O selénio (Se) é um micronutriente essencial para o crescimento, desenvolvimento e normal metabolismo dos animais, incluindo o ser humano. É parte integrante de um conjunto de proteínas, as selenoproteínas, com ação antioxidante (protegendo as membranas celulares contra danos dos radicais livres), envolvidas no metabolismo das hormonas da tiróide, na regulação do crescimento e viabilidade celular, nas funções do sistema imune e na reprodução. É introduzido na dieta alimentar (principalmente nas formas de selenometionina e selenocisteína) através das plantas, e de produtos que delas derivam, que assimilam os compostos de selénio presentes no solo. Uma vez que a quantidade de selénio existente nos solos é muito variável, o teor nos alimentos vai depender da sua origem geográfica e, por consequência, a ingestão de selénio varia entre regiões e países. Baixos níveis de selénio estão associados a um declínio na função imune e problemas cognitivos. A deficiência de Se pode também ocasionar problemas musculares e cardiomiopatia. Concentrações reduzidas foram observadas em indíviduos com crises epiléticas e também em casos de pré-eclampsia. A deficiência de selénio pode também desenvolver-se durante a nutrição parenteral. Atualmente, a Dose Diária Recomendada (DDR) é de 55 μg/dia para homens e mulheres adultos e saudáveis. No entanto, existem evidências clínicas de que a ingestão em doses superiores (200-300 μg/dia) pode ter um papel benéfico na prevenção de alguns tipos de cancro e doenças cardiovasculares, na melhoria da resposta imunológica, como neuroprotetor e na fertilidade. O Se desempenha um papel importante na fertilidade masculina, sendo necessário na biossíntese da testosterona e na formação e normal desenvolvimento dos espermatozóides. Em mulheres grávidas o Se, ajuda a prevenir complicações antes e durante o parto e promove o normal desenvolvimento do feto. Como antioxidante o selénio vai combater os danos provocados pelos radicais livres, impedindo que estes exerçam o seu papel prejudicial no organismo. Sendo o sistema imunológico muito suscetível aos danos provocados pelo stress oxidativo, o Se vai exercer efeitos benéficos combatendo os danos por ele causados. Relativamente à capacidade viral, não é possível saber com exatidão qual a quantidade de Se necessária ou concentração ideal no plasma para evitar a ocorrência e desenvolvimento de infeções virais. No entanto, sabe-se que tem um efeito benéfico em pacientes HIV positivos e em indivíduos infetados com o vírus da hepatite (B ou C) contra a progressão para o neoplasia de fígado. Em teoria, a nível cardiovascular, este elemento pode exercer um efeito protetor, embora alguns estudos epidemiológicos não tenham mostrado uma associação clara entre o risco cardiovascular e os níveis selénio. A nível cerebral o Se vai atuar como neuroprotetor, prevenindo o aparecimento de patologias como demência e doença de Alzheimer. Apesar destes indicadores, a maioria dos países europeus, incluindo Portugal, regista uma deficiente ingestão de selénio por parte da população. A suplementação poderá constituir uma opção para garantir os níveis nutricionais recomendados e/ou ser utilizada com o objetivo de prevenir algumas doenças e o envelhecimento. No entanto o selénio pode também ser tóxico se ingerido em excesso, estando a dose máxima admissível fixada em 400 μg/dia. A intoxicação por selénio é chamada selenose e os sintomas comuns incluem: hálito a alho, distúrbios gastrointestinais, perda de cabelo, descamação das unhas, danos neurológicos e fadiga. Assim, atualmente acredita-se que enquanto indivíduos com baixo nível de Se podem obter benefícios da suplementação, esta pode ser prejudicial aqueles com valores normais ou elevados.

• Stable isotope ratios of organic compounds are valuable tools for determining the geographical origin, identity, authenticity or history of samples from a vast range of sources such as sediments, plants and animals, including humans. • Hydrogen isotope ratios (d2H values) of methoxyl groups in lignin from wood of trees grown in different geographical areas were measured using compound-specificpyrolysis isotope ratio mass spectrometry analysis. • Lignin methoxyl groups were depleted in 2H relative to both meteoric water andwhole wood. A high correlation (r2=0.91) was observed between the d2 H valuesof the methoxyl groups and meteoric water, with a relatively uniform fractionation of –216±19 recorded with respect to meteoric water over a range of d2H values from –110 in northern Norway to + 20‰ in Yemen. Thus, woods from northernlatitudes can be clearly distinguished from those from tropical regions. By contrast, the d2H values of bulk wood were only relatively poorly correlated (r 2 = 0.47) with those of meteoric water. • Measurement of the d 2H values of lignin methoxyl groups is potentially a powerful tool that could be of use not only in the constraint of the geographical origin of lignified material but also in paleoclimate, food authenticity and forensic investigations.

The most consumed squid species worldwide were characterized regarding their concentrations of minerals, fatty acids, cholesterol and vitamin E. Interspecific comparisons were assessed among species and geographical origin. The health benefits derived from squid consumption were assessed based on daily minerals intake and on nutritional lipid quality indexes. Squids contribute significantly to daily intake of several macro (Na, K, Mg and P) and micronutrients (Cu, Zn and Ni). Despite their low fat concentration, they are rich in long-chain omega-3 fatty acids, particularly docosahexaenoic (DHA) and eicosapentanoic (EPA) acids, with highly favorable ω-3/ω-6 ratios (from 5.7 to 17.7), reducing the significance of their high cholesterol concentration (140–549 mg/100 g ww). Assessment of potential health risks based on minerals intake, non-carcinogenic and carcinogenic risks indicated that Loligo gahi (from Atlantic Ocean), Loligo opalescens (from Pacific Ocean) and Loligo duvaucelii (from Indic Ocean) should be eaten with moderation due to the high concentrations of Cu and/or Cd. Canonical discriminant analysis identified the major fatty acids (C14:0, C18:0, C18:1, C18:3ω-3, C20:4ω-6 and C22:5ω-6), P, K, Cu and vitamin E as chemical discriminators for the selected species. These elements and compounds exhibited the potential to prove authenticity of the commercially relevant squid species.

Mycobacterium avium Complex (MAC) comprises microorganisms that affect a wide range of animals including humans. The most relevant are Mycobacterium avium subspecies hominissuis (Mah) with a high impact on public health affecting mainly immunocompromised individuals and Mycobacterium avium subspecies paratuberculosis (Map) causing paratuberculosis in animals with a high economic impact worldwide. In this work, we characterized 28 human and 67 porcine Mah isolates and evaluated the relationship among them by Multiple-Locus Variable number tandem repeat Analysis (MLVA). We concluded that Mah population presented a high genetic diversity and no correlations were inferred based on geographical origin, host or biological sample. For the first time in Portugal Map strains, from asymptomatic bovine faecal samples were isolated highlighting the need of more reliable and rapid diagnostic methods for Map direct detection. Therefore, we developed an IS900 nested real time PCR with high sensitivity and specificity associated with optimized DNA extraction methodologies for faecal and milk samples. We detected 83% of 155 faecal samples from goats, cattle and sheep, and 26% of 98 milk samples from cattle, positive for Map IS900 nested real time PCR. A novel SNPs (single nucleotide polymorphisms) assay to Map characterization based on a Whole Genome Sequencing analysis was developed to elucidate the genetic relationship between strains. Based on sequential detection of 14 SNPs and on a decision tree we were able to differentiate 14 phylogenetic groups with a higher discriminatory power compared to other typing methods. A pigmented Map strain was isolated and characterized evidencing for the first time to our knowledge the existence of pigmented Type C strains. With this work, we intended to improve the ante mortem direct molecular detection of Map, to conscientiously aware for the existence of Map animal infections widespread in Portugal and to contribute to the improvement of Map and Mah epidemiological studies.

The advent of effective combination antiretroviral therapy (ART) in 1996 resulted in fewer patients experiencing clinical events, so that some prognostic analyses of individual cohort studies of human immunodeficiency virus-infected individuals had low statistical power. Because of this, the Antiretroviral Therapy Cohort Collaboration (ART-CC) of HIV cohort studies in Europe and North America was established in 2000, with the aim of studying the prognosis for clinical events in acquired immune deficiency syndrome (AIDS) and the mortality of adult patients treated for HIV-1 infection. In 2002, the ART-CC collected data on more than 12,000 patients in 13 cohorts who had begun combination ART between 1995 and 2001. Subsequent updates took place in 2004, 2006, 2008, and 2010. The ART-CC data base now includes data on more than 70,000 patients participating in 19 cohorts who began treatment before the end of 2009. Data are collected on patient demographics (e.g. sex, age, assumed transmission group, race/ethnicity, geographical origin), HIV biomarkers (e.g. CD4 cell count, plasma viral load of HIV-1), ART regimen, dates and types of AIDS events, and dates and causes of death. In recent years, additional data on co-infections such as hepatitis C; risk factors such as smoking, alcohol and drug use; non-HIV biomarkers such as haemoglobin and liver enzymes; and adherence to ART have been collected whenever available. The data remain the property of the contributing cohorts, whose representatives manage the ART-CC via the steering committee of the Collaboration. External collaboration is welcomed. Le criocère du lis, Lilioceris lilii (Coleoptera : Chrysomelidae), un ravageur de lis et de fritillaires d’origine eurasienne, a été observé pour la première fois en Amérique du Nord en 1943 sur l’Ile de Montréal au Canada. Après y avoir été confiné pendant environ 25 années, ce coléoptère a par la suite progressé rapidement sur le territoire nord-américain. Actuellement, on l’observe dans huit provinces canadiennes et huit états américains. Cette étude a investigué les routes d’invasion utilisées par le criocère du lis au Canada et aux États- Unis avec l’aide de marqueurs génétiques AFLP. Pour ce faire, 516 individus parmi 34 sites en Amérique du Nord et en Europe ont été échantillonnés et analysés. Le premier objectif était de déterminer, en analysant la structure génétique des populations nord-américains, s’il y avait eu une ou plusieurs introductions en provenance d’Europe. Le deuxième objectif était d’identifier l’origine de la ou des populations introduites en Amérique du Nord. Finalement, le troisième objectif consistait à proposer un scénario d’invasion de L. lilii en Amérique du Nord basé sur les données de première mention et de structure génétique des populations échantillonnées. Les résultats démontrent une signature génétique distincte entre les criocères du lis du Canada et ceux des États-Unis, suggérant ainsi deux sources d’introductions indépendantes en Amérique du Nord, soit une première introduction à Montréal, Québec, dans les années 1940 et une seconde aux États-Unis au début des années 1990 à Cambridge, Massachusetts. De plus, les deux populations nord-américaines semblent provenir de différentes régions du nord de l’Europe, ce qui est conséquent avec le scénario suggérant deux sources d’introductions indépendantes. Chacune des populations aurait par la suite progressé respectivement dans leur pays d’introduction selon une dispersion de type stratifiée. En effet, la progression continue de L. lilii dans certaines régions suggère une dispersion naturelle de l’espèce sur le territoire nord-américain, alors que la progression rapide sur de longues distances semble être causée par le transport anthropique de lis contaminés.

Introducción: De todos los casos de cáncer en el mundo el 80% se presentan en países en vía de desarrollo siendo el cáncer de estómago o cáncer gástrico la segunda causa de muerte por cáncer en el mundo con aproximadamente 700.000 muertes cada año. En Colombia, el cáncer gástrico es la primera causa de muerte por tumores malignos en ambos sexos, aún cuando no es la primera neoplasia en frecuencia. Metodología: Estudio observacional descriptivo, de registros de defunción del DANE, Colombia 2000 a 2009. Se analizaron tasas anuales crudas y por grupos de edad, género, procedencia geográfica, estado civil, nivel educativo y área de residencia habitual estableciendo diferencias estadísticas entre las variables y sus categorías. Resultados: En el período estudiado se registraron 43759 defunciones por cáncer gástrico, con mayor frecuencia en hombres 1,5:1. Las tasas de mortalidad por cáncer gástrico ajustadas por grupos etáreos aumentan después de la quinta década de la vida. Se encontraron diferencias estadísticamente significativas en todos los años estudiados y el departamento de residencia habitual del fallecido presentando Cauca (18,11- 19) y Boyacá (14,54-1742) las tasas más altas por 100.000 habitantes. Las tasas más altas se concentran en la zona de la Cordillera de los Andes, al estandarizar por grupos etáreos el Cauca tiene una tasa de 114,98 casos por 100.000 habitantes. Conclusión: El cáncer gástrico es la neoplasia que causa más muertes en Colombia por lo cual es necesario diseñar e implementar programas de detección precoz que vayan dirigidos al control de la mortalidad.

This study attempts to throw some light on the identity of the Matiners of the regions of north-eastern Catalonia by analizing a notebook which was a register of rebels. This notebook gives the age, geographical origin and profession of the rebels. The War of the matiners (1846-49) was a Catalan rebellion framed within the context of popular opposition to the regime of the moderates, and in the midst of a general crisis. Themen who formed the groups of ((trabucaires)) were particulary young (the problem of recriting must not be forgotten), and came from the country, the crafts, and industry, and were militarized above all in the interior regions

Understanding the sources of systematic errors in climate models is challenging because of coupled feedbacks and errors compensation. The developing seamless approach proposes that the identification and the correction of short term climate model errors have the potential to improve the modeled climate on longer time scales. In previous studies, initialised atmospheric simulations of a few days have been used to compare fast physics processes (convection, cloud processes) among models. The present study explores how initialised seasonal to decadal hindcasts (re-forecasts) relate transient week-to-month errors of the ocean and atmospheric components to the coupled model long-term pervasive SST errors. A protocol is designed to attribute the SST biases to the source processes. It includes five steps: (1) identify and describe biases in a coupled stabilized simulation, (2) determine the time scale of the advent of the bias and its propagation, (3) find the geographical origin of the bias, (4) evaluate the degree of coupling in the development of the bias, (5) find the field responsible for the bias. This strategy has been implemented with a set of experiments based on the initial adjustment of initialised simulations and exploring various degrees of coupling. In particular, hindcasts give the time scale of biases advent, regionally restored experiments show the geographical origin and ocean-only simulations isolate the field responsible for the bias and evaluate the degree of coupling in the bias development. This strategy is applied to four prominent SST biases of the IPSLCM5A-LR coupled model in the tropical Pacific, that are largely shared by other coupled models, including the Southeast Pacific warm bias and the equatorial cold tongue bias. Using the proposed protocol, we demonstrate that the East Pacific warm bias appears in a few months and is caused by a lack of upwelling due to too weak meridional coastal winds off Peru. The cold equatorial bias, which surprisingly takes 30 years to develop, is the result of an equatorward advection of midlatitude cold SST errors. Despite large development efforts, the current generation of coupled models shows only little improvement. The strategy proposed in this study is a further step to move from the current random ad hoc approach, to a bias-targeted, priority setting, systematic model development approach.

Early American crania show a different morphological pattern from the one shared by late Native Americans. Although the origin of the diachronic morphological diversity seen on the continents is still debated, the distinct morphology of early Americans is well documented and widely dispersed. This morphology has been described extensively for South America, where larger samples are available. Here we test the hypotheses that the morphology of Early Americans results from retention of the morphological pattern of Late Pleistocene modern humans and that the occupation of the New World precedes the morphological differentiation that gave rise to recent Eurasian and American morphology. We compare Early American samples with European Upper Paleolithic skulls, the East Asian Zhoukoudian Upper Cave specimens and a series of 20 modern human reference crania. Canonical Analysis and Minimum Spanning Tree were used to assess the morphological affinities among the series, while Mantel and Dow-Cheverud tests based on Mahalanobis Squared Distances were used to test different evolutionary scenarios. Our results show strong morphological affinities among the early series irrespective of geographical origin, which together with the matrix analyses results favor the scenario of a late morphological differentiation of modern humans. We conclude that the geographic differentiation of modern human morphology is a late phenomenon that occurred after the initial settlement of the Americas. Am J Phys Anthropol 144:442-453, 2011. The English language has become an international language and is globally used as a lingua franca. Therefore, there has been a shift in English-language education toward teaching English as an interna-tional language (EIL). Teaching from the EIL paradigm means that English is seen as an international language used in communication by people from different linguistic and cultural backgrounds. As the approach to English-language education changes from the traditional native-speaker, target country context, so does the role of culture within English-language teaching. The aim of this thesis is to in-vestigate and analyse cultural representations in two Swedish EFL textbooks used in upper-secondary school to see how they correspond with the EIL paradigm. This is done by focusing on the geograph-ical origin of the cultural content as well as looking at what kinds of culture are represented in the textbooks. A content analysis of the textbooks is conducted, using Kachru’s Concentric Circles of English as the model for the analysis of the geographical origin. Horibe’s model of the three different kinds of culture in EIL is the model used for coding the second part of the analysis. The results of the analysis show that culture of target countries and "Culture as social custom" dominate the cultural content of the textbook. Thus, although there are some indications that the EIL paradigm has influ-enced the textbooks, the traditional approach to culture in language teaching still prevails in the ana-lysed textbooks. Because of the relatively small sample included in the thesis, further studies need to be conducted in order to make conclusions regarding the Swedish context as a whole.

Restriction fragment length polymorphism (RFLP) and sequence analyses of the PCR-amplified 16S-23S rDNA intergenic spacer (ITS) were used for differentiating Acidithiobacillus thiooxidans strains from other related acidithiobacilli, including A. ferrooxidans and A. caldus. RFLP fingerprints obtained with AluI, DdeI, HaeIII, HinfI and MspI enabled the differentiation of all Acidithiobacillus reference strains into species groups. The A. thiooxidans strains investigated (metal mine isolates) yielded identical RFLP patterns to the A. thiooxidans type strain (ATCC 19377(T)), except for strain DAMS, which had a distinct pattern for all enzymes tested. Fourteen A. ferrooxidans mine strains were assigned to 3 RFLP groups, the majority of which were grouped with A. ferrooxidans ATCC 23270(T). The spacer region of one representative strain from each of the RFLP groups obtained was subjected to sequence analysis, in addition to eleven additional A. thiooxidans strains isolated from sediment and water samples, and A. caldus DSM 8584(T). The tRNA(IIe) and tRNA(Ala) genes, present in all strains analyzed, showed high sequence similarity. Phylogenetic analysis of the ITS sequences differentiated all three Acidithiobacillus species. Inter- and infraspecific genetic variations detected were mainly due to the size and sequence polymorphism of the ITS3 region. Mantel tests showed no significant correlation between ITS sequence similarity and the geographical origin of strains. The results showed that the 16S-23S rDNA spacer region is a useful target for the development of molecular-based methods aimed at the detection, rapid differentiation and identification of acidithiobacilli. Location or stock-specific landing data are necessary to improve management of shark stocks, especially those imperiled by overexploitation as a result of the international shark fin trade. In the current absence of catch monitoring directly at extraction sites, genetic stock identification of fins collected from major market supply chain endpoints offers an overlooked but potentially useful approach for tracing the fins back to their geographical, or stock of, origin. To demonstrate the feasibility of this approach, we used mitochondrial control region (mtCR) sequences to trace the broad geographical origin of 62 Hong Kong market-derived Sphyrna lewini fins. Of these fins 21% were derived from the western Atlantic, where this species is listed as 'Endangered' by the International Union for the Conservation of Nature (IUCN). We also show that S. lewini mtCR sequences are geographically segregated in the western Atlantic (overall ΦST = 0.74, n = 177 sharks), indicating that breeding females either remain close to, or home back to, their natal region for parturition. Mixed stock analysis simulations showed that it is possible to estimate the relative contributions of these mitochondrial stocks to fin mixtures in globally sourced trade hubs. These findings underscore the feasibility of using genetic stock identification to source market-derived shark fins to obtain essential and otherwise unavailable data on exploitation levels, and thus to productively inform stock assessment and management of S. lewini and potentially also of other fished shark species. © Inter-Research 2009.
