Advances in the diagnosis of Mycobacterium bovis infection in wildlife hosts may benefit the development of sustainable approaches to the management of bovine tuberculosis in cattle. In the present study, three laboratories from two different countries participated in a validation trial to evaluate the reliability and reproducibility of a real time PCR assay in the detection and quantification of M. bovis from environmental samples. The sample panels consisted of negative badger faeces spiked with a dilution series of M. bovis BCG Pasteur and of field samples of faeces from badgers of unknown infection status taken from badger latrines in areas with high and low incidence of bovine TB (bTB) in cattle. Samples were tested with a previously optimised methodology. The experimental design involved rigorous testing which highlighted a number of potential pitfalls in the analysis of environmental samples using real time PCR. Despite minor variation between operators and laboratories, the validation study demonstrated good concordance between the three laboratories: on the spiked panels, the test showed high levels of agreement in terms of positive/negative detection, with high specificity (100%) and high sensitivity (97%) at levels of 10(5) cells g(-1) and above. Quantitative analysis of the data revealed low variability in recovery of BCG cells between laboratories and operators. On the field samples, the test showed high reproducibility both in terms of positive/negative detection and in the number of cells detected, despite low numbers of samples identified as positive by any laboratory. Use of a parallel PCR inhibition control assay revealed negligible PCR-interfering chemicals co-extracted with the DNA. This is the first example of a multi-laboratory validation of a real time PCR assay for the detection of mycobacteria in environmental samples. Field studies are now required to determine how best to apply the assay for population-level bTB surveillance in wildlife.

Almeida Garrett engajou-se como liberal participando dos acontecimentos políticos de seu país. Após a Revolução de Setembro de 1836, Passos Manuel convida-o para estruturar o teatro português, tendo em vista que é considerado um meio de civilização. Garrett elabora um plano baseado em três pontos fundamentais: a construção de um edifício (futuro Teatro Nacional D. Maria II, inaugurado em 1846); uma escola voltada para formação artística e a criação de um repertório dramático nacional e moderno. Aprovado pela rainha D. Maria II em Decreto de 15 de novembro de 1836, nomeado Inspetor-Geral dos Teatros, para coordenar as atividades, põe em prática o projeto até ser demitido em julho de 1841. A proposta deste trabalho é analisar o teatro garrettiano, sob o ponto de vista político, do período de 1838 a 1843, por esta ser uma fase de intensa atividade do autor na tentativa de restaurar a cena portuguesa. Durante los siglos XVII y XVIII se presentaron varias querellas ante el Tribunal de Justicia Criminal del Nuevo Reino de Granada, en las que se denunciaba que había personas que ejercían los oficios médicos sin tener títulos que los acreditaran como facultativos en las artes curativas. Por ese entonces, se creía que quienes utilizaban yerbas y conjuros como métodos terapéuticos, por lo general mujeres, debían ser juzgadas como yerbateras-envenenadoras, porque no pretendían curar sino matar a quien consumiera sus preparados. El texto establece que los procesos criminales por envenenamiento constituyen un prisma en el que convergen diferentes problemáticas del periodo colonial neogranadino, relacionadas con la salud, los oficios médicos, las enfermedades, las creencias mágico-religiosas, el ideal de mujer en la época, la delincuencia, y las dinámicas de las instituciones españolas, entre otras. De esta manera, se estudió cómo fue la relación entre los aspectos jurídicos, las leyes criminales (dictadas por la Corona) y las conductas “desviadas” (relacionadas con el crimen por envenenamiento) de los habitantes del Nuevo Reino de Granada, entre los siglos XVII y XVIII. Para ello se revistaron desde diferentes perspectivas, varios temas del mundo colonial neogranadino, relacionados con los rumores, la comidilla, los chismes y la importancia de la comunicación hablada en el virreinato; el problema de la honra, como una de las virtudes más sobresalientes de la época y las creencias de la cultura popular con relación al envenenamiento y los diferentes métodos curativos.

David Phips wrote this letter to Colonel Jonathan Snelling from Cambridge on July 12, 1773, to inform him that Massachusetts Governor Thomas Hutchinson had requested the accompaniment of guards during his travels from Milton to Cambridge on July 21, 1773, to attend the Harvard College Commencement exercises. In the letter, Phips informs Snelling that he has issued warrants to the guards, instructing them to congregate at the Sign of the Grey Hound in Roxbury, Massachusetts at eight o'clock on the morning of the 21st. He explains that twelve other men will march, under the command of Sub-Brigadier Sumner, to the Governor's home in Milton to escort him to Roxbury, where the larger party will assemble. These heightened security measures were certainly prompted by political unrest, although this is not stated explicitly in the letter. Phips concludes by saying: "I shall order a dinner for us at Bradish's, where I hope to have the pleasure to dine with you."

Small pen-and-ink and watercolor drawing of Cambridge Green created by Harvard senior John Davis, presumably as part of his undergraduate mathematical coursework. The map surveys Cambridge Commons and includes a few rough outlines of College buildings and the Episcopal church, and notes the burying ground, and the roads to Charlestown, Menotomy, the pond, Watertown, and the bridge. The original handwritten text is faded and was annotated with additional text by Davis including the note "[taken in my Senior year at H. College Septr 1780] Surveyed in concert with classmates, Atkins, Hall 1st, Howard, Payne, &c.- J. Davis." There is a note that "Atkins afterwards took the name of Tying." Davis refers to Dudley Atkins Tyng, Joseph Hall, Bezaleel Howard, and Elijah Paine, all members of the Harvard Class of 1781.
