The microphthalmia transcription factor (MITF), a basic-helix-loop-helix zipper factor, regulates distinct target genes in several cell types. We hypothesized that interaction with the Ets family factor PU.1, whose expression is limited to hematopoietic cells, might be necessary for activation of target genes like tartrate-resistant acid phosphatase (TRAP) in osteoclasts. Several lines of evidence were consistent with this model. The combination of MITF and PU.1 synergistically activated the TRAP promoter in transient assays. This activation was dependent on intact binding sites for both factors in the TRAP promoter. MITF and PU.1 physically interacted when coexpressed in COS cells or in vitro when purified recombinant proteins were studied. The minimal regions of MITF and PU.1 required for the interaction were the basic-helix-loop-helix zipper domain and the Ets DNA binding domain, respectively. Significantly, mice heterozygous for both the mutant mi allele and a PU.1 null allele developed osteopetrosis early in life which resolved with age. The size and number of osteoclasts were not altered in the double heterozygous mutant mice, indicating that the defect lies in mature osteoclast function. Taken in total, the results afford an example of how lineage-specific gene regulation can be achieved by the combinatorial action of two broadly expressed transcription factors.

Argyrodes Simon 1864 is a large, cosmopolitan theridiid genus whose members exhibit a wide range of foraging techniques which usually involve exploiting other spiders, either by using their webs, stealing their food, or preying on them directly. We held a symposium on this genus at the 15th International Congress of Arachnology, Badplaas, South Africa in order to obtain a clearer perspective on the relationship between the phylogeny of the genus and the different foraging techniques. We concluded that Argyrodes forms a monophyletic group within the Theridiidae, and that there are clear monophyletic clades within the genus (already identified as species groups) that appear to share behavioral characteristics. We found no clear indication that foraging behaviors such as kleptoparasitism (stealing food) evolved from araneophagy (eating spiders) or vice versa. However, it appears that species that specialize in either kleptoparasitism or araneophagy use additional techniques in comparison to species that readily use both foraging modes. During our examination of Argyrodes/host interactions we noted the importance of Nephila species as hosts of Argyrodes species around the world and the impact of Argyrodes on Nephila. We also noted the fluid nature of the relationship between Argyrodes and the spiders with which they interact. For example, an Argyrodes/host relationship can change to an Argyrodes/prey relationship, and the type of kleptoparasitic behavior employed by an Argyrodes can change when it changes host species. The importance of eating silk was also noted and identified as an area for further research. We concluded that more work involving international collaboration is needed to fully understand the phylogeny of the genus and the relationships between the different types of foraging behaviors.

Extracellular copper regulates the DNA binding activity of the CopY repressor of Enterococcus hirae and thereby controls expression of the copper homeostatic genes encoded by the cop operon. CopY has a CxCxxxxCxC metal binding motif. CopZ, a copper chaperone belonging to a family of metallochaperones characterized by a MxCxxC metal binding motif, transfers copper to CopY. The copper binding stoichiometries of CopZ and CopY were determined by in vitro metal reconstitutions. The stoichiometries were found to be one copper(l) per CopZ and two copper(l) per CopY monomer. X-ray absorption studies suggested a mixture of two- and three-coordinate copper in Cu(1)CopZ, but a purely three-coordinate copper coordination with a Cu-Cu interaction for Cu(1)(2)CopY. The latter coordination is consistent with the formation of a compact binuclear Cu(l)-thiolate core in the CxCxxxxCxC binding motif of CopY. Displacement of zinc, by copper. from CopY was monitored with 2,4-pyridylazoresorcinol. Two copper(l) ions were required to release the single zinc(II) ion bound per CopY monomer. The specificity of copper transfer between CopZ and CopY was dependent on electrostatic interactions. Relative copper binding affinities of the proteins were investigated using the chelator, diethyldithiocarbamic acid (DDC). These data suggest that CopY has a higher affinity for copper than CopZ. However, this affinity difference is not the sole factor in the copper exchange: a charge-based interaction between the two proteins is required for the transfer reaction to proceed. Gain-of-function mutation of a CopZ homologue demonstrated the necessity of four lysine residues on the chaperone for the interaction with CopY. Taken together, these results suggest a mechanism for copper exchange between CopZ and CopY.

This paper reports descriptive information on the relationship between social competence and the amount and type of peer interaction for nine adolescents with intellectual disability attending a regular high school. Each adolescent's social competence was assessed using the AAMR Adaptive Behavior Scale-School (2nd edn) (ABS-S:2). Naturalistic observations were conducted to obtain information on the amount and type of peer interaction. Data were analysed to determine the relationship between social competence and peer interactions. Social competence was not consistently correlated with the amount of peer interaction. Results from the naturalistic observations showed individual differences in patterns of peer interaction with a tendency towards more frequent interactions with peers who also had intellectual disabilities. These data suggest that social competence did not significantly influence the amount and type of peer interaction. Implications for facilitating peer interactions between adolescents with and without intellectual disability are discussed.

The interaction between genetic and environmental factors for PD was examined in a Chinese population. It was found that although the intron 2 MAOB (GT)(n) repeat polymorphism was not associated with PID in the population, a relationship might have been masked by the protective effect of tea drinking. In individuals who did not drink tea (<1 cup/day), the possession of short length less than or equal to 178 bp (GT), alleles conferred a borderline significant increased risk for PD (adjusted OR = 1.47; C.l. = 1.03-2. 1). As the extent of tea consumption increased, the association between the less than or equal to178 bp allele and PD disappeared. This result suggests that the MAOB gene may be associated with PD in Chinese if the putative protective effect of tea drinking is taken into account. The significance of this finding is unclear as the study may be limited because of its marginal significance and limited numbers. However, it does demonstrate the importance of considering putative positive and negative environmental risk factors in any examination of genetic risk factors for PD. (C) 2003 Elsevier Science Ltd. Solvatochromic UV-Vis shifts of four indicators (4-nitroaniline, 4-nitroanisole, 4-nitrophenol and N,N-dimethy-1-4-nitro aniline) have been measured at 298.15 K in the ternary mixture methano1/1-propanol/acetonitrile (MeOH/1-PrOH/MeCN) in a total of 22 mole fractions, along with 18 additional mole fractions for each of the corresponding binary mixtures, MeOH/1-PrOH, 1-PrOH/MeCN and MeOH/MeCN. These values, combined with our previous experimental results for 2,6-dipheny1-4-(2,4,6-triphenylpyridinium-1-yl)phenolate (Reichardt's betaine dye) in the same mixtures, permitted the computation of the Kamlet-Taft solvent parameters, alpha, beta, and pi*. The rationalization of the spectroscopic behavior of each probe within each mixture's whole mole fraction range was achieved through the use of the Bosch and Roses preferential solvation model. The applied model allowed the identification of synergistic behaviors in MeCN/alcohol mixtures and thus to infer the existence of solvent complexes in solution. Also, the addition of small amounts of MeCN to the binary mixtures was seen to cause a significant variation in pi*, whereas the addition of alcohol to MeCN mixtures always lead to a sudden change in a and The behavior of these parameters in the ternary mixture was shown to be mainly determined by the contributions of the underlying binary mixtures. Las Tecnologías de la Información y la Comunicación (TIC) han alcanzando una fuerte expansión en la última década y se convierten en una estrategia clave para la educación científica y tecnológica. Actualmente se realizan significativas inversiones en computadoras e insumos acordes a esta tecnología lo que no garantiza que éstas mejores disponibilidades sean necesariamente conducentes a propuestas educativas aceptables, ya que la incorporación de las TIC en la educación científica no puede consistir en un mero cambio de soporte sin que esto se acompañe de una renovación constructivista del aprendizaje. También se observa un escaso desarrollo y aplicación de las TIC tanto en la formación de grado de las carreras científicas, como en la formación permanente de los docentes. A partir de lo expuesto, nos planteamos los siguientes problemas: ¿cómo generar cursos de formación docente con alternativas virtuales que garanticen la construcción, actualización y transferencia de conocimientos en las disciplinas científicas? ¿qué diseño de materiales se requiere para potenciar aprendizajes flexibles de calidad?, ¿qué dimensiones, variables e indicadores dan cuenta de procesos de comunicación en estos entornos?, ¿Cuáles son las principales estrategias que promueven los desarrollos tecnológicos? ¿De qué manera se lleva a cabo el seguimiento y evaluación de los desarrollos tecnológicos? Para dar respuesta a estos interrogantes nos planteamos este proyecto que tiene por objetivos: diseñar, desarrollar, implementar y evaluar materiales basados en un modelo constructivista que utiliza como soporte las TIC el marco de la "Enseñanza para la Comprensión". Analizar las relaciones que se establecen en el intercambio comunicativo a través de las TIC, ya sea de manera síncrona o asíncrona e identificar las estrategias de aprendizaje utilizadas por los estudiantes cuando trabajan con entornos virtuales. Se profundizará en los siguientes temas: "La materia y sus cambios", "Biotecnología" y "Dilemas investigativos a los que se enfrentan los científicos". Los dos primeros módulos estarán destinados a la formación de futuros docentes en Ciencias y a la formación permanente de docentes en ejercicio. Los usuarios del tercer módulo serán los estudiantes del ciclo de nivelacion. Se propone desarrollar instrumentos de evaluación, adaptando indicadores que sirvan para evaluar el modelo, el diseño de los materiales, el medio tecnológico, la eficiencia de estos programas con TIC. Se van a definir variables que permitan identificar las estrategias de aprendizaje utilizadas por los usuarios de estos desarrollos tecnológicos y las relaciones que se establecen en el intercambio comunicativo de los entornos virtuales, haciendo hincapié en la lecto-escritura científica. Se utilizará la plataforma Moodle que permite la administración de los estudiantes, las interacciones sociales necesarias para integrar una comunidad de aprendizaje a través de los foros y correo electrónico y la evaluación continua de los aprendizajes. El desarrollo de los contenidos específicos y las actividades de aprendizaje se hará en CD interactivos elaborados con diferentes programas de computación. En el desarrollo de los materiales se incluyen algunas simulaciones interactivas que permiten comprender conceptos que habitualmente requerían experimentación fáctica. El estudio se enmarca en la combinación de dos enfoques metodológicos que integran técnicas y procedimientos de análisis tanto cuantitativos como cualitativos. Consideramos que esta investigación es una contribución teórica importante ya que propone mejorar un modelo para la enseñanza de las Ciencias Naturales introduciendo las TIC. Se espera realizar un aporte novedoso al campo de la Didáctica de las Ciencias con la obtención de los resultados y acciones tendientes a establecer una red de comunicaciones entre los docentes participantes y los investigadores involucrados en el programa, instalada en forma progresiva.

In this study, we explored the predictive role of family interactions and family representations in mothers and fathers during pregnancy for postnatal motherfatherinfant interactions during the first 2 years after birth. Families (N = 42) were seen at the fifth month of pregnancy and at 3 and 18 months after birth. During pregnancy, parents were asked to play with their baby at the first meeting by using a doll in accordance with the procedure of the prenatal Lausanne Trilogue Play (LTP; A. Corboz-Warnery & E. Fivaz-Depeursinge, 2001; E. Fivaz-Depeursinge, F. Frascarolo-Moutinot, & A. Corboz-Warnery, 2010). Family representations were assessed by administering the Family System Test (T. Gehring, 1998). Marital satisfaction and the history of the couple were assessed through self-reported questionnaires. At 3 and 18 months, family interactions were assessed in the postnatal LTP. Infant temperament was assessed through parent reports. Results show that (a) prenatal interactions and child temperament are the most important predictors of family interactions and (b) paternal representations are predictive of family interactions at 3 months. These results show that observational assessment of nascent family interactions is possible during pregnancy, which would allow early screening of family maladjustment. The findings also highlight the necessity of taking into account paternal representations as a significant variable in the development of family interactions.

Metabolic problems lead to numerous failures during clinical trials, and much effort is now devoted to developing in silico models predicting metabolic stability and metabolites. Such models are well known for cytochromes P450 and some transferases, whereas less has been done to predict the activity of human hydrolases. The present study was undertaken to develop a computational approach able to predict the hydrolysis of novel esters by human carboxylesterase hCES2. The study involved first a homology modeling of the hCES2 protein based on the model of hCES1 since the two proteins share a high degree of homology (congruent with 73%). A set of 40 known substrates of hCES2 was taken from the literature; the ligands were docked in both their neutral and ionized forms using GriDock, a parallel tool based on the AutoDock4.0 engine which can perform efficient and easy virtual screening analyses of large molecular databases exploiting multi-core architectures. Useful statistical models (e.g., r (2) = 0.91 for substrates in their unprotonated state) were calculated by correlating experimental pK(m) values with distance between the carbon atom of the substrate's ester group and the hydroxy function of Ser228. Additional parameters in the equations accounted for hydrophobic and electrostatic interactions between substrates and contributing residues. The negatively charged residues in the hCES2 cavity explained the preference of the enzyme for neutral substrates and, more generally, suggested that ligands which interact too strongly by ionic bonds (e.g., ACE inhibitors) cannot be good CES2 substrates because they are trapped in the cavity in unproductive modes and behave as inhibitors. The effects of protonation on substrate recognition and the contrasting behavior of substrates and products were finally investigated by MD simulations of some CES2 complexes.
