Approximate Bayesian computation (ABC) is a highly flexible technique that allows the estimation of parameters under demographic models that are too complex to be handled by full-likelihood methods. We assess the utility of this method to estimate the parameters of range expansion in a two-dimensional stepping-stone model, using samples from either a single deme or multiple demes. A minor modification to the ABC procedure is introduced, which leads to an improvement in the accuracy of estimation. The method is then used to estimate the expansion time and migration rates for five natural common vole populations in Switzerland typed for a sex-linked marker and a nuclear marker. Estimates based on both markers suggest that expansion occurred < 10,000 years ago, after the most recent glaciation, and that migration rates are strongly male biased.

Background: Suppressor of cytokine signaling 3 (SOCS3) is an inducible endogenous negative regulator of signal transduction and activator of transcription 3 (STAT3). Epigenetic silencing of SOCS3 has been shown in head and neck squamous cell carcinoma (HNSCC), which is associated with increased activation of STAT3. There is scarce information on the functional role of the reduction of SOCS3 expression and no information on altered subcellular localization of SOCS3 in HNSCC.Methodology/Principal Findings: We assessed endogenous SOCS3 expression in different HNSCC cell lines by RT-qPCR and western blot. Immunofluorescence and western blot were used to study the subcellular localization of endogenous SOCS3 induced by IL-6. Overexpression of SOCS3 by CMV-driven plasmids and siRNA-mediated inhibition of endogenous SOCS3 were used to verify the role of SOCS3 on tumor cell proliferation, viability, invasion and migration in vitro. In vivo relevance of SOCS3 expression in HNSCC was studied by quantitative immunohistochemistry of commercially-available tissue microarrays. Endogenous expression of SOCS3 was heterogeneous in four HNSCC cell lines and surprisingly preserved in most of these cell lines. Subcellular localization of endogenous SOCS3 in the HNSCC cell lines was predominantly nuclear as opposed to cytoplasmic in non-neoplasic epithelial cells. Overexpression of SOCS3 produced a relative increase of the protein in the cytoplasmic compartment and significantly inhibited proliferation, migration and invasion, whereas inhibition of endogenous nuclear SOCS3 did not affect these events. Analysis of tissue microarrays indicated that loss of SOCS3 is an early event in HNSCC and was correlated with tumor size and histological grade of dysplasia, but a considerable proportion of cases presented detectable expression of SOCS3.Conclusion: Our data support a role for SOCS3 as a tumor suppressor gene in HNSCC with relevance on proliferation and invasion processes and suggests that abnormal subcellular localization impairs SOCS3 function in HNSCC cells.

Background: the soil fungus Rhizoctonia solani anastomosis group 3 (AG-3) is an important pathogen of cultivated plants in the family Solanaceae. Isolates of R. solani AG-3 are taxonomically related based on the composition of cellular fatty acids, phylogenetic analysis of nuclear ribosomal DNA (rDNA) and beta-tubulin gene sequences, and somatic hyphal interactions. Despite the close genetic relationship among isolates of R. solani AG-3, field populations from potato and tobacco exhibit comparative differences in their disease biology, dispersal ecology, host specialization, genetic diversity and population structure. However, little information is available on how field populations of R. solani AG-3 on potato and tobacco are shaped by population genetic processes. In this study, two field populations of R. solani AG-3 from potato in North Carolina (NC) and the Northern USA; and two field populations from tobacco in NC and Southern Brazil were examined using sequence analysis of two cloned regions of nuclear DNA (pP42F and pP89).Results: Populations of R. solani AG-3 from potato were genetically diverse with a high frequency of heterozygosity, while limited or no genetic diversity was observed within the highly homozygous tobacco populations from NC and Brazil. Except for one isolate (TBR24), all NC and Brazilian isolates from tobacco shared the same alleles. No alleles were shared between potato and tobacco populations of R. solani AG-3, indicating no gene flow between them. To infer historical events that influenced current geographical patterns observed for populations of R. solani AG-3 from potato, we performed an analysis of molecular variance (AMOVA) and a nested clade analysis (NCA). Population differentiation was detected for locus pP89 (Phi(ST) = 0.257, significant at P < 0.05) but not for locus pP42F (Phi(ST) = 0.034, not significant). Results based on NCA of the pP89 locus suggest that historical restricted gene flow is a plausible explanation for the geographical association of clades. Coalescent-based simulations of genealogical relationships between populations of R. solani AG-3 from potato and tobacco were used to estimate the amount and directionality of historical migration patterns in time, and the ages of mutations of populations. Low rates of historical movement of genes were observed between the potato and tobacco populations of R. solani AG-3.Conclusion: the two sisters populations of the basidiomycete fungus R. solani AG-3 from potato and tobacco represent two genetically distinct and historically divergent lineages that have probably evolved within the range of their particular related Solanaceae hosts as sympatric species.

Seqüências tipo mitocondriais têm comumente sido encontradas no genoma nuclear de diversos organismos. Quando acidentalmente incluídas em estudos de seqüências mitocondriais, diversas conclusões errôneas podem ser obtidas. No entanto, estes pseudogenes nucleares tipo mitocondriais podem ser usados para a estimativa da taxa relativa de evolução de genes mitocondriais e também como grupo externo em análises filogenéticas. No presente trabalho, seqüências mitocondriais com características do tipo de pseudogene, tais como deleções e/ou inserções e códons de parada, foram encontradas em tamarins (Saguinus spp., Callitrichinae, Primates). A análise filogenética permitiu a estimativa do tempo da migração da seqüência mitocondrial para o genoma nuclear e algumas inferências filogenéticas. A escolha de um grupo externo não adequado (Aotus infulatus) não permitiu uma reconstrução filogenética confiável da subfamília Callitrichinae. A divergência bastante antiga de Cebidae (Callitrichinae, Aotinae e Cebinae) pode ter favorecido o aparecimento de homoplasias, obscurecendo a análise.

Pre-eclampsia (PE) is a complication of human pregnancy characterized by hypertension and proteinuria after 20 weeks of gestation. Its incidence varies from 5% to 7% of pregnancies and is a major cause of morbidity and maternal and fetal mortality. This is a multisystemic disease, with focus on vascular dysfunction and is closely related to the exacerbated activation of the immune system. In addition to increased activation of monocytes and granulocytes, there is an elevated production of proinflammatory cytokines in pregnant women with PE. The nuclear transcription factor-kB (NF-kB) is present in the cells of the immune system and is responsible for transcription of genes related to inflammation. Whereas the PE is associated with intense inflammatory response, the use of substances modulating the activity of NF-kB factor could be useful in alleviating the inflammation present in these patients. Silibinin is the main component of silymarin, a polyphenolic extract obtained from fruits and seeds of Sylibum marianum with potent hepatoprotective, anti-inflammatory and anti-fibrotic activities. The silibinin mechanism of action includes the ability to inhibit NF-kB activation and, consequently, its migration to the nucleus. The objective of this study was to assess whether silibinin modulates the activity of NF-kB and the production of inflammatory cytokines in mononuclear cells of patients with PE. We evaluated 34 pregnant women with PE, 20 normotensive pregnant women (GN) and 15 non-pregnant women (NG). Peripheral blood mononuclear cells (PBMC) were obtained from those groups of women and cultured in the presence or absence of silibinin (5 uM or 50 uM) and stimulated or not with lipopolysaccharide (LPS) for 18 h to obtain supernatant for determination of tumor necrosis factor-alpha (TNF-α) and interleukin-1 (IL-1β) by enzyme immunoassay (ELISA). Osteosarcoma (OS) is the most common primary malignant bone tumor, usually developing in children and adolescents, and is highly invasive and metastatic, potentially developing chemoresistance. Thus, novel effective treatment regimens are urgently needed. This study was the first to investigate the anticancer effects of dehydroxymethylepoxyquinomicin (DHMEQ), a highly specific nuclear factor-kappa B (NF-kappa B) inhibitor, on the OS cell lines HOS and MG-63. We demonstrate that NF-kappa B blockade by DHMEQ inhibits proliferation, decreases the mitotic index, and triggers apoptosis of OS cells. We examined the effects of combination treatment with DHMEQ and cisplatin, doxorubicin, or methotrexate, drugs commonly used in OS treatment. Using the median effect method of Chou and Talalay, we evaluated the combination indices for simultaneous and sequential treatment schedules. In all cases, combination with a chemotherapeutic drug produced a synergistic effect, even at low single-agent cytotoxic levels. When cells were treated with DHMEQ and cisplatin, a more synergistic effect was obtained using simultaneous treatment. For the doxorubicin and methotrexate combination, a more synergistic effect was achieved with sequential treatment using DHMEQ before chemotherapy. These synergistic effects were accompanied by enhancement of chemoinduced apoptosis. Interestingly, the highest apoptotic effect was reached with sequential exposure in both cell lines, independent of the chemotherapeutic agent used. Likewise, DHMEQ decreased cell invasion and migration, crucial steps for tumor progression. Our data suggest that combining DHMEQ with chemotherapeutic drugs might be useful for planning new therapeutic strategies for OS treatment, mainly in resistant and metastatic cases. Continuous enzymatic interesterification is an alternative to chemical interesterification for lipid modification technology which is economically viable for large scale use. A blend of 70% lard and 30% soybean oil was submitted to continuous enzymatic interesterification in a glass tubular bioreactor at flow rate ranging from 0.5 to 4.5 mL/min. The original mixture and the reaction products obtained were examined to determine melting and crystallization behavior by DSC, and analyzed for regiospecific fatty acid distribution. Continuous enzymatic interesterification changed the mixture, forming a new triacylglycerol composition, verified by DSC curves and variation in enthalpy of melting values. The regiospecific distribution of fatty acids was changed by flow variations in the reactor. In the continuous enzymatic interesterification reaction the flow rate of 4.5 mL/min, was more advantageous than slower flow rates, reducing acyl migration and increasing process productivity. Die vorliegende Arbeit wurde im Rahmen eines Stipendiums des interdisziplinärenrnGraduiertenkollegs 826 „Spurenanalytik von Elementspezies: Methodenentwicklung und Anwendungen“, gefördert durch die Deutsche Forschungsgemeinschaft (DFG) und das Land Rheinland-Pfalz, angefertigt. Dabei sollten neue Erkenntnisse über die Wechselwirkung zwischen Neptunium und natürlichem Tongestein mit Hinblick auf die Endlagerung wärmeentwickelnder radioaktiver Abfälle in einem geologischen Tiefenlager gewonnen werden. Auf Grund seiner langen Halbwertszeit von mehr als zwei Millionen Jahren wird Np-237 einen signifikanten Anteil an der Radiotoxizität dieser Abfälle nach Lagerzeiten von mehr als 1000 Jahren haben. Np tritt in Lösung unter umweltrelevanten Bedingungen in den Oxidationsstufen +IV und +V auf. Auf Grund der guten Löslichkeit und daher höheren Mobilität ist Np(V) als die gefährlichere Spezies einzustufen. In den Migrationsstudien wurdernOpalinuston (OPA) aus Mont Terri, Schweiz, als natürliches Referenzmaterial verwendet. Der Fokus dieser Arbeit lag dabei auf der Speziation von Np an der Mineraloberfläche mittels synchrotronbasierter Röntgenabsorptionsspektroskopie (EXAFS/XANES).rnDie Wechselwirkung zwischen Np(V) und OPA wurde zunächst in Batch- und Diffusionsexperimenten in Abhängigkeit verschiedener experimenteller Parameter (u.a. pH, Temperatur, Hintergrundelektrolyt, Einfluss von Huminsäure, Konkurrenz mit U(VI), aerobe/anaerobe Bedingungen) untersucht. Die Untersuchung der Np-Speziation erfolgt zum einen an Pulverproben aus Batch-Experimenten unter aeroben und anaeroben Bedingungen, welche mittels EXAFS-Spektroskopie untersucht wurden. Zum anderen wurden ortsaufgelöste μ-XANES-Messungen an Np-Anreicherungen auf OPA-Dünnschliffen und in Diffusionsproben durchgeführt. Durch Kombination der Spektroskopie mit μ-Rötngenfluoreszenzmapping (XRF) und μ-Röntgenbeugung (XRD) konnten zudem Erkenntnisse über die Elementverteilung von Np und anderen im Opalinuston enthaltenen Elementen sowie über kristalline Mineralphasen im Umfeld von Bereichen erhöhter Np-Konzentration erhalten werden.rnSowohl Sorptionsexperimente als auch die spektroskopischen Untersuchungen zeigten eine teilweise Reduktion von Np(V) zu Np(IV) bei der Wechselwirkung mit OPA. Dabei konnte Pyrit als eine der redoxaktiven Phasen identifiziert werden. In diesem Zusammenhang ist die Bildung schwerlöslicher Np(IV)-Spezies mit Hinblick auf die Endlagerung positiv zu bewerten.

Die vorliegende Arbeit wurde im Rahmen des Verbundprojektes „Rückhaltung endlagerrelevanter Radionuklide im natürlichen Tongestein und salinaren Systemen“ (Förderkennzeichen: 02E10981), welches durch das Bundesministerium für Wirtschaft und Energie (BMWi) gefördert wurde, angefertigt. Ziel war es, erstmals Erkenntnisse zur Wechselwirkung zwischen dem Spaltprodukt Technetium und einem natürlichen Tongestein im Hinblick auf ein Endlager für wärmeentwickelnde radioaktive Abfälle zu erlangen. Hierfür wurde der in der Nordschweiz vorkommende Opalinuston aus Mont Terri als Referenzmaterial verwendet. Das Nuklid Technetium-99 liefert auf Grund seiner langen Halbwertszeit einen signifikanten Beitrag zur Radiotoxizität abgebrannter Brennelemente für mehr als tausend Jahre. Im Falle einer Freisetzung aus den Lagerbehältern wird die Geochemie des Technetiums von seiner Oxidationsstufe bestimmt, wobei lediglich die Oxidationsstufen +IV und +VII von Relevanz sind. Auf Grund seiner hohen Löslichkeit und geringen Affinität zur Sorption an Oberflächen von Mineralien ist Tc(VII) die mobilste und somit auch gefährlichste Spezies. Entsprechend lag der Fokus dieser Arbeit auf Diffusionsexperimenten dieser mobilen Spezies sowie auf dem Einfluss von Eisen(II) auf die Mobilität und die Speziation des Technetiums.rnDie Wechselwirkung zwischen Technetium und Opalinuston wurde in Sorptions- und Diffusionsexperimenten unter Variation verschiedener Parameter (pH-Wert, Zusatz verschiedener Reduktionsmittel, Einfluss von Sauerstoff, Diffusionsweg) untersucht. Im Zuge dieser Arbeit wurden erstmals ortsaufgelöste Untersuchungen zur Speziation des Technetiums an Dünnschliffen und Bohrkernen durchgeführt. Dabei konnten ergänzend zur Speziation auch Informationen über die Elementverteilung und die kristallinen Mineralphasen nahe lokaler Anreicherungen des Radionuklides gewonnen werden. Zusätzlich erlaubten Untersuchungen an Pulverproben die Bestimmung der molekularen Struktur des Technetiums an der Tonoberfläche.rnSowohl die Kombination der oben aufgeführten Sorptionsexperimente mit spektroskopischen Untersuchungen als auch die Diffusionsexperimente zeigten unter Sauerstoffausschluss eine Reduktion von Tc(VII) zu immobilen Tc(IV)-Spezies. Weiterhin konnte die Bildung eines Tc(IV)-Sorptionskomplexes an der Tonoberfläche gezeigt werden. Im Hinblick auf ein Endlager in Tongestein sind diese Ergebnisse positiv zu bewerten.

This study focused on the role of oceanographic discontinuities and the presence of transitional areas in shaping the population structure and the phylogeography of the Raja miraletus species complex, coupled with the test of the effective occurrence of past speciation events. The comparisons between the Atlantic African and the North-Eastern Atlantic-Mediterranean geographic populations were unravelled using both Cytochrome Oxidase I and eight microsatellite loci. This approach guaranteed a robust dataset for the identification of a speciation event between the Atlantic African clade, corresponding to the ex Raja ocellifera nominal species, and the NE Atlantic-Mediterranean R. miraletus clade. As a matter of fact, the origin of the Atlantic Africa and the NE Atlantic-Mediterranean deep split dated about 11.74MYA and was likely due to the synergic influence currents and two upwelling areas crossing the Western African Waters. Within the Mediterranean Sea, particular attention was also paid to the transitional area represented by Adventura and Maltese Bank, that might have contributed in sustaining the connectivity of the Western and the Eastern Mediterranean geographical populations. Furthermore, the geology of the easternmost part of Sicily and the geo-morphological depression of the Calabrian Arc could have driven the differentiation of the Eastern Mediterranean Sea. Although bathymetric and oceanographic discontinuity could represent barriers to dispersal and migration between Eastern and Western Mediterranean samples, a clear and complete genetic separation among them was not detected. Results produced by this work identified a speciation event defining Raja ocellifera and R. miraletus as two different species, and describing the R. miraletus species complex as the most ancient cryptic speciation event in the family Rajidae, representing another example of how strictly connected the environment, the behavioural habits and the evolutionary and ecologic drivers are.

Dendritic cell (DC) migration via lymphatic vessels to draining lymph nodes (dLNs) is crucial for the initiation of adaptive immunity. We imaged this process by intravital microscopy (IVM) in the ear skin of transgenic mice bearing red-fluorescent vasculature and yellow-fluorescent DCs. DCs within lymphatic capillaries were rarely transported by flow, but actively migrated within lymphatics and were significantly faster than in the interstitium. Pharmacologic blockade of the Rho-associated protein kinase (ROCK), which mediates nuclear contraction and de-adhesion from integrin ligands, significantly reduced DC migration from skin to dLNs in steady-state. IVM revealed that ROCK blockade strongly reduced the velocity of interstitial DC migration, but only marginally affected intralymphatic DC migration. By contrast, during tissue inflammation, ROCK blockade profoundly decreased both interstitial and intralymphatic DC migration. Inhibition of intralymphatic migration was paralleled by a strong up-regulation of ICAM-1 in lymphatic endothelium, suggesting that during inflammation ROCK mediates de-adhesion of DC-expressed integrins from lymphatic-expressed ICAM-1. Flow chamber assays confirmed an involvement of lymphatic-expressed ICAM-1 and DC-expressed ROCK in DC crawling on lymphatic endothelium. Overall, our findings further define the role of ROCK in DC migration to dLNs and reveal a differential requirement for ROCK in intralymphatic DC crawling during steady-state and inflammation.

Several studies have linked overexpression of the LIM and SH3 domain protein 1 (LASP1) to progression of breast, colon, liver, and bladder cancer. However, its expression pattern and role in human prostate cancer (PCa) remained largely undefined. Analysis of published microarray data revealed a significant overexpression of LASP1 in PCa metastases compared to parental primary tumors and normal prostate epithelial cells. Subsequent gene-set enrichment analysis comparing LASP1-high and -low PCa identified an association of LASP1 with genes involved in locomotory behavior and chemokine signaling. These bioinformatic predictions were confirmed in vitro as the inducible short hairpin RNA-mediated LASP1 knockdown impaired migration and proliferation in LNCaP prostate cancer cells. By immunohistochemical staining and semi-quantitative image analysis of whole tissue sections we found an enhanced expression of LASP1 in primary PCa and lymph node metastases over benign prostatic hyperplasia. Strong cytosolic and nuclear LASP1 immunoreactivity correlated with PSA progression. Conversely, qRT-PCR analyses for mir-203, which is a known translational suppressor of LASP1 in matched RNA samples revealed an inverse correlation of LASP1 protein and mir-203 expression. Collectively, our results suggest that loss of mir-203 expression and thus uncontrolled LASP1 overexpression might drive progression of PCa.

Listeria monocytogenes rhombencephalitis is a severe progressive disease despite a swift intrathecal immune response. Based on previous observations, we hypothesized that the disease progresses by intra-axonal spread within the central nervous system. To test this hypothesis, neuroanatomical mapping of lesions, immunofluorescence analysis, and electron microscopy were performed on brains of ruminants with naturally occurring rhombencephalitis. In addition, infection assays were performed in bovine brain cell cultures. Mapping of lesions revealed a consistent pattern with a preferential affection of certain nuclear areas and white matter tracts, indicating that Listeria monocytogenes spreads intra-axonally within the brain along interneuronal connections. These results were supported by immunofluorescence and ultrastructural data localizing Listeria monocytogenes inside axons and dendrites associated with networks of fibrillary structures consistent with actin tails. In vitro infection assays confirmed that bacteria were moving within axon-like processes by employing their actin tail machinery. Remarkably, in vivo, neutrophils invaded the axonal space and the axon itself, apparently by moving between split myelin lamellae of intact myelin sheaths. This intra-axonal invasion of neutrophils was associated with various stages of axonal degeneration and bacterial phagocytosis. Paradoxically, the ensuing adaxonal microabscesses appeared to provide new bacterial replication sites, thus supporting further bacterial spread. In conclusion, intra-axonal bacterial migration and possibly also the innate immune response play an important role in the intracerebral spread of the agent and hence the progression of listeric rhombencephalitis.

Strontium-90 activity concentrations in surface soils and areal deposition densities have been studied at a site contaminated by an accidental release to atmosphere from the underground nuclear explosion 'Kraton-3' conducted near the Polar Circle (65.9°N, 112.3°E) within the territory of the former USSR in 1978. In 2001-2002, the ground surface contamination at 14 plots studied ranged from 20 to 15000 kBq/m**2, which significantly exceeds the value of 0.44 kBq/m**2 deduced for three background plots. The zone with substantial radiostrontium contamination extends, at least, 2.5 km in a north-easterly direction from the borehole. The average 137Cs/90Sr ratio in the ground contamination originated from the 'Kraton-3' fallout was estimated to be 0.55, which is significantly different from the ratio of 2.05 evaluated for background plots contaminated mostly from global fallout. Although vertical migration of 90Sr in all undisturbed soil profiles studied is more rapid than that for 137Cs, the depth of percolation of both radionuclides into the ground is mostly limited to the top 10-20 cm, which may be explained, primarily, by permafrost conditions. The horizontal migration rate of radiostrontium in the aqueous phase exceeds the radiocaesium migration rate by many times. This phenomenon seems to be a reason for the significant enrichment of the soil surface layers by radiostrontium at some sites, with variations occurring in accordance with small-scale irregularities of landscape.

La presente tesis se centra en el estudio de los fenómenos de transporte de los isótopos de hidrógeno, y más concretamente del tritio, en materiales de interés para los reactores de fusión nuclear. Los futuros reactores de fusión nuclear necesitarán una Planta de Tritio, con una envoltura regeneradora (breeding blanket) y unos sistemas auxiliares claves para su diseño. Por lo tanto su desarrollo y cualificación son cruciales para demostrar que los reactores de fusión son una opción viable como futura fuente de energía. Se han resaltado los diferentes retos de la difusión y retención de estas especies ligeras para cada sistema de la Planta de Tritio, y se han identificado las necesidades experimentales y paramétricas para abordar las simulaciones de difusión, como factores de transporte como la difusividad, absorción/desorción, solubilidad y atrapamiento. Se han estudiado los fenómenos de transporte y parámetros del T en el metal líquido LiPb, componente del breeding blanket tanto para una planta de fusión magnética como inercial. Para ello se han utilizado dos experimentos con características diversas, uno de ellos se ha llevado a cabo en un reactor de alto flujo, y por lo tanto, en condiciones de irradiación, y el otro sin irradiación. Los métodos de simulación numérica aplicados se han adaptado a los experimentos para las mediciones y para estudiar el régimen de transporte. En el estudio de estos experimentos se ha obtenido un valor para algunos de los parámetros claves en el transporte y gestión del tritio en el reactor. Finalmente se realiza un cálculo de la acumulación y difusión de tritio en una primera pared de tungsteno para un reactor de fusión inercial. En concreto para el proyecto de fusión por láser europeo, HiPER (para sus fases 4a y 4b). Se ha estudiado: la implantación de los isótopos de H y He en la pared de W tras una reacción de fusión por iluminación directa con un láser de 48MJ; el efecto en el transporte de T de los picos de temperatura en el W debido a la frecuencia de los eventos de fusión; el régimen de transporte en la primera pared. Se han identificado la naturaleza de las trampas más importantes para el T y se ha propuesto un modelo avanzado para la difusión con trampas. ABSTRACT The present thesis focuses into study the transport phenomenons of hydrogen isotopes, more specifically tritium, in materials of interest for nuclear fusion reactors. The future nuclear reactors will be provided of a Tritium Plant, with its breeding blanket and its auxiliary systems, all of them essential components for the plant. Therefore a reliable development and coalification are key issues to prove the viability of the nuclear fusion reactors as an energy source. The currently challenges for the diffusion and accumulation of these light species for each system of the TP has been studied. Experimental and theoretical needs have been identified and analyzed, specially from the viewpoint of the parameters. To achieve reliable simulations of tritium transport, parameters as diffusivity, absorption/desorption, solubility and trapping must be reliables. Transport phenomenon and parameters of T in liquid metal have been studied. Lead lithium is a key component of the breeding blanket, either in magnetic or inertial fusion confinement. Having this aim in mind, two experiments with different characteristics have been used; one of them has been realized in a high flux reactor, and hence, under irradiation conditions. The other one has been realized without radiation. The mathematical methods for the simulation have been adapted to the experiments, for the measures and also to study the transport behavior. A value for some key parameters for tritium management has been obtained in these studies. Finally, tritium accumulation and diffusion in a W first wall of an inertial nuclear fusion reactor has been assessed. A diffusion model of the implanted H, D, T and He species for the two initial phases of the proposed European laser fusion Project HiPER (namely, phase 4a and phase 4b) has been implemented using Tritium Migration Analysis Program, TMAP7. The effect of the prompt and working temperatures and the operational pulsing modes on the diffusion are studied. The nature of tritium traps in W and their performance has been analyzed and discussed.
