The nerve terminals of intrinsic muscular fibers of the tongue of adult wistar rats was studied by using silver impregnation techniques, transmission electron microscopy (TEM), and high resolution scanning electron microscopy (HRSEM) to observe the nerve fibers and their terminals. Silver impregnation was done according to Winkelman and Schmit, 1957. For TEM, small blocks were fixed in modified Karnovsky solution, postfixed in 1% buffered osmium tetroxide solution, and embedded in Spurr resin. For HRSEM, the parts were fixed in 2% osmium tetroxide solution with 1/15 M sodium phosphate buffer (pH 7.4) at 4 degrees C for 2 h, according to the technique described by Tanaka, 1989. Thick myelinated nerve bundles were histologically observed among the muscular fibers. The intrafusal nerve fiber presented a tortuous pathway with punctiform terminal axons in clusters contacting the surface of sarcolemma. Several myelinated nerve fibers involved by collagen fibers of the endoneurium were observed in HRSEM in three-dimensional aspects. The concentric lamellae of the myelin sheath and the axoplasm containing neurofilaments interspersed among the mitochondria were also noted. In TEM, myofibrils, mitochondria, rough endoplasmic reticulum, Golgi`s apparatus, and glycogen granules were observed in sarcoplasm. It is also noted that the sarcomeres constituted by myofilaments with their A, I, and H bands and the electron dense Z lines. In areas adjacent to muscular fibers, there were myelinated and unmyelinated nerve fibers involved by endoneurium and perineurium. In the region of the neuromuscular junction, the contact with the sarcolemma of the muscular cell occurs forming several terminal buttons and showing numerous evaginations of the cell membrane. The apparent L-[H-3]glutamate uptake rate (v') was measured in synaptic vesicles isolated from cerebral cortex synaptosomes prepared from autopsied Alzheimer and non-Alzheimer dementia cases, and age-matched controls. The initial synaptosome preparations exhibited similar densities of D-[H-3]aspartate membrane binding sites (B-MAX values) in the three groups. In control brain the temporal cortex D-[H-3]aspartate B-MAX was 132% of that in motor cortex, parallel with the L- [H-3]glutamate v' values (temporal = 139% of motor; NS). Unlike D- [H-3]aspartate B-MAX values, L- [H-3]glutamate v' values were markedly and selectively lower in Alzheimer brain preparations than in controls, particularly in temporal cortex. The difference could not be attributed to differential effects of autopsy interval or age at death. Non-Alzheimer dementia cases resembled controls. The selective loss of vesicular glutamate transport is consistent with a dysfunction in the recycling of transmitter glutamate.

Total petroleum hydrocarbons (TPH) are important environmental contaminants which are toxic to human and environmental receptors. Several analytical methods have been used to quantify TPH levels in contaminated soils, specifically through infrared spectrometry (IR) and gas chromatography (GC). Despite being two of the most used techniques, some issues remain that have been inadequately studied: a) applicability of both techniques to soils contaminated with two distinct types of fuel (petrol and diesel), b) influence of the soil natural organic matter content on the results achieved by various analytical methods, and c) evaluation of the performance of both techniques in analyses of soils with different levels of contamination (presumably non-contaminated and potentially contaminated). The main objectives of this work were to answer these questions and to provide more complete information about the potentials and limitations of GC and IR techniques. The results led us to the following conclusions: a) IR analysis of soils contaminated with petrol is not suitable due to volatilisation losses, b) there is a significant influence of organic matter in IR analysis, and c) both techniques demonstrated the capacity to accurately quantify TPH in soils, irrespective of their contamination levels.

O transporte marítimo sempre assumiu uma importância primordial no transporte, quer de pessoas como de mercadorias. O uso das vias marítimas para o transporte de mercadorias é uma prática utilizada desde os tempos antigos. O transporte marítimo na Europa sempre foi uma forma eficaz e de baixo custo de transportar passageiros e mercadorias de um local para outro. Atualmente, todos os modos de transporte necessitam de melhorar a sua eficiência e reduzir o montante de poluentes que libertam para o meio-ambiente. O transporte marítimo de curta distância (TMCD) é parte essencial no sistema de transporte multimodal Europeu representando cerca de 37% das transações intracomunitárias (tkm). Os resultados alcançados advêm desde que, em 2001, a European Shortsea Network (ESN) em parceria com os Shortsea Promotion Centres (SPC) de cada Estado-Membro da União Europeia (EU) conseguiram efetuar progressos significativos na promoção e desenvolvimento do transporte por esta via. Esta tese de mestrado pretende caraterizar o TMCD na EU e em Portugal e a sua articulação com os outros meios de transporte de mercadorias. Será também analisada a situação Portuguesa ao nível do transporte marítimo de mercadorias fazendo especial relevância à mercadoria contentorizada, às infraestruturas atualmente existentes em Portugal assim como às perspetivas futuras para o setor. A metodologia utilizada inclui a revisão de literatura da investigação efetuada sobre o setor, a recolha de informação primária através do instrumento inquérito e a recolha de informação estatística a partir de fontes secundárias, permitindo a caraterização do setor. O inquérito foi desenvolvido e partilhado com mais de um milhar de empresas a operar em Portugal, nas que se incluem as empresas transportadoras, operadores logísticos, agentes de navegação e as principais empresas exportadoras/importadoras a operar em Portugal, tendo-se obtido 106 respostas válidas. A nível nacional, o inquérito realizado permitiu confirmar que os principais portos logísticos para a movimentação internacional de mercadorias são os portos de Leixões, Lisboa, Sines e Setúbal, estando a maioria das empresas inquiridas localizadas no hinterland destes portos. O inquérito permitiu também concluir que o facto de o transporte rodoviário ser um meio de transporte rápido, de baixo custo para curtas e médias distâncias, de elevada frequência de transporte, de oferecer elevada capacidade e diversos tipos de carga transportada, de oferecer serviços Porta-a-Porta, da elevada flexibilidade e mobilidade oferecida, assim como a rapidez das operações de carga e descarga continuam a contribuir para a elevada competitividade deste modo de transporte de mercadorias, sendo as razões pelo qual este modo é escolhido, pese embora, ser o modo mais poluente por tonelada transportada. Os modos de transporte TMCD e ferroviário apenas são alvo de escolha, como modo de transporte de mercadorias, por parte das empresas mais responsáveis socialmente e ambiental. O inquérito permitiu ainda concluir que, para haver uma melhor articulação entre o TMCD com outros meios de transporte, é necessário que sejam disponibilizados novos serviços, portos e terminais logísticos com serviços privatizados, assim como a redução das tarifas aplicáveis TMCD.

Ship tracking systems allow Maritime Organizations that are concerned with the Safety at Sea to obtain information on the current location and route of merchant vessels. Thanks to Space technology in recent years the geographical coverage of the ship tracking platforms has increased significantly, from radar based near-shore traffic monitoring towards a worldwide picture of the maritime traffic situation. The long-range tracking systems currently in operations allow the storage of ship position data over many years: a valuable source of knowledge about the shipping routes between different ocean regions. The outcome of this Master project is a software prototype for the estimation of the most operated shipping route between any two geographical locations. The analysis is based on the historical ship positions acquired with long-range tracking systems. The proposed approach makes use of a Genetic Algorithm applied on a training set of relevant ship positions extracted from the long-term storage tracking database of the European Maritime Safety Agency (EMSA). The analysis of some representative shipping routes is presented and the quality of the results and their operational applications are assessed by a Maritime Safety expert.

L’objectiu de l’estudi va ser determinar si la força de prensió manual (FPM) era factor pronòstic de mortalitat en malalts amb càncer avançant ingressats a una Unitat de cures pal•liatives (UCP). Es va determinar la força de prensió a l’ingrés i l’evolució de la FPM els primers 12 dies mitjançant un dinamòmetre. Ni la FPM inicial ni a l’evolució va demostrar diferències significatives entre els malalts donats d’alta vius i els que van morir a la UCP. Conclusions: la determinació de FPM no va ser factor pronòstic de supervivència en malalts ingressats per càncer en una UCP.

Several evidences suggest that astrocytes release small transmitter molecules, peptides, and protein factors via regulated exocytosis, implying that they function as specialized neurosecretory cells. However, very little is known about the molecular and functional properties of regulated secretion in astrocytes in the adult brain. Establishing these properties is central to the understanding of the communication mode(s) of these cells and their role(s) in the control of synaptic functions and of cerebral blood flow. In this study, we have set-up a high-resolution confocal microscopy approach to distinguish protein expression in astrocytic structures and neighboring synaptic terminals in adult brain tissue. This approach was applied to investigate the expression pattern of core SNARE proteins for vesicle fusion in the dentate gyrus and CA1 regions of the mouse hippocampus. Our comparative analysis shows that astrocytes abundantly express, in their cell body and main processes, all three protein partners necessary to form an operational SNARE complex but not in the same isoforms expressed in neighbouring synaptic terminals. Thus, SNAP25 and VAMP2 are absent from astrocytic processes and typically concentrated in terminals, while SNAP23 and VAMP3 have the opposite expression pattern. Syntaxin 1 is present in both synaptic terminals and astrocytes. These data support the view that astrocytes in the adult hippocampus can communicate via regulated exocytosis and also indicates that astrocytic exocytosis may differ in its properties from action potential-dependent exocytosis at neuronal synapses, as it relies on a distinctive set of SNARE proteins.

El nostre projecte consisteix en l’anàlisi econòmic de les polítiques públiques de gestió de residus exclusivament municipals. Pretenem, mitjançant l’anàlisi d’un municipi representatiu com és Sant Cugat, dissenyar un sistema de reciclatge -adaptat a aquesta localitat en concret- que sigui el més eficaç possible, incrementant les quantitats recollides i controlant els costos tot i tenint en compte les preferències dels propis usuaris.Amb això, pretenem cercar la fórmula més exacta possible, adaptable a cada tipus de municipi (edificació vertical o horitzontal, extensió, densitat de població, etc.), per tal que els nivells de reciclatge del nostre país arribin a ser equiparables als europeus, avui encara molt llunyans.Tot i que valorem molts sistemes de reciclatge ja existents, ens centrem prioritàriament en l’estudi del Porta A Porta (PAP), el qual creiem el més adequat per a Sant Cugat després d’haver analitzat estadísticament el que opinen els ciutadans mitjançant unes enquestes, així com dades oficials de poblacions on ja hi està implementat.No obstant això, després d’haver comprovat que la satisfacció real dels usuaris del PAP a Vilassar de Mar, on hi funciona des de 2003, no és la que seria d’esperar, hem reflexionat sobre les causes i les solucions possibles a aquests problemes, de manera que ho puguem evitar de cara a crear el sistema per a la població objecte del nostre treball.
