An efficient conjugation method has been developed for the marine Actinomyces sp. isolate M048 to facilitate the genetic manipulation of the chandrananimycin biosynthesis gene cluster. A phi C31-derived integration vector pIJ8600 containing oriT and attP fragments was introduced into strain M048 by bi-parental conjugation from Escherichia coli ET12567 to strain M048. Transformation efficiency was (6.38 +/- 0.41) x 10(-5) exconjugants per recipient spore. Analysis of eight exconjugants showed that the plasmid pIJ8600 was stably integrated at a single chromosomal site (attB) of the Actinomyces genome. The DNA sequence of the attB was cloned and shown to be conserved. The results of antimicrobial activity analysis indicated that the insertion of plasmid pIJ8600 seemed to affect the biosynthesis of antibiotics that could strongly inhibit the growth of E. coli and Mucor miehei (Tu284). HPLC-MS analysis of the extracts indicated that disruption of the attB site resulted in the complete abolition of chandrananimycin A-C production, proving the identity of the gene cluster. Instead of chandrananimycins, two bafilomycins were produced through disruption of the attB site from the chromosomal DNA of marine Actinomyces sp. M048.

本文对256株胶州湾海洋链霉菌进行了抑菌活性的筛选,并选取10株典型菌株进行化学筛选,获得2株有研究价值的菌株。通过大规模发酵,获得纯化的次级代谢产物,进行了结构解析。通过与其他5株活性菌株的16S rRNA基因序列比较,并结合生理生化、形态特征和培养特征分析,探讨了这两株菌的分类地位。 采用液体扩散法,选用金黄色葡萄球菌、大肠杆菌、绿脓杆菌、八叠球菌、隐球菌、白色念珠菌、Mucor miehei (TÜ 284)和Streptomyces viridochromogenes (TÜ57) 8株受试菌进行抑菌活性的筛选,结果22%的菌株显示出对至少一种受试菌具有抑制作用(抑菌圈Æ ³ 8 mm)。根据菌株的形态特征和抑菌活性特点,选择M024、M028、M042、M083、M086、M095、M097、M124、M134和M226 10株链霉菌进行化学筛选。考察了8种培养基和4种培养条件,结果发现菌株M095在Meat extract培养基、pH 6.5、28℃和95 r/min条件下,菌株M097在Meat extract培养基、pH 7.8、 28℃、95 r/min(条件Ⅰ)和M2+培养基、pH 7.8、 35℃、110 r/min(条件Ⅱ)条件下,可供进一步研究。 对菌株M095(24 L规模)和M097(Ⅰ为30 L规模,Ⅱ为14 L规模)进行发酵,采用乙酸乙酯提取和柱层析分离纯化次级代谢产物,通过ESI-MS、EI-MS、1H-NMR和13C-NMR等波谱解析,鉴定出次级代谢产物的结构。发现菌株M095产生一抑菌活性很强的化合物全霉素,首次证实该全霉素具有抑制丝状真菌的作用;菌株M097主要产生10个化合物,其中8个具有不同程度的生物活性,另外两个化合物中,Aloesaponaria Ⅱ为首次从微生物野生菌株(wild strain)中获得,化合物Cui D为一新结构的蒽醌类化合物。 经分子鉴定,初步认为本实验分离的7株活性海洋链霉菌分属于4个链霉菌类群,结合生理生化、形态特征和培养特征分析,认为菌株M095可能为灰色链霉菌的变种,M097可能为球孢类群中的一个新种。

Cystic fibrosis (CF) patients may suffer increased morbidity and mortality through colonisation, allergy and invasive infection from fungi. The black yeast, Exophiala dermatitidis (synonym Wangiella dermatitidis) has been found with increasing frequency in sputum specimens of CF patients, with reported isolation rates ranging from 1.1 to 15.7%. At present, no diagnostic PCR exists to aid with the clinical laboratory detection and identification of this organism. A novel species-specific PCR-based assay was developed for the detection of E. dermatitidis, based on employment of rDNA operons and interspacer (ITS) regions between these rDNA operons. Two novel primers, (designated ExdF & ExdR) were designed in silico with the aid of computer-aided alignment software and with the alignment of multiple species of Exophiala, as well as with other commonly described yeasts and filamentous fungi within CF sputum, including Candida. Aspergillus and Scedosporium. An amplicon of approximately 455 by was generated, spanning the partial ITS I region - the complete 5.8S rDNA region - partial ITS2 region, employing ExdF (forward primer [16-mer], 5'-CCG CCT ATT CAG GTC C-3' and ExdR (reverse primer [16-mer], 5'-TCT CTC CCA CTC CCG C-3', was employed and optimised on extracted genomic DNA from a well characterised culture of E. dermatitidis, as well as with high quality genomic DNA template from a further 16 unrelated fungi, including Candida albicans, C. dubliniensis, C. parapsilosis, C. glabrata, Scedosporium apiospermum, Penicillium sp., Aspergillus fumigatus, Aspergillus versicolor, Pichia guilliermondii, Rhodotorula sp., Trichosporon sp., Aureobasidium pullulans, Fusarium sp., Mucor hiemalis, Bionectria ochroleuca, Gibberella pulicaris. Results demonstrated that only DNA from E. dermatitidis gave an amplification product of the expected sire, whilst none of the other fungi were amplifiable. Subsequent employment of this primer pair detected this yeast from mycological cultures from 2/50 (4%) adult CF patients. These two patients were the only patients who were previously shown to have a cultural history of E. dermatitidis from their sputum. E. dermatitidis is a slow-growing fungus, which usually takes up to two weeks to culture in the microbiology laboratory and therefore is slow to detect conventionally, with the risk of bacterial overgrowth from common co-habiting pan- and multiresistant bacterial pathogens from sputum. namely Pseudomonas aeruginosa and Burkholderia cepacia complex organisms, hence this species-specific PCR assay may help detect this organism from CF sputum more specifically and rapidly. Overall, employment of this novel assay nay help in the understanding of the occurrence. aetiology and epidemiology of E. dermatitidis, as an emerging fungal agent in patients with CF. Soybean oil soapstock was utilized as an alternative carbon source for the production of rhamnolipids by Pseudomonas aeruginosa LBI strain. The chemical composition and properties of the rhamnolipid mixture obtained were determined to define its potential applications. The chemical characterization of the rhamnolipid has revealed the presence of ten different homologues. The monorhamnolipid RhaC(10)C(10) and the dirhamnolipid Rha(2)C(10)C(10) were the main components of the mixture that showed predominance of 44% and 29%, respectively, after 144-h of cultivation. The biosurfactant was able to form stable emulsions with several hydrocarbons and showed excellent emulsification for soybean oil and chicken fat (100%). The rhamnolipid removed 67% of crude oil present in sand samples and presented antimicrobial activity against Bacillus cereus and Mucor miehei at 64 mu g/mL and inhibition of Neurospora crassa, Staphylococcus aureus, and Micrococcus luteus at 256 mu g/mL. The results demonstrated that the rhamnolipid produced in soybean oil soapstock can be useful in environmental and food industry applications.

Using a freeze-etch technique the cytoplasmic and plasma membrane ultrastructure of Paracoccidiodies brasiliensis yeast-phase cells was studied. The multinucleate yeast-phase cells which grow by simultaneous multiple budding, like those of Mucor sp. contain several nuclei, mitochondria, well-developed ER, small vacuoles and lipid droplets. Complex structures with no apparent connexion to the plasma membrane of P. brasiliensis usually lack inveginations, but invaginations which do occur are always rod-shaped which indicates P. brasiliensis to be of either ascomycetous or basidiomycetous origin.

Counts of colony forming units of actinomycetes, bacteria and filamentous fungi were determined in cerrado soil treated with vinasse (processed sugar cane effluent) for 5 yr, using doses of 20 l m-2a-1 and 50 l m-2a-1. A temporary increase in the counts of actinomycetes and bacteria for some months after the addition of vinasse was observed. An increased number of fungi was detected throughout the experiment together with qualitative changes in the population. The most abundant fungi in cerrado soil not treated with vinasse (control soil) were Chaetomium, yeasts, Mucor, Penicillium and Trichoderma, while in vinasse treated soils, the same genera adding Verticillium, with the exception of yeasts, were the most common fungi. -Author

Three species of filamentous fungi, Aspergillus niger, Penicillium fellutanum and Mucor hiemalis, were selected and cultivated in vinasse media with different addition of molasses, pasteurized to 85°C for 30 minutes and with pH = 5.0. The microorganisms, previously adapted to the respective medium for 48 hours, from a solution of 107 spores.ml-1, were cultivated in pure and mixed cultures in Erlenmeyer vessel of 500ml, to 30°C, with constant agitation of 170 rpm, for 24, 48 and 72 hours, with four repetition for each samples. The biomass was separated by vacuum filtration in filter Whatman #1 and dried in oven at 105°C until right weight, the obtained liquid was submited to COD analysis. The datas were statistically analysed using a response surface methodology, to improve the effect on the molasses proportion and culture time, in the biomass production by microorganism in research. According to the obtained results (5.02% of molasses, 55.59h, 70% of spores solution of A. niger and 30% of spores solution of P. fellutanum), cultivating was carried out in Microferm Fermentor New Brunswick for 48 hours at 300 rpm, aired at 1v/v/m, using 5 liters of medium added with 5.0% of molasses on the conditions above described. The average of the results obtained (6.81g.l-1) was higher than the confidence interval (5.937 ; 6.369) and was inside the prediction interval (4.471 ; 7.834) both of them significant at 95% by the statistical test employed.

Fungi producing γ-linolenic acid (GLA) were isolated from soil of the Ecological Station of Juréia-Itatins, SP. This essential fatty acid has aroused great interest due to its increasing by applications in pharmaceutical industry. The GLA production by zygomycetous fungi is an alternative way of comparing seed extraction. Thirty-two zygomycetous strains of Mucorales were isolated, most of them belonging to Mucor genus. The GLA production was evaluated after 4 days of incubation at 25°C on a rotary shaker at 150 rpm in medium containing 2% glucose, and 1% yeast extract, following new medium addition (20%) and incubation for an additional period of 3 days at 12°C, without agitation. The GLA production varied according to the microorganism and the strain.

Dragmacidon reticulatum is a marine sponge of wide occurrence in the Eastern and Western Atlantic. Little is known about D. reticulatum fungal diversity. Filamentous fungi recovered from D. reticulatum were assessed in the present study using a polyphasic taxonomic approach, including classical morphology, molecular biology and MALDI-TOF ICMS. Ninety-eight fungal strains were isolated from two D. reticulatum samples by using six different culture media, which were identified up to the genus level. Sixty-four distinct fungal ribotypes were obtained, distributed among twenty-four different genera belonging to the Ascomycota and Zygomycota. Representatives of Penicillium and Trichoderma were the most diverse and abundant fungi isolated. Amongst Penicillium spp. three isolates belonged to the same ribotype can be considered as a putative new species. Data derived from the present study highlight the importance of using a polyphasic approach to get an accurate identification in order to structure a reliable culture collection. © 2012 Springer-Verlag Berlin Heidelberg.

Background: The increase in opportunistic fungal infections has led to the search for putative sources of contamination in hospital environments. Aim: Ants in a public hospital in Itabuna, north-eastern Brazil were examined for carriage of filamentous fungi. Methods: During a year-long survey, ants from different hospital areas were sampled. Preference was given to locations where it was possible to observe ants actively foraging. The fungi found on the ants' integument were cultured and identified. Findings: A total of 106 ant workers belonging to 12 species in 11 genera were collected. A total of 47 fungal strains was isolated from 40% of the ants (. N = 42). We found 16 fungal species in 13 genera associated with the ant workers. The prevalent fungal genera were . Aspergillus, . Purpureocillium and . Fusarium. The ants . Tapinoma melanocephalum, . Paratrechina longicornis and . Pheidole megacephala were associated with six fungal genera; and four genera of fungi were associated with . Solenopsis saevissima workers. Fungal diversity was higher in the following hospital areas: nursery, hospital beds, breastmilk bank and paediatrics. Conclusion: Ants act as carriers of soil and airborne fungal species, and ant control in hospital areas is necessary to prevent the dissemination of such micro-organisms. © 2012 The Healthcare Infection Society.
