Dans le but de mieux connaître le métabolisme secondaire de la famille des Thymelaeaceae et de découvrir de nouveaux composés naturels à intérêt thérapeutique, 30 extraits provenant de 8 espèces africaines ont été soumis à un criblage chimique et biologique. Les cibles biologiques suivantes ont servi à l?évaluation de l?activité des extraits étudiés : la moisissure phytopathogène Cladosporium cucumerinum, la levure commensale Candida albicans, la bactérie opportuniste Bacillus subtilis, la larve du moustique vecteur de la fièvre jaune Aedes aegypti et l?hôte intermédiaire mollusque de la schistosomiase urinaire Biomphalaria glabrata. Les propriétés antiradicalaires et inhibitrices de l?acétylcholinestérase de ces extraits ont également été dépistées. Des analyses sur CCM avec révélation chimique, ainsi que des expériences LC/DAD-UV, ont permis demettre en évidence la présence de tanins, de flavonoïdes et de xanthones dans les extraits polaires. Sur la base des résultats de ces analyses préliminaires, l?investigation phytochimique des extraits méthanoliques des racines et des parties aériennes de Gnidia involucrata a été entreprise. Cette démarche a permis l?isolement de 8 composés naturels et leur caractérisation complète au moyen de méthodes spectroscopiques (UV, MS, CD, 1H- et 13C-NMR). Les activités de ces produits purs ont été évaluées et il est apparu qu?ils possédaient presque tous des propriétés antiradicalaires intéressantes, supérieures à celles du BHT, un antioxydant de synthèse (E 321) utilisé dans l?industrie alimentaire. Deux benzophénones simples, respectivement O- et C-glucosylées, ont été isolées des parties aériennes de G. involucrata au côté de la mangiférine, une C-glycosylxanthone ubiquitaire. Ces découvertes sont remarquables à plusieurs titres : (1) les benzophénones simples (nonprénylées) sont très rares dans la nature ; (2) c?est la première fois qu?une Oglycosylbenzophénone a été décrite ; (3) aucune xanthone n?avait été mise en évidence auparavant dans la famille et (4) les benzophénones semblent ne pas être que des produits intermédiaires dans la biosynthèse des xanthones. Trois 3,8??-biflavanones du type GB ont été isolées des racines et des parties aériennes de la même plante, dont deux stéréoisomères se trouvant en mélange. Une analyse LC/CD a permis d?attribuer les configurations absolues des quatre carbones asymétriques de chaque molécule. Cette classe de métabolites secondaires est réputée pour ses propriétés analgésiques et sa présence chez les Thymelaeaceae est prometteuse. Des techniques couplées de pointe ont été utilisées dans ce travail et ont montré leur apport inestimable dans le domaine de la recherche phytochimique. Une analyse LC/ MSn a ainsi permis de mettre en évidence on-line trois C-glycosylflavones ? l?isoorientine, l?isovitexine et la vitexine ? dans les extraits méthanoliques bruts de G. involucrata. De plus, les parties aériennes de cette même plante ont servi de matériel pour le développement d?une nouvelle méthode d?analyse d?extraits bruts : la LC/1H-NMR time-slice. Cette approche consiste à « découper » le temps d?analyse par des interruptions régulières du flux LC, durant lesquelles les données NMR nécessaires sont acquises. Le problème de la faible sensibilité relative de la LC/NMR a été partiellement résolu par ce biais et a permis d?envisager l?utilisation de la NMR au sein de systèmes de couplages multiples en série avec d?autres méthodes spectrales (UV, MS, IR, CD,?).<br/><br/>With the aim of acquiring a better knowledge of the secondary metabolism of the family Thymelaeaceae and of the discovering of new natural therapeutics, 30 extracts from 8 African plant species were submitted to chemical and biological screening. The following biological targets were used to estimate the activity of the extracts under study: the phytopathogenic fungus Cladosporium cucumerinum, the commensal yeast Candida albicans, the opportunistic bacteria Bacillus subtilis, larvae of the yellow fever-transmitting mosquito Aedes aegypti and the intermediate snail host of urinary schistosomiasis Biomphalaria glabrata. The antiradical and acetylcholinesterase-inhibiting properties of these extracts were also investigated. TLC analyses followed by chemical detection, together with LC/DAD-UV experiments, showed the presence of tannins, flavonoids and xanthones in the polar extracts. On the basis of these results, a phytochemical investigation of the methanol extracts of the roots and the aerial parts of Gnidia involucrata was undertaken. This procedure led to the isolation of 8 natural products, which were then characterised by spectroscopic means (UV, MS, CD, 1H- and 13C-NMR). The activities of the pure compounds were then further evaluated: almost all of them exhibited very interesting antiradical properties, superior to those of BHT, a synthetic antioxidant (E 321) used in the food industry. Two simple benzophenones, one O- and one C-glycosylated, were isolated from the aerial parts of G. involucrata, together with mangiferin, a ubiquitous C-glycosylxanthone. These findings are of multiple importance: (1) simple (non-prenylated) benzophenones are very rare in nature; (2) it is the first time that an O-glycosylbenzophenone has been described; (3) no xanthones have been previously reported in the family and (4) benzophenones do not seem to be exclusive intermediates in the biosynthesis of xanthones. Three 3,8??-biflavanones of the GB type were isolated from the roots and the aerial parts of the same plant, among them two stereoisomers in mixture. A LC/CD analysis allowed the assignment of the absolute configurations of all four stereocenters in both molecules. This class of secondary metabolite is well known for its analgesic properties and its presence in the Thymelaeaceae is very promising. Advanced hyphenated techniques were used in this work and showed their inestimable contribution to the field of phytochemical research. A LC/MSn analysis, for example, allowed the on-line characterisation of three C-glycosylflavones ? isoorientin, isovitexin and vitexin ? in the crude methanol extracts of G. involucrata. Furthermore, the aerial parts of this plant were used as material for the development of a new analytical method for crude plant extracts: time-slice LC/1H-NMR. This approach consisted in "slicing" the analytical procedure by interrupting the LC flow at given intervals, during which the necessary NMR data were acquired. The relative lack of sensitivity of LC/NMR was partially surmounted by this means, allowing one to envisage the use of NMR in a multiple hyphenated system, together with other spectroscopic methods (UV, MS, IR, CD,?)

The constitutive production of AMPs in shrimps ensures that animals are able to protect themselves from low-level assaults by pathogens present in the environment. As these molecules play important roles in the shrimp immune defense system, the expression level of these AMPs are possible indicators of the immune state of shrimps. The present study also indicates the antiviral property of AMPs, especially ALF, stressing the importance of their up-regulation through the application of immunostimulants/probiotics as a prophylactic strategy in aquaculture. The present study shows that shrimp defense system is equipped enough to evade WSSV infection to a certain extent, when the animals were maintained on marine yeast and probiotic diet, whereas the control diet fed group succumbed to WSSV infection. This study reveals that marine yeast and probiotic supplemented diet can delay the process of WSSV infection and confer greater protection to the animals. Particularly, the protection conferred by marine yeast, C. haemulonii S27 and Bacillus MCCB101 were highly promising imparting greater hope to the aquaculture community to overcome the prevailing disease problems in aquaculture. It may be inferred from the present study that up-regulation of AMP genes could be effected by the application of immunostimulants and probiotics. Also, AMP expression profile could be used as an effective tool for screening immunostimulants and probiotics for application in shrimp culture. Ultimately, it is likely that no single compound or strategy will provide a solution to the problem of disease within aquaculture and that, in reality, a suite of techniques will be required including the manipulation of the rearing environment, addition of probionts as a matter of routine during culture, and the use of immunostimulants and other supplements during vulnerable growth phases. Finally, the development of good management practices, the control of environmental variables, genetic improvement in the penaeid species, understanding of host-virus interaction, modulation of the shrimp immune system, supported by functional genomics and proteomics of this crustacean, as a whole suggests that the control of WSSV is not far.

The antimicrobial activity of irrigating solutions - Endoquil (castor oil detergent), 2% chlorhexidine gluconate solution, and 0.5% NaOCI solution - was evaluated against Gram-positive cocci (Micrococcus luteus, Staphylococcus aureus, Enterococcus faecalis, Staphylococcus epidermidis, Streptococcus mutans, and Streptococcus sobrinus), Gramnegative rods (Escherichia coli and Pseudomonas aeruginosa), and the yeast Candida albicans. Activity was evaluated using the two-layer agar diffusion technique. The base layer was obtained by pouring 10.0 ml of Muller Hinton Medium or 10.0 ml of Brain Heart Infusion agar in a Petri dish. After solidification a 5.0 ml seed layer of Muller Hinton Medium or Brain Heart Infusion agar with inoculum (106/ml) was added. Absorbent paper disks (6.0 mm in diameter) immersed in the solutions were placed at equidistant points. Plates were maintained at room temperature for 2 h for prediffusion of the solutions and incubated at 37°C for 24 h. The candle jar system was used for the Brain Heart Infusion agar plates. All tests were performed in duplicate. After incubation the medium was optimized with 0.05 g% triphenyltetrazolium chlorate gel and inhibition halos were measured. All bacterial strains were inhibited by 2.0% chlorhexidine gluconate. Endoquil was effective against Grampositive microorganisms, and 0.5% NaOCI was effective only against S. aureus. A espécie Pyrostegia venusta é uma liana trepadeira popularmente conhecida como flor-de-São-João. É utilizada na medicina popular para o tratamento de diversas doenças. Na literatura científica, vários compostos bioativos já foram isolados e algumas atividades biológicas importantes, como a atividade antimicrobiana e imunomoduladora, já foram atribuídas a esta espécie. A cárie e a doença periodontalsão as doenças bucais mais prevalentes na população brasileira, sendo o biofilme dental o seu fator etiológico primário. A candidíase oral ocorre devido à infecção por espécies do gênero Candida que, em determinadas situações, podem se comportar como patógenos oportunistas. O objetivo deste trabalho foi realizar um estudo físico, químico e biológico da espécie vegetal Pyrostegia venusta e investigar seu potencial na prevenção das principais doenças bucais. Foram realizados ensaios de citotoxicidade do extrato bruto e frações em células mononucleares do sangue periférico e em macrófagos murinos. Para avaliar a atividade imunomoduladora do extrato bruto e frações, foi realizada dosagem de óxido nítrico (NO) produzido por macrófagos estimulados com lipopolissacarídeo (LPS). Foi realizado ensaio de microdiluição em caldo para determinar a concentração inibitória mínima (CIM), concentração bactericida mínima (CBM) e concentração fungicida mínima (CFM) do extrato bruto, frações e marcadores. Também foram avaliados o efeito na inibição da aderência de Streptococcus mutans em lamínula de vidro, o efeito na redução do pH e a capacidade de inibição do brotamento de Candida albicans do extrato bruto e frações. A espécie Pyrostegia venusta não foi citotóxica em baixas concentrações. A produção de NO foi inibida pelas frações acetato de etila e n-butanólica. O extrato bruto, frações e marcadores apresentaram atividade antimicrobiana contra Streptococcus mutans, Streptococcus mitis, Streptococcus oralis e Candida albicans, capacidade de inibição da aderência e redução do pH. O extrato bruto e frações hexânica, acetato de etila e n-butanólica apresentaram capacidade de inibição do brotamento. A espécie Pyrostegia venusta pode ser uma alternativa na prevenção e tratamento das principais doenças bucais. No entanto, mais estudos são necessários antes de considerá-la realmente promissora.

The increase of the antimicrobial resistance and its propagation around the world are the biggest threats to the public health care and to the treatment of diseases caused by microorganisms. Nowadays the antimicrobial resistance has increased abruptly. The essential oils are volatile and aromatic compounds derived from parts of plants as flowers, leafs, fruits, seeds, roots, sprouts, among others. The activity of extracts and essential oils of several plant species have been recognized and studied by empirical methods since a long time, but its antimicrobial activities were confirmed recently. Medicinal plants are used in folk medicine as medicines, antibiotic, analgesic, sedative and anti-inflammatory. The use of medicinal plants like source of medicines is an alternative of therapeutics for diseases treatment. In Brazil, studies with this goal are very important, once medicinal plants have been used as a choice of treatment and prevention of infections and diseases in health areas. Considering the fact that some products from medicinal plants have antimicrobial properties it is expected that using screening programs, new potential medicaments could be developed. Otherwise, scientific researches focused on determining therapeutic potential of plants are limited, there are lack of scientific studies which confirms the potential antibiotics properties of a large number of plants. The aim of the present study is determinate the antimicrobial activity of 10 medicinal species belonging to CPMA - Collection of Medicinal and Aromatic Plants from CPQBA/UNICAMP. The minimal inhibitory (MIC) and minimal bactericidal or fungicidal concentration (MBC) will be determined against the bacteria Escherichia coli, Pseudomonas aeruginosa, Salmonella choleraesuis, Staphylococcus aureus and the yeast Candida albicans. Furthermore, will be conducted chemical identification and fractionation of essential oils and extract with better activity

Macrophage ingestion of the yeast Candida albicans requires its recognition by multiple receptors and the activation of diverse signaling programs. Synthesis of the lipid mediator prostaglandin E-2 (PGE(2)) and generation of cyclic adenosine monophosphate (cAMP) also accompany this process. Here, we characterized the mechanisms underlying PGE(2)-mediated inhibition of phagocytosis and filamentous actin (F-actin) polymerization in response to ingestion of C. albicans by alveolar macrophages. PGE(2) suppressed phagocytosis and F-actin formation through the PGE(2) receptors EP2 and EP4, cAMP, and activation of types I and II protein kinase A. Dephosphorylation and activation of the actin depolymerizing factor cofilin-1 were necessary for these inhibitory effects of PGE(2). PGE(2)-dependent activation of cofilin-1 was mediated by the protein phosphatase activity of PTEN (phosphatase and tensin homolog deleted on chromosome 10), with which it directly associated. Because enhanced production of PGE(2) accompanies many immunosuppressed states, the PTEN-dependent pathway described here may contribute to impaired antifungal defenses.

A study is made of the benthic foraminifers (size fraction > 63 µm) recovered from 59 upper Eocene through Quaternary sediment samples at DSDP Site 317 (Leg 33), located at a depth of 2598 m in the central part of the Manihiki Plateau (South Pacific). The sediments cored are disturbed in only two samples. The stratigraphic assignements used are based on previous studies of planktic foraminifers and other microfossils. In total, 216 taxa are identified. A cluster analysis based on the 77 species which comprised 5% or more of the entire foraminiferal assemblage in at least one sample suggests the presence of 3 major biostratigraphic zones corresponding approximately to the following ages, zone A: middle Miocene-Quaternary; zones B-C: early Miocene-Oligocene; and zone D: Eocene. The most important faunal turnover occurred between the Eocene and the Oligocene; a less pronounced break took place between the early and the middle Miocene, and an additional minor turnover between the Oligocene and the early Miocene. Eighteen taxa are long-ranging, being recorded from the middle Eocene through the Pliocene-Quaternary. It is concluded that, in general, benthic foraminifers of the bathyal zone are poor worldwide stratigraphic guide fossils; the following taxa are conditionally considered as the most suitable in the Eocene-Quaternary sequence: Aragonia aragonensis, Quadrimorphina profunda, Nuttallides truempyi, Abyssamina poagi, Buliminella grata, Bulimina jarvisi, B. macilenta, Turrilina alsatica, Cibicides notocenicus, C. wuellerstorfi, Pyrgo murrhina. However, most of these species are relatively rare.

Introduction: Licania rigida Benth and Turnera ulmifolia Linn. var. elegans are species of semi-arid regional plants used in the treatment of various diseases. Objectives: The purpose of this study was chemically characterize the extracts and fractions and investigate the antimicrobial and antioxidant potential. Methods: For chemical analysis, were performed spectrophotometric quantification of the total phenolic and characterization of the extracts by chromatographic analysis. Evaluation of antioxidant activity was done by determining the radical scavenging capacity DPPH •. Antimicrobial activity was evaluated by agar diffusion, broth microdilutionand time-kill assays. Results: The extracts and fractions L. rigid and T. ulmifolia showed a high phenolic content, the presence of flavonoids, which were determined as chemical markers. It was observed that the extracts of both species performed as sequestering agents in the trial of antioxidant activity in vitro. The L. rigida extract was the only active front strains of S. aureus 33591 (methicillin-resistant), S. aureus 29213, S. epidermidis 12228, and also against the yeast, Candida albicans, Candida dubliniensis, Candida tropicalis, Candida parapsilosis, Candida rugosa, Candida krusei eTrichosporon asahii. Conclusions: Based on these results it is possibly affirm the antioxidant and antimicrobial activity of L. rigida and attributed the presence of polyphenolic flavonoid like responsible.
