The assemblage of oligochaetes in the Liangzi Lake District, located in middle reaches of the Changjiang River, was studied from May to August, 2001. To establish species composition, richness, and abundance and detect the influence of environmental variables on oligochaete distributional patterns, 45 localities were sampled. All total, 20 species belonging to the families Naididae (eight species), Tubificidae (11 species), and Lumbriculidae (one species) were found. Branchiura sowerbyi, Tubifex sp. 1, and Aulodrilus pluriseta were the dominant species and contributed nearly 70% of the total abundance. The 45 sampling sites were separated into three groups based on composition and relative abundance of benthic oligochaete communities using two-way indictor species analysis associated with detrended correspondence analysis. Canonical correspondence analysis indicated that two plant variables (total plant cover and total submersed macrophyte biomass) were strongly correlated with the faunal gradient (p < 0.05). Other predicator variables were water depth and total nitrogen.

Comparative studies on community structure and biodiversity of macrozoobenthos were carried out in three lake areas with different states of eutrophication (mesotrophic, eutrophic and hypereutrophic) in a shallow subtropic lake (Lake Donghu) in the middle basin of the Yangtze River of China. Thirty-three taxa (including six Mollusca, five Oligochaeta, 15 Chironomidae and seven other zoobenthos) were found during February 1998 to April 1999. The results show that the more eutrophic the lake water, the lower the macrozoobenthos species diversity (measured as species number, diversity index, and K-dominant curves). Abundance of the oligochaete Limnodrilus hoffmeisteri was positively correlated with the degree of eutrophication, which was due mainly to its ability to tolerate low dissolved oxygen. The chironomid Tanypus chinensis also thrived in the hypereutrophic environment; however, it was less endurable to eutrophication than L. hoffmeisteri.

The allatostatins are a family of peptides isolated originally from the cockroach, Diploptera punctata. Related peptides have been identified in Periplaneta americana and the blowfly, Calliphora vomitoria. These peptides have been shown to be potent inhibitors of juvenile hormone synthesis in these species. A peptide inhibitor of juvenile hormone biosynthesis has also been isolated from the moth, Manduca sexta; however, this peptide has no structural homology with the D. punctata-type allatostatins. Investigations of the phylogeny of the D. punctata allatostatin peptide family have been started by examining a number of nonarthropod invertebrates for the presence of allatostatin-like molecules using immunocytochemistry with antisera directed against the conserved C-terminal region of this family. Allatostatin-like immunoreactivity (ALIR) was demonstrated in the nervous systems of Hydra oligactis (Hydrozoa), Moniezia expansa (Cestoda), Schistosoma mansoni (Trematoda), Artioposthia triangulata (Turbellaria), Ascaris suum (Nematoda), Lumbricus terrestris (Oligochaeta), Limax pseudoflavus (Gastropoda), and Eledone cirrhosa (Cephalopoda). ALIR could not be demonstrated in Ciona intestinalis (Ascidiacea). These results suggest that molecules related to the allatostatins may play an important role in nervous system function in many invertebrates as well as in insects and that they also have an ancient evolutionary lineage. Over 11 million tons of nanomaterials (NMs) have been produced in 2012 and predictions point the increase in production. Despite predictions and extended usage via consumer products and industry, the understanding of the potential impact of these materials on the environment is virtually absent. The main aim of this thesis is to understand how a selected group of nanomaterials (metal based particles) may impact soil invertebrates, with special focus on the mechanisms of response. Since a case-by-case Environmental Risk Assessment (ERA) of all the emerging contaminants (particularly NMs) is impossible, among others due to time and cost reasons, to gain understanding on the mechanism of action and response is very important to reach a common paradigm. Understanding the modes of action provides predictive characters in cross particle extrapolation. Besides, it also provides insight for the production of new and sustainable materials. Overall, the effects of the selected NMs (Copper and Silver, Titanium and Zirconium oxides) and the respective salt forms, were investigated at the gene expression (using high-throughput tools, microarray and qPCR technology), biochemical (using enzymatic assays for analysis of oxidative stress markers) and organism (survival and reproduction as in OECD test guidelines) levels, this using standard soil species (Enchytraeus albidus, Enchytraeus crypticus, Eisenia fetida). Gene expression analysis provided valuable information on the mechanisms affected by each of the NMs. The gene expression profile highlighted a (nano)material signature and the effect of the duration of exposure. The functional analyses integrated with the biochemical and organism data, revealed a good understanding power. The biochemical parameters (oxidative stress related) were distinct across the materials and also influenced by duration of exposure and concentration. The standardized organismal responses differed the least between the various materials. The overall outcome is that, in this context of NMs effect assessment, gene expression and enzymatic assays introduced a very important knowledge gap, which could not had been achieved by the standard organismal effects alone. A reoccurring issue with some metal based NMs is the possible dissolution and subsequent release of ions that then causes toxicity e.g. Cu-NPs or Ag-NPs release Cu2+ or Ag+. The oxidation state of the particles was investigated, although this was not the focus of the thesis. The study of fate, e.g. dissolution of NPs, is also only in its beginning and the appropriate techniques are currently being developed. The results showed a specific nanoparticle effect. The UV exposure with titanium dioxide nanoparticles increased its effect.

The aim of the present study was to examine the benthic fauna in a marginal pond lateral to the Paranapanema River and to identify the main controlling factors of its distribution. Considering the small size of the lacustrine ecosystem, we expected that seasonal variations of the benthic community attributes are more important than spatial variations; Methods: Two samplings, one in March and another in August, were carried out at nine sites in the pond. Sediment samples were obtained through a Van Veen grab for invertebrate sorting, granulometric analysis, and for quantification of organic matter in sediment. Other abiotic factors were measured, such as water transparency, dissolved oxygen, pH, electric conductivity, temperature, and depth of sediment sampling sites. Regarding the comparative analysis at spatial scale, no significant variations in density of the benthic invertebrate community were found. Results: In relation to the studied abiotic factors, only depth presented significant differences among sampling sites; All the measured environmental parameters presented significant differences among sampling months, except depth and the physical and chemical characteristics of the sediment. The abundance of Chaoboridae and Chironomidae was the unique attribute with a significant difference in comparing the two months. A higher abundance of taxa occurred in August, especially for Oligochaeta, Nematoda, Chaoboridae, and Chironomidae; Conclusions: Because of the low structural complexity of the studied pond, we concluded that the changes in benthic macroinvertebrate community attributes were mainly due to seasonal effects.

A comparative analysis was conducted of the colonization by benthic macroinvertebrates of rocky and leaf pack substrates, both natural and artificial. This colonization was evaluated by season, with the objective of ascertaining the influence of rainfall on the rate of colonization. The total density of macroinvertebrates after 21 days of colonization was significantly greater in the dry than in the wet season. When the substrate types were compared, artificial leaf pack substrate presented the smallest density for both seasons. In the wet season, Chironomidae, Leptohyphidae, Hydropsychidae, Elmidae, immature stages of Trichoptera, and Hydroptilidae showed a more representative density. In the dry season, Chironomidae, Baetidae and Oligochaeta were the most abundant taxa. The artificial rocky substrate used in this experiment was the most appropriate, due to its resemblance with natural substrate conditions in terms of the maintenance of the structural integrity of the substrate throughout the experimental period. Successional and seasonal effects were of great relevance, playing an important role in the colonization process.

The urea effect on the giant extracellular hemoglobin of Glossoscolex paulistus (HbGp) stability was studied by analytical ultracentrifugation (AUC) and small angle X-ray scattering (SAXS). AUC data show that the sedimentation coefficient distributions curves c (S), at 1.0mol/L of urea, display a single peak at 57 S, associated to the undissociated protein. The increase in urea concentration, up to 4.0mol/L, induces the appearance of smaller species, due to oligomeric dissociation. The sedimentation coefficients and molecular masses are 9.2S and 204kDa for the dodecamer (abcd)3, 5.5S and 69kDa for the tetramer (abcd), 4.1S and 52kDa for the trimer (abc) and 2.0 S and 17kDa for the monomer d, respectively. SAXS data show initially a decrease in the I(0) values due to the oligomeric dissociation, and then, above 4.0mol/L of denaturant, for oxy-HbGp, and above 6.0mol/L for cyanomet-HbGp, an increase in the maximum dimension and gyration radius is observed, due to the unfolding process. According to AUC and SAXS data the HbGp unfolding is described by two phases: the first one, at low urea concentration, below 4.0mol/L, characterizes the oligomeric dissociation, while the second one, at higher urea concentration, is associated to the unfolding of dissociated species. Our results are complementary to a recent report based on spectroscopic observations. © 2012 Elsevier B.V.

The thermal denaturation and aggregation of the HbGp, in the oxy- and cyanomet-forms, was investigated by DSC, AUC, DLS, optical absorption and CD, in the pH range from 5.0 to 7.0. Oxy-HbGp has a denaturation process partially reversible and dependent on the temperature. DSC melting curve is characterized by a single peak with Tc value of 333.4±0.2K for oxy-HbGp, while two peaks with Tc values of 332.2±0.1 and 338.4±0.2K are observed for cyanomet-HbGp, at pH 7.0. In acidic pH oxy- and cyanomet-HbGp are more stable showing higher Tc values and aggregation. AUC data show that, HbGp, at pH 7.0, upon denaturation, remains undissociated at 323K, presenting oligomeric dissociation at 333 (12±3% of tetramer and 88±5% of whole HbGp) and 343K (70±5% of monomer and 30±2% of trimer). DLS data show that the lag period before aggregation is dependent on the temperature and HbGp concentration. Optical absorption and CD results show that the increase of temperature leads to the oxy-HbGp oxidation and aggregation, above 331K, in acidic pH. CD data, for HbGp, present a greater thermal stability in acid medium than at neutral pH, with similar Tc values for both oxidation forms. Our data are consistent with previous studies and represents an advance in understanding the thermal stability of oligomeric HbGp structure. © 2012 Elsevier B.V.
