The correlation between trophocyte size and ovarian development is negative in workers and positive in queens of Melipona quadrifasciata anthidioides. The nurse workers which have the ovaries in a higher developmental stage, present smaller fat body cells them newly-emerged ones. In newly-emerged and nurse workers the trophocytes seem to be delivering their stored products, among which probably vitellogenin. As in workers the cell size variations do not support the occurrence of proteic synthesis or the increasing in reverses storage after the adult emergence, the products released from the trophocytes must come from cellular reserves remaining from the larval phase. This datum is in agreement with the early and brief vitellogenic phase in the ovaries of this caste. In foragers the trophocyte size seen stabilized. In queens it was verified considerable increasing in the trophocyte size from virgin to physogastric queen, as well as the maintenance of the size during all fertile life of the queen.

In social insects, cuticular hydrocarbons are involved in species, kin, caste and nestmate recognition. Gas chromatography and mass spectrometry were used to compare the cuticular hydrocarbon composition of workers, males and queens of Melipona bicolor. The cuticular hydrocarbon composition of this species was found to consist mainly of C23, C25:1, C25, C27:1, C27, C29:1 and C29, which are already present in imagoes that have not yet abandoned the brood cell. This composition varied quantitatively and qualitatively between and within the castes and sexes. The newly emerged workers and young queens (virgins) had similar cuticular hydrocarbon profiles, which were different from those of the males. When the females start executing their tasks in the colony, the cuticular hydrocarbon profile differences appear. The workers have less variety, while the queens conserve or increase the number of cuticular hydrocarbon compounds. The queens have more abdominal tegumentary glands than the workers, which apparently are the source of the new cuticular compounds.

We describe differences in the chemical composition of the Dufour gland secretion of virgin and physogastric queens of Melipona bicolor through gas chromatography and mass spectrometry. The Dufour gland secretion of virgin queens consists only of hydrocarbons, while that of physogastric queens contains, besides these, a variety of other compounds, such as isobutyrate and acetate esters. Such differences may indicate the queen fecundity condition and the oxygenated compounds of the physogastrics secretion may help to increase their attractiveness.

The mandibular gland secretions of newly emerged, nurse and forager workers, virgin and physogastric queens and males of Melipona bicolor were analyzed by gas chromatography-mass spectrometry. The secretion is composed of a blend of hydrocarbons, alcohols, esters, and acids. The secretion is caste-sex specific and also differs with the tasks performed by the workers and the physiological reproductive condition of the queens.

We investigated whether Melipona quadrifasciata worker mandibular gland secretions contribute directly to their cuticular hydrocarbon profile. The mandibular gland secretion composition and cuticular surface compounds of newly emerged worker bees, nurse bees, and foragers were determined by gas chromatography and mass spectrometry and compared. Both the mandibular gland secretions and the cuticular surface compounds of all worker stages were found to be composed almost exclusively of hydrocarbons. Although the relative proportion of hydrocarbons from the cuticular surface and gland secretion was statistically different, there was a high similarity in the qualitative composition between these structures in all groups of bees. © FUNPEC-RP.

The fat body (FB) consists of two types of cells: throphocytes and oenocytes. Throphocytes are related to intermediary metabolism storing lipids, carbohydrates, and proteins while oenocytes play role in the lipids and lipoproteins production. The vitellogenin is the precursor of egg yolk (vitelline) and is synthesized on FB. The aim of this work was to analyze the effects of hormones acting in bee reproduction, as juvenile hormone (JH) and ecdisteroids (20 HE) on FB cells, where vitellogenin is synthesized. For the study were chose nurse workers that in Melipona quadrifasciata anthidioides present activated ovaries and produce eggs, and virgin queens whose ovaries are not yet activated, presenting only previtellogenic follicles. FB trophocytes from these classes of bees were cultivated in media containing different amounts of JH and 20-HE. The effects on trophocytes cytoplasm reserves of lipids, proteins, and activity of acid phosphatase were compared by observing preparations from cultured FB, treated and control, by transmission electron microscopy (TEM). The results showed that the hormones effects are related to the bee's caste and functional ovary stage. The role of acid phosphatase on mobilization of the trophocyte reserves was also determined. © 2012 Wiley Periodicals, Inc.

The yolk protein precursor, vitellogenin (Vg), in bees is synthesized in the fat body trophocytes, delivered to the hemolymph and ultimately absorbed from there during the vitellogenic phase of oocytes in the active ovary. The routes tracing the material exchange that occurs between the trophocytes and the hemolymph, in addition to the transportation from the hemolymph to the ovarian follicles, were marked by alkaline phosphatase and lanthanum nitrate (LN). Active ovaries from nurse workers and physogastric queens, as well as inactive ovaries of virgin queens, were examined by transmission electron microscopy. The LN permitted better visualization of the routes of exchanges between the organs and the hemolymph. Both methods demonstrate the apparent differences between the phases of the ovary and the bee caste. In inactive ovaries of the virgin queens, the routes from the follicular epithelium to the oocyte remain closed; conversely, they are open in active ovaries of the nurse workers and physogastric queens. A criação de abelhas sem ferrão é antiga nas Américas. No entanto, a atividade ainda esta subdesenvolvida, necessitando de melhorias nas técnicas de manejo. Este estudo avaliou alternativas nutricionais para a abelha amazônica Melipona flavolineata, visando substituição da alimentação com mel e pólen fermentado. As seguintes alternativas nutricionais foram comparadas com o mel (T1): xarope de açúcar invertido com minerais (T2) e este xarope acrescido de solução de aminoácidos e vitaminas nas seguintes concentrações: 0,5% (T3), 1,0% (T4) e 10% (T5). Ao pólen fermentado (T1) foram comparados alimentos fermentados com inóculo de pólen fermentado de M. flavolineata, constituídos de: pólen apícola (T2); levedo (T3); e extrato de soja, em duas diferentes concentrações de proteínas (T4, 12% e T5, 18%). O valor nutricional das dietas foi baseado no consumo diário, no peso das operárias, no tamanho dos acinos das glândulas hipofaringeas e no tamanho dos oócitos. Avaliaram-se, tambem, os custos dos ingredientes das dietas. Os resultados obtidos indicaram que dentre as alternativas para o mel, não houve diferenças estatísticas entre o controle e todos os tratamentos. Foram encontradas diferenças apenas entre os tamanhos dos acinos das operárias alimentadas com os tratamentos 2 e 4, sendo o tratamento 4 superior. Os custos dos ingredientes indicaram o tratamento 2 como sendo mais barato que os demais. Dentre as alternativas para o sabura foram encontradas maiores diferenças. O consumo do tratamento 4 foi significativamente superior ao do T3, indicando maior aceitação do extrato de soja em relação ao levedo. Em relação aos demais parâmetros avaliados, o tratamento 5 foi superior a todos os demais e seu custo foi muito inferior ao do T2 e semelhante ao dos T3 e T4. Concluiu-se que as melhores alternativas para a nutrição de M. flavolineata sao o xarope de acucar invertido com minerais, e o pólen fermentado semi-artificial à base de extrato de soja. Colônias recém-formadas, com 500-600 abelhas, devem receber 10 a 20 ml de xarope e 25 g de pólen fermentado semi-artificial semanalmente.

ABSTRACT: Honey produced by three stingless bee species (Melipona flavolineata, M. fasciculata and Apis mellifera) from different regions of the Amazon was analyzed by separating phenolic acids and flavonoids using the HPLC technique. Data were subjected to multivariate statistical analysis (PCA, HCA and DA). Results showed the three species of honey samples could be distinguished by phenolic composition. Antioxidant activity of the honeys was determined by studying the capacity of inhibiting radicals using DPPH assay. Honeys with higher phenolic compound contents had greater antioxidant capacity and darker color.

A longevidade em insetos sociais pode ser condicionada por inúmeros fatores, como por exemplo, as condições ambientais que podem alterar o comportamento de forrageio das abelhas para suprir as necessidades do ninho. O objetivo do presente trabalho foi estudar os fatores que afetam a longevidade das abelhas sem ferrão em ambientes amazônicos. Dessa forma, em três colônias da abelha sem ferrão Melipona fasciculata, foram marcadas individualmente 91 abelhas na estação chuvosa e 109 abelhas na estação seca onde foram registrados diariamente o número de abelhas sobreviventes, a idade de início de forrageio e o tempo gasto em atividades de forrageio, em cada época. Durante a estação chuvosa a longevidade máxima foi de 80 dias enquanto que na estação seca a máxima longevidade foi de 56 dias. A longevidade de operárias foi diferente entre as estações do ano (teste de log-rank p=0,0000), assim como a idade de início de forrageio (U=552; p=0,0000). A longevidade apresentou correlação com a idade de início de forrageio na estação chuvosa (Spearman R=0,23) e seca (Spearman R=0,17), onde observamos que abelhas que forrageavam em idade mais jovem viviam menos do que abelhas que começavam a forragear em idades mais avançadas. Abelhas que forrageavam mais jovens e, no entanto, apresentavam longevidade maior do que outras abelhas, alternavam dias de forrageio e dias dentro do ninho, o que as permitiu aumentar sua longevidade em relação a aquelas que forrageavam vários dias consecutivos. Durante a estação chuvosa apenas um parâmentro da modelagem Weibull foi similar ao observado nas colônias de M. fasciculata, enquanto que durante a estação seca, o padrão de sobrevivência estimado foi totalmente diferente do padrão real. A função de risco apresentou comparativamente diferenças entre as estações, onde as operárias no período seco apresentaram uma probabilidade de morte até duas vezes maior do que no período de chuvas, o que provavelmente torna muito difícil estimar com precisão o padrão de sobrevivência destes insetos. Dessa forma, concluímos que M. fasciculata assim como outras espécies, apresenta longevidade diferenciada entre dois períodos do ano e que a idade de início de forrageio, em parte, pode nos ajudar a compreender o padrão de mortalidade desta espécie de abelha.
