The formation of arsenic-phytochelatin (As-PC) complexes is thought to be part of the plant detoxification strategy for arsenic. This work examines (i) the arsenic (As) concentration-dependent formation of As-PC complex formation and (ii) redistribution and metabolism of As after arrested As uptake in Helianthus annuus. HPLC with parallel ICP-MS/ES-MS detection was used to identify and quantify the species present in plant extracts exposed to arsenate (As(V)) (between 0 and 66.7 micromol As l-1 for 24 h). At As concentrations below the EC50 value for root growth (22 micromol As l-1) As uptake is exponential, but it is reduced at concentrations above. Translocation between root and shoot seemed to be limited to the uptake phase of arsenic. No redistribution of As between root and shoot was observed after arresting As exposure. The formation of As-PC complexes was concentration-dependent. The amount and number of As-PC complexes increased exponentially with concentration up to 13.7 micromol As l-1. As(III)-PC3 and GS-As(III)-PC2 complexes were the dominant species in all samples. The ratio of PC-bound As to unbound As increased up to 1.3 micromol As l-1 and decreased at higher concentrations. Methylation of inorganic As was only a minor pathway in H. annuus with about 1% As methylated over a 32 d period. The concentration dependence of As-PC complex formation, amount of unbound reduced and oxidized PC2, and the relative uptake rate showed that As starts to influence the cellular metabolism of H. annuus negatively at As concentrations well below the EC50 value determined by more traditional means. Generally, As-PC complexes and PC-synthesis rate seem to be the more sensitive parameters to be studied when As toxicity values are to be estimated.

The aim of the study was to determine the time-dependent formation of arsenic-phytochelatin (As-PC) complexes in the roots, stems and leaves of an arsenic-nontolerant plant (Helianthus annuus) during exposure to 66 mol l(-1) arsenite (As(III)) or arsenate (As(V)). We used our previously developed method of simultaneous element-specific (inductively coupled plasma mass spectrometry, ICP-MS) and molecular-specific (electrospray-ionization mass spectrometry, ES-MS) detection systems interfaced with a suitable chromatographic column and eluent conditions, which enabled us to identify and quantify As-PC complexes directly. Roots of As-exposed H. annuus contained up to 14 different arsenic species, including the complex of arsenite with two (gamma-Glu-Cys)(2)-Gly molecules [As((III))-(PC(2))(2)], the newly identified monomethylarsonic phytochelatin-2 or (gamma-Glu-Cys)(2)-Gly CH(3)As (MA((III))-PC(2)) and at least eight not yet identified species. The complex of arsenite with (gamma-Glu-Cys)(3)-Gly (As((III))-PC(3)) and the complex of arsenite with glutathione (GSH) and (gamma-Glu-Cys)(2)-Gly (GS-As((III))-PC(2)) were present in all samples (roots, stems and leaves) taken from plants exposed to As. The GS-As((III))-PC(2) complex was the dominant complex after 1 h of exposure. As((III))-PC(3) became the predominant As-PC complex after 3 h, binding up to 40% of the As present in the exposed plants. No As-PC complexes were found in sap (mainly xylem sap from the root system), in contrast to roots, stems and leaves, which is unequivocal evidence that As-PC complexes are not involved in the translocation of As from root to leaves of H. annuus.

The identification of Diaporthe (anamorph Phomopsis) species associated with stem canker of sunflower (Helianthus annuus) in Australia was studied using morphology, DNA sequence analysis and pathology. Phylogenetic analysis revealed three clades that did not correspond with known taxa, and these are believed to represent novel species. Diaporthe gulyae sp. nov. is described for isolates that caused a severe stem canker, specifically pale brown to dark brown, irregularly shaped lesions centred at the stem nodes with pith deterioration and mid-stem lodging. This pathogenicity of D. gulyae was confirmed by satisfying Koch's Postulates. These symptoms are almost identical to those of sunflower stem canker caused by D. helianthi that can cause yield reductions of up to 40% in Europe and the USA, although it has not been found in Australia. We show that there has been broad misapplication of the name D. helianthi to many isolates of Diaporthe ( Phomopsis) found causing, or associated with, stem cankers on sunflower. In GenBank, a number of isolates had been identified as D. helianthi, which were accommodated in several clades by molecular phylogenetic analysis. Two less damaging species, D. kochmanii sp. nov. and D. kongii sp. nov., are also described from cankers on sunflower in Australia.

El experimento se efectuó en la finca la Concha, departamento de león, con el objetivo, de determinar la influencia de diferente dosis de nitrógeno y fósforo, sobre el crecimiento y desarrollo del girasol, determinar la influencia de diferentes dosis de nitrógeno y fósforo, sobre el rendimiento del girasol, así como la búsqueda de una fertilización adecuada para el cultivo del girasol en la zona la paz centro. Se utilizó la variedad caburé a una distancia de 0.3 m x 0.7m, en un diseño de bloques completamente aleatorizados, distribuido en nueve tratamiento, en la fenología y manuales para controlar los efectos de los tratamientos en los parámetros a evaluar. Se determinó, que el nitrógeno ejerce una influencia significativa en la altura de la planta entre los 43 y 50 d.d.s. entre el testigo y los diferentes tratamientos fertilizados, en la fenología del cultivo, se observó una pequeña diferenciación, durante el estado de antesis del cultivo. La fertilización no tenía influencia sobre la población, el número de capitulo por planta, el diámetro del capítulo y el peso de mil semilla, con el aumento de la fertilización nitrogenada bajaron significativamente el número de semilla por capitulo y el rendimiento. el fosforo no ejerció influencia sobre los factores de rendimiento, pero permitió un menor número de plantas acamadas.

O biodiesel é definido como um mono alquil éster de ácidos graxos de cadeia longa derivado de fontes renováveis tais como óleos vegetais e gorduras animais. Sua importância esta associada ao uso como um combustível alternativo para motores do ciclo Diesel podendo ser utilizado puro ou em misturas com o diesel representando economia de petróleo e menor poluição ambiental. Em geral é obtido por meio da reação de transesterificação na qual os triacilgliceróis, principais constituintes dos óleos e gorduras reagem com álcool, em presença de um catalisador ácido ou básico, produzindo ésteres de ácidos graxos e glicerol. A transesterificação pode ser conduzida por catálise homogênea ou heterogênea. O grande desafio da indústria é otimizar o processo a fim de alcançar um produto e uma rota de produção tecnologicamente eficiente e ambientalmente correta. O objetivo desta pesquisa foi estudar a síntese do biodiesel utilizando o processo de transesterificação do óleo de girassol por catálises homogênea e heterogênea. Foram realizadas reações de transesterificação via rotas metílica e etílica, empregando como catalisador homogêneo alcóxido de potássio e como catalisador heterogêneo a resina comercial de troca iônica Amberlyst 26

High biomass producing plant species, such as Helianthus annuus, have potential for removing large amounts of trace metals by harvesting the aboveground biomass if sufficient metal concentrations in their biomass can be achieved. However, the low bioavailability of heavy metals in soils and the limited translocation of heavy metals to the shoots by mosthigh biomass producing plant species limit the efficiency of the phytoextraction process. Amendment of a contaminated soil with ethylene diamine letraacetic acid (EDTA) or citric acid increases soluble heavy metal concentrations, potentially rendering them more available for plant uptake. This article discusses the effects of EDTA and citric acid on the uptake of heavy metals and translocation to aboveground harvestable plant parts in Helianthus annuus. EDTA was included in the research for comparison purposes in our quest for less persistent alternatives, suitable for enhanced phytoextraction. Plants were grown in a calcareous soil moderately contaminated with Cu, Pb, Zn, and Cd and treated with increasing concentrations of EDTA (0. 1, 1, 3, 5, 7, and 10 mmol kg(-1) soil) or citric acid (0. 01, 0. 05, 0.25, 0.442, and 0.5 mol kg(-1) soil). Heavy metal concentrations in harvested shoots increased with EDTA concentration but the actual amount of phytoextracted heavy metals decreased at high EDTA concentrations, due to severe growth depression. Helianthus annuus suffered heavy metal stress due to the significantly increased bioavailable metal fraction in the soil. The rapid mineralization of citric acid and the high buffering capacity of the soil made citric acid inefficient in increasing the phytoextracted amounts of heavy metals. Treatments that did not exceed the buffering capacity of the soil (< 0.442 mol kg(-1) soil) did not result in any significant increase in shoot heavy metal concentrations. Treatments with high concentrations resulted in a dissolution of the carbonates and compaction of the soil. These physicochemical changes caused growth depression of Helianthus annuus. EDTA and citric acid added before sowing of Helianthus annuus did not appear to be efficient amendments when phytoextraction of heavy metals from calcareous soils is considered.

Remediation of soil pollution is one of the many current environmental challenges. Anthropogenic activity has resulted in the contamination of extended areas of land, the remediation of which is both invasive and expensive by conventional means. Phytoextraction of heavy metals from contaminated soils has the prospect of being a more economic in situ alternative. In addition, phytoextraction targets ecotoxicologically the most relevant soil fraction of these metals, i.e. the bioavailable fraction. Greenhouse experiments were carried out to evaluate the potential of four high biomass crop species in their potential for phytoextraction of heavy metals, with or without with the use of soil amendments (EDTA or EDDS). A calcareous dredged sediment derived surface soil, with high organic matter and clay content and moderate levels of heavy metal pollution, was used in the experiments. No growth depression was observed in EDTA or EDDS treated pots in comparison to untreated controls. Metal accumulation was considered to be low for phytoextraction purposes, despite the use of chelating agents. The low observed shoot concentrations of heavy metals were attributed to the low phytoavailability of heavy metals in this particular soil substrate. The mobilising effects induced by EDTA in the soil were found to be too long-lived for application as a soil amendment in phytoextraction. Although EDDS was found to be more biodegradable, higher effect half lives were observed than reported in literature or observed in previous experiments. These findings caution against the use of any amendment, biodegradable or otherwise, without proper investigation of its effects and the longevity thereof. (C) 2005 Elsevier Ltd. Nowadays, the growing environmental worry leads research the focus the application of alternative materials from renewable resources on the industrial process. The most common vegetable oil extractant using around the world is the hexane, a petroleum derived, toxic and flammable. Based on this fact, the goal of this work was to test vegetable oil extractions from sunflower seeds cultivated on the Rio Grande do Norte State using two extraction process, the mechanical expelling and solvent extraction, this one using hexane and ethanol as a alternative solvent. The solvent extractions were carried out in the Soxhlet extractor in three different extraction times (4, 6, and 8 hours). The effect of solvent and extraction time was evaluated. The mechanical extraction was carried out in a expeller and the sunflower oil obtained was characterized by its physical-chemical properties and compared with sunflower refinery oil. Furthermore this work also explored the pyrolysis reaction carried out by thermogravimetry measurement as alternative route to obtain biofuel. For this purpose the oil samples were heated to ambient temperature until 900°C in heating rate of 5, 10, 20ºC min-1 with the objective evaluated the kinetics parameters such activation energy and isoconversion. The TG/DTG curves show the thermal profile decomposition of triglycerides. The curves also showed that antioxidant presents on the refinery oil not influence on the thermal stability of sunflower oil. The total yield of the extraction s process with hexane and ethanol solvent were compared, and the results indicated that the extraction with ethanol were more efficient. The pyrolysis reaction results indicated that the use of unpurified oil required less energy to obtain the bio-oil

Sowing is a critical time in the cycle of a crop and the seeds are frequently exposed to adverse conditions that may compromise the establishment of seedlings in the field. on this basis, the objective of the present study was to determine the effect of types of environmental stress on the emergence of sunflower, maize and soybean seeds with different levels of vigor. High vigor seeds were artificially aged in order to obtain medium and low vigor seeds and then they were sown in clay soil in plastic boxes and submitted to the following types of environmental stress during the germination process : 1) high temperature (35degreesC), 2) low temperature (15 or 18degreesC), 3) water excess (Psi > -0.0001 MPa), 4) water deficiency (Psi approximately equal to -1.1; -1.2 and -0.6 MPa for sunflower, maize and soybean, respectively), 5) sowing at a depth of 7 cm and 6) pathogenic infection of sunflower seeds with Alternaria helianthi, of maize seeds with Fusarium moniliforme and of soybean seeds with Colletotrichum dematium, var. truncata. The results were compared to those obtained with controls sown under optimal condition. It was concluded that: 1) the effect of seed vigor on emergence depends on the type of enviromental stress to which the seeds are exposed, 2) the stress to which the the seeds demonstrated highest sensitivity varied with species and 3) high temperature stress was the one that most impaired the emergence of the three species.

El topinambur pertenece a la familia de las Asteráceas, es una especie originaria de América del Norte, de la que fundamentalmente se aprovechan sus tubérculos; tiene gran potencial como alimento, materia prima para productos industriales y producción de biocombustibles. En Argentina no hay cultivares de topinambur registrados. Sin embargo hay documentación y testimonios del ingreso de distintas variedades al país a principios del siglo XX. El objetivo de esta tesis fue caracterizar el germoplasma de topinambur que se cultiva, a pequeña escala, en distintas regiones de la Argentina. Se formó una colección de trabajo con introducciones provenientes de 5 provincias del país (Río Negro, Chubut, Buenos Aires, Mendoza y Córdoba), y se condujeron ensayos experimentales en dos ambientes de la provincia de Mendoza. Se evaluaron caracteres morfo³gicos (altura de plantas, dimensiones y ángulo de inserción de hojas, tamaño de tubérculos), feno³gicos (emergencia, inicio, fin y duración de floración, senescencia del cultivo) y rendimiento. Se detectaron diferencias morfo³gicas que permitieron agrupar a las introducciones en dos grupos, correspondiendo uno a introducciones de tubérculos rojos y el otro a introducciones de tubérculos blancos. Se valoró la aptitud hortícola de las introducciones mediante evaluación sensorial; se realizó una prueba de preferencia y percepción de distintos aspectos de la hortaliza (color, olor, sabor y textura), consumida cruda. El nivel de aceptación general de la hortaliza fue bueno. Se evaluó el potencial industrial (obtención de inulina y producción de etanol). Se determinó por HPLC el contenido de inulina de cada introducción, que varió de 18.07 a 22.95 % y se estimó el potencial para producir etanol a partir de los hidratos de carbono fermentables de los tubérculos, que llegó a 4.934 litros por ha, aunque sin diferencias entre introducciones.

Helianthus tuberosus L. presenta potencial para producir etanol, sustituto de la nafta, y que deberá incorporarse a la misma en un 5% a partir del 2010 en Argentina. Hay antecedentes que señalan que a partir de 50 toneladas de tubérculos pueden obtenerse 4500 l de etanol. En este trabajo se comparó el rendimiento de dos variedades de topinambur regado con agua residual urbana o cloacal (AC) y agua subterránea (AS). El ensayo se realizó en la planta de tratamiento de agua cloacal de Obras Sanitarias Mendoza en el Departamento Tunuyán (33°32’89’’ S y 69°00’80’’ O), Mendoza, Argentina. El potencial para producir bioetanol se estimó a partir de la cantidad de hidratos de carbono fermentables en los tubérculos. El rendimiento de tubérculos presentó diferencias entre los tratamientos de riego, siendo de 177750 kg/ha en AC y de 144000 kg/ha en AS. La estimación del potencial para producir etanol generó un valor de 15000 l de alcohol en las parcelas regadas con AC y 13000 l en las regadas con AS. Para obtener 1 l de alcohol a partir de los tubérculos serían necesarios alrededor de 11 kg, relacionado con un valor de sólidos solubles de 16% en los mismos.

Helianthus tuberosus L. produce tubérculos ricos en hidratos de carbono fermentables que pueden ser usados para producir etanol. Para el destino energético el cultivo puede ser regado con aguas residuales urbanas. La densidad poblacional en un cultivo de topinambur afecta los parámetros de crecimiento y rendimiento. Se evaluó el efecto de la densidad de plantación sobre el rendimiento de tubérculos. Se compararon 6 tratamientos de densidad, combinando 2 espaciamientos entre hileras (0,70 y 0,80 m) y 3 espaciamientos entre plantas en la hilera (0,30; 0,40 y 0,50 m), generando stands desde las 25.000 a 47.600 plantas/ha. Se presentaron los mayores rendimientos con distancias entre plantas en la hilera de 0,30 ó 0,40 m; no se presentaron diferencias en respuesta a la distancia entre hileras. Los componentes del rendimiento (rendimiento de tubérculos por planta, número de tubérculos por planta y tamaño medio de los tubérculos) respondieron a cambios en la densidad generados por la distancias entre plantas y fueron indiferentes a la variación de distancia entre hileras. La altura de las plantas, el número de tallos principales, el porcentaje de MS y de SS en los tubérculos no se modificaron ante variaciones de densidad.

El interés por el cultivo de Topinambur (Helianthus tuberosus L.) es creciente a nivel mundial. Como toda hortaliza que se lleva al mercado, o se utiliza como materia prima para la industria, debe estar disponible a lo largo de todo el año. Por esto es importante desarrollar sistemas de almacenamiento que permitan conservar la calidad del producto el mayor tiempo posible. Existen muy pocos antecedentes sobre la conservación poscosecha de tubérculos de topinambur y las variaciones de calidad, que sufre el producto. A través del trabajo de tesis se buscó generar antecedentes sobre los efectos de la fecha de cosecha y la variedad de topinambur sobre las diferentes variables de calidad; la evolución de cada tratamiento conservado en cámara frigorífica y el efecto del lugar/condiciones (cámara frigorífica – campo) y tiempo de conservación sobre la calidad de los tubérculos. Se utilizaron dos variedades de topinambur (tubérculos rojos y blancos). Las variables respuesta ensayadas para definir atributos de calidad fueron: materia seca, sólidos solubles, firmeza, índice de color de piel y de pulpa, presencia de brotes, desarrollo de Penicillium y evolución del peso fresco. Se detectó que el período de cosecha, para garantizar la calidad de los tubérculos y buena evolución en cámara frigorífica durante todo el año, no debería extenderse más allá de los 49 días desde la ocurrencia de la primera helada para la zona de estudio, no observándose diferencias entre variedades. Para consumo en fresco se puede cosechar hasta el 98 días luego de la primera helada, pero estos tubérculos no se conservan correctamente en cámara frigorífica.
