BACKGROUND: Congenital, nonepidermolytic cornification disorders phenotypically resembling human autosomal recessive ichthyosis have been described in purebred dog breeds, including Jack Russell terrier (JRT) dogs. One cause of gene mutation important to humans and dogs is transposon insertions. OBJECTIVES: To describe an autosomal recessive, severe nonepidermolytic ichthyosis resembling lamellar ichthyosis (LI) in JRT dogs due to insertion of a long interspersed nucleotide element (LINE-1) in the transglutaminase 1 (TGM1) gene. METHODS: Dogs were evaluated clinically, and skin samples were examined by light and electron microscopy. Phenotypic information and genotyping with a canine microsatellite marker suggested TGM1 to be a candidate gene. Genomic DNA samples and cDNA generated from epidermal RNA were examined. Consequences of the mutation were evaluated by Western blotting, quantitative reverse transcription-polymerase chain reaction (RT-PCR) and enzyme activity from cultured keratinocytes. RESULTS: Affected dogs had generalized severe hyperkeratosis. Histological examination defined laminated to compact hyperkeratosis without epidermolysis; ultrastructurally, cornified envelopes were thin. Affected dogs were homozygous for a 1980-bp insertion within intron 9 of TGM1. The sequence of the insertion was that of a canine LINE-1 element. Quantitative RT-PCR indicated a significant decrease in TGM1 mRNA in affected dogs compared with wild-type. TGM1 protein was markedly decreased on immunoblotting, and membrane-associated enzyme activity was diminished in affected dogs. CONCLUSIONS: Based on morphological and molecular features, this disease is homologous with TGM1-deficient LI in humans, clinically models LI better than the genetically modified mouse and represents its first spontaneous animal model. This is the first reported form of LI due to transposon insertion.

A 38-year-old woman presented with blurred vision and "jumping" of the right eye for 7 months. Magnetic resonance imaging of the head was normal. Intermittent intorsion of the right eye was noted on examination, consistent with superior oblique myokymia. She was initially treated with carbamazepine but stopped after becoming light-headed. The diagnosis and treatment of superior oblique myokymia are discussed.

Dryers heated by solar energy have been constructed and used in drying whole and half jack fruit almonds. The samples were dried during the day in direct sun and in the conventional solar dryer prepared for this purpose. Another piece of equipment was built for reception and accumulation of sun energy in a body of water, which was used as a heat source for night drying. The drying with the sun energy was compared with artificial drying. The jack fruit almonds were dried whole, half, with pellicle and without it. The storage of solar energy in water was technically viable for use in night drying. The drying by combining solar dryers in the day and night periods were completed in approximately 35 hours, and were equivalent to artificial drying between 40ºC and 70ºC. Almond cut in half and the pellicle removed reduced the drying time.

Sementes de M. maritima germinam na temperatura ótima de 25°C com valores máximos na luz, sendo o processo otimizado em temperatura alternada de 25/30°C. Escarificação mecânica promove a germinação. Em sementes nuas, a germinação é promovida e acelerada tanto na luz quanto no escuro. Temperatura de armazenamento e tempo de estocagem alteram a fotossensibilidade das sementes: sementes estocadas à temperatura ambiente por 90 dias perdem o fotoblastismo positivo enquanto que nas estocadas sob baixa temperatura (10°C) este efeito permanece. O teste do tetrazólio demonstra que aos 180 dias de estocagem 46% das sementes permaneceram viáveis. Análises topoquímicas indicam que a principal fonte de reserva destas sementes, localizada nos cotilédones, é proteica.

Neste trabalho propomos o aproveitamento dos resíduos de laranja gerados após remoção do suco como substrato para a obtenção de enzimas hidrolíticas e oxidativas envolvidas na degradação de materiais lignocelulósicos, tais como: lacase (EC 1.10.3.2), manganês peroxidase (EC 1.11.1.14), xilanase (EC 3.2.1.8) e endo-1,4 glucanase (EC 3.2.1.4), pelo basidiomiceto Pleurotus ostreatus cultivado em estado sólido. O fungo desenvolveu-se bem no resíduo, em diferentes umidades iniciais e sem a necessidade de qualquer suplementação. O meio à base de resíduo de laranja proporcionou a obtenção de elevadas atividades de enzimas com grande potencial de uso industrial, especialmente lacase (74,3 U.g -1 substrato após 15 dias de cultivo) e manganês peroxidase (6,8 U.g -1 substrato após 30 dias de cultivo).
