En el hipotálamo en desarrollo, el ácido gamma-amino butírico (GABA) produce depolarización neuronal, pudiendo incluso disparar potenciales de acción y causar la apertura de canales de calcio dependientes de voltaje. Esto se debe a que la concentración intracelular de Cl- es alta respecto al medio extracelular, por lo que en reposo el potencial de equilibrio de GABA es más positivo que el potencial de membrana. A medida que el desarrollo transcurre, la concentración intracelular de Cl- disminuye y se produce un cambio en la respuesta de depolarizante (etapa excitatoria) a hiperpolarizante (etapa inhibitoria). Se ha demostrado que este cambio ocurre también en neuronas hipotalámicas in vitro. El dimorfismo sexual del cerebro de los vertebrados es consecuencia de la acción del estrógeno aromatizado a partir de andrógenos segregados por el testículo durante el "periodo crítico" del desarrollo cerebral. Evidencias previas de nuestro y otros laboratorios pusieron de manifiesto diferencias en el crecimiento y diferenciación de neuronas que no podían atribuirse a la acción hormonal, ya que ocurren antes que se inicie el brusco aumento de la secreción gonadal, alrededor del día 18 de desarrollo embrionario en la rata (E18). Además de las diferencias morfológicas, encontramos diferencias sexuales en la forma que las neuronas hipotalámicas responden a muscimol, un agonista específico del receptor GABAA. A los 9 días in vitro (9 DIV) la respuesta a muscimol fue hiperpolarizante (etapa inhibitoria) y además fue de mayor amplitud, área y duración en machos respecto a hembras. Esto nos indica que las neuronas provenientes de embriones machos son intrínsecamente diferentes a las de embriones hembra aún antes de la acción organizadora de los esteroides sexuales. En base a estas evidencias nos propusimos continuar nuestros estudios sobre la participación de GABA en la determinación de diferencias sexuales en el cerebro antes de la acción organizadora de los esteroides gonadales. Para ello, en cultivos de neuronas hipotalámicas de E16 separados por sexos, estudiaremos:- la respuesta a muscimol de las neuronas, en la etapa excitatoria (2 DIV) de la acción de GABA.- las composición de subunidades de los receptores GABAA en la etapa excitatoria/inhibitoria de la acción de GABA.- la participación de los receptores GABAA sobre el crecimiento neurítico.- la activación de la vía de las MAP quinasas por muscimol.- la participación de los receptores GABAA sobre el crecimiento axonal inducido por estradiol in vitro.Toda la metodología propuesta es de uso habitual en nuestro laboratorio e involucra herramientas de la electrofisiología y la biología celular-molecular; como patch-clamp, cultivo de neuronas hipotalámicas, Western blot, RT-PCR, entre otras. Esperamos encontrar diferencias sexuales en la amplitud, área y duración de la respuesta de las neuronas hipotalámicas al muscimol a los 2 DIV, y que éstas se deban a una diferente composición de subunidades del receptor GABAA. En cuanto a la participación del receptor GABAA en la neuritogenesis, esperamos encontrar mayor longitud neurítica en neuronas macho como así también una activación sexualmente dimórfica de la vía de las MAP quinasas. Además esperamosque la acción de un antagonista del receptor GABAA interfiera con la axogénesis inducida por estradiol in vitro, característica que muestra diferencia sexual también a favor de los machos, lo que reforzaría nuestra hipótesis. La importancia y originalidad de este proyecto reside en la evaluación de la participación del sistema GABAérgico en la determinación de características que durante el desarrollo, podrían estar involucradas en la determinación de diferencias sexuales permanentes en el cerebro adulto independientemente de la acción de los esteroides sexuales. Hasta la fecha, no ha sido evaluada la influencia de los receptores GABAA en la diferenciación sexual del cerebro antes de la acción organizadora de los esteroides gonadales.

The sequence of pitches which form a musical melody can be transposed or inverted. Since the 1970s, music theorists have modeled musical transposition and inversion in terms of an action of the dihedral group of order 24. More recently music theorists have found an intriguing second way that the dihedral group of order 24 acts on the set of major and minor triads. We illustrate both geometrically and algebraically how these two actions are dual. Both actions and their duality have been used to analyze works of music as diverse as Hindemith and the Beatles.

We prove rigidity and vanishing theorems for several holomorphic Euler characteristics on complex contact manifolds admitting holomorphic circle actions preserving the contact structure. Such vanishings are reminiscent of those of LeBrun and Salamon on Fano contact manifolds but under a symmetry assumption instead of a curvature condition.

The Institute of Public Health in Ireland (IPH) is a partner in the European project DETERMINE, building on its previous involvement in the Closing the Gap project in 2004-2006. In the first year of the project (2007-2008) 15 DETERMINE partners identified policies and actions that have taken place within countries, and at the EU level, to address Social Determinants of Health Inequalities. These policies and actions were identified via a questionnaire, which also identified structures and tools/mechanisms being used in the country to support a 'health in all policy' approach.

The aim of the present article was to present a specific activity approach called the 'life course of a practice' that could be used for the analysis of the long-term dynamic of learning within diverse social practices. The cases of unethical actions (doping) within a population of high-level athletes were examined, considering they could be fruitful exam- 10 ples for this type of analysis. After describing the theoretical construct 'life course of a practice', the places of these unethical actions in the sporting life courses and elements that characterize their emergence in the long-term dynamics are described. Last, targeted prevention measures of unethical actions over a lifetime are presented.

The National Drugs Strategy 2009-16 is a cross cutting area of public policy and service delivery. It is based upon a co-ordinated approach across the full range of Government Departments and Agencies involved in delivering drugs policy. The overall objective of the Strategy is to tackle the harm caused to individuals, families and communities as a result of problem drug and alcohol use through the five pillars of supply reduction, prevention, treatment, rehabilitation and research. Aspirin has always remained an enigmatic drug. Not only does it present with new benefits for treating an ever-expanding list of apparently unrelated diseases at an astounding rate but also because aspirin enhances our understanding of the nature of these diseases processe. Originally, the beneficial effects of aspirin were shown to stem from its inhibition of cyclooxygenase-derived prostaglandins, fatty acid metabolites that modulate host defense. However, in addition to inhibiting cyclooxygenase activity aspirin can also inhibit pro-inflammatory signaling pathways, gene expression and other factors distinct from eicosanoid biosynthesis that drive inflammation as well as enhance the synthesis of endogenous protective anti-inflammatory factors. Its true mechanism of action in anti-inflammation remains unclear. Here the data from a series of recent experiments proposing that one of aspirin's predominant roles in inflammation is the induction of nitric oxide, which potently inhibits leukocyte/endothelium interaction during acute inflammation, will be discussed. It will be argued that this nitric oxide-inducing effects are exclusive to aspirin due to its unique ability, among the family of traditional anti-inflammatory drugs, to acetylate the active site of inducible cyclooxygenase and generate a family of lipid mediators called the epi-lipoxins that are increasingly being shown to have profound roles in a range of host defense responses.

The production of object and action words can be dissociated in aphasics, yet their anatomical correlates have been difficult to distinguish in functional imaging studies. To investigate the extent to which the cortical neural networks underlying object- and action-naming processing overlap, we performed electrostimulation mapping (ESM), which is a neurosurgical mapping technique routinely used to examine language function during brain-tumor resections. Forty-one right-handed patients who had surgery for a brain tumor were asked to perform overt naming of object and action pictures under stimulation. Overall, 73 out of the 633 stimulated cortical sites (11.5%) were associated with stimulation-induced language interferences. These interference sites were very much localized (<1 cm(2) ), and showed substantial variability across individuals in their exact localization. Stimulation interfered with both object and action naming over 44 sites, whereas it specifically interfered with object naming over 19 sites and with action naming over 10 sites. Specific object-naming sites were mainly identified in Broca's area (Brodmann area 44/45) and the temporal cortex, whereas action-naming specific sites were mainly identified in the posterior midfrontal gyrus (Brodmann area 6/9) and Broca's area (P = 0.003 by the Fisher's exact test). The anatomical loci we emphasized are in line with a cortical distinction between objects and actions based on conceptual/semantic features, so the prefrontal/premotor cortex would preferentially support sensorimotor contingencies associated with actions, whereas the temporal cortex would preferentially underpin (functional) properties of objects. Corticosteroids are widely used to treat a diversity of pathological conditions including allergic, autoimmune and some infectious diseases. These drugs have complex mechanisms of action involving both genomic and non-genomic mechanisms and interfere with different signal transduction pathways in the cell. The use of corticosteroids to treat critically ill patients with acute respiratory distress syndrome and severe infections, such as sepsis and pneumonia, is still a matter of intense debate in the scientific and medical community with evidence both for and against its use in these patients. Here, we review the basic molecular mechanisms important for corticosteroid action as well as current evidence for their use, or not, in septic patients. We also present an analysis of the reasons why this is still such a controversial point in the literature.

Purpose. Spanish retina specialists were surveyed in order to propose actions to decrease deficiencies in real-life neovascular age macular degeneration treatment (nv-AMD). Methods. One hundred experts, members of the Spanish Vitreoretinal Society (SERV), were invited to complete an online survey of 52 statements about nv-AMD management with a modified Delphi methodology. Four rounds were performed using a 5-point Linkert scale. Recommendations were developed after analyzing the differences between the results and the SERV guidelines recommendations. Results. Eighty-seven specialists completed all the Delphi rounds. Once major potential deficiencies in real-life nv-AMD treatment were identified, 15 recommendations were developed with a high level of agreement. Consensus statements to reduce the burden of the disease included the use of treat and extend regimen and to reduce the amount of diagnostic tests during the loading phase and training technical staff to perform these tests and reduce the time between relapse detection and reinjection, as well as establishing patient referral protocols to outside general ophthalmology clinics. Conclusion. The level of agreement with the final recommendations for nv-AMD treatment among Spanish retinal specialist was high indicating that some actions could be applied in order to reduce the deficiencies in real-life nv-AMD treatment.
