Incorporation of mevalonate-2-C14, acetate-1-C14, and formate-C14 into the lipids of microorganisms was studied. In the case of four bacteria tested—Agrobacterium tumefaciens, Azotobacter vinelandii, Escherichia coli, and a Pseudomonas species—the various homologues of coenzyme Q present were not labeled with any of the tracers used, although significant amounts of radioactivity were present in the lipids. Both acetate and mevalonate were incorporated into coenzyme Q and sterol of the moulds, Aspergillus niger, Neurospora crassa, Penicillium chrysogenum, and Gibberella fujickuroi, and a yeast, Torulopsis utilis. Mevalonate was incorporated into the side chain but not the ring, whereas acetate was incorporated into both. It appears that the mevalonate pathway for the synthesis of coenzyme Q is operative only in those organisms which also contain other isoprene compounds such as sterol and carotene.

Further purification of indoleacetaldoxime (IAOX) hydro-lyase from Gibberella fujikuroi by DEAE-cellulose chromatography is described. The purified enzyme was activated by dehydroascorbic acid (DHA), ascorbic acid (AA), and pyridoxal phosphate (PALP) and was inhibited by thiol compounds and thiol reagents including phenylthiocyanate. Ferrous ions but not ferric ions activated the purified enzyme. The enzyme was activated by dihydrofolic acid but inhibited by tetrahydrofolic acid. Phenylacetaldoxime, a competitive inhibitor, afforded partial protection of the enzyme from the action of N-ethylmaleimide suggesting the involvement of a thiol function at the active site or substrate-binding site. The inhibition of the enzyme by 2,3-dimercaptopropanol was reversed by DHA, PALP, or frozen storage. KCN inhibition of the enzyme was reversed by PALP. NaBH4 reduction of the purified enzyme in the presence of PALP gave an active enzyme which was further activated by PALP or DHA but not by ferrous ions. These results suggested a "structural" role for PALP in the activity of IAOX hydro-lyase. Dilute solutions of the purified enzyme, obtained during DEAE-cellulose chromatography and concentrated using sucrose, showed enhanced activity upon frozen storage and thawing. The increase in activity of the enzyme during certain culture conditions, the activation and inhibition of the enzyme by several unrelated compounds, and the effect of freezing indicate that IAOX hydro-lyase is probably a metabolically regulated enzyme with a structure composed of subunits.

农药对土壤微生物区系结构和功能的影响以及潜在的生态风险成为人们关注的热点之一。本文以中科院海伦生态站农田黑土作为实验土壤,采用室内模拟的方法,利用传统的(CFU和ergosterol)及分子微生物生态学技术(DGGE,real-time PcR,clolle library)研究了乙草胺、甲胺磷及其复合对黑土真菌的生态效应,并得出以下结果:经8周处理,中、高浓度乙草胺(150和250mgkg-1)对土壤真菌数量、生物量和可培养真菌种群多样性具有长期抑制效应。乙草胺处理8周后可培养真菌和土壤固氮微生物n州基因的种群结构不能得到恢复,而总的真菌结构可基本恢复。甲胺磷对土壤可培养真菌数量和生物量具有促进作用,以高浓度(250mgkg~(-1))尤为显著。高浓度甲胺磷(25omgkg~(-1))对nifH基因多样性有长期抑制效应。甲胺磷处理8周后可培养真菌种群结构不可恢复,而总的真菌和n积基因种群结构可部分恢复。两者复合后对真菌数量,生物量,多样性及n担基因多样性的影响无论是促进还是抑制其作用强度都大于单因子。处理8周后可培养真菌、总的真菌和n州基因三者的种群结构均不可恢复。克隆测序分析发现乙草胺、甲胺磷及其复合可明显促进植物致病真菌(colletolrichum;truncatum,Rhizoctonia zeae,Fusarium oxysporum)的生长,同时使土壤中常见的青霉菌数量减少,使农药处理后具有潜在的植物病害爆发的风险。本试验结果表明,乙草胺、甲胺磷及其复合对土壤真菌数量、结构、多样性和功能基因nifH的多样性及其种群组成有不同程度的影响,甚至产生某些不可逆的长期生态效应。复合处理对土壤真菌的影响要大于两个单因子作用,表现了明显的复合生态效应。一般来说受到午扰的真菌种群结构不容易自然恢复,因此建议在施用这两种农药过程中要避免大量、频繁的单独或复合施用。

A cultivar de trigo BRS 49, desenvolvida pela Embrapa Trigo, foi indicada para semeadura no Estado do Parana, a partir de 1999. Para a determinacao do seu valor de cultivo e uso, foram utilizados dados de 54 experimentos, instalados em nove locais das regioes 6, 7 e 8 do Parana, no periodo de 1996 a 1998. Apresenta como principais caracteristicas: ciclo intermediario, altura media a alta, moderada resistencia ao acamamento, tolerancia ao aluminio toxico no solo, moderada resistencia ao oidio (Blumeria graminis f. sp tritici) e a ferrugem da folha (Puccinia recondita f. sp. tritici), com percentuais de severidade em ensaios inferiores aos das testemunhas Trigo BR 23 e Trigo BR 35, suscetibilidade a giberela (Giberella zeae), cujas notas foram superiores as atribuidas as testemunhas, elevado potencial de rendimento e boa qualidade industrial. No trienio considerado, apresentou rendimento medio de 4.667 kg/ha na Regiao 6, 3.432 kg/ha, na Regiao 7, e 3.930 kg/ha, na regiao 8. Esses valores foram 11%, 11% e 28%, respectivamente, superiores a media das testemunhas (Trigo BR 2, Trigo BR 35 e CEP 24-Industrial). Os percentuais de resposta ao controle fitossanitario foram, em geral, inferiores aos observados nas referidas testemunhas, evidenciando o elevado potencial de rendimento da cultivar com aplicacoes reduzidas de fungicidas. A forca geral de gluten (W), na media de 14 amostras coletadas nas tres Regioes, foi de 260 e a relacao P/L foi de 0,685. Portanto, a cultivar se enquadra na classe Trigo Pao, de acordo com a Instrucao Normativa n.1, de 27 de janeiro de 1999 do MA. A cultivar apresenta caracteristicas de interesse agronomico, principalmente por possibilitar a obtencao de altos rendimentos, sem a necessidade de elevados investimentos com fungicidas.

Cystic fibrosis (CF) patients may suffer increased morbidity and mortality through colonisation, allergy and invasive infection from fungi. The black yeast, Exophiala dermatitidis (synonym Wangiella dermatitidis) has been found with increasing frequency in sputum specimens of CF patients, with reported isolation rates ranging from 1.1 to 15.7%. At present, no diagnostic PCR exists to aid with the clinical laboratory detection and identification of this organism. A novel species-specific PCR-based assay was developed for the detection of E. dermatitidis, based on employment of rDNA operons and interspacer (ITS) regions between these rDNA operons. Two novel primers, (designated ExdF & ExdR) were designed in silico with the aid of computer-aided alignment software and with the alignment of multiple species of Exophiala, as well as with other commonly described yeasts and filamentous fungi within CF sputum, including Candida. Aspergillus and Scedosporium. An amplicon of approximately 455 by was generated, spanning the partial ITS I region - the complete 5.8S rDNA region - partial ITS2 region, employing ExdF (forward primer [16-mer], 5'-CCG CCT ATT CAG GTC C-3' and ExdR (reverse primer [16-mer], 5'-TCT CTC CCA CTC CCG C-3', was employed and optimised on extracted genomic DNA from a well characterised culture of E. dermatitidis, as well as with high quality genomic DNA template from a further 16 unrelated fungi, including Candida albicans, C. dubliniensis, C. parapsilosis, C. glabrata, Scedosporium apiospermum, Penicillium sp., Aspergillus fumigatus, Aspergillus versicolor, Pichia guilliermondii, Rhodotorula sp., Trichosporon sp., Aureobasidium pullulans, Fusarium sp., Mucor hiemalis, Bionectria ochroleuca, Gibberella pulicaris. Results demonstrated that only DNA from E. dermatitidis gave an amplification product of the expected sire, whilst none of the other fungi were amplifiable. Subsequent employment of this primer pair detected this yeast from mycological cultures from 2/50 (4%) adult CF patients. These two patients were the only patients who were previously shown to have a cultural history of E. dermatitidis from their sputum. E. dermatitidis is a slow-growing fungus, which usually takes up to two weeks to culture in the microbiology laboratory and therefore is slow to detect conventionally, with the risk of bacterial overgrowth from common co-habiting pan- and multiresistant bacterial pathogens from sputum. namely Pseudomonas aeruginosa and Burkholderia cepacia complex organisms, hence this species-specific PCR assay may help detect this organism from CF sputum more specifically and rapidly. Overall, employment of this novel assay nay help in the understanding of the occurrence. aetiology and epidemiology of E. dermatitidis, as an emerging fungal agent in patients with CF. Gibberella moniliformis is most commonly associated with maize worldwide and produces high levels of fumonisins, some of the most agriculturally important mycotoxins. Studies demonstrate that molecular methods can be helpful for a rapid identification of Fusarium species and their levels of toxin production. The purpose of this research was to apply molecular methods (AFLP, TEF-1 alpha partial gene sequencing and PCR based on MAT alleles) for the identification of Fusarium species isolated from Brazilian corn and to verify if real time RT-PCR technique based on FUM1 and FUM19 genes is appropriated to estimate fumonisins B(1) and B(2) production levels. Among the isolated strains, 96 were identified as Fusarium verricillioides, and four as other Fusarium species. Concordant phylogenies were obtained by AFLP and TEF-1 alpha sequencing, permitting the classification of the different species into distinct clades. Concerning MAT alleles, 70% of the F. verricillioides isolates carried the MAT-1 and 30% MAT-2. A significant correlation was observed between the expression of the genes and toxin production r=0.95 and r=0.79 (correlation of FUM1 with FB(1) and FB(2), respectively, P < 0.0001): r=0.93 and r =0.78 (correlation of FUM19 with FB(1) and FB(2). respectively, P < 0.0001). Molecular methods used in this study were found to be useful for the rapid identification of Fusarium species. The high and significant correlation between FUM1 and FUM19 expression and fumonisins production suggests that real time RT-PCR is suitable for studies considering the influence of abiotic and biotic factors on expression of these genes. BACKGROUND: The aim of this study was to characterise the mycoflora and the presence of fumonisin in sorghum grains, correlating the results with the environment and abiotic factors. RESULTS: Fifty samples (five collections of ten samples each) of sorghum were analysed. All samples were found to be contaminated with fungi, with higher frequencies of Cladosporium spp. (61.8%) and Helminthosporium spp. (33.4%). Fusarium verticillioides was isolated from 15.1% of the samples, with 38% of them being contaminated with fumonisin B(1) (FB(1)) at levels ranging from 50 to 368.78 ng g(-1). Regarding abiotic factors, temperature, water activity and rainfall showed a positive correlation with the frequency of F. verticillioides and FB(1) production. There was a significant positive correlation between relative air humidity and FB(1) production. The results obtained from sexual crosses between standard F mating tester strains and the isolated strains confirmed that the strains isolated were F. verticillioides. CONCLUSION: It can be concluded that the decrease in F. verticillioides and fumonisin contamination occurred owing to atypical climatic factors during the period of sorghum cultivation, when there was any occurrence of rain and the level of water activity of grains did not reach 0.58. Nematodes cause extensive losses to sugarcane in Brazil and also in other producing regions. Meloidogyne incognita, M. javanica and Pratylenchus zeae are the key species for this culture worldwide. In the present study, the aggressiveness of M. javanica and M. incognita to sugarcane variety SP 911049 was evaluated comparatively,. The following parameters were compared: reproduction factor (RF) of these nematodes, effect of nematodes in the natural incidence of pests, and the influence on the development and technological characteristics of sugarcane. Considering the data of RF, biometrics, natural infestation of pests, mortality of plants, and technological variables, it was concluded that M. javanica was more aggressive to sugarcane, although its rate of multiplication was much smaller than the one of M. incognita.

The aim of this work was to evaluate the population density of Pratylenchus brachyurus and Pratylenchus zeae associated with Brachiaria brizantha, B. decumbens and B. humidicola and their influence on forage availability and quality. The experiment was conducte in the Hisaeda Farm, Terenos, MS, Brazil. Soil, roots and plant aerial part were harvest with ten replications each, in one square meter randomized sets encompassing three treatments: Good, Intermediary and Bad, visually characterized, considering the percentage of green material. P. brachyurus and P. zeae density were evaluated in soil and plant roots. Dry matter of green, dead and re-growth materials, plant nutritional status and forage quality were assessed in the aerial plant part. Soil fertility was determined in all harvested samples. Both nematode species were identified from all samples, with a larger numbe in the roots (between 87-311 P. brachyurus and 1-61 P. zeae.10 g-1) than in the soil (0-8 P. brachyurus and 1-39 P. zeae.200 cm-3), however, no significant differences were found in the number of specimens between treatments. Considering that these forage species are perennial and host Pratylenchus spp, there is a tendency to increase the population of these pathogens over time, becoming a serious phytosanitary problem.
