Ligularia, a highly diversified genus in the eastern Qinghai-Tibet Plateau and adjacent areas, was chosen as a suitable subject in which to study speciation patterns in this 'hot spot' area at the chromosomal level. Chromosome numbers and karyotypes were studied in 23 populations of 14 species, most of which are endemic to this area. The basic number x = 29 was confirmed for all species. Ligularia virgaurea was found to have diploid and triploid cytotypes, 2n = 58 and 87. Other species are only diploid, with 2n = 58. The karyotypes of all populations within any species, and all species spanning most sections and covering most of the morphological range in Ligularia, are very similar to each other, belonging to type 2A according to Stebbin's classification. This karyotype was also found in its close allies, e.g. Cremanthodium, Ligulariopsis, Parasenecio, and Sinacalia. Aneuploid reduction of chromosome number from 2n = 60 to 58 and karyotypic variation was found in Ligularia and its allies. Such a chromosomal pattern with few polyploids infers that variation of karyotype structure at the diploid level seems to be the predominant feature of chromosomal evolution in this group and sympatric speciation via hybridization and polyploidization has played a minor role in its species diversity. All taxa endemic to the Qinghai-Tibet Plateau are hypothesized to have originated in situ or from immediately adjacent areas because of the relatively recent formation of the plateau since the Pliocene, followed by the large-scaled biota extinction and recession caused by the Quaternary ice sheet. However, identification of specific progenitors remains difficult for some endemics, especially some endemic genera. Nannoglottis, with about eight species endemic to this region, is one such genus. Past taxonomic treatments have suggested its relationships with four different tribes of Asteraceae. We intend to identify the closest relatives of Nannoglottis by evaluating the level of monophyly, tribal delimitation, and systematic position of the genus by using molecular data from ndhF gene, trnL-F, and ITS region sequences. We find that all sampled species of Nannoglottis are a well-defined monophyly. This supports all recent taxonomic treatments of Nannoglottis, in which all sampled species were placed in one broadly re-circumscribed genus. Nannoglottis is most closely related to the Astereae, but stands as an isolated genus as the first diverging lineage of the tribe, without close relatives. A tentative relationship was suggested for Nannoglottis and the next lineage of the tribe was based on the ITS topology, the "basal group," which consists of seven genera from the Southern Hemisphere. Such a relationship is supported by some commonly shared plesiomorphic morphological characters. Despite the very early divergence of Nannoglottis in the Astereae, the tribe must be regarded to have its origin in Southern Hemisphere rather than in Asia, because based on all morphological, molecular, biogeographical, and fossil data, the Asteraceae and its major lineages (tribes) are supposed to have originated in the former area. Long-distance dispersal using Southeast Asia as a steppingstone from Southern Hemisphere to the Qinghai-Tibet Plateau is the most likely explanation for this unusual biogeographic link of Nannoglottis. The 23-32-million-year divergence time between Nannoglottis and the other Astereae estimated by DNA sequences predated the formation of the plateau. This estimation is further favored by the fossil record of the Asteraceae and the possible time of origin of the Astereae. Nannoglottis seems to have reached the Qinghai-Tibet area in the Oligocene-Eocene and then re-diversified with the uplift of the plateau. The molecular infragenetic phylogeny of the genus identifies two distinct clades, which reject the earlier infrageneric classification based on the arrangement of the involucral bracts and the length of the ligules, but agree well with the habits and ecological preferences of its current species. The "alpine shrub" vs. "coniferous forest" divergence within Nannoglottis was estimated at about 3.4 million years ago when the plateau began its first large-scale uplifting and the coniferous vegetation began to appear. Most of the current species at the "coniferous forest" clade of the genus are estimated to have originated from 1.02 to 1.94 million years ago, when the second and third uprisings of the plateau occurred, the climate oscillated and the habitats were strongly changed. The assumed evolution, speciation diversity, and radiation of Nannoglottis based on molecular phylogeny and divergence times agree well with the known geological and paleobotanical histories of the Qinghai-Tibet Plateau. Based on the phylogenetic and biogeographical studies of the glyptosternoid fishes in Qinghai-Tibet area, the following hypothesis is proposed: the speciation of this group has a direct relationship with the three uplift intervals of the Qinghai-Tibet Plateau. This process was explained by the theory of vicariance of biogeography. The ancestor of this group was similar to Bagarus and/or Glyptothorax, which still have a wide distribution. At the moment when the Tethys sea closed, the Indian tectonic plate collided with the Eurasian tectonic plate, so the Glyptothorax-like and Bagarus-like ancestors entered Eurasia and gradually became widely distributed. After the Pleistocene, with the enforced colliding, the gradual uplift of the Qinghai-Tibet Plateau brought about the current water environment, and the Glyptosternoids were generated from Glyptothorax-like fish under this environment. The present Glyptosternum, distributed across the Himalayas is the ancestor of Glyptosternoids. In the three uplift intervals of the plateau, the water system of this region was separated gradually and Glyptosternum-like ancestor was isolated in different rivers and evolved into various species. All this resulted in the speciation and formation of the biogeographical pattern of glyptosternoids.

Qinghai-Tibet Railway is the longest and highest plateau railway in the world. A long term monitoring system of the stability of the subgrade in the permafrost regions should be put forward immediately to prevent damage to the railway. As it's very difficult to set up the long-distance automatic monitoring system which contains a lot of measure points along the 550 kilometers railway in the permafrost area, we present a subgrade temperature monitor system based on fiber Bragg grating (FBG). In this paper the principles of the FBG was presented, and the feasibility of the FBG sensors in the permafrost area of Qinghai-Tibet plateau was analysized. We embedded fifteen FBG temperature sensors and thermal resistance temperature sensors. A contrast experiment is made while the two kinds of sensors are arranged in the same position. The result of the experiment shows that the accuracy of the FBG temperature sensors is less than 0.1 degrees C. and the FBG sensors can do well in the measurement of pattern which the temperature varies with the depth of the permafrost soil. The result also shows the stability of the FBG sensors in the bad environmental condition of Qinghai-Tibet plateau, which proves the feasibility of the application of FBG sensors and our monitoring system on the Qinghai-Tibet railway.

Nitrous oxide (N2O) emission was measured in a Kobresia humilis meadow and a Potentilla fruticosa meadow in the Qinghai-Tibet Plateau from June 2003 to July 2006. Five treatments were setup in the two alpine meadows. Two bare soil treatments were setup in the K. humilis meadow (BSK) and in the P. fruticosa meadow (BSP) by removing the above- and belowground plant biomass. Three plant community treatments were setup with one in the K. humilis meadow (herbaceous community in the K. humilis meadow-HCK) and two in the P. fruticosa meadow (herbaceous community in the P. fruticosa meadow-HCP, and shrub community in the P. fruticosa meadow-SCP). Nitrous oxide emission from BSP was estimated to be 38.1 +/- 3.6 mu g m(-2) h(-1), significantly higher than from BSK (30.2 +/- 2.8 mu g m(-2) h(-1)) during the whole experiment period. Rates from the two herbaceous blocks (HCK and HCP) were close to 39.5 mu g m(-2) stop h(-1) during the whole experimental period whereas shrub community (SCP) showed significant high emission rates of N2O. Annual rate of N2O emission was estimated to be 356.7 +/- 8.3 and 295.0 +/- 11.6 mg m(-2) year(-1) from the alpine P. fruticosa meadow and from the alpine K. humilis meadow, respectively. These results suggest that alpine meadows in the Qinghai-Tibetan Plateau are an important source of N2O, contributing an average of 0.3 Tg N2O year(-1). We concluded that N2O emission will decrease, due to a predicted vegetation shift from shrubs to grasses imposed by overgrazing.

Milula, a monotypic genus endemic to the Qinghai-Tibetan Plateau, was found to be nested deeply within Allium by the molecular phylogeny despite the aberrant morphology. It remains unknown what had contributed to the rapid evolution of morphology and origin of this exceptional species. In contrast to a previous report of its karyotypes with 2n = 16 = 8M+8SM (2SAT), similar to most species of Allium, a rather different karyotype, 2n = 20 = 4M +10SM+6T (2SAT), was found in examined 31 individuals from 6 populations of M. spicata distributed in the central Tibet. Karyotypes of 7 Allium species occurring in the Qinghai-Tibetan Plateau were further reported. The basic number x = 8 was confirmed for all of them and their karyotypes consist mainly of metacentric and submetacentric chromosomes with rare subterminal and terminal chromosomes. The karyotype of M. spicata is distinctly different from that of most Allium species occurring in the plateau through a complete comparison of all available species in this region and adjacent areas. However, the same chromosome number and similar karyotypic structure were found in A. fasciculatum of Sect. Bromatorrhiza, indicating a possible close relationship between them. But this similarity is contradictory to the preliminary molecular phylogenetic analysis that Milula was closely related to A. cyathophorum of Sect. Bromatorrhiza with x=8, but the other species with x=10 and 11 in this section were clearly placed in the other clade. We therefore suggested that the paralleling evolution from x=8 to x=9, 10 and 11 with increasing asymmetry of karyotype possibly due to the chromosomal Robertsonian translocation might occur separately in the two recognized phylogenetic lineages of Allium. In addition to aneuploidy and following change of the chromosomal structures, the habitat isolation due to the recent uplift of the Qinghai-Tibetan Plateau and the Quaternary climatic oscillation, plays a greater role in origin of Milula and other endemic species (genera) with aberrant morphology from their progenitors.

Fluid inclusions of protogenous halite, which were collected from two boreholes in the Charhan Salt Lake in the north part of the Qinghai-Xizang Plateau, werea nalyzed for their hydrogen and oxygen isotopes and for their Na, Mg etc. ions.On these grounds, the evolution of lake environment in this region during the last 50 000 years are discussed in this paper. The emphasis is to discuss the time range of extremely arid and cold climate at the last Glacial stage and the geological event of playa associated with such a climate.The guanidine hydrochloride method was used for measurement of hydrogen and oxygen stable isotopes. The measurement of Na, Mg etc. ions were achieved by determination of crystallization temperature of hydrohalite under microscope and then by calculation of chemical compositions of inclusion fluid using a thermodynamic model.The results obtained show that protogenous halite in the Charhan Lake area was formed in three different environment conditions: (1) In fluid inclusions of halite formed in the early period (50 000-30 000 a B. P. ), dD averages -14.9 per mil, d(18)O averages 8.37 per mil, and Mg(2+)ranges from 0.42 to 1.59 mol/L. Their plotting points fall on the right top part of the evaporation line of the present Charhan Lake area, indicating that the Lake water at that time had a higher concentration of brine, and the climate was hot and dry. (2) In fluid inclusions of halite formed in the middle period (30 000-15 000 a B. P.), SD average -66.0 per mil, d(18)O averages 1.00 pr mil, and Mg(2+) 1 mol/L. Their plotting points fall on the left low part of the evaporation line, indicating that the lake water at that time had a concentration of brine lower than that in the early period, and the environment was cold and dry. (3) In fluid inclusions of halite formed in the late period (15 000-present), dD averages 30.8 per mil, d(18)O averages 5.85 per mil, and Mg(2+) M 1 mol/L. Their plotting fall on the evaporation line, indicating that the climate environment at that time was warm and dry, almost the same as the present.The temperature variation of the last 50 000 years in the Charhan Lake area was calculated using the conversion equation proposed by Lorious et al. The time range of the Great ice age of the Last Glacial Stage is about 21 000-15 000 a B.P., which basically coincides with the time of a worldwide low sea level. The temperature in that period was below 0°C and 6-7°C lower than now. Because of lower temperatures, water supply to the lake area decreased rapidly and the concentration of lake water increased sharply. Therefore the Mg(2+) concentration in inclusion fluid reaches or closes to 2mol/L and the Mg/Na ratio varies within a very wide range. These show that the Charhan Lake at that time entered its playa stage. The Charhan Salt Lake is a typical one in the north part of the Qinghai-Xizang Plateau. It can be supposed that the extremely arid and cold climate of the Great Ice Age made most lakes in the north part of the Qinghai-Xizang Plateau enter their playa stage. This event is of importance for formation of salt resources.
