The harsh environment presented by engines, particularly in the exhaust systems, often necessitates the use of robust and therefore low bandwidth temperature sensors. Consequently, high frequencies are attenuated in the output. One technique for addressing this problem involves measuring the gas temperature using two sensors with different time-constants and mathematically reconstructing the true gas temperature from the resulting signals. Such a technique has been applied in gas turbine, rocket motor and combustion research. A new reconstruction technique based on difference equations has been developed and its effectiveness proven theoretically. The algorithms have been successfully tested and proven on experimental data from a rig that produces cyclic temperature variations. These tests highlighted that the separation of the thermocouple junctions must be very small to ensure that both sensors are subjected to the same gas temperatures. Exhaust gas temperatures were recorded by an array of thermocouples during transient operation of a high performance two-stroke engine. The results show that the increase in bandwidth arising from the dual sensor technique allowed accurate measurement of exhaust gas temperature with relatively robust thermocouples. Finally, an array of very fine thermocouples (12.5 - 50 microns) was used to measure the in-cycle temperature variation in the exhaust.

The tailpipe emissions from automotive engines have been subject to steadily reducing legislative limits. This reduction has been achieved through the addition of sub-systems to the basic four-stroke engine which thereby increases its complexity. To ensure the entire system functions correctly, each system and / or sub-systems needs to be continuously monitored for the presence of any faults or malfunctions. This is a requirement detailed within the On-Board Diagnostic (OBD) legislation. To date, a physical model approach has been adopted by me automotive industry for the monitoring requirement of OBD legislation. However, this approach has restrictions from the available knowledge base and computational load required. A neural network technique incorporating Multivariant Statistical Process Control (MSPC) has been proposed as an alternative method of building interrelationships between the measured variables and monitoring the correct operation of the engine. Building upon earlier work for steady state fault detection, this paper details the use of non-linear models based on an Auto-associate Neural Network (ANN) for fault detection under transient engine operation. The theory and use of the technique is shown in this paper with the application to the detection of air leaks within the inlet manifold system of a modern gasoline engine whilst operated on a pseudo-drive cycle. We present photometric and spectroscopic observations at optical and near-infrared wavelengths of the nearby type Ic supernova 2007gr. These represent the most extensive data-set to date of any supernova of this sub-type, with frequent coverage from shortly after discovery to more than one year post-explosion. We deduce a rise time to B-band maximum of 11.5 +/- 2.7 d. We find a peak B-band magnitude of M-B = -16.8, and light curves which are remarkably similar to the so-called "hypernova" SN 2002ap. In contrast, the spectra of SNe 2007gr and 2002ap show marked differences, not least in their respective expansion velocities. We attribute these differences primarily to the density profiles of their progenitor stars at the time of explosion i.e. a more compact star for SN 2007gr compared to SN 2002ap. From the quasi-bolometric light curve of SN 2007gr, we estimate that 0.076 +/- 0.010 M-circle dot of Ni-56 was produced in the explosion. Our near-infrared (IR) spectra clearly show the onset and disappearance of the first overtone of carbon monoxide (CO) between similar to 70 to 175 d relative to B-band maximum. The detection of the CO molecule implies that ionised He was not microscopically mixed within the carbon/oxygen layers. From the optical spectra, near-IR light curves, and colour evolution, we find no evidence for dust condensation in the ejecta out to about +400 d. Given the combination of unprecedented temporal coverage, and high signal-to-noise data, we suggest that SN 2007gr could be used as a template object for supernovae of this sub-class.

Recent searches by unbiased, wide-field surveys have uncovered a group of extremely luminous optical transients. The initial discoveries of SN 2005ap by the Texas Supernova Search and SCP-06F6 in a deep Hubble pencil beam survey were followed by the Palomar Transient Factory confirmation of host redshifts for other similar transients. The transients share the common properties of high optical luminosities (peak magnitudes similar to -21 to -23), blue colors, and a lack of H or He spectral features. The physical mechanism that produces the luminosity is uncertain, with suggestions ranging from jet-driven explosion to pulsational pair instability. Here, we report the most detailed photometric and spectral coverage of an ultra-bright transient (SN 2010gx) detected in the Pan-STARRS 1 sky survey. In common with other transients in this family, early-time spectra show a blue continuum and prominent broad absorption lines of O II. However, about 25 days after discovery, the spectra developed type Ic supernova features, showing the characteristic broad Fe II and Si II absorption lines. Detailed, post-maximum follow-up may show that all SN 2005ap and SCP-06F6 type transients are linked to supernovae Ic. This poses problems in understanding the physics of the explosions: there is no indication from late-time photometry that the luminosity is powered by Ni-56, the broad light curves suggest very large ejected masses, and the slow spectral evolution is quite different from typical Ic timescales. The nature of the progenitor stars and the origin of the luminosity are intriguing and open questions.

Chronic myeloid leukemia (CML) is treated effectively with tyrosine kinase inhibitors (TKIs); however, 2 key problems remain-the insensitivity of CML stem and progenitor cells to TKIs and the emergence of TKI-resistant BCR-ABL mutations. BCR-ABL activity is associated with increased proteasome activity and proteasome inhibitors (PIs) are cytotoxic against CML cell lines. We demonstrate that bortezomib is antiproliferative and induces apoptosis in chronic phase (CP) CD34(+) CML cells at clinically achievable concentrations. We also show that bortezomib targets primitive CML cells, with effects on CD34(+)38(-), long-term culture-initiating (LTC-IC) and nonobese diabetic/severe combined immunodeficient (NOD/SCID) repopulating cells. Bortezomib is not selective for CML cells and induces apoptosis in normal CD34(+)38(-) cells. The effects against CML cells are seen when bortezomib is used alone and in combination with dasatinib. Bortezomib causes proteasome but not BCR-ABL inhibition and is also effective in inhibiting proteasome activity and inducing apoptosis in cell lines expressing BCR-ABL mutations, including T315I. By targeting both TKI-insensitive stem and progenitor cells and TKI-resistant BCR-ABL mutations, we believe that bortezomib offers a potential therapeutic option in CML. Because of known toxicities, including myelosuppression, the likely initial clinical application of bortezomib in CML would be in resistant and advanced disease. (Blood. 2010;115:2241-2250)

The objectives were to determine if the skin secretion of the European yellow-bellied toad (Bombina variegata), in common with other related species, contains a bradykinin inhibitor peptide and to isolate and structurally characterize this peptide. Materials and Methods: Lyophilized skin secretion obtained from this toad was subjected to reverse phase HPLC fractionation with subsequent bioassay of fractions for antagonism of the bradykinin activity using an isolated rat tail artery smooth muscle preparation. Subsequently, the primary structure of the peptide was established by a combination of microsequencing, mass spectroscopy, and molecular cloning, following which a synthetic replicate was chemically synthesised for bioassay. Results: A single peptide of molecular mass 2300.92 Da was resolved in HPLC fractions of skin secretion and its primary structure determined as IYNAIWP-KH-NK-KPGLL-. Database interrogation with this sequence indicated that this peptide was encoded by skin kininogen-1 previously cloned from B. variegata. The blank cycles were occupied by cysteinyl (C) residues and the peptide was located toward the C-terminus of the skin kininogen, and flanked N-terminally by a classical -KR- propeptide convertase processing site. The peptide was named IC-20 in accordance (I = N-terminal isoleucine, C = C-terminal cysteine, 20 = number of residues). Like the natural peptide, its synthetic replicate displayed an antagonism of bradykinin-induced arterial smooth muscle relaxation. Conclusion: IC-20 represents a novel bradykinin antagonizing peptide from amphibian skin secretions and is the third such peptide found to be co-encoded with bradykinins within skin kininogens.

Nebular spectra of supernovae (SNe) offer an unimpeded view of the inner region of the ejecta, where most nucleosynthesis takes place. Optical spectra cover most, but not all, of the emitting elements and therefore offer only a partial view of the products of the explosion. Simultaneous optical-infrared spectra, on the other hand, contain emission lines of all important elements, from C and O through to the intermediate mass elements (IME) Mg, Si, S, Ca and to Fe and Ni. In particular, Si and S are best seen in the IR. The availability of IR data makes it possible to explore in greater detail the results of the explosion. SN 2007gr is the first Type Ic SN for which such data are available. Modelling the spectra with a non-local thermodynamic equilibrium (NLTE) code reveals that the inner ejecta contain similar to 1M(circle dot) of material within a velocity of approximate to 4500 km s(-1). The same mass of Ni-56 derived from the light-curve peak (0.076M(circle dot)) was used to power the spectrum, yielding consistent results. Optical and near-infrared (near-IR) observations of the Type Ic supernova (SN Ic) 2004aw are presented, obtained from -3 to +413 d with respect to the B-band maximum. The photometric evolution is characterized by a comparatively slow post-maximum decline of the light curves. The peaks in redder bands are significantly delayed relative to the bluer bands, the I-band maximum occurring 8.4 d later than that in B. With an absolute peak magnitude of -18.02 in the V band the SN can be considered fairly bright, but not exceptional. This also holds for the U through I bolometric light curve, where SN 2004aw has a position intermediate between SNe 2002ap and 1998bw. Spectroscopically SN 2004aw provides a link between a normal SN Ic like SN 1994I and the group of broad-lined SNe Ic. The spectral evolution is rather slow, with a spectrum at day +64 being still predominantly photospheric. The shape of the nebular [O-I] lambda lambda 6300, 6364 line indicates a highly aspherical explosion. Helium cannot be unambiguously identified in the spectra, even in the near-IR. Using an analytical description of the light-curve peak we find that the total mass of the ejecta in SN 2004aw is 3.5-8.0 M-circle dot, significantly larger than that in SN 1994I, although not as large as in SN 1998bw. The same model suggests that about 0.3 M-circle dot of Ni-56 has been synthesized in the explosion. No connection to a GRB can be firmly established.

Aims. The aim of this paper is to discuss the nature of two type Ic supernovae SN 2007bg and SN 2007bi and their host galaxies. Both supernovae were discovered in wide-field, non-targeted surveys and are found to be associated with sub-luminous blue dwarf galaxies identified in SDSS images.

We report our attempts to locate the progenitor of the peculiar Type Ic SN 2007gr in Hubble Space Telescope (HST) preexplosion images of the host galaxy, NGC 1058. Aligning adaptive optics Altair/NIRI imaging of SN 2007gr from the Gemini ( North) Telescope with the preexplosion HST WFPC2 images, we identify the supernova (SN) position on the HST frames with an accuracy of 20 mas. Although nothing is detected at the SN position, we show that it lies on the edge of a bright source 134 +/- 23 mas (6.9 pc) from its nominal center. On the basis of its luminosity, we suggest that this object is possibly an unresolved, compact, and coeval cluster and that the SN progenitor was a cluster member, although we note that model profile fitting favors a single bright star. We find two solutions for the age of this assumed cluster: 7 -/+ 0.5 Myr and 20 - 30 Myr, with turnoff masses of 28 +/- M-circle dot and 12 - 9 M-circle dot, respectively. Preexplosion ground-based K- band images marginally favor the younger cluster 4 age/higher turnoff mass. Assuming the SN progenitor was a cluster member, the turnoff mass provides the best estimate for its initial mass. More detailed observations, after the SN has faded, should determine whether the progenitor was indeed part of a cluster and, if so, allow an age estimate to within similar to 2 Myr, thereby favoring either a high-mass single star or lower-mass interacting binary progenitor.

Images of the site of the Type Ic supernova (SN) 2002ap taken before explosion were analysed previously by Smartt et al. We have uncovered new unpublished, archival pre-explosion images from the Canada-France-Hawaii Telescope (CFHT) that are vastly superior in depth and image quality. In this paper we present a further search for the progenitor star of this unusual Type Ic SN. Aligning high-resolution Hubble Space Telescope observations of the SN itself with the archival CFHT images allowed us to pinpoint the location of the progenitor site on the groundbased observations. We find that a source visible in the B- and R-band pre-explosion images close to the position of the SN is (1) not coincident with the SN position within the uncertainties of our relative astrometry and (2) is still visible similar to 4.7-yr post-explosion in late-time observations taken with the William Herschel Telescope. We therefore conclude that it is not the progenitor of SN 2002ap. We derived absolute limiting magnitudes for the progenitor of M-B >= -4.2 +/- 0.5 and M-R >= -5.1 +/- 0.5. These are the deepest limits yet placed on a Type Ic SN progenitor. We rule out all massive stars with initial masses greater than 7-8 M-circle dot (the lower mass limit for stars to undergo core collapse) that have not evolved to become Wolf-Rayet stars. This is consistent with the prediction that Type Ic SNe should result from the explosions of Wolf-Rayet stars. Comparing our luminosity limits with stellar models of single stars at appropriate metallicity (Z = 0.008) and with standard mass-loss rates, we find no model that produces a Wolf-Rayet star of low enough mass and luminosity to be classed as a viable progenitor. Models with twice the standard mass-loss rates provide possible single star progenitors but all are initially more massive than 30-40 M-circle dot. We conclude that any single star progenitor must have experienced at least twice the standard mass-loss rates, been initially more massive than 30-40 M-circle dot and exploded as a Wolf-Rayet star of final mass 10-12 M-circle dot. Alternatively a progenitor star of lower initial mass may have evolved in an interacting binary system. Mazzali et al. propose such a binary scenario for the progenitor of SN 2002ap in which a star of initial mass 15-20 M-circle dot is stripped by its binary companion, becoming a 5 M-circle dot Wolf-Rayet star prior to explosion.
