A study conducted in a 450 m² earthen pond to evaluate the production potential of giant freshwater prawn (stocked at 20,000 juveniles/ha) and Indian major carps, catla and rohu (stocked at 5000 juveniles/ha in 2:1 proportion) revealed that in nine months growing period, catla and rohu attained average sizes of 357 and 746 g, respectively, while prawn weighed 48.32 g. The growth of rohu was much faster than catla as indicated by higher relative and absolute weight gains. The total fish production per hectare was estimated to be 2418 kg and prawn production stood at 780 kg with excellent survival of both the fish (>98%) and prawn (>80%).

Effects of three different doses of vitamin D sub(3) on molting, growth, and calcium and phosphate composition of tissue and molt during the grow-out of the giant freshwater prawn Macrobrachium rosenbergii (average weight 10.56 ± 0.20 g), obtained from a grow-out pond, were studied. Intramuscular doses of vitamin D sub(3) (100, 500 and 2000 IU/kg body weight) were given on the 1st, 3rd, 5th, 7th, 9th, 11th, 13th, 15th, 20th, 25th and 30th days. All the experimental animals were fed with a basal diet containing fish meal, shrimp meal, wheat flour, groundnut de-oiled cake, soybean meal and wheat bran at 3% of the body weight. The numbers of molts were recorded as 20±0.50, 29±1.16, 51±1.87, and 30±1.60 in control, 100, 500 and 2000 IU/kg body weight physiological doses, respectively. Maximum growth was recorded in prawns given 500 IU/ kg dose. Survival was between 58.33 ± 9.13 and 77.77 ± 8.61%. The ash content and calcium level increased significantly (p<0.05) and recorded the highest values in 500 IU/kg physiological dose. However, the inorganic phosphate (P sub(i)) content recorded the highest values in tissue in 2000 IU/kg dose (p<0.05, F = 50.60613). There is no significant difference in calcium contents (p>0.05) in both tissue and molt at 500 and 2000 IU/kg doses. It was found that a higher physiological dose (2000 IU/kg) of vitamin D sub(3) increased the rate of mortality. Results have shown that vitamin D sub(3) has a positive impact on the growth and survival of M. rosenbergii and it interferes with the metabolism of Ca and P sub(i), in tissue, and alters molting frequency. Results on physiological dose suggest an alternative and effective dietary supplementation method of vitamin D sub(3) in the grow-out phase of M. rosenbergii.

Observations were made on the larval development of a freshwater prawn, Macrobrachium gangeticum, which revealed that hatching occurred in freshwater but the larvae failed to survive after 2nd molting. Salinity was necessary for survival of the larvae after 2nd molting. The complete larval development involved nine larval stages and the 10th stage was considered to be the post-larva which measured between 4.5 and 5.0 mm in length. All the larval stages were completed within 26 days of hatching. Specimens from each larval stage were taken out and examined under a microscope.

An experiment was conducted with juvenile prawns Macrobrachium malcolmso11ii, (0:76± 0.01 ro 0.94-::tO.Ol g) w evaluate various protein source diets. Six diets containing 20%, 25%, 30%, 35%, 40%,and 45% of crude protein were formulated, and fed to prawns in the form of pellet to evaluate their suitability. The experiment was designed for 60 days and sampling was made at every 15 days interval. At the end of the study period growth, feed conversion ration (FCR) specific growth rate (SGR), feed efficiency and survival were determined for prawns in each dietary treatment. Among the above five feeds poor FCR and higher weight gain observed in 35% protein diet (B-4). Similarly specific growth rate and feed efficiency are also highest with diet containing 35% protein. The dietary protein levels above 35% exerts a decrease in growth of prawn was observed in the present study. The feed efficiency ratio and protein efficiency ratio decreased with the increased dietary protein levels. It is concluded that 35% protein diet could be suitable with optimum protein supply for Macrobrachium malcolmsonii Therefore, above and below this 35% protein level in the formulated feed leads to metabolic stress which lowers the conversion efficiency and wastage of nutrients.

Studies were conducted to observe the effects of different types of feeds on the gonado-somatic index (GSI) and fecundity of freshwater prawn Macrobrachium rogenbergii. Three different treatments (T1 T2 and T3) were designed with three types of feed as follows: (i) Saudi-Bangla Prawn feed 100% - T1 (ii) SaudiBangla prawn feed 50%+ local feed 50%- T2 and (iii) local feed 100%- T3. The results showed that the average value of gonado-somatic index (GSI) was 14.39, 14.35 and 14.36 and the average fecundity of M. rogenbergiiwas 99,741, 98,125 and 97,911 in T1 T2 and T3 respectively. No significant difference (p>00.5) was between gonado-somatic indices (GSI) and fecundities of M. rogenbergii among different feeding trails. The price of Saudi-Bangla prawn feed was very high (Tk. 23/kg) than the local feed (Tk. 14/kg). So, use of local feed was recommended for M. rogenbergii brood rearing.

Effects of three stocking densities, viz., 35, 50 and 65/decimal (1 decimal = 40.48 m2) of juvenile freshwater prawn (Macrobrachium rosenbergii) on prawn and fish production were tested in a polycuture system with silver carp (Hypophthalmichthys molitrix), catla (Catla catla), Grass carp (Ctenophmyngodon idella) and silver barb (Barbodes gonionotus). The fish stocking density was 19/decimal with the species combination of silver carp-10, catlac-6, grass carp-1 and silver barb-2. In a 8-month culture period, the prawn yield 423 ± 144 kg/ha was significantly lower (P< 0.5) with the prawn stocking density of 35/decimal than that of 548 ± 178 kg/ha and 662 ± 243 kg/ha with 50 and 65/decimal respectively. The fish production (1844-1891 kg/ha) did not differ significantly (p <0.05) among the three treatments indicating that prawn stocking densities had no influence on fish yield. The lower mean harvest weight (62 g) and survival rate (67 g) and higher yield (2.67 kg/decimal) with the highest stocking rate of prawn reveals that as density was increased, prawn survival and individual weight at harvest decreased but total yield increased.

Two experiments were conducted to formulate and evaluate test diets using locally available ingredients to find out suitable diets for mono and polyculture of Macrobrachium rosenbergii in ponds. The first experiment was conducted from 1 July 2003 to 29 September 2003 in 12 experimental ponds each measuring 30 square meters behind the Fisheries Faculty Building, BAU campus, Mymensingh. Three experimental diets containing 30% protein were formulated using fish meal, meat and bone meal, mustard oilcake, sesame meal and rice bran and assigned to treatments 1, 2 and 3, respectively. A special shrimp feed (Starter-II) from Saudi-Bangla Fish Feed Ltd. was assigned to treatment 4 (Control). Each treatment had three replications. Juveniles of M. rosenbergii (2.90±0.81g) were stocked at the rate of 4/square meter. Prawns were fed three times daily at the rate of 15% of their body weight at the beginning, which was gradually reduced to 10% and 5% for the last two months. There was no significant (P>0.05) difference between the weight gains of prawns fed diets 1 and 4 (control), but they were significantly (P<0.05) higher than those of diets 2 and 3. The FCR values of diets ranged between 2.61 to 3.36 with diets 1 and 4 showing significantly (P<0.05) lower FCR values. The survival rate of prawns ranged between 68 to 78% with prawns fed diets 1 and 4 showing significantly higher survival rate. The production of prawn ranged from 921 to 1,428 kg/ha/90 days and diet 1 gave the highest production. Treatment 1 gave the highest net profit ofTk. 161,980/ha/90 days. The second experiment was conducted from December, 2003 to April, 2004 to see the growth of over wintered M. rosenbergii juveniles in polyculture with indigenous major carps (catla and rohu) in ponds using formulated diets. Three isoenergetic experimental diets formulated using fish meal, meat and bone meal, mustard oil cake, rice bran, wheat bran and molasses, and a shrimp feed 'Golda special feed' from Saudi-Bangla Fish Feed Ltd. were assigned to treatments 1, 2, 3 and 4 (control), respectively, each treatment had two replications. Eight experimental ponds each measuring 80 square meters in the Field Laboratory Complex of the Faculty of Fisheries, BAU campus, Mymensingh were used. The mean initial weights of M. rosenbergii, catla and rohu were 1.60±0.01, 30.0±0.09 and 25.0±0.08 g, respectively. A total of 160 fish and prawn (20,000/ha) were stocked in each pond at the ratio of 2:1:1 (prawn: catla: rohu). Fish were fed at the rate of 3-5% of their body weight. Prawns fed diet 1, 2 and 4 showed higher weight gains compared to diet 3. The weight gain of catla was significantly higher in T1 while in case of rohu was higher in T1 and T4, respectively. There were no significant differences (P>0.05) in the survival rate of fish as obtained from different treatments. The overall total fish production ranged from 2196 to 2679 kg/ha/5 months. The highest production and the highest profit (Tk. 56,531/ha/5 months) was obtained from T1 and the lowest (Tk. 24,932/ha/5 months) from T4.

An experiment was conducted to evaluate the effects of control of carbon/nitrogen ratio (C/N ratio) by addition of low cost carbohydrate to the water column on water quality and pond ecology in freshwater prawn Macrobrachium rosenbergii post-larvae nursing system. In this experiment, two level of dietary protein 20% and 35% without carbohydrate addition (‘P20' and ‘P35') and with carbohydrate addition (‘P20+CH' and ‘P35+CH') were compared in small ponds of 40 m² area stocked with 20 post-larvae (0.021 ± 0.001g) per m² . Maize flour was used as low cost carbohydrate and applied to the water column followed by the first feeding during the day. The addition of carbohydrate significantly reduced (p< 0.05) ammonia-nitrogen (NH sub(3)-N) and nitrite-nitrogen (NO sub(2) - N) of water in P20 + CH and P35 + CH treatments. It significantly increased (p< 0.05) the total heterotrophic bacteria (THB) population both in water and sediment. Fifty nine genera of plankton were identified belonging to the Bacillariophyceae (11), Chlorophyceae (21), Cyanophyceae (7), Dinophyceae (1), Rotifera (7) and Crustacea (9) without any significant difference (p>0.05) of total phytoplankton and zooplankton among the treatments. Survival rate of prawn was significantly lowest (p<0.05) in P20 and no significant difference (p>0.05) was observed between P20+CH and P35 treatments. Control of C/N ratio by the addition of low-cost carbohydrate to the pond water column benefited the freshwater prawn nursing practices in three ways (1) increased heterotrophic bacterial growth supplying bacterial protein augment the prawn post-larvae growth performances, (2) reduced demand for supplemental feed protein and subsequent reduction in feed cost and (3) reduced toxic NH sub(3)-N and NO sub(2)-N levels in pond nursing system.

A study was carried out at five upazillas (Dumuria, Fakirhat, Pirojpur sadar, Gopalgonj sadar and Kalia) of five southern districts of Khulna, Bagerhat, Pirojpur, Gopalgonj and Narail to understand the comparative production performance and any effect on soil quality in case of year-round golda (Macrobrachium rosenbergii), alternate prawn-paddy and year-round paddy farming. A direct interview of 55 farmers was taken, using pretested questionnaire, and soil samples were taken from selected farms before and after each crop. Among the five upazillas, farmers in Gopalgonj sadar and Kalia are not practicing year-round golda. Rotational golda-paddy farming has been recorded to result in maximum profit, with the highest of Tk. 310,912/ha/year in 2003 at Dumuria. Only paddy farming is less profitable than other two cropping patterns, irrespective of study sites. The nutrient status of soil in rotational golda and paddy farming has been found improved, compared to that of only golda or paddy farming.

Histoarchitecture, seasonal variation and reproductive function of the neuroendocrine structure, brain of freshwater prawn Macrobrachium gangeticum were studied. Three types of NSCs - 'B', 'C' and 'D' were found to be concentrated in four groups in brain. These cells showed larger diameters and higher activity during breeding season. In case of females, the 'C' cells were more active during vitellogenic period. Brain extracts were found to induce gonadal maturation of both males and females.

An electrochemical technique for the real-time detection of hydrogen peroxide (H2O2) was employed to describe respiratory burst activity (RBA) of phagocytes in plasma which can be used to evaluate the ability of immune system and disease resistance. The method is based upon the electric current changes, by redox reaction on platinum electrode of extracellular hydrogen peroxide (H2O2) released from phagocytes stimulated by the zymosan at 680 mV direct current (d.c.). Compared with the control, activation of respiratory burst by zymosan particles results in a high amperometric response, and a current peak was obtained during the whole monitoring process. The peak current was proved by addition Of Cu2+ and other controls, to be the result of intense release of H2O2 from phagocytes. The peak area was calculated and used to evaluate the quantity of effective H2O2, which represents the quantity of H2O2 beyond the clearance of related enzymes in plasma. According to Faraday's law, the phagocytes' ability of prawns to generate effective H2O2 was evaluated from 1.253 x 10(-14) mol/cell to 6.146 x 10(-14) mol/cell, and carp from 1.689 x 10(-15) Mol/Cell to 7.873 x 10(-1)5 mol/cell. This method is an acute and quick detection of extracellular effective H2O2 in plasma and reflects the capacity of phagocytes under natural conditions, which could be applied for selecting species and parents with high immunity for breeding in aquaculture. This thesis Entitled Application of Biofloc technology (BFT) In the Nursery Rearing and Farming of Giant Freshwater Prawn,Macrobrachium Rosenbergii(De Man). Aquaculture, rearing plants and animals under controlled conditions is growing with an annual growth rate of 8.3% in the period 1970-2008 (FAO, 2010). This trend of growth is essential for the supply of protein-rich food for ever increasing world population. But growth and development of aquaculture should be in sustainable manner, preferably without jeopardizing the aquatic environment.In the present study, the application of BFT in the nursery rearing and farming ofgiant freshwater prawn, M. rosenbergii, is attempted. The result of the study is organised into eight chapters. In the first chapter, the subject is adequately introduced. Various types of aquaculture practices followed, development and status of Indian aquaculture, present status of freshwater pravm culture, BF T and its use for the sustainable aquaculture systems, theory of BFT based aquaculture practices, hypothesis, objective and outline of the thesis are described. An extensive review of literature on studies carried out so far on biofloc based aquaculture are given in chapter 2. The third chapter deals with the application of BFT in the primary nursery phase of freshwater prawn. Several workers suggested the need for an intermediate nursery phase in the culture system of freshwater prawn for the successful production. Thirty day experiment was conducted to study the effect of BFT on the water quality, and animal welfare under the various stocking densities. The study concluded that stocking finfishes in biofloc-based monoculture system of freshwater prawns has the potential of increasing total yield. Prawns having a higher commercial value than finfishes besides ensuring economic sustainability. Results showed that prawn yield and survival was better in catla dominated tanks. Based on the results of the study, it is recommended to incorporate 25% rohu and 75% catla in the biofloc-based culture system of giant freshwater prawns. The results of the present study also recommend to stock relatively larger catla for biofloc-based culture system. Fish production was also higher in the 100% catla tank. When catla was added in higher percentages it should ensured that the hiding objects in the culture ponds shall be used in order to reduce the chance of cannibalism among prawns. rohu and catla equally have the ability to harvest the biofloc, catla consumes the planktonic contributes in the floc whereas rohu grazed on the bacterial consortium suspended in the water column. In Chapter 8, recommendations and future research perspectives in the field of biofloc based aquaculture is presented.
