Malária é uma das doenças infecciosas de maior causa de morte no mundo. Modelos experimentais são necessários para melhor compreensão de mecanismos envolvidos na patogênese de doenças e desenvolvimento de novos tratamentos. Galinhas infectadas com Plasmodium gallinaceum fornecem bom modelo de malária devido a proximidade filogenética com o Plasmodium de humano assim como aspectos clínicos comuns, como a malária cerebral. O presente estudo objetivou investigar a participação do óxido nítrico no desenvolvimento da malária aviária, através do tratamento ou não com aminoguanidina (AG - inibidor da enzima Óxido Nítrico Sintase) in vivo de galinhas infectadas experimentalmente com P. gallinaceum. Foi verificado sobrevida, hematologia clássica, bioquímica sérica e patologia nos animais no percurso da infecção. Observou-se maior sobrevida nos animais tratados com AG, apesar de parasitemias mais elevadas. Houve ainda diminuição nos parâmetros hematológicos e aumento no Volume Corpuscular Médio de hemácias, indicando resposta medular para anemia. Linfopenia e trombocitopenia foram detectadas em animais infectados, com menor proporção nos animais tratados. Monócitos, linfócitos e heterófilos apresentaram aumento de tamanho e alterações que indicam ativação. Trombócitos também aumentaram de tamanho durante a infecção e apresentaram morfologia atípica. Os animais tratados mostraram lesões mais brandas nas secções histopatológicas de cérebro, fígado e baço, além de produção diminuída de NO, mesmo em alta parasitemia, em relação aos animais não tratados. Esses resultados confirmam a participação do mediador químico óxido nítrico na patogênese da malária no modelo experimental aviário.

The present study evaluated the liver with homogeneous parenchyma in dogs, with diffuse reduced echogenicity (G1), increased echogenicity (G2) and normal echogenicity (G3) by ultrasound examination associating the findings on animal profile, liver size and hematology and biochemistry results. Blood samples obtained by cephalic or jugular venipunture were submitted for hemogram and ALT, ALP, total proteins, albumin, globulin, urea and creatinine analysis. Among the 30 dogs in G1, 30 (100%) presented portal venous and gall bladder wall clarity, 23 (76.67%) presented normal liver size and edges, 15 (50%) were from 1 to 6 years old and 8 (26.67%) belonged to the lhasa apso breed. No predispositions were found according to gender, as well as no significant alterations in biochemical and hematological exams were observed in G1. The most prevalent disease found in this group was gastroenteritis (43.33%). Among the 30 dogs in G2, 27 (90%) presented hepatomegaly and rounded hepatic edges, 18 (60%) were over 9 years old, 16 (53.33%) were female and 9 (30%) belonged to the poodle breed. The laboratorial findings related to this group were increased ALT and ALP serum activity, decreased levels of total protein, albumin, globulin, erythrocytes and hematocrit, as well as leukocytosis with neutrophilia and a left shift, eosinopenia, lymphopenia and monocytosis. The most prevalent diseases found in this group were metabolic disorders (54%), such as diabetes mellitus and hyperadrenocorticism, and chronic hepatopathies (17%) due to prolonged and continuous use of corticoid and anticonvulsive drugs. Among the 30 dogs in the control group (G3), normal liver size and edges were presented in 22 (73.33%). In this group, no alterations were seen in laboratorial exams.

The leukocyte count interpretation on the blood helps on understanding about the possible dysfunction showed by the animal. In general, an abnormal leukogram allows the identification of pathological process. A lot of events lead to an increase in the number of neutrophils, among then, inflammatory process, infection and stress. Dependent on the stress mediator that the animal is submited, the leukocyte alteration will be different. Is important to know how the stress acts on the animal organism, changing the hematological parameters, in order to avoid mistakes on diseases‟ diagnosis. It‟s known that after a stress situation, it‟s expected that stress leukogram appears on the hemogram of the animal. However there are different kinds of stress with different responses. It‟s believed that when the animal is under an acute stress situation, a clinical picture of physiologic leukocytosis occurs which is mediated by the epinephrine. This is characterized by neutrophilia, lymphocytosis, monocytosis and eosinofilia. The physiologic leukocytosis is a transient alteration in the leukogram that occurs within minutes of the stimulus and due to fleeting effect of catecolamines it resolves within 20 to 30 minutes. However, if the animal is under chronicle stress, as solitude, it „s expected to find the stress leukogram on the leukogram, that can be noted especially in dogs. The stress leukogram is characterized by leukocytosis, neutrophilia, lymphopenia, monocytosis and eosinopenia. This kind of stress is induced by the glucocorticoids. It´s important to understand that stress leukogram takes time to occur. The variation on time depends on the animal species, although, in general, the glucocorticoides have a peak effects among 4 to 8 hours, which can last from 24 hours to 2 or 3 days

Canine Distemper is a contagious, severe and multisystemic disease caused by a virus from Morbillivírus genus. The virus is distributed worldwide and it presents a high lethality rate, affecting mainly dogs. The diagnosis is based on clinical signs associated with hematological fi ndings. The observation of Lentz bodies in erythrocytes and leukocytes is the defi nitive diagnosis for the disease. The aim of this work was to test the hypothesis that the hematological profi le in dogs positive for canine distemper differs according to blood cell type presenting Lentz bodies. For this purpose, 25 dogs positive for the disease were evaluated at the Veterinary Hospital “Luís Quintiliano de Oliveira” UNESP, Araçatuba city. The diagnosis was based on the observation of Lentz bodies in blood smears. Fromthe total, 64% of dogs presented anemia, 16% leucopenia and 12% leukocytosis. Lymphopenia occurred in 76% of dogs. Viral inclusions were observed solely in neutrophils (32%), lymphocytes (28%) and erythrocytes (12%). Concomitant observation occurred in lymphocytes and erythrocytes (4 %), in lymphocytes and neutrophils (12%), in neutrophils and monocytes (4%) and in neutrophils and erythrocytes (4%). In an isolated case Lentz bodies were observed simultaneously in neuthrophils, monocytes and lymphocytes. In conclusion, hematological profi le is not associated with the presence of viral inclusion in a particular cell type.

Background: Antibodies directed against endothelial cell surface antigens have been described in many disorders and have been associated with disease activity. Since the most prominent histopathologic feature in mixed connective tissue disease (MCTD) is the widespread and unique proliferative vascular lesion, our aim was to evaluate the frequency of anti-endothelial cell antibodies (AECA) in this condition. Objectives: To evaluate the frequency of AECA in this disease and assess its clinical and laboratory associations. Methods: Seventy-three sera from 35 patients with MCTD (Kasukawa's criteria), collected during a 7 year period, were tested for immunoglobulins G and M (IgG and IgM) AECA by cellular ELISA, using HUVEC (human umbilical vein endothelial cells). Sera from 37 patients with systemic lupus erythematosus (SLE), 22 with systemic sclerosis (SSc) and 36 sera from normal healthy individuals were used as controls. A cellular ELISA using HeLa cells was also performed as a laboratory control method. Results: IgG-AECA was detected in 77% of MCTD patients, 54% of SLE patients, 36% of SSc patients and 6% of normal controls. In MCTD, IgG-AECA was associated with vasculitic manifestations, disease activity and lymphopenia, and was also a predictor of constant disease activity. Immunosuppressive drugs were shown to reduce IgG-AECA titers. Since antibodies directed to HeLa cell surface were negative, AECA was apparently unrelated to common epitopes present on epithelial cell lines. Conclusions: AECA are present in a large proportion of patients with MCTD and these antibodies decrease after immunosuppressive treatment. IMAJ 2012; 14:84-87

Once rare, septic shock (SS) due to disseminated fungal infections has been increasingly reported due to a growing number of immunocompromised patients, but remains rare in non-immune-compromised individuals. In paracoccidioidomycosis, it has been described in only three patients with the severe, acute form of the disease. We describe the development of a refractory, fatal septic shock due to a severe disseminated chronic form of paracoccidioidomycosis in an older woman without any other microbial insults. A striking event in the evolution of her case was the severe depletion of lymphocytes from the peripheral blood and lymphoid organs. Lymphocyte depletion due to apoptosis is described in the late phase of sepsis and can contribute both to immunosuppression and the progression of SS. The possible mechanisms involved in the induction of SS in the chronic form of paracoccidioidomycosis are discussed.

Objective. To assess the immunogenicity and safety of non-adjuvanted influenza A H1N1/2009 vaccine in patients with juvenile autoimmune rheumatic disease (ARD) and healthy controls, because data are limited to the adult rheumatologic population. Method's. A total of 237 patients with juvenile ARD [juvenile systemic lupus erythematosus (JSLE), juvenile idiopathic arthritis (JIA), juvenile dermatomyositis (JDM), juvenile scleroderma, and vasculitis] and 91 healthy controls were vaccinated. Serology for anti-H1N1 was performed by hemagglutination inhibition assay. Seroprotection rate, seroconversion rate, and factor-increase in geometric mean titer (GMT) were calculated. Adverse events were evaluated. Results. Age was comparable in patients and controls (14.8 +/- 3.0 vs 14.6 +/- 3.7 years, respectively; p = 0.47). Three weeks after immunization, seroprotection rate (81.4% vs 95.6%; p = 0.0007), seroconversion rate (74.3 vs 95.6%; p < 0.0001), and the factor-increase in GMT (12.9 vs 20.3; p = 0.012) were significantly lower in patients with juvenile ARD versus controls. Subgroup analysis revealed reduced seroconversion rates in JSLE (p < 0.0001), JIA (p = 0.008), JDM (p = 0.025), and vasculitis (p = 0.017). Seroprotection (p < 0.0001) and GMT (p < 0.0001) were decreased only in JSLE. Glucocorticoid use and lymphopenia were associated with lower seroconversion rates (60.4 vs 82.9%; p = 0.0001; and 55.6 vs 77.2%; p = 0.012). Multivariate logistic regression including diseases, lymphopenia, glucocorticoid, and immunosuppressants demonstrated that only glucocorticoid use (p = 0.012) remained significant. Conclusion. This is the largest study to demonstrate a reduced but adequate immune response to H1N1 vaccine in patients with juvenile ARD. It identified current glucocorticoid use as the major factor for decreased antibody production. The short-term safety results support its routine recommendation for patients with juvenile ARD. The leaves of the Cashew plant (Anacardium occidentale L.) are used by the folk medicine in South America and West Africa. This plant is rich in flavonoids, which are polyphenolic compounds widespread in plants, and that have diverse physiological effects. In a sub-acute toxicity assay it was found that an ethanolic extract of Cashew leaves elicited lymphopenia in rats. The extract was also found to be cytotoxic and to induce apoptosis in Jurkat (acute lymphoblastic leukemia) cells. The crude ethanolic extract was fractionated and resolved by HPLC. One of the four fractions obtained led to the isolation of the biflavonoid agasthisflavone. [H-3]-thymidine incorporation assays and flow cytometry analysis showed that the isolated compound displayed a high anti-proliferative effect in Jurkat cells with an IC50 of 2.4 mu g/ml (4.45 mu M). The effect of agathisflavone on the acute promyelocytic leukemia cell line HL60, Burkitt lymphoma Raji cells and Hep-2 laryngeal carcinoma cells was also tested. The two latter ones were only mildly affected by agathisflavone. It is also shown that agathisflavone induces apoptosis in Jurkat cells and it this proposed that this is the likely mechanism of agathisflavone specific cytotoxicity. (C) 2010 Elsevier GmbH. Case Description-A 9-year-old spayed female green iguana (Iguana iguana) was evaluated because of a distended coelom and weight loss. History included a single episode of egg binding and subsequent bilateral ovariosalpingectomy.;Clinical Findings-Physical examination revealed a mass within the coelomic cavity. Ultrasonography revealed a large, irregular mass with hypoechoic regions and coelomic effusion. Clinicopathologic derangements included heterophilia, monocytosis, lymphopenia, basophilia, hypocholesterolemia, hypoproteinemia, and hypercalcemia. Results of cytologic evaluation of the mass were suggestive of malignant epithelial neoplasia, but neoplastic cells were not found in the effusion. An ovarian tumor was suspected on the basis of clinical signs, clinicopathologic findings, and results of cytologic evaluation of the mass.;Treatment and Outcome-Surgical exploration revealed a large left ovary, a normal-appearing contralateral ovary, and a mass in the fat body, all of which were removed and submitted for histologic examination. The histologic diagnosis was granulosa cell tumor with metastasis to the fat body. The patient died 11 months after evaluation, and disseminated granulosa cell tumor was confirmed at necropsy; histologic examination at that time also identified systemic mastocytosis.;Clinical Relevance-Granulosa cell tumors are uncommon in reptiles, and this was the first granulosa cell tumor described antemortem cytologically, histologically, and ultrastructurally in an iguana. Findings in this iguana underscored concerns associated with incomplete oophorectomy of iguanas; cytologic and histopathologic findings were similar to those observed in other domestic animals. Oophorectomy should be considered as an alternative to standard ovariosalpingectomy to avoid potential complications in pet reptiles, and use of microsurgical instruments and vascular clips is advised. Detection of persistent infection with BovineViral Diarrhea Virus (BVDV) is essential for both epidemiological and clinical reasons. In addition to the classical virological methods such as virus isolation in tissue culture, ELISA and RT-PCR, immunohistochemistry of skin biopsies has become a useful and reliable tool. Assuming that the presence of BVDV antigen in skin structures is restricted to persistent infection, this method could differentiate from transient infection. In order to answer this question, 6 calves were experimentally infected orally with a non-cytopathic genotype 1 BVDV strain belonging to the subtype k.The calves developed fever, mucopurulent nasal discharge, coughing and leucopenia with relative lymphopenia. Immunohistochemistry of skin biopsies taken daily up to day 13-post infection did not reveal any evidence of BVDV infection. BVDV was, however, isolated from blood samples on cell cultures. Anti-NS3-antibody-ELISA and serum neutralization tests showed that all six calves seroconverted. We conclude that in acute BVDV infections, with genotype 1 and the subtypes found in Switzerland (b, e, h and k) viral antigen is not found in epidermal structures of the skin. In contrast, persistently infected animals test positive for BVD viral antigen by immunohistochemistry of the skin.
