Hypothyroidism is a complex clinical condition found in both humans and dogs, thought to be caused by a combination of genetic and environmental factors. In this study we present a multi-breed analysis of predisposing genetic risk factors for hypothyroidism in dogs using three high-risk breeds-the Gordon Setter, Hovawart and the Rhodesian Ridgeback. Using a genome-wide association approach and meta-analysis, we identified a major hypothyroidism risk locus shared by these breeds on chromosome 12 (p = 2.1x10-11). Further characterisation of the candidate region revealed a shared ~167 kb risk haplotype (4,915,018-5,081,823 bp), tagged by two SNPs in almost complete linkage disequilibrium. This breed-shared risk haplotype includes three genes (LHFPL5, SRPK1 and SLC26A8) and does not extend to the dog leukocyte antigen (DLA) class II gene cluster located in the vicinity. These three genes have not been identified as candidate genes for hypothyroid disease previously, but have functions that could potentially contribute to the development of the disease. Our results implicate the potential involvement of novel genes and pathways for the development of canine hypothyroidism, raising new possibilities for screening, breeding programmes and treatments in dogs. This study may also contribute to our understanding of the genetic etiology of human hypothyroid disease, which is one of the most common endocrine disorders in humans.

Herein we report the clinical, histopathological, and molecular features of a cancer syndrome with predisposition to uterine leiomyomas and papillary renal cell carcinoma. The studied kindred included 11 family members with uterine leiomyomas and two with uterine leiomyosarcoma. Seven individuals had a history of cutaneous nodules, two of which were confirmed to be cutaneous leiomyomatosis. The four kidney cancer cases occurred in young (33- to 48-year-old) females and displayed a unique natural history. All these kidney cancers displayed a distinct papillary histology and presented as unilateral solitary lesions that had metastasized at the time of diagnosis. Genetic-marker analysis mapped the predisposition gene to chromosome 1q. Losses of the normal chromosome 1q were observed in tumors that had occurred in the kindred, including a uterine leiomyoma. Moreover, the observed histological features were used as a tool to diagnose a second kindred displaying the phenotype. We have shown that predisposition to uterine leiomyomas and papillary renal cell cancer can be inherited dominantly through the hereditary leiomyomatosis and renal cell cancer (HLRCC) gene. The HLRCC gene maps to chromosome 1q and is likely to be a tumor suppressor. Clinical, histopathological, and molecular tools are now available for accurate detection and diagnosis of this cancer syndrome.

The herpes simplex virus type 1 origin of DNA replication, oriS, contains three copies of the recognition sequence for the viral initiator protein, origin binding protein (OBP), arranged in two palindromes. The central box I forms a short palindrome with box III and a long palindrome with box II. Single-stranded oriS adopts a conformation, oriS*, that is tightly bound by OBP. Here we demonstrate that OBP binds to a box III–box I hairpin with a 3′ single-stranded tail in oriS*. Mutations designed to destabilize the hairpin abolish the binding of OBP to oriS*. The same mutations also inhibit DNA replication. Second site complementary mutations restore binding of OBP to oriS* as well as the ability of mutated oriS to support DNA replication. OriS* is also an efficient activator of the hydrolysis of ATP by OBP. Sequence analyses show that a box III–box I palindrome is an evolutionarily conserved feature of origins of DNA replication from human, equine, bovine, and gallid alpha herpes viruses. We propose that oriS facilitates initiation of DNA synthesis in two steps and that OBP exhibits exquisite specificity for the different conformations oriS adopts at these stages. Our model suggests that distance-dependent cooperative binding of OBP to boxes I and II in duplex DNA is succeeded by specific recognition of a box III–box I hairpin in partially unwound DNA.

Mechanical treatments such as shredding or extrusion are applied to municipal solid wastes (MSW) to produce refuse-derived fuels (RDF). In this way, a waste fraction (mainly composed by food waste) is removed and the quality of the fuel is improved. In this research, simultaneous thermal analysis (STA) was used to investigate how different mechanical treatments applied to MSW influence the composition and combustion behaviour of fuel blends produced by combining MSW or RDF with wood in different ratios. Shredding and screening resulted in a more efficient mechanical treatment than extrusion to reduce the chlorine content in a fuel, which would improve its quality. This study revealed that when plastics and food waste are combined in the fuel matrix, the thermal decomposition of the fuels are accelerated. The combination of MSW or RDF and woody materials in a fuel blend has a positive impact on its decomposition.

Baltic sediments have been studied by Behrens, Munthe, Küppers, Spethmann, Apstein, Sjöstedt, Pratje and the writer. The following types of sediments have been observed: varved and non-varved late-glacial clays, gray and black, post-glacial muds, and sands. The organic content of late-glacial clays ordinarily is less than 1.3 per cent, and of post-glacial muds more than 3 per cent. Sediments containing intermediate quantities are scarce. This can be explained as a result of the changed balance between organic and inorganic sedimentation when the glacial period ended; the abundance of fresh detritus then suddenly ceased and inorganic sedimentation became very much slower than before; consequently, the relative amount of organic detritus increased. As most of the material was not subjected to biological analysis, it has not been possible to distinguish different ages among post-glacial sediments.

Repetitions in child-directed speech (CDS) have been shown to vary over time, and are suggested to affect first language acquisition. Correlations between verbal contents of repetitions in CDS and children’s language development have been suggested. The verbal contents of repetitions in Swedish CDS have not yet been investigated. The aim of this study was to examine the verbal contents of repetitions in Swedish CDS during the child’s first 2 years and possible changes in proportions of repetitions during the same time span. Verbal contents of repetitions in parents’ speech in 10 parent-child dyads as the children were 3, 6, 9, 12 and 24 months old were investigated focusing on word classes, sentence types and whole-constituent change. The results were compared to the children’s productive vocabularies at the age of 30 months. Possible occurrences of item-based constructions and frequent frames in the repetitions were also examined. The overall results revealed patterns concerning change in verbal contents in repetitions over time and correlations between verbal contents in repetitions and child language development. Two proposals were made: parents adjust the complexity of their speech to linguistic developmental stages of their children, and linguistic variation in the input increases as the child grows older.

Denna studie är en kvalitativ litteraturstudie av bilddidaktisk forskning, vars syfte är att undersöka huruvida bildundervisningen ger möjlighet till utveckling och lärande som även främjar lärande i akademiska ämnen, utöver lärandet enbart i bildämnet. Bildämnet anses vara ett viktigt ämne i skolan som främjar elevers individuella utveckling och kan ge dem utvecklingsmöjligheter även i akademiska ämnen genom att det inkluderar både praktiska och teoretiska arbetsmetoder. Bilddidaktisk forskning diskuterar därför om det finns särskilda effekter som bildämnet ger möjlighet att utveckla hos eleven, vilka kan påverka elevers resultat i akademiska ämnen positivt. Metoden som har använts är en innehållsanalys av relevant bilddidaktik forskning, och avser undersöka och jämföra eventuella likheter och olikheter. Materialet för studien består av artiklar, forskningsrapporter och avhandlingar som behandlar bildundervisningens utvecklingsmöjligheter och eventuella transfereffekter från bildämnet till akademiska ämnen. Resultaten av studien visar att forskningen inte har funnit några direkta transfereffekter från bildämnet till specifika akademiska ämnen och att eventuella bevis är svåra att påvisa, men att bildämnet ändå spelar en viktig roll för elevens individuella utveckling genom att det inkluderar flera kognitiva förmågor, vilket skapar helhetsammanhang och bidrar till större ökad social och kulturell förståelse hos eleven.

Vesienhoidon keskeisenä tavoitteena on estää jokien, järvien ja rannikkovesien sekä pohjavesien tilan heikkeneminen sekä pyrkiä kaikkien vesien vähintään hyvään tilaan. Erinomaisiksi tai hyviksi arvioitujen vesien tilaa ei saa heikentää. Tavoitteen saavuttamiseksi suunnitellaan ja toteutetaan vesien tilaa parantavia toimenpiteitä ja seurataan niiden vaikutuksia. Vesienhoidossa otetaan huomioon myös merenhoidon, tulvariskien hallinnan sekä luonnonsuojelun tavoitteet. Vesienhoitoa suunnitellaan vesienhoitoalueittain, joita on Manner-Suomessa seitsemän. Vesienhoitoalue muodostuu yhdestä tai useammasta vesistöalueesta. Närpiönjoen vesistöalue kuuluu Kokemäenjoen-Saaristomeren-Selkämeren vesienhoitoalueeseen. Vesienhoidon suunnittelu etenee kuuden vuoden jaksoissa. Ensimmäiset vuoteen 2015 ulottuvat toimenpideohjelmat laadittiin laajassa yhteistyössä vuosien 2008–2009 aikana. Den här studien syftar till att ta reda på vad elever i årskurs 9 och årskurs 1 på gymnasiet anser om bildämnet. Intresset för de olika åldrarna kommer av att eleverna i årskurs 9 läser bild som ett obligatoriskt ämne medan eleverna i årskurs 1 har gjort ett aktivt val för att läsa bild, och studien önskar att belysa eventuella skillnader i elevernas uppfattningar om bildämnet på grund av detta. Studiens utgångspunkt tar avstamp i det sociokulturella perspektivets syn på lärande, estetiska ämnens betydelse för identitetsskapande, skolans uppdrag genom läroplanernas övergripande syften och mål och elevers egna åsikter om ämnet. Genom dessa perspektiv presenteras bildämnets bakgrund i samhället och tidigare forskning inom området, vilket sedan ställs mot elevernas egna utsagor om bildämnets betydelse. Studien bygger på kvalitativa intervjuer där eleverna har intervjuats i mindre grupper och har diskuterat och reflekterat över bildämnets betydelse. Resultatet visar att eleverna menar att bildämnet har störst betydelse som en kreativ paus bland annan undervisning, men att bildämnet också har en viss betydelse för annat lärande, personligt uttryck och som grundläggande kunskaper i framtiden.

Drawing on ethnographic data collected in Italian courts and prosecution offices, this dissertation offers new perspectives on legal decision-making by highlighting the importance of emotions for constructing and evaluating legal narratives. Focusing on criminal cases, it describes and dissects how judges and prosecutors use emotions in reflection and action tied to lay narratives and legal constraints. The analysis shows that legal professionals engage in different types of emotional dynamics when dealing with stories; first, they develop gut feelings, which are either endorsed or kept at distance by means of emotional reflexivity, to comply with legal ideals of objectivity and impartiality. Second, empathy emerges as a crucial tool to direct the interaction with lay people and to interpret legal prerequisites, such as credibility, and intent. Finally, the dissertation shows that lay stories lead legal professionals to become passionate and committed towards the correct application of the law, the restoration of the moral order, and the achievement of justice. In light of the empirical findings, this thesis strives to develop a theoretical understanding of legal decision-making as narrative work that includes emotional dynamics consistent with rational, objective action.
