Background: Peroxiredoxins have diverse functions in cellular defense-signaling pathways. 2-Cys-peroxiredoxins (2-Cys-Prx) reduce H2O2 and alkyl-hydroperoxide. This study describes the purification and characterization of a genuine 2-Cys-Prx from Vigna unguiculata (Vu-2-Cys-Prx). Methods: Vu-2-Cys-Prx was purified from leaves by ammonium sulfate fractionation, chitin affinity and ion exchange chromatography. Results: Vu-2-Cys-Prx reduces H2O2 using NADPH and DTT. Vu-2-Cys-Prx is a 44 kDa (SDS-PAGE)/46 kDa (exclusion chromatography) protein that appears as a 22 kDa molecule under reducing conditions, indicating that it is a homodimer linked intermolecularly by disulfide bonds and has a pI range of 4.56-4.72; its NH2-terminal sequence was similar to 2-Cys-Prx from Phaseolus vulgaris (96%) and Populus tricocarpa (96%). Analysis by ESI-Q-TOF MS/MS showed a molecular mass/pI of 28.622 kDa/5.18. Vu-2-Cys-Prx has 8% alpha-helix, 39% beta-sheet, 22% of turns and 31% of unordered forms. Vu-2-Cys-Prx was heat stable, has optimal activity at pH 7.0, and prevented plasmid DNA degradation. Atomic force microscopy shows that Vu-2-Cys-Prx oligomerized in decamers which might be associated with its molecular chaperone activity that prevented denaturation of insulin and citrate synthase. Its cDNA analysis showed that the redox-active Cys(52) residue and the amino acids Pro(45), Thr(49) and Arg(128) are conserved as in other 2-Cys-Prx. General significance: The biochemical and molecular features of Vu-2-Cys-Prx are similar to other members of 2-Cys-Prx family. To date, only one publication reported on the purification of native 2-Cys-Prx from leaves and the subsequent analysis by N-terminal Edman sequencing, which is crucial for construction of stromal recombinant 2-Cys-Prx proteins. The cowpea (Vigna unguiculata L. Walp) is a major food crops in northeastern Brazil. In Rio Grande do Norte, the cowpea, vigna beans or cowpea, as it is known, has great socioeconomic importance as a source of nutrients in food, with great emphasis among agricultural products. To improve productivity and resistance to pests, two cultivars were developed exclusively by EMPARN (Agricultural Research Corporation of Rio Grande do Norte), for breeding. The samples were provided by EMPARN, two improved (Potiguar and Laugh year) and two landraces (Rib of beef and Canapu). The seeds were ground and made into flour samples and the determination of moisture and ash by graviméticos methods, lipids by Soxhlet extraction, fibers with determiner fiber, carbohydrates by difference and minerals by ICP-OES were performed except the match analyzed by UVvis. The results showed a high fiber content (55.55% and 55.32% and 50.04% improved samples and 50.32% creole samples) and protein (25.52% and 25.27% improved and 27 samples, 23% and 24.99% creole samples). Comparing the results of the mineral content, the improved cultivars stood out in relation to Ca, Co, P, Mg, Mo and Na. Creole cultivars showed better results for Cu, Cr (low), Fe, Mn, Ni, K and Zn. The results will be important in future nutritional research and to build a table of Brazilian chemical composition of foods.

Chitin is an important structural component of the cellular wall of fungi and exoskeleton of many invertebrate plagues, such as insects and nematodes. In digestory systems of insects it forms a named matrix of peritrophic membrane. One of the most studied interaction models protein-carbohydrate is the model that involves chitin-binding proteins. Among the involved characterized domains already in this interaction if they detach the hevein domain (HD), from of Hevea brasiliensis (Rubber tree), the R&R consensus domain (R&R), found in cuticular proteins of insects, and the motif called in this study as conglicinin motif (CD), found in the cristallography structure of the β-conglicinin bounded with GlcNac. These three chitin-binding domains had been used to determine which of them could be involved in silico in the interaction of Canavalia ensiformis and Vigna unguiculata vicilins with chitin, as well as associate these results with the WD50 of these vicilins for Callosobruchus maculatus larvae. The technique of comparative modeling was used for construction of the model 3D of the vicilin of V. unguiculata, that was not found in the data bases. Using the ClustalW program it was gotten localization of these domains in the vicilins primary structure. The domains R&R and CD had been found with bigger homology in the vicilins primary sequences and had been target of interaction studies. Through program GRAMM models of interaction ( dockings ) of the vicilins with GlcNac had been gotten. The results had shown that, through analysis in silico, HD is not part of the vicilins structures, proving the result gotten with the alignment of the primary sequences; the R&R domain, although not to have structural similarity in the vicilins, probably it has a participation in the activity of interaction of these with GlcNac; whereas the CD domain participates directly in the interaction of the vicilins with GlcNac. These results in silico show that the amino acid number, the types and the amount of binding made for the CD motif with GlcNac seem to be directly associates to the deleterious power that these vicilins show for C. maculatus larvae. This can give an initial step in the briefing of as the vicilins interact with alive chitin in and exert its toxic power for insects that possess peritrophic membrane

Estudou-se o nivel de dano economico de Empoasca Kraemeri Ross & Moore em Vigna unguiculata (L.), em condicoes de campo, em Petrolina, Pernambuco. Neste estudo, correlacionou-se a percentagem de reducao de produtividade de V. unguiculata com o nivel medio de infestacao de E. Kraemeri, no periodo e 20 a 60 dias apos o plantio.

Estudou-se a resistência de 50 variedades de Vigna unguiculataWalp ao ataque de Liriomyza sativae Blanchard, sob condições de irrigação por sulco. As variedades que sofreram os menores e os maiores danos causados pelo inseto foram: VITA-3 e 5F-PI-112, respectivamente, observando-se susceptibilidades intermediárias das variedades estudadas entre estes dois extremos.

O feijão-de-metro é um importante alimento, rico em proteína, vitaminas, fibras e minerais. Muito consumido na região Norte do país, principalmente no estado do Pará, onde é conhecido como feijão-verde. Os materiais utilizados atualmente são os mesmos introduzidos no início do século passado. São comercializadas sementes de uma única cultivar registrada no Registro Nacional de Cultivares (RNC) e de outra que não tem registro. Este trabalho foi realizado com o objetivo de avaliar linhagens de feijão-de-metro em Belém-PA. Foram avaliados 66 acessos no delineamento de blocos de Federer, com duas testemunhas comuns. O experimento foi conduzido na Embrapa Amazônia Oriental, em delineamento de Blocos Federer. Foram avaliados caracteres qualitativos e quantitativos. Com base na produtividade e nos caracteres cor, comprimento e aspecto geral da vagem, importantes para a aceitação comercial, destacaram-se com produtividade superior à 1,6 kg/2m2 os seguintes acessos: China town (PI 487501), TVO 2141 (MNC 1618), CP-74 (PI 487550), Dagupan pangasinan (PI 487486).

A survey was conducted to identify viruses affecting dry bean (Phaseolus vulgaris) and cowpea (Vigna unguiculata) in Lebanon. Three hundred and thirty four samples exhibiting virus-like symptoms were collected from 13 different locations during the fall growing season of 1984. Samples were stored at 20 deg C until they were tested by the enzyme-linked immunosorbent assay (ELISA) for the presence of blackeye cowpea mosaic virus (B1CMV), bean yellow mosaic virus (BYMV), bean common mosaic virus (BCMV) and cucumber mosaic virus (CMV). In preliminary tests, the extraction buffer 0.1M phosphate + O.1MEDTA, pH 7.4 was found to be far better than the standard extraction buffer and, accordingly, was used for virus extraction for all field samples. Results obtained indicated that around 50% of the bean samples tested were infected with B1CMV. Incidence of BCMV, BYMV and CMV in the samples tested were 4,4 and 1.7%, respectively. B1CMV was detected in 10 locations, whereas, BYMV, BCMV and CMV were found in 1,4 and 4 locations, respectively. Mixed infections such as BCMV, BICMV, BCMV+CMV, BYMV+CMV and BICMV+BCMV+CMV were detected. In 35% of the samples assayed, the causal virus was not identified

Se llevó a cabo un estudio con el objetivo de evaluar el comportamiento agronómico de especies y variedades de Vignas forrajeras (Vigna unguiculata, Vigna unguiculata, Vigna unguiculata, Vigna unguiculata y Vigna radiata). El Ensayo se realizó durante el período Agosto – Diciembre, 2009.en la Finca Santa Rosa, Universidad Nacional Agraria. Managua, Nicaragua. Los tratamiento consistieron en cuatro variedades de Vigna unguiculata de reciente introducción y un testigo de Vigna radiata.. El diseño utilizado fue un BCA (Bloques completo al Azar) con tres repeticiones. Las variables de estudio para cada tratamiento fueron, porcentajes de germinación, Altura de plantas (cm), Número de hojas (Conteo visual), Porcentaje de cobertura. Se realizaron análisis de varianza (ANDEVA) y separaciones de medias, usando Duncan (P<0.05). Para el análisis estadístico las variables codificadas en porcentajes se transformaron, según, arco seno 2 p (Arco seno de dos veces la raíz cuadrada de la proporción). Los resultados encontrados demuestran diferencias significativas (P ≤ 0.05) para todas las variables de estudio. El porcentaje de germinación varío de 61 % a 82 % para Vigna unguiculata CV Vun 1 y Vigna radiata CV Mungo respectivamente, mientras la Altura de plantas vario de 17.31 cm. a 33.32 cm. para Vigna unguiculata CV Vun 2 y V.unguiculata CV Vun 1. El mayor número de hojas se presentó en Vigna unguiculata CV Vun Naranja y el menor lo presentó Vigna radita CV Mungo con 10 y 4 hojas respectivamente. La mayor cobertura del suelo se presentó en Vigna unguiculata CV Vun Naranja con 42.17 % y la menor en Vigna unguiculata CV Vun 2 con 18.48 %. Se concluye que la especie Vigna unguiculata y los cultivares Vun Naranja y Vun 1 son las que presentaron un mejor comportamiento agronómico y pueden pasar a otras etapas de evaluación para su utilización en la alimentación animal.

Con la finalidad de estudiar el comportamiento del crecimiento, desarrollo y rendimiento de los cultivos de sorgo (Sorghum bicolor. L. Moench) híbrido D-55 y maíz (Zea mays L) variedad NB-30, bajo los efectos de diferentes cultivos leguminosos: Maní (Arachis hypogaea L. ), caupí (Vigna unguiculata L. Walp), soya (Glycine max L. Merril) y frijol común ( Phaseolus vulgaris L.) como cultivos antecesores, se realizó un experimento de campo, utilizando un diseño de Bloques Completos al Azar con doce tratamientos y cuatro repeticiones realizados durante la siembra de primera y postrera de 1991, en el Centro Experimental La Compañía; Carazo, cuyo suelos son de topografía plana, textura media, franco limoso ( Typic Durandept ). De los resultados obtenidos se llegó a la conclusión que los diferentes cultivos leguminosos influyeron sobre el crecimiento, desarrollo y rendimiento de los cultivos de sorgo y maíz, ya que estos fueron afectados de manera significativa por los cultivos antecesores. Por otro lado se comprobó que el sorgo y el maíz, cuando tienen como cultivo antecesor a las leguminosas, sus rendimientos fueron superiores que cuando se tiene al sorgo y al maíz como cultivo antecesor. De las leguminosas, el cultivo de maní tuvo un mayor efecto sobre el rendimiento del sorgo y maíz. (4,391 y 1,675 kg/ha respectivamente). Se comprobó cuantitativamente el efecto negativo de utilizar a los cultivos de sorgo y maíz como cultivos antecesores a su mismo cultivo, ya que los rendimientos disminuyen considerablemente, siendo estos los siguientes: sorgo-sorgo obtuvo 2,170 kg/ha., maíz-sorgo dio un rendimiento de 2,373 kg/ha, maíz-maíz presentó 762 kg/ha, y finalmente el tratamiento sorgo-maíz rindió 674 kg/ha.
