Rocuronium (ROC) is a neuromuscular blocking agent used in surgical procedures which is eliminated primarily by biliary excretion. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for analysis of ROC in human plasma. Separation of ROC and IS (verapamil) was performed using an endcapped C-18 column and a mixture of water:acetonitrile:trifluoracetic acid (50:50:0.1, v/v) as mobile phase. Aliquots of 100 mu L of human plasma were extracted at pH 3, using dichloromethane. The lower limit of quantification of 5 ng/mL shows the high sensitivity of this method. Intra- and inter-assay precision (as relative standard deviation) was all <= 14.2% and accuracy (as relative standard error) did not exceed 10.1%. The validated method was successfully applied to quantify ROC concentrations in patients under surgical procedures up to 6 h after the administration of the 0.4-0.9 mg/kg ROC. The pharmacokinetic parameter estimations of ROC showed AUC/dose of 563 mu g min/mL, total clearance of 2.5 mL/min/kg, volume of distribution at steady state of 190 mL/kg and mean residence time of 83 min. An high performance liquid chromatography (HPLC) method for the enantioselective determination of donepezil (DPZ), 5-O-desmethyl donepezil (5-ODD), and 6-O-desmethyl donepezil (6-ODD) in Czapek culture medium to be applied to biotransformation studies with fungi is described for the first time. The HPLC analysis was carried out using a Chiralpak AD-H column with hexane/ethanol/methanol (75:20:5, v/v/v) plus 0.3 % triethylamine as mobile phase and UV detection at 270 nm. Sample preparation was carried out by liquid-liquid extraction using ethyl acetate as extractor solvent. The method was linear over the concentration range of 100-10,000 ng mL(-1) for each enantiomer of DPZ (r a parts per thousand yenaEuro parts per thousand 0.9985) and of 100-5,000 ng mL(-1) for each enantiomer of 5-ODD (r a parts per thousand yenaEuro parts per thousand 0.9977) and 6-ODD (r a parts per thousand yenaEuro parts per thousand 0.9951). Within-day and between-day precision and accuracy evaluated by relative standard deviations and relative errors, respectively, were lower than 15 % for all analytes. The validated method was used to assess DPZ biotransformation by the fungi Beauveria bassiana American Type Culture Collection (ATCC) 7159 and Cunninghamella elegans ATCC 10028B. Using the fungus B. bassiana ATCC 7159, a predominant formation of (R)-5-ODD was observed while for the fungus C. elegans ATCC 10028B, DPZ was biotransformed to (R)-6-ODD with an enantiomeric excess of 100 %.

Mitogen-activated protein kinase (MAPK) cascades are conserved eukaryotic signaling modules consisting of a MAPK, a MAPKK and a MAP3K. MAPK cascades are involved in many cellular responses including proliferation, differentiation, apoptosis, stress and immune responses. ^ The first part of this thesis describes the cloning and biochemical analysis of JNKK2, a member of MAPKK gene family. Our results demonstrate that JNKK2 is a specific JNK activator and activates the JNK-dependent signal transduction pathway in vivo by inducing c-Jun and ATF2-mediated gene expression. We also found that JNKK2 is specifically activated by a MAP3K MEKK2 through formation of MEKK2-JNKK2-JNK1 triple complex module. JNKK2 is likely to mediate specific upstream signals to activate JNK cascade. ^ The second part of this thesis describes biochemical and gene disruption analysis of MEKK3, a member of MAP3K gene family. We showed that overexpression of MEKK3 strongly activates both JNK and p38 MAPKs but only weakly activates ERK. MEKK−/− embryos die at about embryonic day (E) 11. MEKK3−/− embryos displayed defects in blood vessel development in the yolk sacs, and in the myocardium and endocardium development at E9.5. The angiogenesis in the head, intersomitic region and placenta was also abnormal. These results demonstrate that MEKK3, a member of MAP3K MEKK/STE11 subgene family, is essential for early embryonic cardiovascular development. Furthermore, it was found that disruption of MEKK3 did not alter the expression of vascular endothelial growth factor-1 (VEGF-1), angiopoietin-1, -2 and their respective receptors Flt-1, Flk-1, Tie-1, Tie-2. Finally, MEKK3 was shown to activate myocyte-specific enhancer factor 2C (MEF2C), a crucial transcription factor for early embryonic cardiovascular development through the p38 MAPK cascade, suggesting that MEF2C is one of the key targets of the MEEKK3 signaling pathway during early embryonic cardiovascular development. ^

A selenium-containing catalytic antibody (Se-4A4), prepared by converting reactive serine residues of a monoclonal antibody (4A4) raised against a GSH derivative into selenocysteines, acts as a mimic of cytosolic glutathione peroxidase (cGPX). To clarify the mechanism of action of this catalytic antibody, detailed studies on kinetic behaviour and biological activity were carried out. A rate of acceleration (k(cat)/K-m/k(uncat)) 10(7)-fold that of the uncatalytic reaction is observed. Under similar conditions, the turnover number (k(cat)) of Se-4A4 is 42% of that of the natural rabbit liver cGPX. The Se-4A4 reaction involves a Ping Pong mechanism, which is the same as that of the natural cGPX. The selenocysteine residue is located in the binding site of the antibody and is shown to be crucial for this activity. Of the thiol compounds tested, only GSH is able to serve as substrate for Se-4A4. It was demonstrated, using the free-radical-damage system (hypoxanthine/xanthine oxidase) of cardiac mitochondria, that Se-4A4 can protect mitochondria from free-radical damage at least 10(4)-fold more effectively than the natural cGPX.

Macroporosity(>100µm) in bone void fillers is a known prerequisite for tissue regeneration, but recent literature has highlighted the added benefit of microporosity(0.5 - 10µm). The aim of this study was to compare the in vitro performances of a novel interconnective microporous hydroxyapatite (HA) derived from red algae to four clinically available macroporous calcium phosphate (CaP) bone void fillers. The use of algae as a starting material for this novel void filler overcomes the issue of sustainability, which overshadows continued use of scleractinian coral in the production of some commercially available materials, namely Pro-OsteonTM and Bio-Coral®. This study investigated the physicochemical properties of each bone voidfiller material using x-ray diffraction, fourier transform infrared spectroscopy, inductive coupled plasma, and nitrogen gas absorption and mercury porosimetry. Biochemical analysis, XTT, picogreen and alkaline phosphatase assays were used to evaluate the biological performances of the five materials. Results showed that algal HA is non-toxic to human foetal osteoblast (hFOB) cells and supports cell proliferation and differentiation. The preliminary in vitro testing of microporous algal-HA suggests that it is comparable to the four clinically approved macroporous bone void fillers tested. The results demonstrate that microporous algal HA has good potential for use in vivo and in new tissue engineered strategies for hard tissue repair.

Detection of growth-promoter use in animal production systems still proves to be an analytical challenge despite years of activity in the field. This study reports on the capability of NMR metabolomic profiling techniques to discriminate between plasma samples obtained from cattle treated with different groups of growth-promoting hormones (dexamethasone, prednisolone, oestradiol) based on recorded metabolite profiles. Two methods of NMR analysis were investigated—a Carr–Purcell–Meiboom–Gill (CPMG)-pulse sequence technique and a conventional 1H NMR method using pre-extracted plasma. Using the CPMG method, 17 distinct metabolites could be identified from the spectra. 1H NMR analysis of extracted plasma facilitated identification of 23 metabolites—six more than the alternative method and all within the aromatic region. Multivariate statistical analysis of acquired data from both forms of NMR analysis separated the plasma metabolite profiles into distinct sample cluster sets representative of the different animal study groups. Samples from both sets of corticosteroid-treated animals—dexamethasone and prednisolone—were found to be clustered relatively closely and had similar alterations to identified metabolite panels. Distinctive metabolite profiles, different from those observed within plasma from corticosteroid-treated animal plasma, were observed in oestradiol-treated animals and samples from these animals formed a cluster spatially isolated from control animal plasma samples. These findings suggest the potential use of NMR methodologies of plasma metabolite analysis as a high-throughput screening technique to aid detection of growth promoter use.

The loggerhead turtle (Caretta caretta) is an endangered marine reptile for whom assessing population health requires knowledge of demographic parameters such as individual growth rate. In Cape Verde, as within several populations, adult female loggerhead sea turtles show a size-related behavioral and trophic dichotomy. While smaller females are associated with oceanic habitats, larger females tend to feed in neritic habitats, which is reflected in their physiological condition and in their offspring. The ratio of RNA/DNA provides a measure of cellular protein synthesis capacity, which varies depending on changes in environmental conditions such as temperature and food availability. The purpose of this study was to evaluate the combined use of morphometric data and biochemical indices as predictors of the physiological condition of the females of distinct sizes and hatchlings during their nesting season and how temperature may influence the physiological condition on the offspring. Here we employed biochemical indices based on nucleic acid derived indices (standardized RNA/DNA ratio-sRD, RNA concentration and DNA concentration) in skin tissue as a potential predictor of recent growth rate in nesting females and hatchling loggerhead turtles. Our major findings were that the physiological condition of all nesting females (sRD) decreased during the nesting season, but that females associated with neritic habitats had a higher physiological condition than females associated with oceanic habitats. In addition, the amount of time required for a hatchling to right itself was negatively correlated with its physiological condition (sRD) and shaded nests produced hatchlings with lower sRD. Overall, our results showed that nucleic acid concentrations and ratios of RNA to DNA are an important tool as potential biomarkers of recent growth in marine turtles. Hence, as biochemical indices of instantaneous growth are likely temperature-, size- and age-dependent, the utility and validation of these indices on marine turtles stocks deserves further study.

En el presente estudio se analizan los efectos de los fotoperiodos ambientales de otoño e invierno y los fotoperiodos experimentales de 24, 12 y 0 horas de luz artificial sobre la calidad del semen de machos reproductores porcinos de raza Landrace. El estudio se realizó sobre 30 machos postpuberales de 8 meses de edad y testados con el fin de comprobar la homogeneidad. Los machos fueron distribuidos aleatóriamente en los 3 grupos de luz artificial durante 3 meses. El tratamiento de 12 horas de luz artificial fue considerado como grupo control. Previamente al inicio de cada tratamiento, se sometió a cada grupo de machos al fotoperiodo ambiental correspondiente a la época del año; así, se caracterizó también la calidad seminal en otoño e invierno, manteniendo la temperatura constante. La nave experimental que acogió a los machos tiene una superficie de 100 m2 y una altura de 3,5 m. Un pasillo central divide la nave en dos hileras de 5 y 6 celdas respectivamente. En una de las celdas pequeñas se instaló el maniquí y fue utilizada para las extracciones de semen. La iluminación artificial se consiguió con la instalación de 6 lámparas fluorescentes en el techo del pasillo central que proporcionaron una luz homogénea superior a 200 lux. Así mismo, la nave se mantuvo en todo momento a 21±1ºC y la humedad relativa osciló entre el 60-75%. A todos los verracos se les proporcionó una dieta nutritiva y equilibrada y se les sometió a un regimen de extracciones de semen de 2 veces por semana, habiendo sido previamente entrenados en la monta del maniquí. Les muestras de semen fueron recogidas según la técnica de la mano enguantada (Martín, 1982; Daza, 1992) y se analizaron los siguientes parámetros: el volumen y el pH seminales, la concentración, la vitalidad y la motilidad espermáticas, la resistencia acrosómica de los espermatozoides, la morfología espermàtica a partir de la frecuencia de los espermatozoides maduros, inmaduros y aberrantes, la producción testicular y el número de dosis seminales. Se analizó, además, bioquímicamente el plasma seminal al principio y al final de cada tratamiento experimental de luz artificial a partir de la concentración de proteína total, de la identificación de residuos fosforilados de proteína y del contenido de azúcares. También se determinaron los índices de fertilidad y prolificidad. El volumen y el pH de los eyaculados se utilizaron como marcadores del estado funcional de las glándulas sexuales accesorias; la concentración espermàtica como un indicador de la actividad testicular (Pinart y col., 1999). La vitalidad y la motilidad espermáticas fueron estimadores del grado de diferenciación del espermatozoide tanto a nivel testicular como epididimario; la resistencia acrosómica fue utilizada para valorar el nivel de diferenciación de la membrana acrosómica durante la espermiogénesis y/o maduración epididimària (Briz i col., 1996; Pinart i col., 1999). Referente a la morfología espermática, los espermatozoides inmaduros fueron marcadores de anomalías en la maduración de éstos a lo largo del conducto epididimario y los espermatozoides aberrantes se utilizaron como marcadores de una diferenciación defectuosa a nivel de testículo (anomalías primarias) y a nivel de conducto epididimario (anomalías secundarias) (Briz i col., 1996). La concentración de proteína total se utilizó para valorar la integridad funcional de las membranas del espermatozoide y la actividad de las glándulas sexuales accesorias. La identificación de proteínas con residuos de tirosina fosforilados fue un estimador de la viabilidad celular y la actividad de las glándulas sexuales, y el contenido de azúcares como un indicador de la producción de las vesículas seminales. La determinación del volumen y el pH de los eyaculados se realizó en las instalaciones de la granja a partir de semen fresco el mismo día de la extracción. El resto de parámetros se analizaron en el laboratorio durante las 48 horas posteriores a la extracción a partir de semen diluido en BTS (diluyente de Bestville) (Daza, 1992) y transportado y conservado a 15ºC. Las muestras fueron previamente filtradas con el fin de eliminar la tapioca. El estudio estadístico de los resultados obtenidos se realizó a partir del análisis de la varianza (ANOVA) con un nivel de significación de =0,05. En cuanto al estudio comparativo de los fotoperiodos ambientales estacionales se ha observado un incremento significativo del pH del eyaculado en los machos expuestos a otoño (P0,0001), mientras que el volumen seminal se mantiene en valores similares en ambos tratamientos (P=0,1650). La concentración espermàtica, la producción espermàtica y el número de dosis seminales que se pueden preparar a partir de un eyaculado se duplica en los verracos sometidos al fotoperiodo de primavera (P0,0001). La vitalidad y la motilidad espermáticas no experimentan cambios significativos entre tratamientos (P=0,3440 y P=0,9220, respectivamente). La resistencia osmótica de los acrosomas desciende únicamente en los machos expuestos a condiciones estacionales de otoño (P0,0001). En referencia a la morfología espermàtica aunque no se observan diferencias entre primavera y otoño (P0,05), sí se detecta un incremento de los porcentajes de espermatozoides inmaduros y aberrantes en ambos fotoperiodos estacionales, y en especial en los machos expuestos a condiciones fotoperiódicas de otoño. Según los resultados obtenidos en este estudio la calidad seminal de los verracos es inferior en el fotoperiodo de otoño debido a un descenso de la concentración y la producción espermáticas, un aumento del pH seminal, una disminución de la resistencia de la membrana acrosómica y a un incremento en la frecuencia de espermatozoides inmaduros y aberrantes. Parece ser, pues, que en el otoño tiene lugar la disminución de la producción testicular, cambios en la actividad de las glándulas sexuales accesorias y disfunciones en el proceso de diferenciación testicular y epididimària de los espermatozoides y especialmente del acrosoma. En relación a los resultados obtenidos en el estudio de los diferentes fotoperiodos artificiales se observa que la iluminación continua provoca un aumento significativo del volumen del eyaculado en el primer y segundo mes de tratamiento (P0,0001), disminuyendo en el tercer mes. La oscuridad absoluta no modifica este parámetro (P0,05). En cuanto al pH seminal la iluminación continua provoca un incremento progresivo del valor del pH a lo largo del periodo experimental (P0,0001), mientras que la oscuridad absoluta tiene un efecto más irregular. La exposición de los machos a iluminación continua y a oscuridad absoluta se manifiesta en un descenso de la concentración y la producción espermáticas que se mantiene hasta el segundo mes de tratamiento (P0,0001), observándose un incremento en el tercer mes de exposición de los machos a oscuridad absoluta (P=0,1010). De todas maneras, este descenso es mas severo en los machos sometidos a iluminación continua ya que no presentan recuperación. La vitalidad y la motilidad espermáticas no se ven alteradas por la iluminación continua y la oscuridad absoluta, ni tampoco el contenido de los azúcares mayoritarios del plasma seminal (P0,005). La glucosa aparece como un azúcar minoritario y sí que presenta concentraciones inferiores en los tratamientos experimentales de luz continua y de oscuridad absoluta (P0,0001 y P=0,0002, respectivamente). La resistencia osmótica de los acrosomas desciende en ambos tratamientos artificiales extremos de luz continua y oscuridad total (P0,0001), aunque en los machos expuestos a iluminación continua se produce una recuperación a partir del segundo mes de tratamiento (P=0,4930). Dado que tampoco se han observado diferencias significativas en las concentraciones de proteína total (P0,05), es probable que las anomalías de la membrana acrosómica se originen durante el proceso de espermiogénesis y/o maduración epididimària. La exposición de los verracos a oscuridad absoluta no altera la morfología espermàtica de los eyaculados, aunque se observa un aumento de la frecuencia de espermatozoides con anomalías en la forma de la cola en el primer mes (P0,0001), y un aumento de la frecuencia de espermatozoides inmaduros con gota distal y de espermatozoides con anomalías en el número de colas en el tercer mes de experimentación (P=0,0030 y P0,0001). La luz continua, sin embargo, provoca un incremento de la frecuencia de espermatozoides inmaduros con gota distal (P0,0001) y de espermatozoides con anomalías en la forma de la cola (P=0,0040) ya en el primer mes. El fotoperiodo provoca un descenso de la fertilidad de los machos expuestos a oscuridad absoluta en el tercer mes de tratamiento (P0,0001) y un incremento de ésta en los machos sometidos a iluminación continua (P=0,0005). La prolificidad no se ve modificada por ambas condiciones extremas de luz artificial (P0,05). Así pues, los resultados obtenidos demuestran que el fotoperiodo afecta la actividad testicular, provoca alteraciones en la actividad de las glándulas sexuales accesorias, altera el proceso de expulsión de la gota citoplasmática y provoca anomalías en el proceso de diferenciación de la cola tanto a nivel testicular como epididimario, siendo los verracos expuestos a luz continua más sensibles a estos parámetros que los verracos sometidos a oscuridad absoluta. El fotoperiodo, sin embargo, no altera de forma esencial la integridad de las membranas del espermatozoide ni la capacidad fecundante de éste.

This review article addresses recent advances in the analysis of foods and food components by capillary electrophoresis (CE). CE has found application to a number of important areas of food analysis, including quantitative chemical analysis of food additives, biochemical analysis of protein composition, and others. The speed, resolution and simplicity of CE, combined with low operating costs, make the technique an attractive option for the development of improved methods of food analysis for the new millennium.

The risk of malnutrition is high among elderly population, yet few studies have measured indicators of nutritional status among Australian aged-care residents. To determine the relationship between nutritional status and bone density, hand grip strength, and the timed-up and go test, in a group of Australian aged-care residents. Anthropometric and biochemical analysis measured in subjects recruited to be part of a six month multivitamin supplementation study. One hundred and fifteen subjects participated (68% female). The mean (SD) age and body weight was 80.2(10.6) years, and 66.5(15.0) kg, respectively. Eleven percent were underweight (body mass index, BMI, <or =20.0 kg/m(2)), and 20% were obese BMI >or =30 kg/m(2)). Low serum 25-hydroxy-vitamin D (25(OH)D, <or =50 nmol/L) concentrations were found among 79% of subjects. After adjustment for body weight, there was an association between serum 25(OH)D and bone density (heel ultrasound) (r=.204, p=.027). Low serum zinc <or =10.7 micromol/L) concentrations were found among 46% of subjects; this group had a slower timed up and go time compared with those with higher zinc concentrations (n=19, 44.6 +/- 5.6 seconds vs. n=27, 30.0 +/- 3.3 seconds, p=.020). There were no associations between nutritional markers and hand grip strength. In this group, more than (3/4) of subjects had low serum 25(OH)D, and 46% had low zinc concentrations. Serum 25(OH)D was associated a lower bone density and zinc with a slower walking time. This indicates that the elderly in long term residential care facilities are at high risk for poor nutritional status, potentially increasing morbidity and mortality.

Few studies to date have examined age-related changes in markers of immune status in healthy older individuals. The immune status of 93 healthy individuals aged 55–70 years was assessed by two- and three-color flow cytometry and biochemical analysis. There were significant age effects (p ≤.05) on monocyte phagocytic activity and cluster of differentiation (CD) 3/human leukocyte antigen-D-related (HLA-DR) late-activated T lymphocytes (% expression). There was a significant (p ≤ 0.1) Age x Sex interaction in absolute counts (x 109/L) of CD3/CD8 total cytotoxic T lymphocytes (CTL), the CD4 T- helper to CD8 CTL ratio, the CD3/CD4/CD45RA naïve T helper to CD3/CD4/CD45RO memory T helper lymphocyte ratio, and interleukin (IL)-1ß (% expression) by activated monocytes. The study shows that alterations in markers of immune status occur between 55 and 70 years, and provides reference values for the lymphocyte measures in healthy men and postmenopausal women in this age group. The study further highlights the need for sex-specific reference ranges for such markers.

Background: Studies have confirmed that the rate of mental illness is no higher in rural Australians than that of urban Australians. However, the rate of poor mental health outcomes, and in particular suicide, is significantly raised in rural populations. This is thought to be due to lack of early diagnosis, health service access, the distance-decay effect, poor physical health determinants and access to firearms. Research conducted by the National Centre for Farmer Health between 2004 and 2009 reveals that there is a correlation between obesity and psychological distress among the farming community where suicide rates are recognised as high. Chronic stress overstimulates the regulation of the hypothalamic-pituitary-adrenal (HPA) axis that is associated with abdominal obesity. Increasing physical activity may block negative thoughts, increase social contact, positively influence brain chemistry and improve both physical and mental health. This paper describes the design of the Farming Fit study that aims to identify the effect of physical activity on psychological distress, obesity and health behaviours such as diet patterns and smoking in farm men and women.
Methods/Design: For this quasi-experimental (convenience sample) control-intervention study, overweight (Body Mass Index ≥25 kg/m2) farm men and women will be recruited from Sustainable Farm Families™ (SFF) programs held across Victoria, Australia. Baseline demographic data, health data, depression anxiety stress scale (DASS) scores, dietary information, physical activity data, anthropometric data, blood pressure and biochemical analysis of plasma and salivary cortisol levels will be collected. The intervention group will receive an exercise program and regular phone coaching in order to increase their physical activity. Analysis will evaluate the impact of the intervention by longitudinal data (baseline and post intervention) comparison of intervention and control groups.
Discussion: This study is designed to examine the effect of physical activity on psychological health and other comorbidities such as obesity, impaired glucose tolerance, hypertension and dyslipidaemia within a high-risk cohort. The outcomes of this research will be relevant to further research and service delivery programs, in particular those tailored to rural communities.

Background:
Acute kidney injury (AKI) is a major complication for infants following an asphyxic insult at birth. We aimed to determine if kidney structure and function were affected in an animal model of birth asphyxia and if maternal dietary creatine supplementation could provide an energy reserve to the fetal kidney, maintaining cellular respiration during asphyxia and preventing AKI.

Methods:
Pregnant spiny mice were maintained on normal chow or chow supplemented with creatine from day 20 gestation. On day 38 (term ~39 d), pups were delivered by cesarean section (c-section) or subjected to intrauterine asphyxia. Twenty-four hours after insult, kidneys were collected for histological or molecular analysis. Urine and plasma were also collected for biochemical analysis.

Results:
AKI was evident at 24 h after birth asphyxia, with a higher incidence of shrunken glomeruli (P < 0.02), disturbance to tubular arrangement, tubular dilatation, a twofold increase (P < 0.02) in expression of Ngal (early marker of kidney injury), and decreased expression of the podocyte differentiation marker nephrin.
