Les complications vasculaires telles que l’augmentation de la contractilité et la prolifération cellulaire sont les complications les plus communes observées dans le diabète et l’hyperglycémie chronique est un facteur important dans ces processus. La voie de signalisation de Gαq joue un rôle important dans la régulation du tonus vasculaire et l’altération de celle-ci peut contribuer aux complications vasculaires observées dans les cas de diabète et d’hyperglycémie. Il a été observé que les taux et l’activité des protéines kinase C (PKC) et du diacylglycérol (DAG) sont augmentés dans ces conditions. Cependant, aucune étude n’a démontré l’implication de Gαq/11 et des PLCβ, molécules de signalisation en amont de PKC/DAG. Plusieurs études révèlent que l’augmentation des taux et de l’activité des PKC et du DAG induite par l’hyperglycémie dans des cellules du muscle lisse vasculaire (CMLV) est attribuée à l’augmentation du stress oxydatif. De plus, les niveaux de certains peptides vasoactifs, tels que l’angiotensine II et l’endothéline-1, augmentés dans les conditions de diabète/d’hyperglycémie, peuvent contribuer à l’augmentation du stress oxydatif observée. Le travail présenté dans cette thèse avait pour but d’examiner les effets de l’hyperglycémie sur les niveaux d’expression protéique de Gαq/11 et de ses molécules associées, ainsi que d’étudier le mécanisme moléculaire par lequel l’hyperglycémie module la voie de signalisation de Gαq dans les CMLV. Dans la première étude, nous avons examiné si l’hyperglycémie pouvait moduler l’expression des protéines Gαq, Gα11, PLCβ1 et PLCβ2. Le prétraitement des CMLV A10 avec 26 mM de glucose durant 72 heures augmente l’expression des protéines Gαq, Gα11, PLCβ-1 et PLCβ-2 en comparaison avec les CMLV témoins. Le traitement avec des antagonistes aux récepteurs AT1 de l’Ang II, et ETA/ETB de l’ET-1, atténue la hausse de Gαq, de Gα11, de PLCβ1 et de PLCβ2 induite par l’hyperglycémie. De plus, la formation d’IP3 stimulée par l’ET-1 était plus élevée dans les CMLV exposée à 26 mM de glucose. Le traitement des CMLV A10 avec l’Ang II et l’ET-1 augmente également les niveaux d’expression des protéines Gα q/11 et PLCβ. Cette augmentation de l’expression est restaurée au niveau des CMLV témoins par les antagonistes des récepteurs AT1, ETA et ETB. Ces résultats suggèrent que l’augmentation de l’expression des protéines Gαq/11 et PLCβ dans les CMLV induite par l’hyperglycémie est attribuée à l’activation des récepteurs AT1, ETA et ETB. Dans la seconde étude, nous avons examiné l’implication du stress oxydatif dans l’augmentation des niveaux d’expression des protéines Gαq/11 et PLCβ et de leur signalisation induite par l’hyperglycémie. Nous avons également déterminé le mécanisme responsable de l’augmentation du stress oxydatif induite par l’hyperglycémie. L’augmentation de l’expression des protéines Gαq/11 et PLCβ des CMLV A10 exposées à 26 mM de glucose est revenue au niveau basal après un traitement avec l’antioxydant diphenyleneiodonium (DPI), et la catalase, un chélateur du peroxyde d’hydrogène, mais pas par le 111Mn-tetralis(benzoic acid porphyrin) (MnTBAP) ni par l’acide urique, des chélateurs du peroxynitrite. De plus, l’augmentation de la formation d’IP3 stimulée par l’ET-1 dans les CMLV exposées à 26 mM de glucose est revenue au niveau basal après un traitement avec le DPI et la catalase. Ces résultats suggèrent que l’augmentation du stress oxydatif induite par l’hyperglycémie contribue à l’augmentation de l’expression des protéines Gαq/11 et les molécules associées à la voie de signalisation de Gq. De plus, l’augmentation de la production d’anion superoxyde (O2-), de l’activité de la NADPH oxydase et de l’expression des protéines p22(phox) et p47(phox) induite par l’hyperglycémie est revenue à un niveau basal après un traitement avec les antagonistes des récepteurs AT1, ETA et ETB. Ces résultats suggèrent que l’hyperglycémie augmente les niveaux endogènes de l’Ang II et de l’ET-1, ce qui augmente le stress oxydatif par la formation d’O2- et de H2O2 et peut contribuer à l’augmentation des niveaux de Gq/11α et de leurs molécules de signalisation. Puisqu’il a été observé que l’hyperglycémie transactive les récepteurs aux facteurs de croissance tels que le récepteur au facteur de croissance épidermique (EGF-R) et le récepteur au facteur de croissance dérivé des plaquettes (PDGF-R), nous avons entrepris d’examiner, dans la troisième étude, l’implication d’EGF-R et de PDGF-R dans l’augmentation des niveaux de Gαq/11, de PLCβ et de leur signalisation induite par l’hyperglycémie. L’augmentation des niveaux d’expression des protéines Gαq, Gα11, PLCβ-1 et PLCβ-2 induite par l’hyperglycémie est revenue au niveau basal après un traitement avec les inhibiteurs d’EGF-R (AG1478) et de PDGF-R (AG1295) et par l’inhibiteur de c-Src, PP2. L’augmentation de la phosphorylation d’EGF-R et de PDGF-R induite par l’hyperglycémie a été abolie par AG1478, AG1295 et PP2. De plus, l’augmentation des niveaux de Gαq/11, et de PLCβ induite par l’hyperglycémie est atténuée par l’inhibiteur des MAPK, le PD98059, et par l’inhibiteur d’AKT, le wortmannin. L’augmentation de la phosphorylation d’ERK et d’AKT était également atténuée par AG1478 et AG1295. Ces résultats suggèrent que la transactivation des récepteurs aux facteurs de croissance induite par c-Src peut contribuer à l’augmentation des niveaux de Gα q/11/PLC et de leur signalisation par la voie des MAPK/PI3K. En conclusion, les études présentées dans cette thèse indiquent que l’hyperglycémie augmente les niveaux de Gαq/11 et de PLCβ. Nous avons émis des évidences qui démontrent que l’augmentation endogène de l’Ang II et de l’ET-1 par l’hyperglycémie peut contribuer à l’augmentation de la production d’O2- et de H2O2 résultant ainsi en une augmentation du stress oxydatif qui pourrait être responsable de l’augmentation de Gαq/11/PLC et de leur signalisation dans les conditions d’hyperglycémie. Finalement, nous avons démontré que la transactivation des récepteurs aux facteurs de croissance induite par l’hyperglycémie peut être responsable de l’augmentation de Gαq/11/PLC et les molécules associées à la voie de signalisation de Gq dans les cas de diabète et d’hyperglycémie.

The present study was designed to investigate the protective effect of glucose, oxygen and epinephrine resuscitation on impairment in the functional role of GABAergic, serotonergic, muscarinic receptors, PLC, BAX, SOD, CAT and GPx expression in the brain regions of hypoxia induced neonatal rats. Also, the role of hormones - Triiodothyronine (T3) and insulin, second messengers – cAMP, cGMP and IP3 and transcription factors – HIF and CREB in the regulation of neonatal hypoxia and its resuscitation methods were studied. Behavioural studies were conducted to evaluate the motor function and cognitive deficit in one month old control and experimental rats. The efficient and timely supplementation of glucose plays a crucial role in correcting the molecular changes due to hypoxia, oxygen and epinephrine. The sequence of glucose, epinephrine and oxygen administration at the molecular level is an important aspect of the study. The additive neuronal damage effect due to oxygen and epinephrine treatment is another important observation. The corrective measures by initial supply of glucose to hypoxic neonatal rats showed from the molecular study when brought to practice will lead to healthy intellectual capacity during the later developmental stages, which has immense clinical significance in neonatal care.

This thesis Entitled Neuronal degeneration in streptozotocin induced diabetic rats: effect of aegle marmelose and pyridoxine in pancreatic B cell proliferation and neuronal survival. Diabetes mellitus, a chronic metabolic disorder results in neurological dysfunctions and structural changes in the CNS. Antioxidant therapy is a challenging but necessary dimension in the management of diabetes and neurodegenerative changes associated with it. Our results showed regional variation and imbalance in the expression pattern of dopaminergic receptor subtypes in diabetes and its role in imbalanced insulin signaling and glucose regulation. Disrupted dopaminergic signaling and increased hyperglycemic stress in diabetes contributed to the neuronal loss. Neuronal loss in diabetic rats mediated through the expression of pattern of GLUT-3, CREB, IGF-1, Akt-1, NF,B, second messengers- cAMP, cGMP, IP3 and activation of apoptotic factors factors- TNF-a,caspase-8. Disrupted dopaminergic receptor expressions and its signaling in pancreas contributed defective insulin secretion in diabetes. Activation of apoptotic factors- TNF- a,caspase-8 and defective functioning of neuronal survival factors, disrupted second messenger signaling modulated neuronal viability in diabetes. Hyperglycemic stress activated the expression of TNF-a,caspase-8, BAX and differential expression of anti oxidant enzymes- SOD and GPx in liver lead to apoptosis. Treatment of diabetic rats with insulin, Aegle marmelose and pyridoxine significantly reversed the altered dopaminergic neurotransmission, GLUT3, GLUT2, IGF-1 and second messenger signaling. Antihyperglycemic and antioxidant activity of Aegle marmelose and pyridoxine enhanced pancreatic B cell proliferation, increased insulin synthesis and secretion in diabetic rats. Thus our results conclude the neuroprotective and regenerating ability of Aegle marmelose and pyridoxine which in turn has a novel therapeutic role in the management of diabetes.

The present study deals with the Cholinergic Receptor subtypes functional regulation in spinal cord injured monoplegic rats: Effect of 5-HT GABA and bone marrow cells.Spinal cord injury causes permanent and irrevocable motor deficits and neurodegeneration. Disruption of the spinal cord leads to diminished transmission of descending control from the brain to motor neurons and ascending sensory information. Behavioural studies showed deficits in motor control and coordination in SCI rats. Cholinergic system plays an important role in SCI, the evaluation of which provides valuable insight on the underlying mechanisms of motor deficit that occur during SCI. The cholinergic transmission was studied by assessing the muscarinic and nicotinic receptors; cholinergic enzymes- ChAT and AChE; second messenger enzyme PLC; transcription factor CREB and second messengers - IP3, cAMP and cGMP. We observed a decrease in the cholinergic transmission in the brain and spinal cord of SCI rats. The disrupted cholinergic system is the indicative of motor deficit and neuronal degeneration in the spinal cord and brain regions. SCI mediated oxidative stress and apoptosis leads to neuronal degeneration in SCI rats. The decreased expression of anti oxidant enzymes – SOD, GPx and neuronal cell survival factors - BDNF, GDNF, IGF-1, Akt and cyclin D2 along with increased expression of apoptotic factors – Bax, caspase-8, TNFa and NF-kB augmented the neuronal degeneration in SCI condition. BMC administration in combination with 5-HT and GABA in SCI rats showed a reversal in the impaired cholinergic neurotransmission and reduced the oxidative stress and apoptosis. It also enhanced the expression of cell survival factors in the spinal cord region. In SCI rats treated with 5-HT and GABA, the transplanted BMC expressed NeuN confirming that 5-HT and GABA induced the differentiation and proliferation of BMC to neurons in the spinal cord. Neurotrophic factors and anti-apoptotic elements in SCI rats treated with 5-HT and GABA along with BMC rendered neuroprotective effects accompanied by improvement in behavioural deficits. This resulted in a significant reversal of altered cholinergic neurotransmission in SCI. The restorative and neuro protective effects of BMC in combination with 5-HT and GABA are of immense therapeutic significance in the clinical management of SCI.

The onset of spontaneous seizures triggers a cascade of molecular and cellular events that eventually leads to neuronal injury and cognitive decline. The present study investigated the effect of Withania somnifera (WS) root extract and Withanolide A (WA) in restoring behavioural deficit by inhibiting oxidative stress induced alteration in glutamergic neurotransmission. The subdued performance in behavioural tests shows impaired motor coordination and memory. Histopathological investigations revealed significant neuronal loss in hippocampus of epileptic rats indicating glutamate mediated excitotoxicity. The treatment with WS and WA restored behavioural deficit and ameliorated neuronal loss. An altered redox homeostasis leading to oxidative stress is a hallmark of TLE. The antioxidant potential was afflicted in epileptic rats, evident from altered activity of SOD and CAT, down regulation of SOD and GPX expression and enhanced lipid peroxidation. The antioxidant property of WS and WA restored altered antioxidant capacity. Alteration in GDH activity and down regulation of GLAST expression resulted in enhanced glutamate content in the brain regions. The metabolism of glutamate was altered in the form of down regulated GAD expression. The alteration in synthesis, transport and metabolism resulted in further increase of the glutamate concentration at the synapse leading to glutamate mediated excitotoxicity. The decreased NMDA and AMPA receptor binding and down regulated NMDA R1, NMDA 2B and AMPA (GluR2) mRNA expression indicated altered glutamergic receptor function. The treatment with WS and WA reversed altered glutamergic receptor function, synthesis, transport and metabolism. The enhanced levels of second messenger IP3 responsible for Ca2+ mediated toxicity was reversed after treatment with WS and WA. Neurotoxics concentration of glutamate resulted in up regulation of pro apoptotic factors Bax and Caspase 8 and down regulation of anti apoptotic factor Akt resulting in neuronal death. The treatment with WS and WA resulted in activation of Akt and down regulation of Bax and caspase 8 leading to blocking of apoptotic pathway. The treatment with WS and WA resulted in reduced seizure frequency and amelioration of associated alterations suggesting the therapeutic role of Withania somnifera in temporal lobe epilepsy

In the present study, the initial phase was directed to confirm the effects of curcumin and vitamin D3 in preventing or delaying diabetes onset by studying the blood glucose and insulin levels in the pre-treated and diabetic groups. Behavioural studies were conducted to evaluate the cognitive and motor function in experimental rats. The major focus of the study was to understand the cellular and neuronal mechanisms that ensure the prophylactic capability of curcumin and vitamin D3. To elucidate the mechanisms involved in conferring the antidiabetogenesis effect, we examined the DNA and protein profiles using radioactive incorporation studies for DNA synthesis, DNA methylation and protein synthesis. Furthermore the gene expression studies of Akt-1, Pax, Pdx-1, Neuro D1, insulin like growth factor-1 and NF-κB were done to monitor pancreatic beta cell proliferation and differentiation. The antioxidant and antiapoptotic actions of curcumin and vitamin D3 were examined by studying the expression of antioxidant enzymes - SOD and GPx, and apoptotic mediators like Bax, caspase 3, caspase 8 and TNF-α. In order to understand the signalling pathways involved in curcumin and vitamin D3 action, the second messengers, cAMP, cGMP and IP3 were studied along with the expression of vitamin D receptor in the pancreas. The neuronal regulation of pancreatic beta cell maintenance, proliferation and insulin release was studied by assessing the adrenergic and muscarinic receptor functional regulation in the pancreas, brain stem, hippocampus and hypothalamus. The receptor number and binding affinity of total muscarinic, muscarinic M1, muscarinic M3, total adrenergic, α adrenergic and β adrenergic receptor subtypes were studied in pancreas, brain stem and hippocampus of experimental rats. The mRNA expression of muscarinic and adrenergic receptor subtypes were determined using Real Time PCR. Immunohistochemistry studies using confocal microscope were carried out to confirm receptor density and gene expression results. Cell signalling alterations in the pancreas and brain regions associated with diabetogenesis and antidiabetogenesis were assessed by examining the gene expression profiles of vitamin D receptor, CREB, phospholipase C, insulin receptor and GLUT. This study will establish the anti-diabetogenesis activity of curcumin and vitamin D3 pre-treatment and will attempt to understand the cellular, molecular and neuronal control mechanism in the onset of diabetes.Administration of MLD-STZ to curcumin and vitamin D3 pre-treated rats induced only an incidental prediabetic condition. Curcumin and vitamin D3 pretreated groups injected with MLD-STZ exhibited improved circulating insulin levels and behavioural responses when compared to MLD-STZ induced diabetic group. Activation of beta cell compensatory response induces an increase in pancreatic insulin output and beta cell mass expansion in the pre-treated group. Cell signalling proteins that regulate pancreatic beta cell survival, insulin release, proliferation and differentiation showed a significant increase in curcumin and vitamin D3 pre-treated rats. Marked decline in α2 adrenergic receptor function in pancreas helps to relent sympathetic inhibition of insulin release. Neuronal stimulation of hyperglycemia induced beta cell compensatory response is mediated by escalated signalling through β adrenergic, muscarinic M1 and M3 receptors. Pre-treatment mediated functional regulation of adrenergic and cholinergic receptors, key cell signalling proteins and second messengers improves pancreatic glucose sensing, insulin gene expression, insulin secretion, cell survival and beta cell mass expansion in pancreas. Curcumin and vitamin D3 pre-treatment induced modulation of adrenergic and cholinergic signalling in brain stem, hippocampus and hypothalamus promotes insulin secretion, beta cell compensatory response, insulin sensitivity and energy balance to resist diabetogenesis. Pre-treatment improved second messenger levels and the gene expression of intracellular signalling molecules in brain stem, hippocampus and hypothalamus, to retain a functional neuronal response to hyperglycemia. Curcumin and vitamin D3 protect pancreas and brain regions from oxidative stress by their indigenous antioxidant properties and by their ability to stimulate cellular free radical defence system. The present study demonstrates the role of adrenergic and muscarinic receptor subtypes functional regulation in curcumin and vitamin D3 mediated anti-diabetogenesis. This will have immense clinical significance in developing effective strategies to delay or prevent the onset of diabetes.

In früheren Experimenten konnte gezeigt werden, dass die sekundären Botenstoffe cAMP, cGMP und IP3 in der olfaktorischen Signaltransduktionskaskade bei Manduca sexta involviert sind. Während cGMP Perfusionen in die Antenne die Pheromonwahrnehmung tageszeitabhängig adaptierten, führten cAMP Perfusionen zu einer tageszeitabhängigen Sensitisierung, ähnlicher der von Octopamin (OA). Daher wurde hypothetisiert, dass eine tageszeitabhängige Oszillation antennaler OA Level sowie der intrazelluläre Kalziumkonzentration in einer Schwankung von sekundären Botenstoffen resultieren könnte. Diese Hypothese wurde mittels biochemischen Nachweißverfahren in der Antenne von M. sexta und Rhyparobia maderae überprüft. Tatsächlich konnten in der Antenne des Tabakschwärmers tageszeitabhängige Unterschiede in der OA-, cAMP- und IP3-, aber nicht in der cGMP Konzentration, nachgewiesen werden. Während die cAMP- und OA Oszillationen einander sehr ähnelten und die Maxima in der Paarungsphase aufzeigten, korrelierte der IP3 Verlauf sehr stark mit dem Flug- bzw. Fressverhalten. Diese Korrelationen konnten auch in der Madeira Schabe beobachtet werden, in der darüber hinaus gezeigt werden konnte, dass antennale cAMP- und IP3 Level von dem circadianen Uhrwerk gesteuert werden. Zudem wurde herausgefunden, dass OA die cAMP- und teilweise auch die IP3- Spiegel reguliert. Demgegenüber beeinflusste Kalzium die Konzentration aller untersuchten sekundären Botenstoffe. Daher wird angenommen, dass die intrazelluläre Kalziumkonzentration aber auch der antennale OA Level kritische Faktoren bei der Regelung der olfaktorischen Sensitivität sind. Da Oszillationen von sekundären Botenstoffen in mutmaßlichen, peripheren Schrittmacher nachgewiesen wurden, wurde untersucht, ob sie auch im circadianen Schrittmacher der Madeira Schabe oszillieren und ob das Neuropeptid pigment-dispersing factor (PDF), ein entscheidender Kopplungsfaktor des Uhrwerks in Insekten, diese Rhythmen generieren könnte. Es konnte gezeigt werden, dass PDF die cAMP Synthese steigert. Darüber hinaus wurden bimodale cAMP Oszillationen unter licht-dunkel Bedingungen beobachtet, welche unter konstanten Umweltbedingungen verblieben. Daher wird angenommen, dass PDF Freisetzung zelluläres cAMP erhöht über das das circadiane Uhrwerk synchronisiert wird.

Calcium is an important second messenger in the rat pineal gland, as well as cAMP. They both contribute to melatonin synthesis mediated by the three main enzymes of the melatonin synthesis pathway: tryptophan hydroxylase, arylalkylamine N-acetyltransferase and hydroxyindole-O-methyltransferase. The cytosolic calcium is elevated in pinealocytes following alpha(1)-adrenergic stimulation, through IP3-and membrane calcium channels activation. Nifedipine, an L-type calcium channel blocker, reduces melatonin synthesis in rat pineal glands in vitro. With the purpose of investigating the mechanisms involved in melatonin synthesis regulation by the L-type calcium channel, we studied the effects of nifedipine on noradrenergic stimulated cultured rat pineal glands. Tryptophan hydroxylase, arylalkylamine N-acetyltransferase and hydroxyindole-O-methyltransferase activities were quantified by radiometric assays and 5-hydroxytryptophan, serotonin, N-acetylserotonin and melatonin contents were quantified by HPLC with electrochemical detection. The data showed that calcium influx blockaded by nifedipine caused a decrease in tryptophan hydroxylase activity, but did not change either arylalkylamine N-acetyltransferase or hydroxyindole-O-methyltransferase activities. Moreover, there was a reduction of 5-hydroxytryptophan, serotonin, N-acetylserotonin and melatonin intracellular content, as well as a reduction of scrotonin and melatonin secretion. Thus, it seems that the calcium influx through L-type high voltage-activated calcium channels is essential for the full activation of tryptophan hydroxylase leading to melatonin synthesis in the pineal gland. Muscarinic (mAChRs) and nicotinic acetylcholine receptors (nAChRs) are involved in various physiological processes, including neuronal development. We provide evidence for expression of functional nicotinic and muscarinic receptors during differentiation of P19 carcinoma embryonic cells, as an in vitro model of early neurogenesis. We have detected expression and activity alpha(2)-alpha(7), beta(2), beta(4) nAChR and M1-M5 mAChR subtypes during neuronal differentiation. Nicotinic alpha(3) and beta(2) mRNA transcription was induced by addition of retinoic acid to P19 cells. Gene expression Of alpha(2), alpha(4)-alpha(7), beta(4) nAChR subunits decreased during initial differentiation and increased again when P19 cells underwent final maturation. Receptor response in terms of nicotinic agonist-evoked Ca2+, flux was observed in embryonic and neuronal-differentiated cells. Muscarinic receptor response, merely present in undifferentiated P19 cells, increased during neuronal differentiation. The nAChR-induced elevation of intracellular calcium ([Ca2+](i)) response in undifferentiated cells was due to Ca2+ influx. In differentiated P19 neurons the nAChR-induced [Ca2+](i) response was reduced following pretreatment with ryanodine, while the mAChR-induced response was unaffected indicating the contribution of Ca2+ release from ryanodine-sensitive stores to nAChR- but not mAChR-mediated Ca2+ responses. The presence of functional nAChRs in embryonic cells suggests that these receptors are involved in triggering Ca2+ waves during initial neuronal differentiation. (C) 2007 Elsevier Ltd. A proteína ácida fibrilar glial (GFAP) é uma proteína da classe dos filamentos intermediários, exclusivamente expressa em astrócitos no sistema nervoso central (SNC). A função específica da fosforilação desta proteína é ainda desconhecida. No entanto, tem sido demonstrado que o equilíbrio dinâmico entre o estado fosforilado e desfosforilado de sítios específicos da GFAP pode regular a polimerização e despolimerização dos filamentos intermediários durante eventos de estruturação do citoesqueleto glial. Nosso grupo de pesquisa demonstrou que a fosforilação da GFAP em hipocampo de ratos jovens (P12-P16) é estimulada no mesmo nível por glutamato, via um receptor glutamatérgico metabotrópico do grupo II (mGluR II), e pela ausência de Ca2+ externo (presença de EGTA). Entretanto, o tratamento simultâneo com glutamato e EGTA não resulta em efeito sinergístico, sugerindo um mesmo mecanismo de ação para estas duas situações estimulatórias da fosforilação da GFAP (WofchuK & Rodnight, 1994; Kommers et al., 1999; Rodnight et al., 1997). Este mecanismo provavelmente não envolve reservas intracelulares de Ca2+ associadas a receptores de IP3, uma vez que mGluRs II estão envolvidos com o mecanismo de transdução de sinal via adenilato ciclase e não via hidrólise de fosfoinositídios. Uma hipótese proposta é de que o glutamato, via mGluR, bloqueia canais de Ca2+ tipo L, inibindo uma cascata de desfosforilação dependente de Ca2+, associada a GFAP (Rodnight et al., 1997). Interessantemente, os receptores rianodina (RyRs) presentes nas reservas intracelulares de Ca2+ reguladas por tais receptores estão associados com canais de Ca2+ tipo L (Chavis et al., 1996). Com base nestes dados, buscou-se neste trabalho avaliar se a modulação glutamatérgica da fosforilação da GFAP em fatias de hipocampo de ratos jovens envolve as reservas intracelulares de Ca2+ reguladas por RyRs e se o Ca2+ proveniente destas reservas atua de maneira semelhante ao Ca2+ oriundo do espaço extracelular. Nossos resultados mostraram que há uma evidente participação do Ca2+ proveniente das reservas intracelulares reguladas por RyRs no mecanismo modulatório da fosforilação da GFAP via ativação de mGluRs em fatias de hipocampo de ratos jovens, uma vez que a cafeína e a rianodina (agonistas de RyRs) revertem totalmente o efeito estimulatório do agonista glutamatérgico metabotrópico 1S,3R-ACPD sobre a fosforilação da proteína e este efeito da cafeína é inibido por dantrolene (antagonista de RyRs). Talvez o Ca2+ oriundo das reservas reguladas por RyRs tenha o mesmo papel do Ca2+ proveniente do espaço extracelular, ou seja, desencadeia uma cascata de desfosforilação associada à GFAP mediada pela calcineurina, uma vez que quelando o Ca2+ intracelular livre com BAPTA-AM, após a mobilização destas reservas, tal efeito não ocorre. A participação de receptores adenosina (AdoRs) e do AMP cíclico (AMPc) ainda permanece a ser estudada. Entretanto, é sabido que em ratos jovens a ativação de mGluRs aumenta a formação de AMPc potenciando o efeito de outros tipos de receptores, como os AdoRs e, provavelmente, isto é mediado por um mGluR II (Schoepp & Johnson, 1993; Winder & Conn, 1996). Neste trabalho mostrou-se justamente o possível envolvimento de tais mecanismos de transdução de sinal na modulação da fosforilação da GFAP, pois a adenosina deaminase (enzima que metaboliza adenosina endógena) e a forscolina (agente que estimula a enzima adenilato ciclase) alteraram o nível de fosforilação da GFAP. Estes resultados evidenciam o envolvimento das reservas intracelulares de Ca2+ reguladas por RyRs no mecanismo de transdução de sinal que modula o estado de fosforilação GFAP mediado pela ativação de mGluRs.

We studied the development of the insulin secretion mechanism in the pancreas of fetal (19- and 21-day-old), neonatal (3-day-old), and adult (90-day-old) rats in response to stimulation with 8.3 or 16.7 mM glucose, 30 mM K+, 5 mM theophylline (Theo) and 200 µM carbamylcholine (Cch). No effect of glucose or high K+ was observed on the pancreas from 19-day-old fetuses, whereas Theo and Cch significantly increased insulin secretion at this age (82 and 127% above basal levels, respectively). High K+ also failed to alter the insulin secretion in the pancreas from 21-day-old fetuses, whereas 8.3 mM and 16.7 mM glucose significantly stimulated insulin release by 41 and 54% above basal levels, respectively. Similar results were obtained with Theo and Cch. A more marked effect of glucose on insulin secretion was observed in the pancreas of 3-day-old rats, reaching 84 and 179% above basal levels with 8.3 mM and 16.7 mM glucose, respectively. At this age, both Theo and Cch increased insulin secretion to close to two-times basal levels. In islets from adult rats, 8.3 mM and 16.7 mM glucose, Theo, and Cch increased the insulin release by 104, 193, 318 and 396% above basal levels, respectively. These data indicate that pancreatic B-cells from 19-day-old fetuses were already sensitive to stimuli that use either cAMP or IP3 and DAG as second messengers, but insensitive to stimuli such as glucose and high K+ that induce membrane depolarization. The greater effect of glucose on insulin secretion during the neonatal period indicates that this period is crucial for the maturation of the glucose-sensing mechanism in B-cells.

Trehalose ist der Hauptblutzucker in der Hämolymphe der meisten Insekten. Trehalose wird im Fettkörper synthetisiert, dem wichtigsten Organ des Intermediärstoffwechsels bei Insekten. Wie die Homöostase des Blutzuckers reguliert wird, ist nicht vollständig geklärt. Die Produktion von Trehalose erfordert eine grundlegende Umschaltung im Stoffwechsel des Fettkörpers, die mehrere wichtige Stoffwechselwege betrifft, so dass die Fettkörperzellen (Trophocyten) von der Speicherung und Katabolisierung von Zucker zur Mobilisierung von Reservestoffen (Glykogen, Fett, Protein) und Trehalosesynthese umschalten. Am Fettkörper und isolierten Trophocyten der Argentinischen Schabe (Blaptica dubia) wurden Stoffwechseleffekte und Elemente der Signalkette des hypertrehalosämischen Hormons Bld HrTH untersucht. Inkubation isolierter Fettkörperloben mit Bld HrTH verringerte innerhalb von 60 min den Glykogengehalt (um 13,4 %) und steigerte die Konzentration der Hexosephosphate Glucose-6-phosphat und Fructose-6-phosphat, die Substrat sowohl für die Trehalosesynthese als auch für die Glykolyse sind. Pyruvat, Glycerin-3-phosphat, Citrat und insbesondere Fructose-1,6-bisphosphat (+750 %) waren ebenfalls erhöht. Der Glykolyseaktivator/Gluconeogeneseinhibitor Fructose-2,6-bisphosphat wird durch Bld HrTH vermindert. Da Trehalosesynthese und Glykolyse um dieselben Substrate (Glucosephosphate) konkurrieren, fördert der hormoninduzierte Abfall des Glykolyseaktivators Fructose-2,6-bisphosphat die Trehalogenese.Es ist gelungen, Trophocyten zu isolieren und die Signaltransduktion von Bld HrTH an einheitlichen Zellen und auch an Einzelzellen zu studieren. Hauptziel dieser Arbeit war es, die Funktion von Ca2+ im Signalweg des Bld HrTH genauer zu untersuchen. Die isolierten Zellen reagierten auf das Neuropeptid mit einer deutlichen Steigerung der Trehalosesynthese (+133,7 %) und einer Senkung des Fructose-2,6-bisphosphat-Gehaltes (-30,2 %). Sie bieten somit ein geeignetes System zur Untersuchung der Wirkungsmechanismen von Bld HrTH auf zellulärem Niveau. Ca2+ aus dem Extrazellulärraum und aus intrazellulären Speichern spielen bei der Signaltransduktion eine Rolle. Während extrazelluläres Ca2+ insbesondere für die Senkung des Fructose-2,6-bisphosphat-Gehaltes wichtig war, wurde Ca2+ aus zellulären Speichern insbesondere für die Trehalosesynthese benötigt, wobei sich jedoch beide Wege wechselseitig beeinflussen. Erstmals konnten an isolierten Trophocyten Änderungen von Ca2+ mikrofluorometrisch an Einzelzellen studiert werden. Das hypertrehalosämische Hormon ruft einen schnellen und starken Anstieg der intrazellulären Ca2+-Konzentration ([Ca2+]i) hervor. Die Untersuchungen deuten auf einen Signalweg über IP3 und Diacylglycerin hin, entsprechend der Phosphoinositidkaskade. Eine Beteiligung des biogenen Amins Octopamin, von cAMP oder von Stickstoffmonoxid (NO) an der Signaltransduktion scheint hingegen unwahrscheinlich. Der Zuckergehalt im Medium scheint ebenfalls auf die Trehalogenese zu wirken. Bei hohen Konzentrationen von Glucose oder Trehalose wurde eine Hemmung der Trehalosesynthese beobachtet, die als Rückkopplungshemmung gedeutet werden kann. Bei Hunger wird das im Fettkörper gespeicherte Glykogen stark reduziert. Außerdem scheint die Zahl der symbiontischen Mikroorganismen in den Mycetocyten verringert.

Through the years, several studies reported the involvement of nuclear lipid signalling as highly connected with cell cycle progression. Indeed, nuclear Phosphatidylinositol-4,5-Biphosphate (PIP2) hydrolisis mediated by Phospholipases C (PLC), which leads to production of the second messengers Diacylglycerol (DAG) and Inositol-1,4,5-Triphosphate (IP3), is a fundamental event for both G1/S and G2/M checkpoints. In particular, we found that nuclear DAG production was mediated by PLCbeta1, enzyme mainly localized in the nucleus of K562 human erythroleukemia cells. This event triggered the activation and nuclear translocation of PKCalpha, which, in turn, resulted able to affect cell cycle via modulation of Cyclin D3 and Cyclin B1, two important enzymes for G1/S transition and G2/M progression respectively.

2-Aminoethyl diphenylborinate (2-APB) is a known modulator of the IP3 receptor, the calcium ATPase SERCA, the calcium release-activated calcium channel Orai and TRP channels. More recently, it was shown that 2-APB is an efficient inhibitor of the epithelial calcium channel TRPV6 which is overexpressed in prostate cancer. We have conducted a structure-activity relationship study of 2-APB congeners to understand their inhibitory mode of action on TRPV6. Whereas modifying the aminoethyl moiety did not significantly change TRPV6 inhibition, substitution of the phenyl rings of 2-APB did. Our data show that the diaryl borinate moiety is required for biological activity and that the substitution pattern of the aryl rings can influence TRPV6 versus SOCE inhibition. We have also discovered that 2-APB is hydrolyzed and transesterified within minutes in solution.
