通过秋水仙素诱导获得同源四倍体水稻10个株系,包括6个恢复系、3个保持系和1个不育系,这些株系具有加倍的染色体组。田间观察表明10个株系具有特殊的农艺性状:茎杆变粗壮、植株颜色加深、叶片变厚、叶宽适度增加、分蘖数减少、有效分蘖的比率下降等。根尖有丝分裂鉴定表明,同源四倍体水稻10个株系具有正常的有丝分裂,观察细胞的染色体数目皆为2n=48。花粉母细胞减数分裂鉴定表明10个株系具有比较理想的减数分裂行为,后期I染色体滞后、末期I微核生成和末期II异常小孢子比率较低,能较好的完成减数分裂过程,其中后期I染色体滞后比率约为10%-20%,末期I微核生成比率约为1%-6%,末期II异常小孢子比率约为1%-8%。这提示,染色体联合和分离不规则导致三价体、单价体 和落后染色体等产生,并进一步导致在后期和末期不均横分离产生异常小孢子,这可能是同源四倍体株系结实率不高的原因之一。 同源四倍体水稻正常胚囊为蓼型,变异胚囊具有多种类型,其比率显著高于二倍体对照,变化范围为39.62%-69.85%。按变异胚囊的结构特点和形成方式,分为四种类型:退化型,结构变异型,无融合生殖型和反足细胞增殖型。退化型胚囊的平均比率为29.17%,包括小胚囊(15.04%)和完全退化胚囊(14.13%),前者仍有较小胚囊腔而后者胚囊腔缺失。结构变异胚囊包括结构缺失、结构重复和位置异常,反映了蓼型胚囊八核七细胞结构的变异,其在各株系的平均比率为18.96%。无融合生殖胚囊发生比率极低,平均比率为1.77%,类型包括反足胚和卵细胞胚。反足细胞增殖胚囊是反足细胞团频繁增殖形成,伴随上述三种变异发生使异常胚囊的多样性进一步增加,其在各株系的平均比率为10.62%。相关分析表明,同源四倍体水稻结实可能主要来自三部分:正常胚囊、正常型小胚囊和反足细胞增殖型胚囊。这三种胚囊具有相对完整的蓼型结构,可能具有较好的育性,其对结实率的贡献程度估计值分别为72.44%、15.12%、12.44%。此外,完全退化型胚囊和位置异常型胚囊对结实率分别表现出显著(-0.66)和极显著(-0.92)的负相关,这表明二者可能是结实性的抑制因素。 Ten autotetraploid strains, which include six restoring lines, three maintaining lines and a sterile line, are derived from artificial induction by colchicine treatments. Variations of agronomical traits are observed which include large organs, sturdy plants, long panicle length and deep leaf color and so on. It has been confirmed that autotetraploid strains exhibit normal chromosome behaviors in mitosis and the chromosome numbers are all 48. Moreover, abnormal chromosome behaviors are investigated in meiosis including univalent, trivalent, quatrivalent, chromosome lagging and microkernel and so on. It evaluates that the percentage of chromosome lagging in anaphase I is about 10%-20%, the percentage of microkernel in telophase I is about 1%-6% and the percentage of abnormal microspore in telophase II is about 1%-8%. In all, abnormal behaviors of chromosomes could induce univalent, trivalent and et al. and subsequently induce infertile microspore. That may be one of the causes of low seed sets in autotetraploid strains. Embryo sacs of autotetraploid strains are formed according to the Polygonum type. However, these strains exhibit variations of abnormal embryo sacs at high frequencies (39.62% - 69.85%). The variations are frequently involved in the spikelets of the main panicles and the first tillers, leading to obvious decreases of the percentages of normal embryo sacs among the strains. Four types of abnormal embryo sacs are classified basing on their different structures and origins: degenerated embryo sac (DES), structure variation (SV), apomixis (Apo) and antipodal cell proliferation (ACP). Embryo sacs of DES (29.17%) exhibit small embryo sacs (15.04%) or no embryo sac (14.13%), the former showing embryo sacs with decreased size and the latter showing no sac. Embryo sacs of AS (18.96%) include three subtypes: structure deletion, structure duplication and location variation, which suggests abnormalities of the eight nuclei, seven celled pattern of the Polygonum type. Embryo sacs of Apo (only 1.77%) include two origins of apomictic embryos: antipodal embryo and egg embryo. Embryo sacs of ACP are observed frequently (10.62%) in autotetraploid strains which subsequently increase the variations of abnormal embryo sacs. It evaluates by the Pearson’s correlation analysis that seed set is probably contributed by three origins of embryo sacs: normal embryo sacs, small embryo sacs (normal pattern) and embryo sacs of ACP. These three origins exhibit comparatively good structure of the Polygonum type and could account for seed set at a percentage of 72.44%, 15.12%, 12.44%, respectively. Moreover, the subtype of no embryo sac (NES) negatively related to seed set at the P>0.01 level (-0.92) and the subtype of location variation (LV) negatively related to seed set at the P>0.05 level (-0.66). Which suggest the two subtypes may have strong stress on seed set.

Chez les plantes à fleurs, l’ovaire est l’organe reproducteur femelle et il interagit de façon importante avec les gamètes mâles durant la croissance, le guidage, la réception et la rupture du tube pollinique ainsi que la fusion des gamètes. Le processus débute lorsque de nombreux gènes de l’ovule sont activés à longue distance lors de la réception du pollen sur le stigmate. Afin d’explorer les signaux provenant de l’ovule ayant un impact important sur les interactions pollen–pistil, particulièrement les molécules sécrétées impliquées dans la signalisation espècespécifique, l’expression génique des ovules sous forme d’ARNm ainsi et la sécrétion protéique ont été étudiées chez Solanum chacoense, une espèce diploïde de pomme de terre sauvage. S. chacoense a subi beaucoup d’hybridation interspécifique avec d’autres espèces sympathiques de solanacées, facilitant ainsi grandement l’étude des interactions pollen–ovule de façon espècespécifique ainsi que leur évolution. Dans ce projet, des ovules provenant de trois conditions différentes ont été comparés: des ovules matures de type sauvage, des ovules légèrement immatures, récoltés deux jours avant l’anthèse et des ovules provenant du mutant frk1 pour lesquels le sac embryonnaire est absent. Un séquençage d’ARN à haut débit a d’abord été effectué sur les ovules de type sauvage de S. chacoense afin de générer un assemblage de référence comprenant 33852 séquences codantes. D’autres séquençages ont été effectués sur les trois conditions d’ovules et sur les feuilles afin de faire une analyse d’expression différentielle des gènes. En comparaison avec les ovules de type sauvage, 818 gènes sont réprimés dans les ovules du mutant frk1. Un sous-groupe de 284 gènes, étaient également sous-exprimés dans les ovules légèrement immatures, suggérant un rôle spécifique dans les stades tardifs de la maturation du sac embryonnaire (stade de développent FG6 à FG7) ainsi que du guidage du tube pollinique, puisque ni les ovules du mutant frk1 ni ceux légèrement immatures ne sont capables d’attirer les tubes polliniques lors d’essais de croissance semi in vivo. De plus, 21% de ces gènes sont des peptides riches en cystéines (CRPs). En utilisant un transcriptome assemblé de novo provenant de deux proches parents de S. chacoense, S. gandarillasii et S. tarijense, une analyse d’orthologie a été effectuée sur ces CRPs, révélant une grande variabilité et une évolution rapide chez les solanacées. De nouveaux motifs de cystéine uniques à cette famille ont également été découverts. En comparant avec des études similaires chez Arabidopsis, le sac embryonnaire de S. chacoense montre un transcriptome fortement divergent, particulièrement en en ce qui a trait à la catégorisation fonctionnelle des gènes et de la similarité entre les gènes orthologues. De plus,même si la glycosylation n’est pas requise lors du guidage mycropylaire du tube pollinique chez Arabidopsis, Torenia ou le maïs, des extraits d’ovules glycosylés de S. chacoense sont capables d’augmenter la capacité de guidage de 18%. Cette étude est donc la première à montrer une corrélation entre glycosylation et le guidage du tube pollinique par l’ovule. En complément à l’approche transcriptomique, une approche protéomique portant sur les protéine sécrétées par l’ovule (le secrétome) a été utilisée afin d’identifier des protéines impliquées dans l’interaction entre ovule et tube pollinique. Des exsudats d’ovules matures (capables d’attirer le tube pollinique) et d’ovules immatures (incapables d’attirer le tube pollinique) ont été récoltés en utilisant une nouvelle méthode d’extraction par gravité permettant de réduire efficacement les contaminants cytosoliques à moins de 1% de l’échantillon. Un total de 305 protéines sécrétées par les ovules (OSPs) ont été identifiées par spectrométrie de masse, parmi lesquelles 58% étaient spécifiques aux ovules lorsque comparées avec des données de protéines sécrétées par des tissus végétatifs. De plus, la sécrétion de 128 OSPs est augmentée dans les ovules matures par rapport aux ovules immatures. Ces 128 protéines sont donc considérées en tant que candidates potentiellement impliquées dans la maturation tardive de l’ovule et dans le guidage du tube pollinique. Cette étude a également montré que la maturation du sac embryonnaire du stade FG6 au stade FG7 influence le niveau de sécrétion de 44% du sécrétome total de l’ovule. De façon surprenante, la grande majorité (83%) de ces protéines n’est pas régulée au niveau de l’ARN, soulignant ainsi l’importance de cette approche dans l’étude du guidage du tube pollinique comme complément essentiel aux études transcriptomiques. Parmi tous les signaux sécrétés par l’ovule et reliés au guidage, obtenus à partir des approches transcriptomiques et protéomiques décrites ci-haut, nous avons spécifiquement évalué l’implication des CRPs dans le guidage du tube pollinique par l’ovule chez S. chacoense, vu l’implication de ce type de protéine dans les interactions pollen-pistil et le guidage du tube pollinique chez d’autres espèces. Au total, 28 CRPs étaient présentes dans les ovules capables d’attirer le tube pollinique tout en étant absentes dans les ovules incapables de l’attirer, et ce, soit au niveau de l’ARNm et/ou au niveau du sécrétome. De celles-ci, 17 CRPs ont été exprimées dans un système bactérien et purifiées en quantité suffisante pour tester le guidage. Alors que des exsudats d’ovules ont été utilisés avec succès pour attirer par chimiotactisme le tube pollinique, les candidats exprimés dans les bactéries n’ont quant à eux pas été capables d’attirer les tubes polliniques. Comme l’utilisation de systèmes d’expression hétérologue eucaryote peut permettre un meilleur repliement et une plus grande activité des protéines, les candidats restants seront de nouveau exprimés, cette fois dans un système de levure ainsi que dans un système végétal pour produire les peptides sécrétés. Ceux-ci seront ensuite utilisés lors d’essais fonctionnels pour évaluer leur capacité à guider les tubes polliniques et ainsi isoler les attractants chimiques responsable du guidage du tube pollinique chez les solanacées comme S. chacoense.

Mast cells are present in the eye of chick embryos from the 14th day onward, displaying metachromatic granules, mainly in the iris anterior surface and pectinate ligament. Ultrastructurally these cells show electron-dense granules and a few thin and short cytoplasmic projections in close contact with fibroblasts. Sometimes these contacts are extensive, with long fibroblast projections partially involving the mast cells. Gap junctions between mast cells and fibroblasts are observed only in the eyes of 16- and 20-day-old embryos. These intercellular specializations are represented by a close apposition of cytoplasmic membranes with an extension up to 300 nm. Gap junctions between mast cells and fibroblasts were not observed previously in vivo or in vitro, although in vitro studies have shown that a number of functionally critical interactions may occur between these cells. Our morphological findings suggest that, in vivo, fibroblasts interact with mast cells and may influence their maturation.

Incubating eggs (1,800 total) produced by a commercial flock of Cobb broiler breeders were used to determine the effects of storage duration (3 and 18 d) on gas partial pressure, thyroid hormones, and hatching parameters. Partial pressure of oxygen (pO2) and carbon dioxide (pCO2) were measured on d 18 and at internal pipping (IP) during incubation. Blood samples were collected for determination of triiodothyronine (T3), thyroxine (T4), and corticosterone concentrations in the embryos at IP and in newly hatched chicks. From 464 to 510 h of incubation, eggs were checked individually every 2 h to determine the timing and duration of IP, external pipping (EP), and total hatching time. At 18 d of incubation and at IP, pCO2 was greater in air cell of eggs stored for 3 d compared to those stored for 18 d (P < 0.05), but pO2 was greater in eggs stored for 18 d. At IP, T3 and corticosterone levels were higher in plasma of the embryos of eggs stored for 3 d compared to those stored for 18 d, but it was the reverse in newly hatched chicks (P < 0.05). Embryos from eggs stored for 18 d required more time to complete IP compared to embryos of eggs stored for only 3 d (P < 0.05), whereas the duration of EP was not affected by storage. The overall longer incubation was, however, not only due to prolonged IP but also to later occurrence of IP. It was concluded that prolonged IP as a result of long storage may be related to the late increase in corticosterone level, which may be a necessary stimulus for higher T 3/T4 ratio, late increase in pCO2 level, and decrease in pO2. The effect of long storage was a delay in hatching and a continuous increase in T3 due to higher corticosterone levels between IP and hatching, which may be an indication of the more stressful event of hatching of embryos from eggs stored longer. Differences in pCO2, pO2, T3, T4, and corticosterone levels in the incubating eggs may be manifestations of these changes culminating in altered hatching parameters and consequently differences in chick quality and growth potentials.

Altered pressure in the developing left ventricle (LV) results in altered morphology and tissue material properties. Mechanical stress and strain may play a role in the regulating process. This study showed that confocal microscopy, three-dimensional reconstruction, and finite element analysis can provide a detailed model of stress and strain in the trabeculated embryonic heart. The method was used to test the hypothesis that end-diastolic strains are normalized after altered loading of the LV during the stages of trabecular compaction and chamber formation. Stage-29 chick LVs subjected to pressure overload and underload at stage 21 were reconstructed with full trabecular morphology from confocal images and analyzed with finite element techniques. Measured material properties and intraventricular pressures were specified in the models. The results show volume-weighted end-diastolic von Mises stress and strain averaging 50–82% higher in the trabecular tissue than in the compact wall. The volume-weighted-average stresses for the entire LV were 115, 64, and 147Pa in control, underloaded, and overloaded models, while strains were 11, 7, and 4%; thus, neither was normalized in a volume-weighted sense. Localized epicardial strains at mid-longitudinal level were similar among the three groups and to strains measured from high-resolution ultrasound images. Sensitivity analysis showed changes in material properties are more significant than changes in geometry in the overloaded strain adaptation, although resulting stress was similar in both types of adaptation. These results emphasize the importance of appropriate metrics and the role of trabecular tissue in evaluating the evolution of stress and strain in relation to pressure-induced adaptation.

The six-layered neuron structure in the cerebral cortex is the foundation for human mental abilities. In the developing cerebral cortex, neural stem cells undergo proliferation and differentiate into intermediate progenitors and neurons, a process known as embryonic neurogenesis. Disrupted embryonic neurogenesis is the root cause of a wide range of neurodevelopmental disorders, including microcephaly and intellectual disabilities. Multiple layers of regulatory networks have been identified and extensively studied over the past decades to understand this complex but extremely crucial process of brain development. In recent years, post-transcriptional RNA regulation through RNA binding proteins has emerged as a critical regulatory nexus in embryonic neurogenesis. The exon junction complex (EJC) is a highly conserved RNA binding complex composed of four core proteins, Magoh, Rbm8a, Eif4a3, and Casc3. The EJC plays a major role in regulating RNA splicing, nuclear export, subcellular localization, translation, and nonsense mediated RNA decay. Human genetic studies have associated individual EJC components with various developmental disorders. We showed previously that haploinsufficiency of Magoh causes microcephaly and disrupted neural stem cell differentiation in mouse. However, it is unclear if other EJC core components are also required for embryonic neurogenesis. More importantly, the molecular mechanism through which the EJC regulates embryonic neurogenesis remains largely unknown. Here, we demonstrated with genetically modified mouse models that both Rbm8a and Eif4a3 are required for proper embryonic neurogenesis and the formation of a normal brain. Using transcriptome and proteomic analysis, we showed that the EJC posttranscriptionally regulates genes involved in the p53 pathway, splicing and translation regulation, as well as ribosomal biogenesis. This is the first in vivo evidence suggesting that the etiology of EJC associated neurodevelopmental diseases can be ribosomopathies. We also showed that, different from other EJC core components, depletion of Casc3 only led to mild neurogenesis defects in the mouse model. However, our data suggested that Casc3 is required for embryo viability, development progression, and is potentially a regulator of cardiac development. Together, data presented in this thesis suggests that the EJC is crucial for embryonic neurogenesis and that the EJC and its peripheral factors may regulate development in a tissue-specific manner.

In contrast to animals and lower plant species, sperm cells of flowering plants are non-motile and are transported to the female gametes via the pollen tube, i.e. the male gametophyte. Upon arrival at the female gametophyte two sperm cells are discharged into the receptive synergid cell to execute double fertilization. The first players involved in inter-gametophyte signaling to attract pollen tubes and to arrest their growth have been recently identified. In contrast the physiological mechanisms leading to pollen tube burst and thus sperm discharge remained elusive. Here, we describe the role of polymorphic defensin-like cysteine-rich proteins ZmES1-4 (Zea mays embryo sac) from maize, leading to pollen tube growth arrest, burst, and explosive sperm release. ZmES1-4 genes are exclusively expressed in the cells of the female gametophyte. ZmES4-GFP fusion proteins accumulate in vesicles at the secretory zone of mature synergid cells and are released during the fertilization process. Using RNAi knock-down and synthetic ZmES4 proteins, we found that ZmES4 induces pollen tube burst in a species-preferential manner. Pollen tube plasma membrane depolarization, which occurs immediately after ZmES4 application, as well as channel blocker experiments point to a role of K(+)-influx in the pollen tube rupture mechanism. Finally, we discovered the intrinsic rectifying K(+) channel KZM1 as a direct target of ZmES4. Following ZmES4 application, KZM1 opens at physiological membrane potentials and closes after wash-out. In conclusion, we suggest that vesicles containing ZmES4 are released from the synergid cells upon male-female gametophyte signaling. Subsequent interaction between ZmES4 and KZM1 results in channel opening and K(+) influx. We further suggest that K(+) influx leads to water uptake and culminates in osmotic tube burst. The species-preferential activity of polymorphic ZmES4 indicates that the mechanism described represents a pre-zygotic hybridization barrier and may be a component of reproductive isolation in plants.

The in vitro development of hamster preimplantation embryos is supported by non-glucose energy substrates. To investigate the importance of embryonic metabolism, influence of succinate and malate on the development of hamster 8-cell embryos to blastocysts was examined using a chemically defined protein-free modified hamster embryo culture medium-2 (HECM-2m). There was a dose-dependent influence of succinate on blastocyst development; 0.5 mM succinate was optimal (85.1% ± 3.9 vs. 54.5% ± 3.5). In succinate-supplemented HECM-2m, blastocyst development was reduced by omission of lactate (68.5% ± 7.2), but not pyruvate (85.8% ± 6.2) or glutamine (84.1% ± 2.1). Succinate along with either glutamine or lactate or pyruvate poorly supported blastocyst development (28%-58%). Malate also stimulated blastocyst development; 0.01 mM malate was optimal (86.3% ± 2.8). Supplementation of both succinate and malate to HECM-2m supported maximal (100%) blastocyst development, which was inhibited 4-fold by the addition of glucose/phosphate. The mean cell numbers (MCN) of blastocysts cultured in succinate-supplemented HECM-2m was higher (28.3 ± 1.1) than it was for those cultured in the absence of glutamine or pyruvate (range 20-24). The MCN was the highest (33.4 ± 1.6) for blastocysts cultured in succinate-malate-supplemented HECM-2m followed by those in succinate (28.3 ± 1.1) or malate (24.7 ± 0.5) supplemented HECM-2m. Embryo transfer experiments showed that 29.8% (±4.5) of transferred blastocysts cultured in succinate-malate-supplemented HECM-2m produced live births, similar (P > 0.1) to the control transfers of freshly recovered 8-cells (33.5% ± 2.0) or blastocysts (28.9% ± 3.0). These data show that supplementation of succinate and malate to HECM-2m supports 100% development of hamster 8-cell embryos to high quality viable blastocysts and that non-glucose oxidizable energy substrates are the most preferred components in hamster embryo culture medium.
