Feathers are composed of a structure that, whilst being very light, is able to withstand the large aerodynamic forces exerted upon them during flight. To explore the contribution of molecular orientation to feather keratin mechanical properties, we have examined the nanoscopic organisation of the keratin molecules by X-ray diffraction techniques and have confirmed a link between this and the Young's modulus of the feather rachis. Our results indicate that along the rachis length, from calamus to tip, the keratin molecules become more aligned than at the calamus before returning to a state of higher mis-orientation towards the tip of the rachis. We have also confirmed the general trend of increasing Young's modulus with distance along the rachis. Furthermore, we report a distinct difference in the patterns of orientation of beta-keratin in the feathers of flying and flightless birds. The trend for increased modulus along the feathers of volant birds is absent in the flightless ostrich.

This study focuses on the restoration of chalk grasslands over a 6-year period and tests the efficacy of two management practices, hay spreading and soil disturbance, in promoting this process for phytophagous beetles. Restoration success for the beetles, measured as similarity to target species-rich chalk grassland, was not found to be influenced by either management practice. In contrast, restoration success for the plants did increase in response to hay spreading management. Although the presence of suitable host plants was considered to dictate the earliest point at which phytophagous beetles could successfully colonized, few beetle species colonized as soon as their host plants became established. Morphological characteristics and feeding habits of 27 phytophagous beetle species were therefore tested to identify factors that limited their colonization and persistence. The lag time between host plant establishment and colonization was greatest for flightless beetles. Beetles with foliage-feeding larvae both colonized at slower rates than seed-, stem-, or root-feeding species and persisted within the swards for shorter periods. Although the use of hay spreading may benefit plant communities during chalk grassland restoration, it did not directly benefit phytophagous beetles. Without techniques for overcoming colonization limitation for invertebrate taxa, short-term success of restoration may be limited to the plants only.

Within the healthy population, there is substantial, heritable, and interindividual variability in the platelet response. We explored whether a proportion of this variability could be accounted for by interindividual variation in gene expression. Through a correlative analysis of genome-wide platelet RNA expression data from 37 subjects representing the normal range of platelet responsiveness within a cohort of 500 subjects, we identified 63 genes in which transcript levels correlated with variation in the platelet response to adenosine diphosphate and/or the collagen-mimetic peptide, cross-linked collagen-related peptide. Many of these encode proteins with no reported function in platelets. An association study of 6 of the 63 genes in 4235 cases and 6379 controls showed a putative association with myocardial infarction for COMMD7 (COMM domain-containing protein 7) and a major deviation from the null hypo thesis for LRRFIP1 [leucine-rich repeat (in FLII) interacting protein 1]. Morpholino-based silencing in Danio rerio identified a modest role for commd7 and a significant effect for lrrfip1 as positive regulators of thrombus formation. Proteomic analysis of human platelet LRRFIP1-interacting proteins indicated that LRRFIP1 functions as a component of the platelet cytoskeleton, where it interacts with the actin-remodeling proteins Flightless-1 and Drebrin. Taken together, these data reveal novel proteins regulating the platelet response.

The distribution, abundance and biomass of seabirds in Western Port, Victoria, were surveyed between April 1991 and August 1994. Individuals were counted along an 81-km series of transects from a boat at approximately monthly intervals. A total of 25 seabird taxa were recorded, of which 18 and 15 were common to those recorded by an earlier study in Port Phillip Bay and waters south of Phillip Island, respectively. The most numerous species by far was the Short-tailed Shearwater (Puffinus tenuirostris) followed by the Silver Gull (Larus novaehollandiae), Little Penguin (Eudyptula minor) and Crested Tern (Sterna bergii). Distribution within Western Port was not uniform, with pursuit divers such as cormorant and grebe species being recorded mostly in the shallow Eastern Arm. In contrast, surface-seizing (e.g. albatrosses), surface-plunging (e.g. Crested Terns), shallow-plunging (Australasian Gannet, Morus serrator) and pursuit-plunging (e.g. shearwaters) species predominated in the deeper Western Arm of Western Port. These species were also seasonally abundant, with peak numbers for most occurring in late summer–early autumn, which coincides with the reported influx of juvenile clupeoid fish into Western Port. Average biomass (686 ± 395 kg) comprised mostly Short-tailed Shearwaters, Little Penguins and Pied Cormorants (Phalacrocorax varius). Biomass density (8.5 kg km–2) was similar to that reported for Port Phillip Bay (8.1 kg km–2) but lower than off the southern coast of Phillip Island (9.9 kg km–2).

Huge aggregations of flightless locust nymphs pose a serious threat to agriculture when they reach plague proportions but provide a very visible and nutritious resource for native birds. Locust outbreaks occur in spring and summer months in semiarid regions of Australia. Fenitrothion, an organophosphate pesticide, is sprayed aerially to control locust plagues. To evaluate fenitrothion exposure in birds attending locust outbreaks, we measured total plasma cholinesterase (ChE), butrylcholinesterase (BChE), and acetylcholinesterase (AChE) activities in four avian species captured pre- and postfenitrothion application and ChE reactivation in birds caught postspray only. Eleven of 21 plasma samples from four species had ChE activity below the diagnostic threshold (two standard deviations below the mean ChE activity of prespray samples). Granivorous zebra finches (Taeniopygia guttata) and insectivorous white-winged trillers (Lalage sueurii) had significantly lower mean plasma total ChE, BChE, and AChE activity postspray, while other insectivores, white-browed (Artamus superciliosus) and masked woodswallows (Artamus personatus), did not. Cholinesterase was reactivated in 19 of the 73 plasma samples and in one of three brain samples. We conclude that native bird species are exposed to fenitrothion during locust control operations. This exposure could have detrimental impacts, as both locust outbreaks and avian reproductive events are stimulated by heavy summer rainfall, leading to co-occurrence of locust control and avian breeding activities.

1. We estimated nitrogen (N) and phosphorus (P) loading into wetlands by carnivorous waterbirds with alternative physiological models using a food-intake and an excreta-production approach. The models were applied for non-breeding and breeding Dutch inland carnivorous waterbird populations to quantify their contribution to nutrient loading on a landscape scale.

2. Model predictions based on food intake exceeded those based on excretion by 59–62% for N and by 2–36% for P, depending on dietary assumptions. Uncertainty analysis indicated that the intake model was most affected by errors in energy requirement, while the excretion model was dependent on faecal nutrient composition.

3. Per capita loading rate of non-breeders increased with body mass from 0.3–0.8 g N day−1 and 0.15 g P day−1 in little gulls Larus minutus to 4.5–11.5 g N day−1 and 2.1–3.2 g P day−1 in great cormorants Phalacrocorax carbo. For breeding birds, the estimated nutrient loading by a family unit over the entire breeding period ranged between 17.6–443.0 g N and 8.6 g P for little tern Sterna albifrons to 619.6–1755.6 g N and 316.2–498.1 g P for great cormorants.

4. We distinguished between external (i.e. importing) and internal (i.e. recycling) nutrient loading by carnivorous waterbirds. For the Netherlands, average external-loading estimates ranged between 38.1–91.5 tonnes N and 16.7–18.2 tonnes P per year, whilst internal-loading estimates ranged between 53.1–140.5 tonnes N and 25.2–39.2 tonnes P and per year. The average contribution of breeding birds was estimated to be 17% and 32% for external and internal loading respectively. Most important species were black-headed gull Larus ridibundus and mew gull Larus canus for external loading, and great cormorant and grey heron Ardea cinerea for internal loading.

5. On a landscape scale, loading by carnivorous waterbirds was of minor importance for freshwater habitats in the Netherlands with 0.26–0.65 kg N ha−1 a−1 and 0.12–0.16 kg P ha−1 a−1. However, on a local scale, breeding colonies may be responsible for significant P loading.

Previous anecdotal reports have suggested that Black-faced Cormorant Phalacrocorax fuscescens breeds only in winter in southeastern Australia, but detailed reports confirming this are lacking. Here we examine the timing of breeding in Black-faced Cormorants at Notch Island in northern Bass Strait in 2006. Peak laying occurred during winter (ca 26 July). The diet of Black-faced Cormorants was predominantly fish (97% of identified prey) and varied between breeding and post-breeding periods. Black-faced Cormorants consumed a total of 14 different species with four species having a frequency of occurrence in the diet of ?5% during the breeding season and six species during the post-breeding period. We provide data for the first time on the chronology of breeding of Black-faced Cormorants in one year and give a preliminary description of their diet based on pellet analyses. We propose that late winter breeding may be a strategy to avoid the high ambient temperatures in northern Bass Strait during summer, the associated higher thermoregulatory costs for adults and the increased mortality for chicks.

One way of measuring pre-existing knowledge of a threatened species and its circumstances is to measure the degree of surprise expressed by stakeholders in relation to factual statements regarding the species. Beach-goers (n = 684) were surveyed in regard to their knowledge of the beach-dwelling, threatened, Hooded Plover Thinornis rubricollis, a coastal obligate in south eastern Australia. Principle components analysis revealed that respondents’ degree of knowledge could be categorized as involving ‘chick (flightless young) ecology’ and ‘human impacts’ (threatening processes). Respondents were more surprised by aspects of chick ecology than by threatening processes (F1,514 = 460.446, p < 0.001). Prior knowledge of the species was associated with less surprise at factual statements. Therefore, priorities for further education should focus on linking threats with chick ecology, particularly because an understanding that chicks are not stationary within fenced areas is critical to the interpretation and effectiveness of current signage used to mitigate human impacts.

Myxobolus cerebralis, the cause of whirling disease in salmonids, has dispersed to waters in 25 states within the USA, often by an unknown vector. Its incidence in Yellowstone cutthroat trout Oncorhynchus clarkii bouvieri within the highly protected environment of Yellowstone Lake, Yellowstone National Park, is a prime example. Given the local abundances of piscivorous birds, we sought to clarify their potential role in the dissemination of M. cerebralis. Six individuals from each of three bird species (American white pelican Pelecanus erythrorhynchos, double-crested cormorant Phalacrocorax auritus, and great blue heron Ardea herodias) were fed known-infected or uninfected rainbow trout O. mykiss. Fecal material produced during 10-d periods before and after feeding was collected to determine whether M. cerebralis could be detected and, if so, whether it remained viable after passage through the gastrointestinal tract of these birds. For all (100%) of the nine birds fed known-infected fish, fecal samples collected during days 1–4 after feeding tested positive for M. cerebralis by polymerase chain reaction. In addition, tubificid worms Tubifex tubifex that were fed fecal material from known-infected great blue herons produced triactinomyxons in laboratory cultures, confirming the persistent viability of the parasite. No triactinomyxons were produced from T. tubifex fed fecal material from known-infected American white pelicans or double-crested cormorants, indicating a potential loss of parasite viability in these species. Great blue herons have the ability to concentrate and release viable myxospores into shallow-water habitats that are highly suitable for T. tubifex, thereby supporting a positive feedback loop in which the proliferation of M. cerebralis is enhanced. The presence of avian piscivores as an important component of aquatic ecosystems should continue to be supported. However, given the distances traveled by great blue herons between rookeries and foraging areas in just days, any practices that unnaturally attract them may heighten the probability of M. cerebralis dispersal and proliferation within the Greater Yellowstone Ecosystem.

In order to determine potential definitive hosts of the digenetic trematode, Bolbophorus damnificus, two American White Pelicans (Pelecanus erythrorhynchos), two Double-crested Cormorants (Phalacrocorax auritus), two Great Blue Herons (Ardea herodias), and two Great Egrets (Ardea alba) were captured, treated with praziquantel, and fed channel catfish (Ictalurus punctatus) infected with B. damnificus metacercariae. Patent infections of B. damnificus, which developed in both American White Pelicans at 3 days post-infection, were confirmed by the presence of trematode ova in the feces. Mature B. damnificus trematodes were recovered from the intestines of both pelicans at 21 days post-infection, further confirming the establishment of infection. No evidence of B. damnificus infections was observed in the other bird species studied. This study provides further evidence that Double-crested Cormorants, Great Blue Herons, and Great Egrets do not serve as definitive hosts for B. damnificus.

A survey of catfish producers by the United States Department of Agriculture, Centers for Epidemiology and Animal Health (CEAH) in 1996 indicated that the two primary sources of catfish losses in commercial operations were disease (45%) and wildlife (37%) (CEAH 1997a). A variety of avian and mammalian predators are amracred to aquaculture facilities in the United States (Parkhurs: er al. 1992) because ponds and open raceways provide a constant and readily accessible food supply for these animals. However, the mere presence of these predators arcund aquaculture faciliries does not necessarily mean that significant depredation problems are occurring. At catfish farms, three species or species groups of birds are primarily cited by catfish producers as causing most depredation problems (Wywialowski 1999). These include doublecrested cormorants, wading birds (herons and egrets), and American white pelicans, in order of importance to catfish producers (Wywialowski 1993). Although all of these species consume catfish, their biology, distribution, dietary preferences dictare the extent of depredation problems they cause and the approaches needed to alleviate their depredations. With the exception of total bird exclusion from ponds, there are no simple solutions for resolving all bird depredation problems in catfish aquaculture. Thus, in most cases, an integrated management approach to alleviating bird depredations must be considered.

Photorezeptorzellen sind aus funktionell und morphologisch unterschiedlichen Kompartimenten aufgebaut: Einem lichtsensitiven Außensegment, das über ein nichtmotiles modifiziertes Cilium mit dem metabolisch aktiven Innensegment verbunden ist. Die Membranen des Außensegments werden kontinuierlich erneuert. Diese Prozesse erfordern die Translokation von Proteinen der Signaltransduktionskaskade vom Syntheseort im Innensegment in das Außensegment. Der intrazelluläre Transport der Proteine erfolgt durch das Verbindungscilium als einzige direkte cytoplasmatische Brücke zwischen den beiden Kompartimenten. Die Cytoskelettproteine des Verbindungsciliums sind maßgeblich an diesen Transportprozessen, sowie an der Ausbildung der Disk-Membranen und Funktion des Ciliums als Diffusionsbarriere beteiligt. Trotz dieser, für die Aufrechterhaltung der Photorezeptorzelle wichtigen Funktionen, sind bislang nur wenige molekulare Strukturkomponenten des Verbindungsciliums bekannt und funktionell charakterisiert. Um weitere Proteinkomponenten des Ciliums zu identifizieren, wurde eine biochemisch-molekularbiologische Strategie angewandt. Detergenzextrahierte Cilienapparate von Rinderphotorezeptorzellen wurden zur Immunisierung eines Kaninchens eingesetzt. Das affinitätsgereinigte Antiserum, mit Antikörpern gegen Epitope der unterschiedlichen Proteine des Verbindungsciliums, wurde anschließend zur Durchmusterung einer Rattenretina cDNA-Expressionsbank verwendet. Positive Klone wurden isoliert, sequenziert, und deren 3´- und 5´-terminale cDNA-Sequenzen in Datenbankrecherchen analysiert. Neben Klonen, die für Fragmente von bereits bekannten photorezeptorspezifischen Proteinen kodieren, Klonen mit Homologien zu EST´s, und Klonen ohne Homologien zu in Datenbanken enthaltenen Einträgen, wurden 8 cDNA-Klone isoliert, die für bisher unbekannte Cytoskelettproteine des Verbindungsciliums kodieren. Zwei dieser Proteine wurden näher charakterisiert: Das Mikrotubuli-Bindungsprotein EB2p und das Aktin-Bindungsprotein Flightless (Flip). Indirekte Immunofluoreszenzmarkierungen mit Antikörpern gegen EB1p, die mit EB2p kreuzreagieren, wurden EB-Proteine im Verbindungscilium und Basalkörper der Rezeptorzellen lokalisiert. Funktionell trägt Rn EB2p vermutlich zur Stabilisierung der axonemalen Mikrotubuli bei, und dürfte durch Interaktion mit cytoplasmatischem Dynein an retrograden Transportprozessen im Verbindungscilium beteiligt sein. Das Aktin-Bindungsprotein Flightless ist ein cytoplasmatisches Protein mit einer N-terminalen LRR-Domäne, die Protein-Protein- und Protein-Lipid-Interaktionen vermittelt. C-terminal weist Flip zwei Segmente mit Homologien zu Gelsolin auf. Indirekte Immunofluoreszenzmarkierungen und immuno-elektronenmikroskopische Analysen zeigen, daß Flip im Verbindungscilium in der subzellulären Domäne zwischen den axonemalen Mikrotubulipaarringen und der Plasmamembran lokalisiert ist. Im Außensegment der Photorezeptorzellen liegt Flip an den Disk-Membranen assoziiert vor. Flip verfügt vermutlich über zwei Funktion: Membran-assoziiert dürfte es Aktinfilamente an den Membranen der Außensegmente verankern. Im Verbindungscilium hingegen könnte es die ciliären Aktinfilamente modifizieren, die Transportwege für aktin-assoziierte Motorproteine sind. Flip und EB2p sind möglicherweise an den intrazellulären Transportprozessen durch das Verbindungscilium beteiligt.Die in der vorliegenden Arbeit angewandte Methode konnte erfolgreich zur Isolierung bislang unbekannter Proteine des Verbindungsciliums eingesetzt werden. Darüberhinaus wurde Flip auch in den ciliären Sinneszellen des Riechepithels und den mechanorezeptiven Haarzellen des Innenohrs identifiziert. Erkenntnisse über die molekulare Zusammensetzung des ciliären Cytoskeletts von Photorezeptorzellen können daher auch auf andere ciliäre Sinneszellen angewandt werden. Dies ermöglicht einen besseren Einblick in die allgemeine Funktion cilärer Cytoskelettstrukturen sensorischer Zellen.

Papillomaviruses (PV) are double stranded (ds) DNA viruses that infect epithelial cells within the skin or mucosa, most often causing benign neoplasms that spontaneously regress. The immune system plays a key role in the defense against PVs. Since these viruses infect keratinocytes, we wanted to investigate the role of the keratinocyte in initiating an immune response to canine papillomavirus-2 (CPV-2) in the dog. Keratinocytes express a variety of pattern recognition receptors (PRR) to distinguish different cutaneous pathogens and initiate an immune response. We examined the mRNA expression patterns for several recently described cytosolic nucleic acid sensing PRRs in canine monolayer keratinocyte cultures using quantitative reverse transcription-polymerase chain reaction. Unstimulated normal cells were found to express mRNA for melanoma differentiation associated gene 5 (MDA5), retinoic acid-inducible gene I (RIG-I), DNA-dependent activation of interferon regulatory factors, leucine rich repeat flightless interacting protein 1, and interferon inducible gene 16 (IFI16), as well as their adaptor molecules myeloid differentiation primary response gene 88, interferon-β promoter stimulator 1, and endoplasmic reticulum-resident transmembrane protein stimulator of interferon genes. When stimulated with synthetic dsDNA [poly(dA:dT)] or dsRNA [poly(I:C)], keratinocytes responded with increased mRNA expression levels for interleukin-6, tumor necrosis factor-α, interferon-β, RIG-I, IFI16, and MDA5. There was no detectable increase in mRNA expression, however, in keratinocytes infected with CPV-2. Furthermore, CPV-2-infected keratinocytes stimulated with poly(dA:dT) and poly(I:C) showed similar mRNA expression levels for these gene products when compared with expression levels in uninfected cells. These results suggest that although canine keratinocytes contain functional PRRs that can recognize and respond to dsDNA and dsRNA ligands, they do not appear to recognize or initiate a similar response to CPV-2.

Understanding how the environment influences patterns of diversity is vital for effective conservation management, especially in a changing global climate. While assemblage structure and species richness patterns are often correlated with current environmental factors, historical influences may also be considerable, especially for taxa with poor dispersal abilities. Mountain-top regions throughout tropical rainforests can act as important refugia for taxa characterised by low dispersal capacities such as flightless ground beetles (Carabidae), an ecologically significant predatory group. We surveyed flightless ground beetles along elevational gradients in five different subregions within the Australian Wet Tropics World Heritage Area to investigate (1) whether the diversity and composition of flightless ground beetles are elevationally stratified, and, if so, (2) what environmental factors (other than elevation per se) are associated with these patterns. Generalised linear models and model averaging techniques were used to relate patterns of diversity to environmental factors. Unlike most taxonomic groups, flightless ground beetles increased in species richness and abundance with elevation. Additionally, each subregion consisted of distinct assemblages containing a high level of regional endemic species. Species richness was most strongly positively associated with the historical climatic conditions and negatively associated with severity of recent disturbance (treefalls) and current climatic conditions. Assemblage composition was associated with latitude and current and historical climatic conditions. Our results suggest that distributional patterns of flightless ground beetles are not only likely to be associated with factors that change with elevation (current climatic conditions), but also factors that are independent of elevation (recent disturbance and historical climatic conditions). Variation in historical vegetation stability explained both species richness and assemblage composition patterns, probably reflecting the significance of upland refugia at a geographic time scale. These findings are important for conservation management as upland habitats are under threat from climate change.
