Two-hybrid technology provides a simple way to isolate small peptide aptamers that specifically recognize and strongly bind to a protein of interest. These aptamers have the potential to dominantly interfere with specific activities of their target proteins and, therefore, could be used as in vivo inhibitors. Here we explore the ability to use peptide aptamers as in vivo inhibitors by expressing aptamers directed against cell cycle regulators in Drosophila. We expressed two peptide aptamers, each of which specifically recognizes one of the two essential cyclin-dependent kinases (Cdks), DmCdk1 and DmCdk2, in Drosophila. Expression of each Cdk aptamer during organogenesis caused adult eye defects typical of those caused by cell cycle inhibition. Co-overexpression of DmCdk1 or DmCdk2 resulted in suppression of the eye phenotypes, indicating that each aptamer interacts with a Cdk target in vivo and suggesting that these peptides disrupt normal eye development by inhibiting Cdk function. Moreover, the specificity of each aptamer for one of the two Cdks as determined in two-hybrid assays was retained in Drosophila. Combined, our results demonstrate that peptide aptamers generated by yeast two-hybrid methods can serve as inhibitory reagents to target specific proteins in vivo.

Two-hybrid methods have augmented the classical genetic techniques biologists use to assign function to genes. Here, we describe construction of a two-bait interaction trap that uses yeast cells to register more complex protein relationships than those detected in existing two-hybrid systems. We show that such cells can identify bridge or connecting proteins and peptide aptamers that discriminate between closely related allelic variants. The protein relationships detected by these cells are analogous to classical genetic relationships, but lend themselves to systematic application to the products of entire genomes and combinatorial libraries. We show that, by performing logical operations on the phenotypic outputs of these complex cells and existing two-hybrid cells, we can make inferences about the topology and order of protein interactions. Finally, we show that cells that register such relationships can perform logical operations on protein inputs. Thus these cells will be useful for analysis of gene and allele function, and may also define a path for construction of biological computational devices.

The field of natural language processing (NLP) has seen a dramatic shift in both research direction and methodology in the past several years. In the past, most work in computational linguistics tended to focus on purely symbolic methods. Recently, more and more work is shifting toward hybrid methods that combine new empirical corpus-based methods, including the use of probabilistic and information-theoretic techniques, with traditional symbolic methods. This work is made possible by the recent availability of linguistic databases that add rich linguistic annotation to corpora of natural language text. Already, these methods have led to a dramatic improvement in the performance of a variety of NLP systems with similar improvement likely in the coming years. This paper focuses on these trends, surveying in particular three areas of recent progress: part-of-speech tagging, stochastic parsing, and lexical semantics.

Recibido 27 de julio de 2011 • Aceptado 26 de agosto de 2011 • Corregido 28 de octubre de 2011 El debate epistemológico de la investigación científico-social durante años ha consistido en confrontar los métodos, tratando de hacer valer más uno que el otro a partir de la gran teoría que lo soporta. Aunque propuestas más recientes recuperan métodos híbridos o planteamientos epistemológicos más diversos, la postura de la dualidad ha prevalecido. Descriptiva o explicativa; fenomenológica o positivista; clásica o emergente; los paradigmas de investigación son la manera en cómo tenemos acceso a la realidad investigada y así obtener principios y respuestas. Ello define los métodos. Aún cuando es posible agrupar y categorizar estos métodos en cuantitativos y cualitativos, el entramado epistemológico que establecen las estructuras de cualquier metodología permitirá el abordaje de un fenómeno mejor que otro, sin que esto pondere necesariamente alguno de manera definitoria. Dentro de la educación, tanto métodos cuantitativos como cualitativos han ayudado a abordar fenómenos de tal manera que han producido resultados favorables para el bienestar del quehacer educativo. Este ensayo, producto de una revisión bibliográfica y una trayectoria investigativa y educativa, pretende esclarecer el complejo panorama inicial de los investigadores educativos novatos antes de revisar y recuperar una postura epistemológica.

The paper compares three different methods of inclusion of current phasor measurements by phasor measurement units (PMUs) in the conventional power system state estimator. For each of the three methods, comprehensive formulation of the hybrid state estimator in the presence of conventional and PMU measurements is presented. The performance of the state estimator in the presence of conventional measurements and optimally placed PMUs is evaluated in terms of convergence characteristics and estimator accuracy. Test results on the IEEE 14-bus and IEEE 300-bus systems are analyzed to determine the best possible method of inclusion of PMU current phasor measurements.

A study of environmental chloride and groundwater balance has been carried out in order to estimate their relative value for measuring average groundwater recharge under a humid climatic environment with a relatively shallow water table. The hybrid water fluctuation method allowed the split of the hydrologic year into two seasons of recharge (wet season) and no recharge (dry season) to appraise specific yield during the dry season and, second, to estimate recharge from the water table rise during the wet season. This well elaborated and suitable method has then been used as a standard to assess the effectiveness of the chloride method under forest humid climatic environment. Effective specific yield of 0.08 was obtained for the study area. It reflects an effective basin-wide process and is insensitive to local heterogeneities in the aquifer system. The hybrid water fluctuation method gives an average recharge value of 87.14 mm/year at the basin scale, which represents 5.7% of the annual rainfall. Recharge value estimated based on the chloride method varies between 16.24 and 236.95 mm/year with an average value of 108.45 mm/year. It represents 7% of the mean annual precipitation. The discrepancy observed between recharge value estimated by the hybrid water fluctuation and the chloride mass balance methods appears to be very important, which could imply the ineffectiveness of the chloride mass balance method for this present humid environment.

We have successfully extended our implicit hybrid finite element/volume (FE/FV) solver to flows involving two immiscible fluids. The solver is based on the segregated pressure correction or projection method on staggered unstructured hybrid meshes. An intermediate velocity field is first obtained by solving the momentum equations with the matrix-free implicit cell-centered FV method. The pressure Poisson equation is solved by the node-based Galerkin FE method for an auxiliary variable. The auxiliary variable is used to update the velocity field and the pressure field. The pressure field is carefully updated by taking into account the velocity divergence field. This updating strategy can be rigorously proven to be able to eliminate the unphysical pressure boundary layer and is crucial for the correct temporal convergence rate. Our current staggered-mesh scheme is distinct from other conventional ones in that we store the velocity components at cell centers and the auxiliary variable at vertices. The fluid interface is captured by solving an advection equation for the volume fraction of one of the fluids. The same matrix-free FV method, as the one used for momentum equations, is used to solve the advection equation. We will focus on the interface sharpening strategy to minimize the smearing of the interface over time. We have developed and implemented a global mass conservation algorithm that enforces the conservation of the mass for each fluid.
