bacillus)和节杆菌(Arthrobacter)则富集成为优势菌。 从张士灌区Cd污染土壤中分离出32株Cd抗性细菌,研究了Cd抗性细菌和Cd抗性基因cadA的分布特征。这32株Cd抗性细菌分别归属于拟杆菌门(Bacteroidetes)(37.5%)、变形菌门(Proteobacteria) (37.5%)、放线菌门(Actinobacteria)(9.4%) 和厚壁菌门(Firmicutes)(15.6%)。在液体LB培养基中对Cd的抗性浓度都大于2 mmol L-1,对Zn抗性浓度介于5~13 mmol L-1。首次从Cetobacillus属的Cd抗性菌株S1基因组DNA中扩增出cadA基因的部分片断。在芽孢杆菌属(Bacillus)的4株菌N7,N9,N10和N11的基因组DNA中扩增出cadA基因的部分片断。序列分析结果表明这5株菌的cadA基因序列相似性为99%~93%,它们与坚强芽孢杆菌(Bacillus firmus) cadA 基因序列(M90750)相似性为94%~92%。系统发育分析结果表明这5株菌的cadA都与Bacillus firmus cadA 基因有着较近的亲缘关系。不同属的Cd抗性细菌间cadA基因的高度相似性揭示了cadA基因能在不同种属间转移的特性。

OLAND两阶段生物脱氮系统是一项正在开发且极具应用前景的处理高氨氮、低COD废水的新技术。在实验室水平,对OLAND系统限氧亚硝化阶段MBR反应器和厌氧氨氧化阶段SBR反应器的启动和运行进行了系统研究。MBR反应器控制参数在DO0.1-0.3mg/L,pH7.8±0.1,温度30±0.5℃,SRT无穷大的条件下,可实现在较高的容积负荷By=IO00mgN/L,水力停留时间HRT:ld下稳定的亚硝酸型硝化,使NH4+-N和NO2-N的出水比例达到理想的比值(1:1.20±0.20),保证厌氧氨氧化阶段SBR反应器的理想进水;SBR反应器在完全厌氧、pH7.8-8.2,温度30±0.5℃,SRT无穷大的条件下,无需外加任何有机碳源,在较高总氮负荷550mgN/L下,可实现NH4+-N和NO2--N同时稳定的去除,二者的消耗比例为1:(1.21±0.05),总氮去除率高达92%。采用巢式PCR、DGGE、 FISH等分子技术对MBR反应器硝化菌群随溶解氧的动态变化规律和SBR反应器厌氧氨氧化菌群结构、组成进行了研究,并探讨了硝化菌群与氮素组成变化之间的内在联系。结果表明:在限氧亚硝化阶段硝化菌群中氨氧化菌受溶解氧浓度的影响较大,其种群结构从反应器启动初期到稳定后期发生了非常明显的变化。亚硝酸氧化菌NOB的种群组成受溶氧影响并不明显,从启动初期到稳定后期其种群结构无明显变化。硝化菌群中氨氧化菌 AOB与亚硝酸氧化菌NOB的数量比例关系随溶解氧的降低而不断升高,从最初的3.6:1升高到稳定后期的5.5:1。MBR反应器优势硝化菌群主要由A、B、C三类氨氧化菌和硝化杆菌D、硝化螺菌F组成,其中优势菌C为维持MBR反应器稳定出水比例的主要功能菌。硝化菌群组成和结构的变化,带来了不同N素之间组成和比例的规律性变化。厌氧氨氧化阶段基本由厌氧氨氧化菌AnAOB和ANAMMOX两类菌组成,二者空间结构紧密,在反应器中的活菌数量比例分别为55%和42%。厌氧氨氧化菌AnAOB种群多样性相对比较丰富,主要由条带I和H所代表的两种优势菌组成;ANAMMOX菌种群多样性变化较小,其种群主要由优势菌K和J组成。对MBR和SBR反应器中的优势菌进行了克隆、测序和系统发育学分析,结果表明:限氧亚硝化阶段MBR反应器启动初期的优势菌A属于Nitrosomonadaceae科,是否是一个新属,还有待于进一步鉴定。运行中期优势菌B与Nitroso)nonaseurooaea亲缘关系最近,同源性高达99.1%,暂命名为Nilrosomonas sp.BI。稳定后期优势菌C与Nitrosomonas eutroPha亲缘关系最近,同源性为96.3%,暂命名为Nifrosomonas sp.cl。硝化杆菌属优势菌D与Nitrobacter alkalicus、Nitrobacter hambllrgensts和Nitlobac招rwinograsky 亲缘关系较近,同源性分别为95.5-97%、96.5~97%和95.8~96.8%。SBR反应器中AnAOB优势菌I与MBR反应器优势菌B亲缘关系最近,同源性高达98.7%,与Nitlosomonas euroPaea同源性为98.3%。根据序列比较和生理特性分析,优势菌I与优势菌B应为Nitrosomonas属两个不同的种,暂将优势菌I命名为Nitrosomonas sp.II。优势菌H与MBR反应器优势菌C亲缘关系最近,同源性达97.9%,与Nitrosomonas eutropha同源性为96.3%。结合其生理特性分析,二者应为Nitrosomonas属两个不同的种,暂将优势菌H命名为Nitrosomollas sp.Hl。ANAMMox优势菌K与未培养的 Planctomycete和已鉴定的另一种ANAMMoX菌尤uenenia sf况ttgartiensis亲缘关系较近,同源性分别为99.8%和96.6%。优势菌J与Gen bank收录的所有菌的相似性均低于76%,说明该菌是OLAND系统厌氧氨氧化阶段比较独特的菌,是迄今为止在厌氧反应过程中未发现的一个新菌。

The intermediate steps in the biosynthesis of the ADP-L-glycero-D-manno-heptose precursor of inner core lipopolysaccharide (LPS) are not yet elucidated. We isolated a mini-Tn10 insertion that confers a heptoseless LPS phenotype in the chromosome of Escherichia coli K-12. The mutation was in a gene homologous to the previously reported rfaE gene from Haemophilus influenzae. The E. coli rfaE gene was cloned into an expression vector, and an in vitro transcription-translation experiment revealed a polypeptide of approximately 55 kDa in mass. Comparisons of the predicted amino acid sequence with other proteins in the database showed the presence of two clearly separate domains. Domain I (amino acids 1 to 318) shared structural features with members of the ribokinase family, while Domain II (amino acids 344 to 477) had conserved features of the cytidylyltransferase superfamily that includes the aut gene product of Ralstonia eutrophus. Each domain was expressed individually, demonstrating that only Domain I could complement the rfaE::Tn10 mutation in E. coli, as well as the rfaE543 mutation of Salmonella enterica SL1102. DNA sequencing of the rfaE543 gene revealed that Domain I had one amino acid substitution and a 12-bp in-frame deletion resulting in the loss of four amino acids, while Domain II remained intact. We also demonstrated that the aut::Tn5 mutation in R. eutrophus is associated with heptoseless LPS, and this phenotype was restored following the introduction of a plasmid expressing the E. coli Domain II. Thus, both domains of rfaE are functionally different and genetically separable confirming that the encoded protein is bifunctional. We propose that Domain I is involved in the synthesis of D-glycero-D-manno-heptose 1-phosphate, whereas Domain II catalyzes the ADP transfer to form ADP-D-glycero-D-manno-heptose.

A synthetic version of the metal-regulated gene A (mrgA) promoter from Bacillus subtilis, which in this Gram-positive bacterium is negatively regulated by manganese, iron, cobalt, or copper turned out to promote high level of basal gene expression that is further enhanced by Co(II), Cd(II), Mn(II), Zn(II), Cu(II), or Ni(II), when cloned in the Gram-negative bacterium Cupriavidus metallidurans. Promoter activity was monitored by expression of the reporter gene coding for the enhanced green fluorescent protein (EGFP), and cellular intensity fluorescence was quantified by flow cytometry. Expression levels in C. metallidurans driven by the heterologous promoter, here called pan, ranged from 20- to 53-fold the expression level driven by the Escherichia coli lac promoter (which is constitutively expressed in C. metallidurans), whether in the absence or presence of metal ions, respectively. The pan promoter did also function in E. coli in a constitutive pattern, regardless of the presence of Mn(II) or Fe(II). In conclusion, the pan promoter proved to be a powerful tool to express heterologous proteins in Gram-negative bacteria, especially in C. metallidurans grown upon high levels of toxic metals, with potential applications in bioremediation. he present model of agriculture is based on intensive use of industrial inputs, due to its rapid response, but it brings harmful consequences to the environment, and it is necessary the use of modern inputs. And an alternative is the use of rock biofertilizers in agriculture, a product easy to use, with higher residual effect and does not harm the environment. The objective of study was to evaluate the inoculation and co-inoculation of different microorganisms in the solubilization of rock phosphate and potash ground microbial evaluating the best performance in the production of biofertilizers comparing with rocks pure in soil chemical properties and, verify effect of inoculation of the bacterium Paenibacillus polymyxa in the absorption of minerals dissolved in the development of cowpea (Vigna unguiculata [L.] Walp.). The first bioassay was conducted in Laboratory (UFRN) for 72 days in Petri dishes, where the rock powder was increased by 10% and sulfur co-inoculated and inoculated with bacterial suspension of Paenibacillus polymyxa grown in medium tryptone soy broth, Ralstonia solanacearum in medium Kelman, Cromobacterium violaceum in medium Luria-Bertani and Acidithiobacillus thiooxidans in medium Tuovinen and Kelly,and fungi Trichoderma humatum and Penicillium fellutanum in malt extract. Every 12 days, samples were removed in order to build up the release curve of minerals. The second bioassay was conducted in a greenhouse of the Agricultural Research Corporation of Rio Grande do Norte in experimental delineation in randomized block designs, was used 10 kg of an Yellow Argissolo Dystrophic per pot with the addition of treatments super phosphate simple (SS), potassium chloride (KCl), pure rock, biofertilizers in doses 40, 70, 100 and 200% of the recommendation for SS and KCl, and a control, or not inoculated with bacteria P. polymyxa. Were used seeds of cowpea BRS Potiguar and co-inoculated with the bacterial suspension of Bradyrhizobium japonicum and P. polymyxa. The first crop was harvested 45 days after planting, were evaluated in the dry matter (ADM), macronutrients (N, P, K, Ca, Mg) and micronutrients (Zn, Fe, Mn) in ADM. And the second at 75 days assessing levels of macro end micronutrients in plants and soil, and the maximum adsorption capacity of P in soil. The results showed synergism in co-inoculations with P. polymyxa+R. solanacearum and, P. polymyxa+C. violaceum solubilizations providing higher P and K, respectively, and better solubilization time at 36 days. The pH was lower in biofertilizers higher doses, but there was better with their addition to P at the highest dose. Significant reduction of maximum adsorption capacity of phosphorus with increasing dose of biofertilizer. For K and Ca was better with SS+KCl, and Mg to pure rock. There was an effect of fertilization on the absorption, with better results for P, K and ADM with SS+KCL, and N, Ca and Mg for biofertilizers. Generally, the P. polymyxa not influence the absorption of the elements in the plant. In treatments with the uninoculated P. polymyxa chemical fertilizer had an average significantly higher for weight and number of grains. And in the presence of the bacteria, biofertilizers and chemical fertilizers had positive values in relation to rock and control. The data show that the rocks and biofertilizers could meet the need of nutrients the plants revealed as potential for sustainable agriculture

This review deals with a comparative analysis of seven genome sequences from plant-associated bacteria. These are the genomes of Agrobacterium tumefaciens, Mesorhizobium loti, Sinorhizobium meliloti, Xanthomonas campestris pv campestris, Xanthomonas axonopodis pv citri, Xylella fastidiosa, and Ralstonia solanacearum. Genome structure and the metabolism pathways available highlight the compromise between the genome size and lifestyle. Despite the recognized importance of the type III secretion system in controlling host compatibility, its presence is not universal in all necrogenic pathogens. Hemolysins, hemagglutinins, and some adhesins, previously reported only for mammalian pathogens, are present in most organisms discussed. Different numbers and combinations of cell wall degrading enzymes and genes to overcome the oxidative burst generally induced by the plant host are characterized in these genomes. A total of 19 genes not involved in housekeeping functions were found common to all these bacteria.

The objective of this study was to evaluate the effect of incorporation of neem (Azadirachta indica) leaves into the soil for controlling bacterial wilt of tomato caused by Ralstonia solanacearum. The experiment was conducted in a greenhouse of the State University of Maranhao (Brazil). Dry and fresh neem leaves were incorporated in the soil in different amounts (0, 20, 40, 60, 80 and 100 g) and kept in it for different periods of time (0, 15, 30, 45 and 60 days). After each of these periods, seedlings inoculated with R. solanacearum were transplanted in the amended soil. Results showed positive effects in the disease control by incorporating neem leaves, with a reduction of wilting symptoms up to 100% with dry leaves and 78% with fresh leaves.

Antimicrobial activity of 45 extracts of medicinal plants were tested on Xanthomonas campestris pv. vesicatoria, Ralstonia solanacearum and Clavibacter michiganense subsp. michiganense. Some assays were done to verify the capability of these plants extrats to show an antibiosis. Five extracts (EAFQ, SM1, SM12, SM16, SA1) shown positive activity. The extract EAFQ expressed bactericide activity on C. michiganense. It suggests the possibility of using these actives substances in natura or as a model to synthesize industrialized products, intendind field utilization.

Most of the interspecific rootstocks (Lycopersicum esculentum × L. hirsutum) used in grafted Spanish tomato crops are resistant to Meloidogyne nematodes, but the 'Mi' resistance gene does not work well at high soil temperatures. Ralstonia solanacearum is a bacterial disease usual in tropical areas, but recently identified with low incidence in several European countries. This disease could be controlled by grafting tomato on Solanum torvum, which is also resistant to Meloidogyne. However, S. torvum and tomato have low grafting affinity, which could be improved using an intermediate rootstock. Some cultivars of eggplant have a relatively good affinity with tomato and complete affinity with S. torvum. In this study we compared two tomato cultivars (one resistant to Verticillium dalihae, Fusarium oxysporum v. lycopersici race 2 and Meloidogyne spp., and one non-resistant) grafted onto 'Beaufort' (Lycopersicum esculentum × L. hirsutum), 'Torvum Vigor' (Solanum torvum) and also with an intermediate grafting of eggplant ('Cristal') between tomato and S. torvum, with nongrafted plants as controls. This arrangement was carried out in two cropping cycles (winter-spring and summer-autumn). In both cycles, plants grafted onto S. torvum, both single or double grafted, yielded less than those grafted onto 'Beaufort' or nongrafted plants. In the spring cycle, no differences were found between single and double-grafted plants using S. torvum rootstocks, but in the autumn cycle double grafted plants had higher yields than the single grafted plants. The severity of nematode infections, in terms of reducing yields, and/or hypothetical infections of Ralstonia, will determine the utility of this technique in tomato production.

A diverse set of phage lineages is associated with the bacterial plant-pathogen genomes sequenced to date. Analysis of 37 genomes revealed 5,169 potential genes (approximately 4.3 Mbp) of phage origin, and at least 50 had no function assigned or are nonessential to phage biology. Some phytopathogens have transcriptionally active prophage genes under conditions that mimic plant infection, suggesting an association between plant disease and prophage transcriptional modulation. The role of prophages within genomes for cell biology varies. For pathogens such as Pectobacterium, Pseudomonas, Ralstonia, and Streptomyces, involvement of prophage in disease symptoms has been demonstrated. In Xylella and Xanthomonas, prophage activity is associated with genome rearrangements and strain differentiation. For other pathogens, prophage roles are yet to be established. This review integrates available information in a unique interface (http://propnav.esalq.usp.br) that may be assessed to improve research in prophage biology and its association with genome evolution and pathogenicity. © Copyright ©2013 by Annual Reviews.
