Connectivity mapping is the process of establishing connections between different biological states using gene-expression profiles or signatures. There are a number of applications but in toxicology the most pertinent is for understanding mechanisms of toxicity. In its essence the process involves comparing a query gene signature generated as a result of exposure of a biological system to a chemical to those in a database that have been previously derived. In the ideal situation the query gene-expression signature is characteristic of the event and will be matched to similar events in the database. Key criteria are therefore the means of choosing the signature to be matched and the means by which the match is made. In this article we explore these concepts with examples applicable to toxicology.

Studies on persistence and degradation of the synthetic pyrethroid insecticides, permethrin and fenvalerate, were carried out under natural environmental conditions of the Niagara Peninsula. Permethrin and fenvalerate were treated on apple foliage atrat~s of 0.21 kg(AI)!ha and 0.14 kg(AI)/ha, respectively. The initial cis- and trans-permethrin spray deposits were found to be 13.5 ppm and 19.2 ppm, respectively and 38.0 ppm was observed for the fenvalerate treated sample. Twenty-three days and 84 days after spray application, permethrin residues were 4.0 ppm and 2.7 ppm for the cis-isomer, whereas they were 7.9 ppm and 4.7 ppm for the trans-isomer, respectively. Residues of fenvalerate 23 days and 84 days after spray application were 13.4 ppm and 8.0 ppm, respectively. The values of observed half-life of cis-permethrin, trans-permethrin and fenvalerate were found to be 42 days, 46 days and 51 days, respectively. Studies were extended to quantitatively determine some of the major degradation compounds of permethrin and fenvalerate, which were expected to be produced as results of ester cleavage of the parent compounds. A permethrin treated sample, 84 days after initial spray application, showed 0.25 and 0.8 ppm of cis- and trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylic acid (C12CA (18), respectively. These two acids were not found as free acids, but found as conjugated compounds. The other expected degradation compounds, 3-phenoxybenzyl alcohol (PBalc (~)),3-phenoxybenz.aldehyde (PBald (38)) and 2- (4-chlorophenyl) isovaleric acid (CPIA (31)) were not detected by the methods employed in this study. The results indicate that these degradation compounds were not present, or, if they were present, their concentrations were too low to detect by the methods used.

Les pyréthrinoïdes sont des insecticides largement utilisés. La population générale y est exposée par l’alimentation tandis que les travailleurs agricoles y sont exposés lors de tâches diverses en champs. Leurs effets neurotoxiques, immunitaires et endocriniens potentiels en font des composés à surveiller pour assurer la santé de la population. La mesure de biomarqueurs d’exposition, qui consiste à quantifier la concentration dans l’organisme d’une substance ou de ses métabolites, permet d’estimer les doses absorbées. Les biomarqueurs peuvent également être des molécules répondant à un stress physiologique, identifiées comme des biomarqueurs d’effets. Pour raffiner les stratégies de biosurveillance de l’exposition, on se doit de bien connaître la toxicocinétique d’un xénobiotique; actuellement, les études de biosurveillance considèrent rarement la variabilité temporelle, intra-invidivuelle et inter-individuelle, qui pourrait influencer l’estimation de l’exposition. L’objectif de la thèse est donc d’appliquer une approche cinétique pour l’évaluation de l’exposition aux pyréthrinoïdes en conditions contrôlées et en milieu de travail. Dans un volet exploratoire, l’effet de cette exposition sur des changements métaboliques précoces a également évalué. Trois métabolites finaux (cis-DCCA, trans-DCCA et 3-PBA) de deux pyréthrinoïdes les plus utilisés, soient la perméthrine et la cyperméthrine, ont été mesurés dans le plasma et l’urine de six volontaires oralement exposés à une dose équivalente à la dose de référence. Une demi-vie moyenne (t½) d’élimination apparente du trans-DCCA, cis-DCCA et 3-PBA dans le plasma de 5,1, 6,9 et 9,2 h, respectivement, a été obtenue après exposition orale à la cyperméthrine, comparativement à 7,1, 6,2 et 6,5 h après exposition à la perméthrine. Dans l’urine, la demi-vie d'élimination apparente (t½) était de 6,3, 6,4 et 6,4 h pour le trans-DCCA, cis-DCCA et 3-PBA, respectivement, après administration de la cyperméthrine comparé à 5,4, 4,5 et 5,7 h après administration de la perméthrine. Les profils temporels étaient semblables suite à l’exposition à la cyperméthrine et perméthrine. Ensuite, une étude en milieu agricole a été réalisée avec la participation de travailleurs pour évaluer leur exposition et raffiner les stratégies de biosurveillance. La variabilité intra-individuelle dans les niveaux de biomarqueurs d’exposition chez plusieurs travailleurs était plus importante que la variabilité inter-individuelle. Les échantillons urinaires ont également été utilisés pour identifier des modifications du métabolome pouvant fournir de nouveaux biomarqueurs d’effets précoces. Chez les travailleurs, une augmentation de l'hippurate urinaire (p <0,0001) a été observée après exposition aux pyréthrinoïdes, un biomarqueur de la conjugaison de l’acide benzoïque. En conclusion, cette étude a permis de mieux documenter la cinétique de biomarqueurs d’exposition aux pyréthrinoïdes dans des conditions contrôlées et réelles afin de raffiner les stratégies de biosurveillance. Elle a aussi contribué à renseigner sur les niveaux d’exposition agricole québécois et sur les paramètres professionnels associés à une plus forte exposition. Ce projet s’insère dans une démarche d’analyse de risque en santé au travail.

This thesis embodies findings on a taxonomical investigation of a group of lower marine invertebrates belonging to the category coelomata. Bryozoans are well known both in fossil and recent taxonomical history. They comprise of about 5,000 living and 16000 fossil species. Bryozoans are well known for their taxonomic abundance and structural diversity,representing the various ecological niches ranging from the intertidal to the abyssal benthic. At a time when global marine biological diversity has become a concern of not only to the scientists but also to the policy makers,an understanding of species diversity and abundance are cardinal aspects of biological studies. Geological time scales which is known that by Pre-Cambrian, marine invertebrate diversity reach the maximum and this diversity has become more comprehensive as time advanced. Taxonomists a vanishing species of scientists have become more concerned in discerning patterns of species diversity. The basic tool for this is identification fo animals. with this idea in mind a detailed study of taxonomy of bryozoan was undertaken . The major part of this thesis is devoted to describe various species of bryozoans with detailed description and ecotypical variations.The pattern of distribution and abundance which are important aspects of animal groups have also been documented. Possible effects of heavy metal contamination on the tolerance and growth of bryozoans, a few species of which have been eliminated from the chronically polluted areas of Cochin backwaters have also been documented.

Instead of developing easily degradable, and low-priced insecticides, we are going after highly sophisticated chemicals. Here, an attempt is being made to develop safer formulations of insecticides of botanical origin. Different parts of the plants were chosen based on their use in countryside and villages The dried plant materials were extracted with petroleum ether, and were applied on Tribolium castaneum. The results were statistically analysed. The active principles from Croton tigilium and Leea sambucina, the most potential plants, were isolated using Column Chromatography, TLC, and Hydrolysis. The isolated principles were analysed spectroscopically ( UV-Vis., IR, NMR, and MS ) to identify their chemical nature. The active principles from Leea and Croton were identified as a cholisterate derivative and a phorbol derivative respectively. In order to ascertain the environmental combatibility of the principles, degradation by soil bacteria was studied. The isolated principles were made into three type of formulations using stabilizers .The formulations were applied on Snake gourd semilooper, Pulse beetle, and mosquito larvae. Also the biocidal activity of the formulations was studied. Both Leea derivative and Croton derivative could be formulated effectively and were effective against a variety of pests. They are eco-friendly, as there is no artificial chemicals involved.

The title of the study is ''Toxicology Literature: An Informetric Analysis".In the field of Toxicology, the interdisciplinary research resulted in 'information fragmentation' of the basic subject to environmental, medical and economic toxicology. The interest in collaborative research resulted in the transdisciplinary growth of Toxicology which ultimately resulted in the scatter of literature.For the purpose of present study Toxicology is defined as the physical and chemical aspects of all poisons affecting environmental, economical and medical aspects of human life. Informetrics is "the use and development of a variety of measures to study and analyse several properties of information in general and documents in particular."The present study fled light on the main fields of Toxicology research as well as the important primary journals through which the results are being published. The authorshippattern, subject-wise scatter, country-wise, language-wise and growth pattern, self-citation, bibliographic coupling of the journals were studied. The study will be of great use in forrnulatinq the acquisition policy of documents in a library. The present study is useful in identifying obsolate journals so that they can be discarded from the collection

Objetivos Mediante un estudio descriptivo transversal, establecer la presencia de una relación entre los niveles de exposición a plaguicidas con la presencia de hallazgos clínicos neurológicos periféricos en personas expuestas ocupacionalmente a estos. Resultados La muestra poblacional fue de 59 trabajadores, en el área rural del municipio del Espinal, Tolima. El 52% de la población refirió usar organofosforados, 32 % carbamatos, 16% organoclorados, y 66% otros grupos de plaguicidas. Los niveles sanguíneos de carbamatos estuvieron presentes en el 7% de la población, organofosforados en el 66% y organoclorados en el 100%. En el 42% de la población se estableció sospecha de neuropatía a partir de los síntomas y en el 92% a partir de los signos. Se estableció asociación estadísticamente significativa entre los niveles sanguíneos de determinados organoclorados con la sospecha de neuropatía periférica a partir de síntomas; la presencia de sensación de pérdida de la fuerza en extremidades y la presencia de alteraciones en los reflejos osteotendinosos. Conclusiones El presente estudio permitió identificar los plaguicidas a los cuales está expuesta la población agrícola del municipio del Espinal así como corrobora el efecto de bioacumulación y biomagnificación de los organoclorados. Adicionalmente, se detectó la presencia de hallazgos sugestivos de compromiso neurológico periférico, con asociaciones significativas entre ciertas anormalidades y determinados organoclorados. Este estudio es una aproximación a la relación entre la exposición a plaguicidas de la población agrícola de la zona rural del municipio del Espinal con la secuelas neurológicos periféricas asociadas a esta.
